A Novel 3D High-Throughput Phenotypic Drug Screening Pipeline to Identify Drugs with Repurposing Potential for the Treatment of Ovarian Cancer.

Ovarian cancer (OC) poses a significant clinical challenge due to its high recurrence rates and resistance to standard therapies, particularly in advanced stages where recurrence is common, and treatment is predominantly palliative. Personalized treatments, while effective in other cancers, remain underutilized in OC due to a lack of reliable biomarkers predicting clinical outcomes. Accordingly, precision medicine approaches are limited, with PARP inhibitors showing efficacy only in specific genetic contexts. Drug repurposing offers a promising, rapidly translatable strategy by leveraging exis

Chemical genetics and drug screening in Drosophila cancer models.

Drug candidates often fail in preclinical and clinical testing because of reasons of efficacy and/or safety. It would be time- and cost-efficient to have screening models that reduce the rate of such false positive candidates that appear promising at first but fail later. In this regard, it would be particularly useful to have a rapid and inexpensive whole animal model that can pre-select hits from high-throughput screens but before testing in costly rodent assays. Drosophila melanogaster has emerged as a potential whole animal model for drug screening. Of particular interest have been drugs t

DCMiC: a double-cylinder micro-chamber platform for high-throughput drug screening and modeling of microenvironmental resistance in Ewing sarcoma.

We fabricated a double-cylinder micro-chamber (DCMiC) platform using stereolithography-printed master molds, followed by PDMS replica molding and integration into a 96-well plate format for scalable and reproducible generation of Ewing sarcoma spheroids. The simple yet novel DCMiC design stabilizes spheroids during media exchange, enabling reliable long-term culture and high-throughput drug screening. Using this platform, we screened 11 small-molecule compounds previously shown to target vulnerabilities relevant to Ewing sarcoma, including epigenetic regulators, DNA damage response, growth sig

Insights into the structure-activity relationship of the anticancer compound ZJ-101, a derivative of marine natural product superstolide A: A role played by the lactone moiety.

Compound ZJ-101, a structurally simplified analog of the marine natural product superstolide A, was previously developed in our laboratory. In the subsequent structure-activity relationship study, a new analog ZJ-109 was designed and synthesized to probe the importance of the lactone moiety of the molecule by replacing the lactone in ZJ-101 with a lactam. The biological evaluation showed that ZJ-109 is about 8-12 times less active against cancer cells in vitro than ZJ-101, suggesting that the lactone moiety of the molecule is important for its anticancer activity.

Succinylated Apoptolidins from Amycolatopsis sp. ICBB 8242.

Two new apoptolidins, 2'-O-succinyl-apoptolidin A (11) and 3'-O-succinyl-apoptolidin A (12), were isolated from the culture broth of an Indonesian Amycolatopsis sp. ICBB 8242. These compounds inhibit the proliferation and viability of human H292 and HeLa cells. However, in contrast to apoptolidin A (1), they do not inhibit cellular respiration in H292 cells. It is proposed that apoptolidins are produced and secreted in their succinylated forms and 1 is the hydrolysis product of 11 and 12.

Signaling and drug sensitivity.

Even though alterations in receptor and nonreceptor kinases are involved in the development of human cancer, many cancer cell lines still retain their responsiveness to growth factors. We have investigated the hypothesis that cellular signaling events regulate the sensitivity of cancer cells to chemotherapeutic agents. In 2008 human ovarian carcinoma cells, activation of a number of different transduction pathways resulted in a 2 to 4-fold increase in the sensitivity to cisplatin. These signaling events include pathways activated by the epidermal growth factor (EGF) receptor, tumor necrosis fa

Synthesis and Preliminary Evaluation of Tanshinone Mimic Conjugates for Mechanism of Action Studies.

Human antigen R (HuR) is an RNA binding protein (RBP) belonging to the ELAV (Embryonic Lethal Abnormal Vision) family, which stabilizes mRNAs and regulates the expression of multiple genes. Its altered expression or localization is related to pathological features such as cancer or inflammation. Dihydrotanshinone I (DHTS I) is a naturally occurring, tetracyclic ortho-quinone inhibitor of the HuR-mRNA interaction. Our earlier efforts led to the identification of a synthetic Tanshinone Mimic (TM) 2 with improved affinity for HuR. Here we report five new TM probes 3-5 bearing a detection-promotin

The ITCC-P4 PDX Platform Enables Preclinical Testing of Pediatric Cancers.

