Dissecting Discoidin Domain Receptor 2 Dynamics in Fibrosis with a Programmable Collagen-Mimetic Probe.

Collagen receptors orchestrate vital extracellular matrix signaling, yet the inability to distinguish functional receptor activation from mere expression in real-time has obscured our understanding of fibrotic progression. Here, we develop a fluorescent triple-helical collagen-mimetic peptide (CMP) probe, [GVMGFO]3, designed to selectively target the ligand-engaging conformation of Discoidin Domain Receptor 2 (DDR2)─the sole receptor tyrosine kinase family that signals collagen. In pulmonary fibrosis, this probe identifies active fibrotic niches in vivo and directly tethers to disease-driving

Evaluation of Discoidin domain receptor 1 (DDR1) in junctional epithelial permeability and wound healing.

BACKGROUND: Epithelium is the periodontal first line of defense against microbes. Discoidin domain receptor 1 (DDR1) is a collagen receptor expressed in epithelium. Ddr1 knockout (Ddr1-/-) mice develop periodontitis-like defects, including junctional epithelium (JE) downgrowth, bacterial invasion, and alveolar bone loss. The objective of this study was to investigate epithelial responses in the absence of DDR1. We hypothesized that Ddr1-/- mice exhibit increased JE permeability and delayed epithelial wound healing. METHODS: Epithelium was analyzed in Ddr1-/- and wild-type (Ddr1+/+) mice. JE p

Discoidin domain receptor 1 activation links extracellular matrix to podocyte lipotoxicity in Alport syndrome.

BACKGROUND: Discoidin domain receptor 1 (DDR1) is a receptor tyrosine kinase that is activated by collagens that is involved in the pathogenesis of fibrotic disorders. Interestingly, de novo production of the collagen type I (Col I) has been observed in Col4a3 knockout mice, a mouse model of Alport Syndrome (AS mice). Deletion of the DDR1 in AS mice was shown to improve survival and renal function. However, the mechanisms driving DDR1-dependent fibrosis remain largely unknown. METHODS: Podocyte pDDR1 levels, Collagen and cluster of differentiation 36 (CD36) expression was analyzed by Real-tim

Collagen I-discoidin domain receptor 1 signalling promotes inflammation development during keloid formation.

BACKGROUND: Keloid formation is a chronic inflammatory skin disease, characterized by abnormal fibroproliferative scars. There are no effective treatments. Many factors are involved in keloid formation, primarily a highly active inflammatory response, wound tension and hereditary susceptibility. Collagen signalling plays important roles in various diseases, including fibrotic diseases and tumours, but its function in keloid inflammation remains unknown. OBJECTIVES: To study the roles and mechanisms of collagen signalling in inflammation development during keloid formation. METHODS: Normal an

Deficiency of myeloid discoidin domain receptor 2 aggravates melanoma lung and bone metastasis.

Melanoma, one of the most prevalent cancers worldwide, frequently metastasizes to the lung and bones. Tumor-associated macrophages play essential roles in melanoma metastasis but the underlying mechanism remains obscure. We previously demonstrated that specific knockout of Ddr2, a receptor tyrosine kinase, exacerbates systemic inflammation via modulating macrophage repolarization. To investigate whether myeloid Ddr2 regulates melanoma growth and metastasis, we injected B16BL6 melanoma cells into Ddr2LysM (cKO) mice via subcutaneous neck, tail vein, and left ventricle, respectively. We found th

Structural mapping of hot spots within human CASPR2 discoidin domain for autoantibody recognition.

Accumulating evidence has showed that anti-CASPR2 autoantibodies occur in a long list of neurological immune disorders including limbic encephalitis (LE). Belonging to the well-known neurexin superfamily, CASPR2 has been suggested to be a central node in the molecular networks controlling neurodevelopment. Distinct from other subfamilies in the neurexin superfamily, the CASPR subfamily features a unique discoidin (Disc) domain. As revealed by our and others' recent studies, CASPR2 Disc domain bears a major epitope for autoantibodies. However, structural information on CASPR2 recognition by aut

Discoidin domain receptor 1 kinase activity is required for regulating collagen IV synthesis.

