Assumption of linearity in dose--effect relationships.

As a basis for establishing radiation protection standards, a substantial amount of quantitative information is now available on the frequency with which malignant diseases are induced in man by moderately high doses of radiation. Such estimates can now be made not only for irradiation of the whole body but also for exposure of a number of body organs individually. The frequency with which cancers might follow the much lower doses involved in occupational or environmental exposure to radiation, however, cannot be derived from any available epidemiological surveys. It can at present only be inf

Modulation of radiation-induced chromosome aberrations by DMSO, an OH radical scavenger. 1: Dose-response studies in human lymphocytes exposed to 220 kV X-rays.

Human G0 lymphocytes were exposed to 220 kV X-radiation in the presence or absence of DMSO, an efficient selective scavenger of OH radicals. Our studies demonstrate that DMSO affects a concentration-dependent modulation of induced asymmetrical aberrations in human lymphocytes exposed to approximately 3.0 Gy, with maximum protectible fractions of approximately 70 percent at DMSO concentrations of greater than or equal to 1 M. The dose dependency for dicentrics in lymphocytes acutely exposed to X-ray doses of 0.51 to 4.98 Gy in the absence of DMSO is adequately described by the linear-quadratic

Normal tissue radiosensitivity: prediction on deterministic or stochastic basis?

For the same standardized physical radiation dose there is considerable variation, among different patients, of the magnitude of early and late normal tissue reactions. Technical and clinical factors account for about one third only of these variations. Genetic or epigenetic differences between patients account for the greater proportion of interpatient radiosensitivity differences. Attempts have been made to correlate differences in the tissue radiosensitivity with the in vitro radiosensitivity of fibroblasts or lymphocytes as well as with other biological cellular processes related to cell d

Normal tissue complications after radiation therapy.

This paper describes the biological mechanisms of normal tissue reactions after radiation therapy, with reference to conventional treatments, new treatments, and treatments in developing countries. It also describes biological reasons for the latency period before tissue complications arise, the relationship of dose to incidence, the effect of increasing the size of the irradiated volume, early and late tissue reactions, effects of changes in dose fractionation and dose rate, and combined chemotherapy and radiotherapy responses. Examples are given of increases in knowledge of clinical radiobio

How to avoid phototoxic reactions in photopatch testing with chlorpromazine.

The purpose of this study was to determine the concentration, vehicle, and UVA dose suitable for avoiding phototoxic reactions while revealing photoallergic reactions to chlorpormazine (CPZ) in photopatch testing. Photopatch tests were performed on 27 volunteers with CPZ concentrations between 0.05% and 1% in water or Plastibase with UVA doses of 4 or 7 J/cm2. Photopatch tests were also carried out on 1 patient with photoallergic contact dermatitis to CPZ and an increased UVA sensitivity. The same concentration of CPZ caused phototoxic reactions less frequently and less intensely in Plastibase

Time course of PUVA erythema.

Optimization of the PUVA regime requires knowledge of the time-course and dose-response characteristics of PUVA erythema. Although existing guidelines recommend determination of the minimal phototoxic dose (MPD) 72 hours after UVA exposure and treatment administered 72 hours apart, recent evidence suggests that maximal PUVA erythema occurs beyond this time point, and hence the current assessment time underestimates the phototoxic effect of PUVA. This report reviews the current literature, including the authors' own experience of the characteristics of PUVA erythema, and recommends that the opt

Distribution and behavior of isoselenouronium salts in the body. VI. Comparative study of distribution and radioprotectivity of selenium derivatives.

The protective effect of AESe, 2-ASe, and Se-MEG during irradiation with a dose rate of 500 R/min was followed. The protective effect of 2-ASe is substantially lower during the dose rate of 20 R/min and even during an increased dose. AESe and Se-MEG are noneffective. The time of duration of the protective effect of 2-ASe corresponds to the period of its high concentration in the organism after the administration, i.e. ca. 45 min.

DNA analysis of cattle parasitic protozoan Tritrichomonas foetus after photodynamic therapy.

Photodynamic therapy (PDT) is a modality of therapy that involves the activation of photosensitive substances and the generation of cytotoxic oxygen species and free radicals to promote the selective destruction of target tissues. This study analyzed the application of PDT to Tritrichomonas foetus, a scourged and etiological agent of bovine trichomoniasis, a sexually transmitted infectious disease. As it is an amitochondrial and aerotolerant protozoan, it produces energy under low O2 tension via hydrogenosome. T. foetus from an axenic culture was incubated with photosensitizer tetrasulfonated

Radiation-related thyroid autoimmunity and dysfunction.

