[Development of traditional "drug-drug co-processing" technique: a review].

"Drug-drug co-processing"(DDCP) is a distinctive technique in the field of traditional Chinese medicine(TCM) processing. By synergistically processing target materials with specific drugs or medicinal juices, it demonstrates significant advantages in toxicity reduction, efficacy enhancement, and the modulation of medicinal properties. This study systematically reviews the historical evolution of DDCP and innovatively constructs a tripartite technical classification framework encompassing dry drug processing, medicinal juice processing, and fractional processing. The study finds that

Kidney Pathophysiology, Toxicology, and Drug-Induced Injury in Drug Development.

Anatomically, the kidneys are paired, bean-shaped (in most mammals), excretory organs that lie in the retroperitoneum. High blood flow to the kidneys, together with high oxygen consumption, makes them more vulnerable to exposure, via the circulation, and subsequent injury related to high concentrations of xenobiotics and chemicals. In preclinical drug development and safety assessment of new investigational drugs, changes in kidney structure and/or function following drug administration in experimental laboratory animals need to be put in context with interspecies differences in kidney functio

Alzheimer's disease drug development pipeline: 2026.

INTRODUCTION: Discovery and development of new therapies for Alzheimer's disease (AD) are urgently needed to address the world's growing population of individuals on the AD pathophysiological continuum. Clinicaltrials.gov is a resource for studying drugs in development for treatment of AD. RESULTS: There are currently 158 drugs in 192 AD trials. Of the agents in trials, 39% are small molecule disease targeting therapies (DTTs); 34% are biologic DTTs; 18% are cognition enhancing symptom targeted therapies (STTs); and 10% are STTs being developed to treat neuropsychiatric symptoms of AD. Structural barriers to antifungal drug development.

Fungal diseases represent a growing yet under-recognized global health threat, with mortality comparable to major infectious diseases but a disproportionately weak therapeutic pipeline. This review examines the antifungal innovation gap as a systemic phenomenon shaped by intertwined scientific, economic, and societal constraints. While multidrug-resistant pathogens underscore the urgent need for new antifungal agents, progress is hindered by fungal-human cellular similarity, high development costs, limited commercial incentives, and toxicity concerns. Agricultural fungicide practices that driv

Targeting S1P signaling for drug development.

Sphingosine-1-phosphate (S1P), a downstream metabolite of the sphingolipid pathway, exerts diverse biological functions, is implicated in a wide spectrum of diseases, and has been shown to have central roles in regulating cell proliferation, migration, and immune cell trafficking. Five agonists of its receptor have been approved for the treatment of multiple sclerosis, and an inhibitor targeting its production is currently undergoing clinical trials for cancer therapy. Here, we discuss the modulators and their preliminary structure-activity relationships in terms of the generation, transportat

Tuberculosis drug development; fluoroquinolone structural tailoring.

Tuberculosis (TB) is a contagious infectious disease caused by the bacillus Mycobacterium tuberculosis (Mtb). It is transmitted through small particles in the air (<5 µm) expelled by active tuberculosis patients; when inhaled by a new host, they can potentially cause infection. Nowadays, TB is still the major cause of morbidity and mortality by a single infectious agent, this is further exacerbated by the worldwide emergence of multidrug-resistant strains of Mtb. Thus, effective methods of diagnosis, prophylaxis, and new pharmacological therapies must be carried out in order to control this di

Cancer drug development: The missing links.

Impact statementThe success rate for cancer drugs which enter into phase 1 clinical trials is utterly less. Why the vast majority of drugs fail is not understood but suggests that pre-clinical studies are not adequate for human diseases. In 1975, as per the Tufts Center for the Study of Drug Development, pharmaceutical industries expended 100 million dollars for research and development of the average FDA approved drug. By 2005, this figure had more than quadrupled, to $1.3 billion. In order to recover their high and risky investment cost, pharmaceutical companies charge more for their product

Xemilofiban/orbofiban: insight into drug development.