Discoidin domain receptor 1 (DDR1) is a receptor tyrosine kinase that binds to and is activated by collagens. DDR1 expression increases following kidney injury and accumulating evidence suggests that it contributes to the progression of injury. To this end, deletion of DDR1 is beneficial in ameliorating kidney injury induced by angiotensin infusion, unilateral ureteral obstruction, or nephrotoxic nephritis. Most of the beneficial effects observed in the DDR1-null mice are attributed to reduced inflammatory cell infiltration to the site of injury, suggesting that DDR1 plays a pro-inflammatory e

Discoidin domain receptor 1: isoform expression and potential functions in cirrhotic human liver.

Discoidin domain receptor 1 (DDR1) is a receptor tyrosine kinase that binds and is activated by collagens. Transcriptional profiling of cirrhosis in human liver using a DNA array and quantitative PCR detected elevated mRNA expression of DDR1 compared with that in nondiseased liver. The present study characterized DDR1 expression in cirrhotic and nondiseased human liver and examined the cellular effects of DDR1 expression. mRNA expression of all five isoforms of DDR1 was detected in human liver, whereas DDR1a demonstrated differential expression in liver with hepatitis C virus and primary bilia

Discoidin domain receptor 1 mediates collagen-induced inflammatory activation of microglia in culture.

Discoidin domain receptor 1 (DDR1) is a nonintegrin collagen receptor tyrosine kinase with an extracellular domain homologous to discoidin 1 of a soil-living amoeba Dictyostelium discoideum. We have previously demonstrated that DDR1 mediates collagen-induced nitric oxide production in J774A.1 murine macrophages. Because collagen is one of the main components of extracellular matrix in the central nervous system, we hypothesized that collagen also induces inflammatory activation of brain microglia, and DDR1 may mediate collagen-induced microglial activation. Using BV-2 mouse microglial cells an

Supermeres Containing the Discoidin Domain Receptor 1 Extracellular Domain Promote Collagen Alignment and Immune Exclusion in Microsatellite-Stable Colorectal Cancer.

UNLABELLED: Immune checkpoint blockade (ICB) is an effective treatment for microsatellite instability-high (MSI-H) colorectal cancers that are highly infiltrated by CD8+ T cells. Microsatellite-stable (MSS) colorectal cancers are unresponsive to ICB, at least in part, due to the paucity of intratumoral CD8+ T cells. We recently identified discoidin domain receptor 1 (DDR1) as one of four genes associated with CD8+ T-cell exclusion in MSS colorectal cancer. There are conflicting reports about the presence and role of the cleaved ectodomain (cECD) of DDR1 in mouse models of breast and pancreatic

Genetic and pharmacological tools to study the role of discoidin domain receptors in kidney disease.

Following injury the kidney undergoes a repair process, which results in replacement of the injured tissue with little evidence of damage. However, repetitive injuries or inability of the kidney to stop the repair process result in abnormal deposition of extracellular matrix (ECM) components leading to fibrosis and organ dysfunction. The synthesis/degradation of ECM components is finely regulated by several factors, including discoidin domain receptors (DDRs). These are receptor tyrosine kinases that are activated by collagens. Upon activation, DDRs control several cell functions that, when ex

Discoidin Domain Receptor 1 in Colonic Epithelial Cells: A Paracrine Driver of Colonic Fibrosis.

INTRODUCTION: This study investigated the role and potential mechanisms of discoidin domain receptor 1 (DDR1) in colon fibrogenesis. METHODS: We employed the DSS-induced chronic colitis and fibrosis model to evaluate the therapeutic potential of DDR1 knockout on colonic fibrosis. In vitro experiments involved generating human normal colonic epithelial cells (HIEC line) with DDR1 overexpression by lentivirus transfection. Human colonic fibroblasts were exposed to conditioned medium (CM) from the stably transfected cells that had been treated with transforming growth factor-beta 1 (TGF-β1). The

A network map of discoidin domain receptor 1(DDR1)-mediated signaling in pathological conditions.

