Pneumocystosis associated with canine distemper virus infection in a mink.

An adult mink from a farm experiencing 100% mortality in affected animals was submitted for diagnostic examination. Clinical history included signs of respiratory disease, oculonasal discharge, and thickening of footpads. Canine distemper virus and Pneumocystis carinii were identified in lung tissue, suggesting immunosuppresion and secondary infection due to morbillivirus disease.

Unusual Canine Distemper Virus Infection in Captive Raccoons (Procyon lotor).

Canine morbillivirus, also known as canine distemper virus (CDV), is the causative agent of canine distemper (CD), which is a serious contagious disease of canines, large felids, and, occasionally, raccoons. This study included seven raccoons from the Timisoara Zoological Garden, Romania. CDV was detected using RT-qPCR on blood samples, but several other exams were also performed-clinical, bacteriological, immunohistochemistry (IHC) and histopathology, toxicological screening, and necropsy-which confirmed CDV infection. Severe digestive disorders (diarrhea and frequent hematemesis) were observ

Antiviral Screen against Canine Distemper Virus-Induced Membrane Fusion Activity.

Canine distemper virus (CDV), a close relative of the human pathogen measles virus (MeV), is an enveloped, negative sense RNA virus that belongs to the genus Morbillivirus and causes severe diseases in dogs and other carnivores. Although the vaccination is available as a preventive measure against the disease, the occasional vaccination failure highlights the importance of therapeutic alternatives such as antivirals against CDV. The morbilliviral cell entry system relies on two interacting envelope glycoproteins: the attachment (H) and fusion (F) proteins. Here, to potentially discover novel e

A canine distemper virus epidemic in Serengeti lions (Panthera leo).

Canine distemper virus (CDV) is thought to have caused several fatal epidemics in canids within the Serengeti-Mara ecosystem of East Africa, affecting silver-backed jackals (Canis mesomelas) and bat-eared foxes (Otocyon megalotis) in 1978 (ref. 1), and African wild dogs (Lycaon pictus) in 1991 (refs 2, 3). The large, closely monitored Serengeti lion population was not affected in these epidemics. However, an epidemic caused by a morbillivirus closely related to CDV emerged abruptly in the lion population of the Serengeti National Park, Tanzania, in early 1994, resulting in fatal neurological d

SEROLOGIC RESPONSE TO CANINE DISTEMPER VACCINATION IN CAPTIVE LINNAEUS'S TWO-TOED SLOTHS ( CHOLOEPUS DIDACTYLUS) AFTER A FATAL CANINE DISTEMPER VIRUS OUTBREAK.

Canine distemper virus (CDV) affects many wild and captive, nondomestic species worldwide but has not been previously reported in Xenarthra. Paucity of information on vaccination safety and efficacy presents challenges for disease prevention in captive collections. CDV infections and subsequent mortalities in five captive Linnaeus's two-toed sloths ( Choloepus didactylus) in eastern Tennessee are reported. Clinical signs included oculonasal discharge, oral ulcerations, and diarrhea, and the diagnosis was confirmed by necropsy, histopathology, immunohistochemistry, virus isolation, and polymera

Study on immunogenicity of recombinant ferritin hemagglutinin of canine distemper virus.

BACKGROUND: Canine distemper virus (CDV) hemagglutinin protein (H), as one of the surface glycoproteins of the virus, helps attach the virus to the host cell through its interaction with cell receptors, makes hemagglutinin protein a key target for the development of neutralizing antibodies. METHODS: In this study, H protein sequences were analyzed by conservation analysis and prediction of multiple immune cell epitopes, three possible epitope sequences were identified. The epitopes were ligated to Ferritin vector with linkers to construct pET30a-Ferritin-Hemagglutinin-Canine distemper virus (

Induction of chronic neurologic disease in mice with canine distemper virus.

The capacity of a mouse-adapted strain of canine distemper virus (CDV) to induce central nervous system (CNS) disease in weanling mice was investigated. Lethality of infection was found to be mouse-strain-dependent. In sensitive strains, an acute meningoencephalomyelitis developed. Brain tissue from acutely ill animals demonstrated numerous foci of viral antigen, and extracts yielded infectious virus. Mice of resistant strains, notably the SJL strain, survived the effects of acute infection, appeared well for several weeks, and then began to develop signs of subacute CNS disease. Preliminary h

Epitope mapping and a candidate vaccine design from canine distemper virus.

BACKGROUND: Canine distemper (CD) is a worldwide spread disease that has been described in 12 families of mammals, especially in the Carnivora order, being better studied in domestic canines where vaccination represents the best means of control. CD is controlled by vaccination, but many cases of the disease still occur in vaccinated animals. AIM: The aim of this work was to study antigen-specific epitopes that can subsidize the development of a new vaccine approach. METHODS: Mapping of T cell reactive epitopes for CD virus (CDV) was carried out through enzyme-linked immunospot assays using

Increased frequency of canine distemper virus-specific antibodies in multiple sclerosis.

BACKGROUND AND PURPOSE: Canine distemper virus (CDV) is a candidate agent in the etiology of multiple sclerosis (MS). Elevated anti-CDV levels were previously found in the sera from MS patients compared with controls. We now investigated whether there was an age-related association with the presence of antibodies specific to CDV-hemagglutinin (H) protein in MS. METHODS: Sera from patients with MS, other neurological diseases, and inflammatory and/or autoimmune diseases, and healthy individuals were screened for anti-CDV in an ELISA using linear peptides of the CDV-H protein as antigen. A recombinant canine distemper virus expressing interleukin-7 enhances humoral immunity.

