Ecdysone regulates phagocytic cell fate of epithelial cells in developing Drosophila eggs.

Acquisition of nonprofessional phagocytic cell fate plays an important role in sculpting functional metazoan organs and maintaining overall tissue homeostasis. Though physiologically highly relevant, how the normal epithelial cells acquire phagocytic fate is still mostly unclear. We have employed the Drosophila ovary model to demonstrate that the classical ecdysone signaling in the somatic epithelial follicle cells (AFCs) aids the removal of germline nurse cells (NCs) in late oogenesis. Our live-cell imaging data reveal a novel phenomenon wherein collective behavior of 4-5 AFCs is required for

Systemic positive feedback regulation of ecdysone biosynthesis via sugar metabolism in insects.

Insects are recognized as the most diverse species in the world. They are classified into three separate groups based on their developmental characteristics: ametabola, hemimetabola, and holometabola. Holometabolous insects have attracted the attention of many developmental biologists because of their dramatic metamorphosis as a key model for studying regulatory mechanisms of development. Central to this process is ecdysone, a steroid hormone that governs insect growth and maturation. Extensive research has clarified ecdysone's structure, functions, and signaling pathways. In particular, the r

Could Crustaceans' Ovary Synthesize Ecdysone? First Evidence in a Crab Species During Intermolt.

The function of ecdysone in promoting molting is widely known in arthropods. Moreover, in insects, ecdysone synthesis in the ovary is involved in reproduction and development. In crustaceans, some studies conducted in species with the molt linked to mating have shown that ecdysone produced by the Y organ stimulates ovarian maturation, but scarce evidence about the capacity of the ovary itself to synthesize ecdysone is currently available. The present study provides the first evidence of the expression of a putative shadow gene (sad, cytochrome P450 CYP315A1), involved in the ecdysone synthesis

Beta-Ecdysone protects osteocytes from excess glucocorticoids via Akt1-mediated regulation of Connexin43.

Glucocorticoid-induced osteoporosis (GIOP) is a leading cause of secondary osteoporosis (OP). β-Ecdysone (βEcd), a naturally occurring estrogen analog, was evaluated for its ability to mitigate the effects of glucocorticoids (GCs) on osteocytes, the crucial cells in bone remodeling. In GIOP mouse model induced by dexamethasone (Dex), micro-CT, biomechanical testing, silver nitrate staining, and hematoxylin-eosin (HE) staining were employed, demonstrating that βEcd effectively attenuated Dex-induced decreases in bone mass and strength, and alleviated Dex induced reduction in osteocyte dendrite

Juvenile hormone potentiates ecdysone receptor-dependent transcription in a mammalian cell culture system.

Insect development is guided by the combined actions of ecdysteroids and juvenile hormones (JHs). The transcriptional effects of ecdysteroids are mediated by a protein complex consisting of the ecdysone receptor (EcR) and its heterodimeric partner, Ultraspiracle (USP), but a corresponding JH receptor has not been defined conclusively. Given that the EcR ligand binding domain (LBD) is similar to that of the JH-responsive rat farnesoid-X-activated receptor (FXR), we sought to define experimental conditions under which EcR-dependent transcription could be promoted by JH. Ecdysteroid-mimicking compounds act as both agonists and antagonists to the crustacean ecdysone receptor.

To characterize the potential endocrine-disrupting chemicals (EDCs) in the environment that interact with the crustacean ecdysone receptor (EcR), we established a method involving in silico modeling/molecular docking and in vitro reporter gene assay. Cherry shrimp (Neocaridina davidi) EcR (NdEcR) and retinoid X receptor (NdRxR) were identified and cloned for use in this method. A theoretical 3D model of NdEcR ligand-binding domain (LBD) was built in silico based on sequence homology with the established X-ray structure of insect EcR. The interaction of the NdEcR LBD with ecdysteroids, diacylhy

Germline cysts require Ecdysone receptor for proper timing of encapsulation in the Drosophila ovary.

Oocytes are packaged into ovarian follicles, each containing a maturing germ cell surrounded by a layer of somatic cells. This conserved arrangement is essential for proper oocyte development and reproductive success. Continuous coordination and bi-directional signaling from somatic cells to germ cells are necessary for proper oocyte packaging. In Drosophila, although germ cell packaging has been presumed to be controlled by somatic follicle cells enveloping passive germ cells, recent studies suggest that germ cells themselves produce motor forces that drive somatic encapsulation. Here, in sup

Identification of Ecdysone Hormone Receptor Agonists as a Therapeutic Approach for Treating Filarial Infections.

