Effect of Dopamine D2 Receptor Antagonists on [18F]-FEOBV Binding.

The interaction of dopaminergic and cholinergic neurotransmission in, e.g., Parkinson's disease has been well established. Here, D2 receptor antagonists were used to assess changes in [18F]-FEOBV binding to the vesicular acetylcholine transporter (VAChT) in rodents using positron emission tomography (PET). After pretreatment with either 10 mg/kg haloperidol, 1 mg/kg raclopride, or vehicle, 90 min dynamic PET scans were performed with arterial blood sampling. The net influx rate (Ki) was obtained from Patlak graphical analysis, using a metabolite-corrected plasma input function and dynamic PET

The Universal 3D QSAR Model for Dopamine D2 Receptor Antagonists.

In order to search for novel antipsychotics acting through the D2 receptor, it is necessary to know the structure-activity relationships for dopamine D2 receptor antagonists. In this context, we constructed the universal three-dimensional quantitative structure-activity relationship (3D- QSAR) model for competitive dopamine D2 receptor antagonists. We took 176 compounds from chemically different groups characterized by the half maximal inhibitory concentration (IC50)from the CHEMBL database and docked them to the X-ray structure of the human D2 receptor in the inactive state. Selected docking

Acetaminophen improves tardive akathisia induced by dopamine D2 receptor antagonists.

Tardive akathisia is a movement disorder characterized by internal restlessness with an uncontrollable urge to move, leading to repetitive movements. It is a common side effect of long-term treatment with dopamine D2 receptor antagonists. In the present study, we analyzed the FDA Adverse Event Reporting System and IBM MarketScan Research Database to find a drug that can be used concomitantly with dopamine D2 receptor antagonists and still reduce the risk of akathisia. Acetaminophen was determined to be the most effective akathisia-suppressing drug. In an experimental validation of the hypothes

Distinct Dopamine D₂ Receptor Antagonists Differentially Impact D₂ Receptor Oligomerization.

Dopamine D₂ receptors (D₂R) are known to form transient homodimer complexes, of which the increased formation has already been associated with development of schizophrenia. Pharmacological targeting and modulation of the equilibrium of these receptor homodimers might lead to a better understanding of the critical role played by these complexes in physiological and pathological conditions. Whereas agonist addition has shown to prolong the D₂R dimer lifetime and increase the level of dimer formation, the possible influence of D₂R antagonists on dimerization has remained rather unexplored. Here,

Role of dopamine receptors in vesicourethral function. A urodynamic study with dopamine receptor antagonist metoclopramide.

The effect of intravenous administration of 20 mg metoclopramide, a dopamine receptor antagonist upon cystometry, urethral pressure profile and external urethral sphincter electromyography (EMG) was investigated in 10 patients with neurogenic bladder dysfunction. No significant change was noticed in detrusor reflex activity or in peak urethral pressure. This suggests that dopamine receptors are not important in vesicourethral function. The increase in sphincter EMG activity observed was probably due to the cholinergic effect of metoclopramide.

Modulation of dopamine uptake in rat nucleus accumbens: effect of specific dopamine receptor antagonists and sigma ligands.

The ability of dopamine (DA) antagonists and sigma receptor ligands to alter [(3)H]-DA uptake was examined using synaptosomes prepared from the nucleus accumbens of female rats. Pre-incubation with compounds having a high affinity for sigma (rimcazole, haloperidol, and spiperone) receptors produced dose dependent inhibition of (3)H-DA uptake. Sulpiride, a pure DA D(2) antagonist had no effect. In contrast, DA uptake was potentiated in response to (+)-3-(3-hydroxyphenyl)-N-(1-propyl)piperidine, a mixed sigma receptor antagonist and DA D(2) receptor agonist. Similarly, SKF-10,047, a selective si

Effects of dopamine receptor antagonists on ongoing maternal behavior in rats.

The effects of different peripheral doses of four dopamine (DA) receptor antagonists on general activity and maternal behavior were examined in lactating female rats. Administration of the classic D1-like and D2-like DA receptor blocker haloperidol (0.1 and 0.05 mg/kg) disrupted pup retrieval and nest-building behaviors and reduced motor activity. Pimozide (0.5 and 0.2 mg/kg), which has more affinity for DA D2-like receptors, mildly disrupted pup retrieval while showing no significant influence on open-field behaviors. The putative DA D(4) receptor blocker, clozapine (1.5 and 1.0 mg/kg) reduce

Development of uterine adenomyosis after treatment with dopamine antagonists in mice.