A number of studies have reported on the successful use of intravenous glycoprotein IIb/IIIa receptor antagonists in patients with unstable angina or undergoing percutaneous interventions. The promise of interrupting the aggregation of platelets in the setting of unstable plaques on a chronic basis had led to the evaluation of several oral agents for longer-term administration. The development program of two of these agents, xemilofiban and orbofiban, will be reviewed and evaluated to understand the selection process of therapeutic targets for use based upon complex pharmacokinetic and pharmac

Leukemia: A model for drug development.

Early attempts at preclinical model development for cancer drug development relied heavily on mouse leukemias and lymphomas to detect agents with antitumor activity. These models were applied clinically, and the concepts of combination chemotherapy, remission induction, and maintenance treatment all developed in leukemia. Subsequently, the predominant impact of cytogenetics on probability of response to treatment and survival was first illustrated in leukemia. The power of a single drug to change the natural history of a disease was noted in acute myelogenous leukemia, in which a previously in

Novel drug development for geographic atrophy.

Geographic atrophy (GA), the advanced stage of dry age-related macular degeneration, has transitioned from an untreatable condition to a field of active therapeutic development. This review outlines the key molecular pathways driving GA progression, including complement system dysregulation, oxidative stress, and chronic inflammation. A marked important therapeutic milestone with the first FDA approvals of complement inhibitors, Pegcetacoplan and Avacincaptad pegol, demonstrated a modest but statistically significant slowing of lesion growth. However, these anatomic benefits have not consisten

Progress in Anti-Mammarenavirus Drug Development.

Mammarenaviruses are prevalent pathogens distributed worldwide, and several strains cause severe cases of human infections with high morbidity and significant mortality. Currently, there is no FDA-approved antiviral drugs and vaccines against mammarenavirus and the potential treatment option is limited to an off-label use of ribavirin that shows only partial protective effect and associates with side effects. For the past few decades, extensive research has reported potential anti-mammarenaviral drugs and their mechanisms of action in host as well as vaccine candidates. This review describes c

Alzheimer's Disease: Advances in Drug Development.

As of 2018, Alzheimer's disease (AD) is the most common form of neurodegenerative dementia. It contributes to a progressive neuron loss, deterioration of memory, and cognitive impairment. Current therapies may provide a symptomatic benefit, but do not treat the underlying process. Ongoing researches focus on understanding the causal mechanisms and finding neuropathological hallmarks of AD. Therapeutic approaches targeting senile plaques or neurofibrillary tangles have not yet resulted in a significant cognitive improvement. However, recent data according to the analysis of AD clinical trials (

Advances in Alzheimer's disease drug development.

Alzheimer's disease (AD) is the foremost cause of dementia worldwide. Clinically, AD manifests as progressive memory impairment followed by a gradual decline in other cognitive abilities leading to complete functional dependency. Recent biomarker studies indicate that AD is characterized by a long asymptomatic phase, with the development of pathology occurring at least a decade prior to the onset of any symptoms. Current FDA-approved treatments target neurotransmitter abnormalities associated with the disease but do not affect what is believed to be the underlying etiology. In this review, we

Role of neuroimaging in drug development.

The development of new molecular imaging techniques has bridged the gap between preclinical and clinical research. During the last decade, the developments in imaging strategies have taken a great leap by the advancements in new imaging scanners, development of pharmaceutical drugs, diagnostic agents, and new therapeutic regimens that made significant improvements in health care. The knowledge gained from imaging techniques in preclinical research can be applicable to the patients. Similarly, the problems from clinical studies with humans can be tested and studied in preclinical studies. The a

Stent implantation has become the new standard angioplasty procedure. In-stent re-stenosis remains the major limitation of coronary stenting. Re-stenosis is related to patient-, lesion- and procedure-specific factors. Patient-specific factors can not be influenced to any extent. Procedure-specific factors are affected by implantation technique and stent characteristics. Design and material influence vascular injury and humoral and cellular response. Radiation has been shown to have inhibitory effects on smooth muscle cell growth and neo-intima formation, but in clinical trials the outcome has