Discoidin domain receptor 1 (DDR1) is one of the receptors that belong to a family of non-integrin collagen receptors. In common, DDR1 is predominantly found in epithelial and smooth muscle cells and its mainly involved in organogenesis during embryonic development. However, it's also overexpressed in several pathological conditions, including cancer and inflammation. The DDR1 is reported in numerous cancers, including breast, prostate, pancreatic, bladder, lung, liver, pituitary, colorectal, skin, gastric, glioblastoma, and inflammation. DDR1 activates through the collagen I, IV, IGF-1/IGF1R,

Discoidin domain receptor 2: An emerging pharmacological drug target for prospective therapy against osteoarthritis.

Discoidin domain receptor2 (DDR2), a cell membrane tyrosine kinase on chondrocytes surface plays main role in cell-ECM interaction during the progressive degeneration of articular cartilage in osteoarthritis. The degraded component of ECM, type II collagen upon DDR2 binding provokes synthesis of matrix metalloproteinases (MMPs), responsible for severe destruction of joint tissues. DDR2 knockout has been investigated to decline the expression of MMP-1 and 13. Previously, various molecules were effective in preclinical level against different targets in OA, but found to be collapsed in clinical

Selective Role of Discoidin Domain Receptor 2 in Murine Temporomandibular Joint Development and Aging.

Temporomandibular joint (TMJ) disorders are often associated with development of osteoarthritis-like changes in the mandibular condyle. Discoidin domain receptor 2 (DDR2), a collagen receptor preferentially activated by type I and III collagen found in the TMJ and other fibrocartilages, has been associated with TMJ degeneration, but its role in normal joint development has not been previously examined. Using Ddr2 LacZ-tagged mice and immunohistochemistry, we found that DDR2 is preferentially expressed and activated in the articular zone of TMJs but not knee joints. To assess the requirement fo

Discoidin domain receptor 1 on bone marrow-derived cells promotes macrophage accumulation during atherogenesis.

RATIONALE: We described a critical role for the discoidin domain receptor (DDR)1 collagen receptor tyrosine kinase during atherosclerotic plaque development. Systemic deletion of Ddr1 in Ldlr(-/-) mice accelerated matrix accumulation and reduced plaque size and macrophage content. However, whether these effects reflected an independent role for macrophage DDR1 during atherogenesis remained unresolved. METHODS: In the present study, we performed sex-mismatched bone marrow transplantation using Ddr1(+/+);Ldlr(-/-) and Ddr1(-/-);Ldlr(-/-) mice to investigate the role of macrophage DDR1 during at

Consistent and selective expression of the discoidin domain receptor-1 tyrosine kinase in human brain tumors.

OBJECTIVE: Few molecular targets are both consistently and selectively expressed in a majority of central nervous system (CNS) neoplasms. Receptor tyrosine kinases have been implicated in brain tumor oncogenesis. We previously isolated one such receptor, discoidin domain receptor-1 (DDR1), from high-grade pediatric brain tumors. Here, we analyze the cellular origin and distribution of DDR1 expression in human brain tumors and its expression in tumor cells relative to surrounding brain. METHODS: By use of a digoxigenin-labeled DDR1 riboprobe, we investigated the expression of DDR1 messenger ri

Discoidin domain receptor 1 contributes to colorectal cancer progression by promoting cell migration and invasion.

Colorectal cancer (CRC) progression is strongly influenced by interactions between tumor cells and the collagen-rich extracellular matrix. Discoidin domain receptor 1 (DDR1), a collagen-activated receptor tyrosine kinase, has been associated with poor prognosis in several epithelial malignancies, including CRC. However, the mechanisms by which DDR1 contributes to metastatic initiation remain incompletely understood. Here, we investigated the role of DDR1 in regulating CRC cell migration and invasion within type I collagen-rich microenvironments. DDR1 overexpression in low-invasive HT-29 cells

Discoidin domain receptor 2 is an important modulator of BMP signaling during heterotopic bone formation.

Bone morphogenetic proteins are essential for bone regeneration/fracture healing but can also induce heterotopic ossification (HO). Understanding accessory factors modulating BMP signaling would provide both a means of enhancing BMP-dependent regeneration while preventing HO. This study focuses on the ability of the collagen receptor, discoidin domain receptor 2 (DDR2), to regulate BMP activity. As will be shown, induction of bone formation by subcutaneous BMP2 implants is severely compromised in Ddr2-deficient mice. In addition, Ddr2 deficiency attenuates HO in mice expressing the ACVR1 mutat