UNLABELLED: Cancer is the leading cause of disease-related deaths among children in high-income countries. Tumor heterogeneity and the lack of mechanism of action-based therapeutic options are key challenges to overcome to improve the survival of pediatric patients with cancer. To address these challenges, we formed the European Union-Innovative Medicines Initiative 2 (EU-IMI2) funded public-private partnership "Innovative Therapies for Children with Cancer Paediatric Preclinical Proof-of-Concept Platform" (ITCC-P4), which built a large repertoire of patient-derived xenograft (PDX) models repr

Protocol for establishing a chemoresistant orthotopic triple-negative breast cancer patient-derived xenograft model for preclinical drug testing.

Here, we present a protocol for establishing patient-derived xenograft (PDX) mouse models of chemoresistant triple-negative breast cancer (TNBC). We describe steps for tumor collection, surgical implantation, and serial expansion. We then detail procedures for histopathological validation and in vivo therapeutic assessment to confirm the maintenance of chemoresistance across passages. These models provide a clinically relevant platform to investigate mechanisms of treatment resistance, evaluate novel therapeutic strategies in preclinical settings, and predict therapeutic responses in refractor

The potential of personalized cancer spheroids in precision therapy.

The clinical implementation of precision oncology remains challenged by intratumoral heterogeneity, complex tumor microenvironment interactions, and the limited predictive value of conventional preclinical models. 3D cancer spheroids have emerged as physiologically relevant platforms that closely mimic in vivo tumor architecture, cellular heterogeneity, hypoxic gradients, and treatment resistance mechanisms. This review highlights the growing role of cancer spheroids in drug panel testing and their application in personalized therapeutic decision-making across diverse malignancies, including b

Preclinical and clinical studies of Coriolus versicolor polysaccharopeptide as an immunotherapeutic in China.

Conventional cancer treatments include surgery, chemotherapy, and radiation therapy. In recent years, immunotherapy in cancer care has been gaining momentum. Interestingly, an immunotherapeutic regime that employs polysaccharopeptide (PSP), a unique peptide-containing polysaccharide isolated from Coriolus versicolor, has already become a routine clinical practice in Japan since 1977 and in China since 1987. Coriolus versicolor is one of the most well-known traditional food and medicinal mushrooms in China for thousands of years. Medically used PSP is mostly obtained from the extraction of cult

Synthesis and anti-proliferative activity of aromatic substituted 5-((1-benzyl-1H-indol-3-yl)methylene)-1,3-dimethylpyrimidine-2,4,6(1H,3H,5H)-trione analogs against human tumor cell lines.

Based on previous SAR studies on N-benzylindole and barbituric acid hybrid molecules, we have synthesized a series of aromatic substituted 5-((1-benzyl-1H-indol-3-yl)methylene)-1,3-dimethylpyrimidine-2,4,6(1H,3H,5H)-trione analogs (3a-i) and evaluated them for their in vitro growth inhibition and cytotoxicity against a panel of 60 human tumor cell lines. Compounds 3c, 3d, 3f and 3g were identified as highly potent anti-proliferative compounds against ovarian, renal and breast cancer cell lines with GI50 values in low the nanomolar range. The 4-methoxy-N-benzyl analog (3d) was the most active c

Cytotoxicity assays for cancer drug screening: methodological insights and considerations for reliable assessment in drug discovery.

The importance of cytotoxicity assays in in vitro drug discovery investigations has led to their rising profile. Drugs and other substances can disrupt cell membranes, limit protein synthesis, and bind irreversibly to receptors, all of which lead to cell death in cancer cells. To precisely measure the cell death resulting from these damages, one must choose a cytotoxicity test that meets specific criteria. A systematic search strategy was used to gather grey literature from 2001 to 2024, utilizing databases such as PubMed and Google Scholar. Specific keywords related to colorimetric, fluoromet

Analogues of the Potent Antitumor Compound Leiodermatolide from a Deep-Water Sponge of the Genus Leiodermatium.

Two new analogues of the potent antitumor compound leiodermatolide, which we call leiodermatolides B and C, have been isolated from specimens of a deep-water sponge of the genus Leiodermatium collected off Florida. The compounds were purified using standard chromatographic methods, and the structures defined through interpretation of the HRMS and 1D and 2D NMR data. Leiodermatolide B (2) lacks the C-21 hydroxy group found in leiodermatolide and has equal potency as the parent compound, providing a simpler analogue for possible clinical development. It inhibits the proliferation of the AsPC-1 h