Canine distemper (CD) causes gastrointestinal and respiratory and/or neurological signs and results in high morbidity and mortality, remaining a threat to carnivores around the world. Live-attenuated vaccines have been widely used to reduce the number of CD outbreaks, but efforts are still needed to improve immune efficiency. Interleukin-7 (IL-7) has been reported to boost host immunity by recruiting follicle helper T (TFH) or germinal center (GC) B cells. Here, we constructed a recombinant canine distemper virus (rCDV) by reverse genetics and evaluated the properties of six intergenic sites f

Canine distemper virus infection of weanling mice. Pathogenesis of CNS disease.

Intracerebral inoculation of canine distemper virus (CDV) into weanling BALB/c, ICR an SJL mice produced acute encephalitis 2 - 3 weeks later. All animals that became ill exhibited lethargy an hind-limb paralysis followed by death. BALB/c mice were more susceptible to CDV than ICR or SJL mice. Pathologic changes were characterized primarily by meningitis and focal areas of parenchymal inflammation, necrosis and microglial hypertrophy. CDV was readily isolated from infected mouse brain 10 - 23 days after infection, and peak virus titers occurred at the height of neurologic disease 14 - 17 days

Immunogenic evaluation of a recombinant rabies virus expressing canine distemper virus H gene and canine parvovirus VP2 gene.

In clinical practice, canine infectious diseases including rabies, canine distemper, and canine parvovirosis pose substantial health threats to canines. In this study, we constructed a recombinant rabies virus (RABV) rHEP-Flury(H+VP2) expressing the canine distemper virus (CDV) H gene and canine parvovirus (CPV) VP2 gene, and evaluated its growth characteristics and immunogenicity. The recombinant virus was generated by inserting the CDV H gene and CPV VP2 gene between the G and L genes of HEP-Flury strain. We found that the recombinant virus maintained high genetic stability after 20 consecut

Coinfection with Canine Distemper Virus and Yellow Fever Virus in a Neotropical Primate in Brazil.

We describe a natural coinfection with canine distemper virus (CDV) and yellow fever virus in a free-ranging neotropical primate of the genus Callithrix, found dead in the northeastern region of Brazil. The laboratory diagnosis included histopathology, immunohistochemistry, rRT-PCR, and phylogenetic analyses. The CDV sequences from this primate in Brazil represent a divergent lineage in Rio Grande do Norte, closely related to genotypes EU1/South America 1 and South America 2. To our knowledge, this is the first report of natural coinfection by CDV and yellow fever virus in a neotropical primat

Antibody study in canine distemper virus nucleocapsid protein gene-immunized mice.

The gene for the nucleocapsid (N) protein of canine distemper virus was cloned into the pMD-18T vector, and positive recombinant plasmids were obtained by enzyme digestion and sequencing. After digestion by both EcoRI and KpnI, the plasmid was directionally cloned into the eukaryotic expression vector pcDNA; the positive clone pcDNA-N was screened by electrophoresis and then transfected into COS-7 cells. Immunofluorescence analysis results showed that the canine distemper virus N protein was expressed in the cytoplasm of transfected COS-7 cells. After emulsification in Freund's adjuvant, the r

Canine distemper virus L gene: sequence and comparison with related viruses.

We cloned the genomic RNA of canine distemper virus (CDV) and determined the nucleotide sequence of the large (L) protein-coding gene. The L gene is 6573 nucleotides long and contains a single open reading frame coding for a polypeptide of 2161 amino acids (MW 246,354). The precise 5' end of the viral genome consists of a 38-nucleotide leader region. The CDV L protein shows over 77% amino acid similarity with its morbilliform relative measles virus (MV) with nearly 67% of their amino acids conserved. The sequence homology of 11 negative strand viruses L proteins is compared and relatedness was

Serological detection of infection with canine distemper virus, canine parvovirus and canine adenovirus in communal dogs from Zimbabwe.

Domestic dogs are common amongst communities in sub-Saharan Africa and may serve as important reservoirs for infectious agents that may cause diseases in wildlife. Two agents of concern are canine parvovirus (CPV) and canine distemper virus (CDV), which may infect and cause disease in large carnivore species such as African wild dogs and African lions, respectively. The impact of domestic dogs and their diseases on wildlife conservation is increasing in Zimbabwe, necessitating thorough assessment and implementation of control measures. In this study, domestic dogs in north-western Zimbabwe wer

Overcoming the Barrier of the Respiratory Epithelium during Canine Distemper Virus Infection.

Canine distemper virus (CDV) is a highly contagious pathogen and is known to enter the host via the respiratory tract and disseminate to various organs. Current hypotheses speculate that CDV uses the homologous cellular receptors of measles virus (MeV), SLAM and nectin-4, to initiate the infection process. For validation, here, we established the well-differentiated air-liquid interface (ALI) culture model from primary canine tracheal airway epithelial cells. By applying the green fluorescent protein (GFP)-expressing CDV vaccine strain and recombinant wild-type viruses, we show that cell-free