Development of uterine adenomyosis was studied in SHN mice treated with psychotherapeutic drugs, sulpiride and perphenazine, and gastroenteric drug, metoclopramide, which act as dopamine antagonists to increase prolactin release from the pituitary gland. Administration of these drugs twice daily for 40-70 or 40-90 days of age induced an elevation in serum level of prolactin. Furthermore, the treated mice showed a prolongation of metestrous plus diestrous phase and a high incidence of uterine adenomyosis compared with vehicle-treated control mice. These results indicate that hyperprolactinemia

Effects of dopamine antagonists injected through vortex veins on intraocular pressure.

In order to avoid the factors of the corneal barrier and tearing washout on absorption of eye drops, dopamine and antagonists were injected into rabbit eyeballs through vortex veins. Many dopamine antagonists, such as haloperidol, moperone, metoclopramide, trifluperidol, lenperone, and fluoropipamide, lowered the intraocular pressure (IOP), whereas a few of them, such as chlofluperol and trifluoperazine, raised the IOP. Furthermore, it was found that dopamine agonists also could either increase or decrease the IOP. The ocular hypertensive effect of dopamine was blocked by haloperidol as expect

Behavioural correlates to the dopamine D-1 and D-2 antagonists.

The acute dopamine (DA) receptor blockade of neuroleptics can be demonstrated in mice by antagonism of stereotypies induced by the DA-agonist methylphenidate and in rats by antagonism of stereotypies induced by the DA-agonists amphetamine or apomorphine. Neuroleptics such as the thioxanthene, cis(Z)-flupentixol, the phenothiazine, fluphenazine, the butyrophenone, haloperidol and the benzamide clebopride are equipotent behaviourally as well as clinically. Also the D-1 receptor-antagonist SCH 23390 has the same pharmacological effects. In a series of experiments where the methylphenidate-induced

Dopamine antagonists do not block testosterone-induced decrease in luteinizing hormone.

These studies were designed to test the hypothesis that testosterone might lower circulating levels of luteinizing hormone (LH) in castrated male rats by activating dopaminergic neuronal systems. Blood samples were drawn from conscious rats through an indwelling cannula during the fourth and eight hours after subcutaneous administration of testosterone (1 or 5 mg/kg, in corn oil). Testosterone caused a dose-dependent decrease in plasma LH levels eight hours after administration. This decrease was not blocked by prior administration of the dopaminergic antagonists haloperidol and flupenthixol.

OATP1A2 and OATP2B1 Are Interacting with Dopamine-Receptor Agonists and Antagonists.

Interaction with the dopaminergic system in the central nervous system is either therapeutically intended or it is a side effect. In both cases, dopamine-receptor agonists (DRA) like the ergoline derivative bromocriptine and dopamine-receptor antagonists (DRAn) like metoclopramide have to cross the blood-brain barrier (BBB). The organic anion transporting polypeptides (OATP) 1A2 and 2B1 are cellular uptake carriers for a variety of endogenous and xenobiotic compounds. As both transporters are expressed in endothelial cells of the BBB, the aim of the present study was to determine whether the D

Dopamine receptor antagonists modulate glucose uptake in rat pheochromocytoma (PC12) cells.

A variety of dopaminergic ligands were evaluated for their ability to alter glucose transport in PC12 cells. Certain antipsychotic drugs which targeted D2 dopamine receptors, such as pimozide, fluphenazine and chlorpromazine, inhibited glucose uptake (with IC50's in the range of 2-40 microM). By contrast, haloperidol and sulpiride (also D2 antagonists) showed marginal activity. The atypical antipsychotic drug, clozapine (a D4 antagonist), also effectively inhibited glucose transport by the cells. Ligands specific for D1 receptors did not interfere with glucose uptake. Time course studies revea

Effects of AMPA receptor antagonists on dopamine-mediated behaviors in mice.

