Bioinformatics analysis of genes associated with disulfidptosis in spinal cord injury.

Research findings indicate that programmed cell death (PCD) plays a pivotal role in the pathophysiology of spinal cord injury (SCI), and a recently discovered form of cell death, disulfidptosis, has emerged as a novel phenomenon. However, the characterization of disulfidptosis-related genes in SCI remains insufficiently explored. We retrieved SCI-related data from the Gene Expression Omnibus (GEO) database and identified three key genes associated with disulfidptosis in human SCI (CAPZB, SLC3A2, and TLN1), whose mediated signaling pathways are closely intertwined with SCI. Subsequent functiona

Prognostic and therapeutic implications of disulfidptosis-related genes in multiple myeloma.

BACKGROUND: Multiple myeloma (MM), a malignancy of plasma cells in the bone marrow, urgently requires novel prognostic biomarkers. However, the prognostic significance of disulfidptosis-related genes and their association with treatment response in MM remain unclear. METHODS: Transcriptomic data from MM samples were obtained from the Gene Expression Omnibus (GEO) database. A disulfidptosis-related prognostic model was constructed using LASSO-Cox regression analysis. The performance of the model was evaluated, and its clinical relevance to treatment response was subsequently assessed. Finally,

Pathogenesis of Osteoarthritis: Mechanisms of Action of Disulfidptosis and Targeted Therapeutic Strategies.

The onset of osteoarthritis (OA) involves the interplay of mechanical stress, inflammatory responses, and metabolic disorders. Pro-inflammatory cytokines, including IL-1β, TNF-α, and IL-6, drive cartilage degradation, synovitis, and subchondral bone remodeling through activation of NF-κB and MAPK signaling pathways. Recent studies have identified disulfidptosis, a novel form of programmed cell death, and suggest its potential involvement in OA pathogenesis. This death modality is triggered by abnormal intracellular accumulation of disulfide bonds, dependent on high SLC7A11 expression and NADPH

Disulfidptosis: A Metabolic Cell Death Mechanism with Therapeutic Potential in Cancer.

Disulfidptosis is a newly identified form of regulated cell death (RCD) first described in 2023, representing a significant advance in understanding programmed cell death pathways. This unique cell death modality is characterized by abnormal intracellular accumulation of disulfide bonds and disruption of redox homeostasis, leading to cytoskeletal collapse without caspase activation. Disulfidptosis is primarily triggered by glucose deprivation in cells with high expression of solute carrier family 7 member 11 (SLC7A11). Under these conditions, insufficient NADPH supply prevents the effective re

Experimental Detection Methods and Clinical Translational Challenges of Disulfidptosis in Cancer.

Disulfidptosis was originally characterized as a programmed cell death modality reliant on SLC7A11 overexpression, which ultimately induces cytoskeleton collapse. This cell death pattern offers innovative research perspectives and therapeutic strategies for the treatment of drug-resistant tumors with elevated SLC7A11 expression. Nevertheless, the recent identification of non-canonical disulfidptosis pathways, including mitochondrial stress and TrxR1 inhibition, indicates that the upstream and downstream metabolic regulatory networks governing disulfidptosis are far more intricate than previous

Oxygen pump microneedles coupling cuproptosis and disulfidptosis to enhance melanoma immunotherapy.

Cuproptosis and disulfidptosis are new types of programmed cell death with great potential for tumor immunotherapy, but their efficacy is severely restricted by the hypoxic microenvironment and metabolic reprogramming of tumors. Herein, we develop an oxygen-pump microneedle system (CPGH@OMN) co-loading glucose oxidase (GOx) and copper-polyphenol nanoparticles (CP), using sodium percarbonate (SPC) as an in situ oxygen generator and hyaluronic acid (HA) as a tumor-targeting carrier. SPC-mediated oxygen generation acts as a booster to promote deep drug penetration, and downregulates hypoxia-induc

BACKGROUND: Parkinson's disease (PD), a globally prevalent neurodegenerative disorder, has been implicated with oxidative stress (OS) as a central pathomechanism. Excessive reactive oxygen species (ROS) trigger neuronal damage and may induce disulfidptosis-a novel cell death modality not yet characterized in PD pathogenesis. METHOD: Integrated bioinformatics analyses were conducted using GEO datasets to identify PD-associated differentially expressed genes (DEGs). These datasets were subjected to: immune infiltration analysis, gene set enrichment analysis (GSEA), weighted gene co-expression n

Targeting Disulfidptosis with Potentially Bioactive Natural Products in Metabolic Cancer Therapy.