The thyroid gland is vulnerable not only to external radiation but also to internal radiation, because the thyroid cells can incorporate radioactive iodine when synthesizing thyroid hormones. Since radiation-induction of thyroid neoplasia, including thyroid cancer, is well recognized, the data on radiation-related thyroid autoimmunity and dysfunction are summarized and reviewed. High-dose irradiation, irrespective of being external or internal, is strongly associated with a risk of hypothyroidism (with the prevalence ranging from 2.4% to 31%) and of Graves' hyperthyroidism (with the prevalence

Distribution and behaviour of isoselenium salts in organism. III. Radioprotective effect of selenium derivatives.

Radioprotective properties of Se-2-aminoethylioselenouronium bromide hydrobromide, 2-aminoselenoazoline and of selenium analogue of mercaptoethylguanidine were followed. Radio-protective effectiveness comparable with classical thiol radioprotectiva was manifested by 2-aminoselenoazoline. In the other two compounds there was on the contrary found a synergism of toxicity and radiation.

[Changes in lymphocyte size after chronic exposure to radiation and chemical factors].

Results of the analysis of changes in peripheral blood lymphocytes size under chronic exposure to external gamma radiation and pesticide chlorophos++ in combination and separately are presented. It has been found out that under exposure of animals to radiation or the pesticide it is small and big lymphocytes respectively which most significantly suffer quantitatively. Under the joint radiation ++-chemical exposure of the organism the number of both types of cells is reduced simultaneously.

Effect of sub-lethal photodynamic inactivation on the antibiotic susceptibility and biofilm formation of clinical Staphylococcus aureus isolates.

BACKGROUND: A promising approach to kill antibiotic-resistant bacteria uses light in combination with a photosensitizer to induce a phototoxic reaction. A major concern with the use of any non-antibiotic antimicrobial treatment is that exposure of bacteria to sub-lethal concentrations will lead to the development of resistance to antibiotics. This study aimed to determine the effect of sub-lethal photodynamic inactivation (PDI) on the antibiotic susceptibility and biofilm formation of clinical Staphylococcus aureus isolates. METHODS: Forty clinical S. aureus isolates were exposed to PDI with

Reactive oxygen species in cell responses to toxic agents.

This review first summarizes experimental data on biological effects of different concentrations of ROS in mammalian cells and on their potential role in modifying cell responses to toxic agents. It then attempts to link the role of steadily produced metabolic ROS at various concentrations in mammalian cells to that of environmentally derived ROS bursts from exposure to ionizing radiation. The ROS from both sources are known to both cause biological damage and change cellular signaling, depending on their concentration at a given time. At low concentrations signaling effects of ROS appear to p

[Reduction of radiation-induced early skin damage (mouse foot) by O-(beta-hydroxyethyl)-rutoside (author's transl)].

The effect of bioflavonoid, O-(beta-hydroxyethyl)-rutoside (HR) on early radiation-induced skin damage was examined, using the mouse foot system; the response to radiation is not species specific and comparison with the clinical situation is therefore possible. The aim was to see whether HR, which is highly effective in protecting against late damage, is also able to reduce early effects. Early reactions were considered to be erythema, swelling and ulceration and occurring up to 30 days after irradiation. It was found that HR significantly reduces early damage, both after a single dose and aft

Low doses of ionizing radiation can prevent radiation-induced colonic epithelial hyporesponsiveness to muscarinic agonists.

PURPOSE: Colonic epithelium hyporesponsiveness to different secretagogues occurs after exposure to ionizing radiation, increasing susceptibility to bacterial translocation and intraluminal toxins. Growing evidence suggests that the biological effects of radiation might be hormetic in nature. We investigated if exposure to low doses of ionizing radiation (LDR) can prevent colon hyposecretion due to subsequent larger doses. METHODS: Rats were exposed to LDR (0.05 Gy) 24 h prior to 6 Gy, high dose radiation (HDR). The cyclic adenosine monophosphate (cAMP)-mediated pathway was explored using fors

Dose rate effectiveness and potentially lethal damage repair in normal and double-strand break repair deficient murine cells by gamma-rays and 5-fluorouracil.

SCID (severe combined immunodeficiency) fibroblasts established from C.B 17-scid/scid embryos showed higher sensitivity to high (1.105 Gy/min) and low (0.00069 Gy/min) dose rate gamma-rays and also to 5-fluorouracil, a cancer sedative producing double-strand breaks, than wildtype cells from C.B17- +/+ embryos. Furthermore, SCID cells were deficient in repairing DNA damage induced by high dose rate gamma-rays even after dose fractionation and after 24 h recovery periods, while wildtype cells showed an apparent repair ability on DNA damage after these gamma-ray exposures. This is the first repor

[Halothane on irradiated liver tissue].

The results of animal experiments show that distinction must be made between acute and chronic tissue changes produced by irradiation of the liver. The pathologico-morphological findings obtained by irradiation of the liver combined with repeated halothane administration during short-term experiments are scanty and dosage-dependent. Long-term observation has revealed higher-dosage irradiation to be followed by minor inflammation which, if combined with regular halothane administration, progresses to produce the patho-anatomical picture of chronic hepatitis with diffuse cell damage and focal ne
