Single-Cell Transcriptomics Reveals the Ameliorative Effect of Oridonin on Septic Liver Injury.

Sepsis is a life-threatening syndrome leading to hemodynamic instability and potential organ dysfunction. Oridonin, commonly used in Traditional Chinese Medicine (TCM), exhibits significant anti-inflammation activity. To explore the protective mechanisms of oridonin against the pathophysiological changes, the authors conducted single-cell transcriptome (scRNA-seq) analysis on septic liver models induced by cecal ligation and puncture (CLP). They obtained a total of 63,486 cells, distributed across 11 major cell clusters, and concentrated their analysis on four specific clusters (hepatocytes/He

Biotechnological interventions of in vitro propagation and production of valuable secondary metabolites in Stevia rebaudiana.

Plant cell and tissue culture makes provision of a sustainable and nature-friendly strategy for the production of secondary metabolites, and modern progress in gene editing and genome engineering provides novel possibilities to improve both the qualitative and quantitative aspects of such phytochemicals. The ever-expanding quest for plant-based medicine to treat diabetes facilitates large-scale cultivation of Stevia rebaudiana to enhance the yield of its much-coveted low-calorie sweetener glycosides. The potential to process stevia as a "natural" product should enhance the acceptance of stevio

Antibacterial activity of trichorabdal A from Rabdosia trichocarpa against Helicobacter pylori.

Rabdosia trichocarpa is used in a popular home-made remedy for gastric and stomachic complaints in Japan. A diterpene, trichorabdal A from R. trichocarpa, showed a very strong in vitro antibacterial activity against Helicobacter pylori. With regard to the antibacterial activity of the extract and constituents of R. trichocarpa against H. pylori, the effect of traditional use of this plant for the treatment of gastritis is probably due to the suppression of H. pylori since the extract of R. trichocarpa particularly inhibits the growth of H. pylori. It could be a promising native herb treatment

Mutations on ent-kaurene oxidase 1 encoding gene attenuate its enzyme activity of catalyzing the reaction from ent-kaurene to ent-kaurenoic acid and lead to delayed germination in rice.

Rice seed germination is a critical step that determines its entire life circle, with seeds failing to germinate or pre-harvest sprouting both reduce grain yield. Nevertheless, the mechanisms underlying this complex biological event remain unclear. Previously, gibberellin has been shown to promote seed germination. In this study, a delayed seed germination rice mutant was obtained through screening of the EMS induced mutants. Besides of delayed germination, it also shows semi-dwarfism phenotype, which could be recovered by exogenous GA. Through re-sequencing on the mutant, wild-type and their

Discovery of lysosome-targeted covalent anticancer agents based on isosteviol skeleton.

Covalent drugs play corresponding bioactivities by forming covalent bonds with the target, which possess many significant pharmacological advantages including high potency, ligand efficiency, and long-lasting effects. However, development of covalent inhibitors is a challenge due to their presumed indiscriminate reactivity. Here, we report the discovery of series of lysosome-targeting covalent anticancer agents by introducing nitrogenous bases to the modified isosteviol skeleton in order to minimize the toxicity and increase the selectivity. By introducing the electrophilic α, β-unsaturated ke

Preventive and chronic mineralocorticoid receptor antagonism is highly beneficial in obese SHHF rats.

BACKGROUND AND PURPOSE: Mineralocorticoid receptor (MR) activation contributes to heart failure (HF) progression. Its overactivity in obesity is thought to accelerate cardiac remodelling and HF development. Given that MR antagonists (MRA) are beneficial in chronic HF patients, we hypothesized that early MRA treatment may target obesity-related disorders and consequently delay the development of HF. EXPERIMENTAL APPROACH: Twenty spontaneously hypertensive HF dyslipidaemic obese SHHF(cp/cp) rats and 18 non-dyslipidaemic lean SHHF(+/+) controls underwent regular monitoring for their metabolic an

Clerodane Diterpenes and Structural Modification of 12-Oxo-Hardwickiic Acid from the Roots of Croton krabas: α‑Glucosidase Inhibitory Activity, Molecular Docking, and ADMET.

The roots of Croton krabas have traditionally been used as a water decoction for treatment of bloating and flatulence. The preliminary bioactivity screening revealed that the crude n-hexane and EtOAc extracts exhibited significant α-glucosidase inhibitory activity. Accordingly, the first phytochemical investigation of the roots of C. krabas led to the isolation of two previously undescribed clerodane diterpenes (1 and 2), together with eight known compounds (3-10). The structures and stereochemistry were elucidated by extensive analysis of spectroscopic data (UV, IR, 1H NMR, 13C NMR, and 2D NM

Oridonin targets PRDX1 to promote apoptosis by inducing ROS-mediated ER stress and modulating autophagy.

Oridonin is a bioactive diterpenoid derived from the widely used traditional Chinese medicinal herb Rabdosia rubescens, exhibits broad-spectrum anti-cancer activity, with several derivatives currently in clinical trials. However, the molecular mechanism underlying its anticancer effects, especially its direct target proteins, remain to be fully elucidated. Here, we found that Oridonin promoted intracellular reactive oxygen species (ROS) accumulation, which in turn induced endoplasmic reticulum (ER) stress-mediated apoptosis. Moreover, ER stress was instrumental in inducing autophagy after Orid