BACKGROUND: Metabolic cancers are defined by metabolic reprogramming. Although this reprograming drives rapid tumour growth and invasion, it also reveals specific metabolic vulnerabilities that can be therapeutically exploited in cancer therapy. A novel form of programmed cell death, known as disulfidptosis, was identified last year; tumour cells with high SLC7A11 expression undergo disulfidptosis when deprived of glucose. Natural products have attracted increasing attention and have shown potential to treat metabolic cancers through diverse mechanisms. METHODS: We systematically searched ele

Disulfide-Bridged Ru Complex-Mediated Photo-Disulfidptosis for Colorectal Cancer Therapy.

Despite numerous therapeutic strategies targeting specific programmed cell death pathways, effectively eliminating malignant colorectal cancer (CRC) remains a significant challenge. Disulfidptosis, a newly identified form of cell death, is characterized by rapid NADPH depletion and abnormal disulfide bond formation in cytoskeletal proteins in cells with high SLC7A11 expression under glucose starvation. Given the aberrant expression of SLC7A11 and the hypermetabolic phenotype of CRC cells, targeting the disulfidptosis pathway offers a promising therapeutic approach for CRC treatment. Here, we d

The AMPK activator A769662 promotes ferroptosis and suppresses disulfidptosis by inhibiting SLC7A11.

AMP-activated protein kinase (AMPK) plays a critical role in maintaining cellular energy homeostasis. Activation of AMPK negatively regulates ferroptosis, a form of cell death caused by iron-dependent accumulation of lipid peroxides. In the present study, we unexpectedly found that A769662, widely used as a selective AMPK activator, inhibited SLC7A11-mediated cystine uptake, thereby decreasing intracellular reduced glutathione (GSH) levels and promoting ferroptosis in glioblastoma cells. Deletion of the AMPKα1 gene or treatment with an AMPK inhibitor compound C did not inhibit A769662-induced

BACKGROUND: Disulfidptosis and ferroptosis are different programmed cell death modes, which are closely related to the development of a variety of diseases, but the relationship between them and ulcerative colitis (UC) is still unclear. Therefore, our study aimed to explore the molecular subtypes and biomarkers associated with disulfidptosis-related ferroptosis (DRF) in UC. METHODS: We used Pearson analysis to identify DRF genes. Then, we classified 140 UC samples into different subtypes based on the DRF genes and explored the biological and clinical characteristics between them. Next, the hu

Nanoparticle-based strategies targeting disulfidptosis, anoikis, and PANoptosis for enhanced cancer therapy.

The evasion of regulated cell death (RCD) is a distinct hallmark of cancer that drives tumor progression, metastatic dissemination, and therapeutic resistance. Recent discoveries of non-canonical cell death pathways, namely disulfidptosis, anoikis, and PANoptosis, have provided promising new directions for overcoming resistance to conventional therapies. Disulfidptosis is characterized by intracellular disulfide stress and actin cytoskeleton collapse under glucose starvation; anoikis serves as an extracellular matrix detachment-induced barrier to metastasis; and PANoptosis acts as a highly reg

Disulfidptosis links the pathophysiology of ulcerative colitis and immune infiltration in colon adenocarcinoma.

Ulcerative colitis (UC), a chronic inflammatory bowel disease, significantly increases the risk of colon adenocarcinoma (COAD). Disulfidptosis, a novel form of programmed cell death, has been implicated in various diseases, including UC. This study investigates the expression of disulfidptosis-related genes, particularly CD2AP and MYH10, in UC and COAD. Through analysis of public datasets, we found MYH10 significantly upregulated and CD2AP downregulated in UC compared to healthy controls, with consistent patterns in COAD. Immune infiltration analysis revealed correlations between these genes a