Current data indicate that dopaminergic and glutamatergic neurotransmitter systems interact. The role of alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) glutamate receptor subtypes in modulating dopamine neurotransmission, however, remains unclear. The noncompetitive AMPA antagonists, GYKI 52466 (5-40 mg/kg) and LY300164 (1-6 mg/kg), and the competitive AMPA antagonists, LY326325 (5-80 mg/kg) and NBQX (10-80 mg/kg), were compared to the dopamine antagonist, haloperidol (0.03-1.0 mg/kg), for their ability to inhibit dopamine-mediated behaviors after i.p. administration in mice.

Dopamine antagonists in tic-related and psychotic spectrum obsessive compulsive disorder.

Serotonin uptake inhibitors (SUIs) have been established as the first-line pharmacotherapy of obsessive compulsive disorder (OCD). However, approximately one half of patients who receive an adequate trial with these agents remain clinically unchanged. The addition of drugs that enhance serotonin (5-HT) neurotransmission, such as lithium and buspirone, to ongoing treatment in SUI-refractory patients has generally proved to be an ineffective strategy. The addition of dopamine antagonists to the regimens of SUI-resistant patients appears to be a useful approach for OCD patients with a comorbid ch

Tachykinin gene expression in rat limbic nuclei: modulation by dopamine antagonists.

The content and nature of the preprotachykinin (PPT; i.e., substance P/neurokinin A-encoding) messenger RNAs (mRNAs) present in rat brain striatum and limbic tissues were determined by RNA protection experiments. The rank order of PPT mRNA concentration was striatum greater than nucleus accumbens much greater than bed nucleus of the stria terminalis greater than hypothalamus, amygdala and septum. The proportion of beta-(full length) to gamma-(minus exon 4) PPT mRNA was invariant (40/60) among the tissues tested. Because these brain regions receive prominent dopaminergic innervations, the effec

Calcium alters the sensitivity of intact horizontal cells to dopamine antagonists.

Horizontal cells of the carp retina possess dopamine receptors linked to adenylate cyclase. Isolated, intact horizontal cells respond to micromolar concentrations of dopamine, whereas nanomolar concentrations of haloperidol, (+)-butaclamol, and flupenthixol block the dopamine response. Preincubation in Ringer's solution containing increased levels of Ca2+ (5-110 mM) decreases the sensitivity of the cells to these antagonists by 1,000-10,000 times. Dopamine sensitivity of the cells is not affected by Ca2+ levels in the preincubation medium. Preincubation of the cells in Ringer's solution contai

Parthenolide acts as a cocaine antagonist in the human dopamine transporter.

In this work, I describe the effects of parthenolide, a naturally occurring sesquiterpene lactone, on radiolabeled cocaine binding and on radiolabeled dopamine uptake in the human dopamine transporter expressed on HEK-293 cells (HEK/DAT cells). Parthenolide displaces radiolabeled cocaine in a concentration-dependent manner and does not display any significant inhibition of radiolabeled dopamine uptake. Further, parthenolide seems to alleviate the inhibition of dopamine uptake by 5 μM unlabeled cocaine at sub-toxic parthenolide concentrations.

Olanzapine: a serotonin-dopamine-receptor antagonist for antipsychotic therapy.

The pharmacology, pharmacokinetics, clinical efficacy, adverse effects, drug interactions, dosage and administration, and cost of olanzapine are reviewed. Olanzapine is a serotonin-dopamine-receptor antagonist indicated for use in the treatment of schizophrenia and other psychotic disorders. The affinity of olanzapine for neuroreceptors is similar to that of clozapine. The drug is well absorbed from the GI tract; food has no effect. Olanzapine is more effective than placebo and equal to haloperidol in reducing psychotic symptoms on two rating scales. However, unlike typical dopamine-receptor a

The influence of dopamine agonists and antagonists on gastric lesions in mice.

The dopamine agonist bromocriptine and L-dopa significantly inhibited whereas dopamine antagonist haloperidol aggravated the gastric lesions induced by pylorus ligation in mice as found earlier for rats. Furthermore, the successful use of a dopamine antagonist alone for the induction of gastric lesions also in mice was demonstrated, since the gastric lesions were induced by a single dose of haloperidol without any additional noxious treatment. Bromocriptine successfully inhibited both the gastric lesion-potentiating as well as the gastric lesion-inducing effect of haloperidol.