BACKGROUND: A homologue of the ecdysone receptor has previously been identified in human filarial parasites. As the ecdysone receptor is not found in vertebrates, it and the regulatory pathways it controls represent attractive potential chemotherapeutic targets. METHODOLOGY/ PRINCIPAL FINDINGS: Administration of 20-hydroxyecdysone to gerbils infected with B. malayi infective larvae disrupted their development to adult stage parasites. A stable mammalian cell line was created incorporating the B. malayi ecdysone receptor ligand-binding domain, its heterodimer partner and a secreted luciferase

Mating-induced ecdysone in the testis disrupts soma-germline contacts and stem cell cytokinesis.

Germline maintenance relies on adult stem cells to continually replenish lost gametes over a lifetime and respond to external cues altering the demands on the tissue. Mating worsens germline homeostasis over time, yet a negative impact on stem cell behavior has not been explored. Using extended live imaging of the Drosophila testis stem cell niche, we find that short periods of mating in young males disrupts cytokinesis in germline stem cells (GSCs). This defect leads to failure of abscission, preventing release of differentiating cells from the niche. We find that GSC abscission failure is ca

Aedes Dronc: a novel ecdysone-inducible caspase in the yellow fever mosquito, Aedes aegypti.

Caspases are cysteinyl-aspartate-specific proteases known for their role in apoptosis. Here, we describe the characterization of Aedes Dronc, a novel caspase in the yellow fever mosquito, Aedes aegypti. Aedes Dronc is predicted to contain an N-terminal caspase recruitment domain and is a homologue of Drosophila Dronc and human caspase-9. An increase in transcripts and caspase activity coincides with developmental changes in the mosquito, suggesting that Aedes Dronc plays a role in developmental apoptosis. Exposure of third instar larvae to ecdysone resulted in a significant increase in both tr

Dual functions of the Aedes aegypti ecdysone receptor in dengue virus replication and reproduction control.

BACKGROUND: The ecdysone receptor (EcR) is a central regulator of mosquito physiology, best known for its role in vitellogenesis. However, its contribution to antiviral defense and dengue virus (DENV) replication in Aedes aegypti remains poorly understood. METHODS: RNA interference was used to silence Aedes aegypti EcR (AaEcR). Effects on DENV replication, immune gene expression, ovarian development, vitellogenin (Vg) synthesis, and target of rapamycin (TOR) pathway activity were assessed using molecular, cellular, and phenotypic analyses. RESULTS: Silencing AaEcR markedly suppressed DENV re

Arpc2 integrates ecdysone and juvenile hormone metabolism to influence metamorphosis and reproduction in Tribolium castaneum.

BACKGROUND: Actin-related protein 2/3 complex regulates actin polymerization and the formation of branched actin networks. However, the function and evolutionary relationship of this complex subunit 2 (Arpc2) has been poorly understood in insects. RESULTS: To address these issues, we performed comprehensive analysis of Arpc2 in Tribolium castaneum. Phylogenetic analysis revealed that Arpc2 was originated from one ancestral gene in animals but evolved independently between vertebrates and insects after species differentiation. T. castaneum Arpc2 has a 906-bp coding sequence and consists of 4 e

Biochemical mode of action and differential activity of new ecdysone agonists against mosquitoes and moths.

THQ (1-aroyl-4-(arylamino)-1,2,3,4-tetrahydroquinoline) compounds were identified by FMC Corporation in cell-based assays that used ecdysone receptors from Drosophila melanogaster, Heliothis virescens, or Plodia interpunctata. THQ compounds showed weak insecticidal activity against H. virescens and, therefore, were not developed further. Several ecdysone agonists based on THQ chemotype have been synthesized and tested for their activity against a number of EcRs in transactivation assays. The THQ compound, RG-120768, activated AaEcR (EcR from A. aegypti) but did not activate EcRs cloned from ot

The Drosophila gap gene giant regulates ecdysone production through specification of the PTTH-producing neurons.

In Drosophila melanogaster, hypomorphic mutations in the gap gene giant (gt) have long been known to affect ecdysone titers resulting in developmental delay and the production of large (giant) larvae, pupae and adults. However, the mechanism by which gt regulates ecdysone production has remained elusive. Here we show that hypomorphic gt mutations lead to ecdysone deficiency and developmental delay by affecting the specification of the PG neurons that produce prothoracicotropic hormone (PTTH). The gt¹ hypomorphic mutation leads to random loss of PTTH production in one or more of the 4 PG neuron

Ailanthone exerts insecticidal activity as a non-orthosteric 20E antagonist by promoting ecdysone receptor degradation.

Ailanthone, a natural quassinoid from Ailanthus altissima, has been reported to exhibit insecticidal activity, yet its molecular mechanism remains elusive. Here, we show that ailanthone acts as a non-orthosteric inhibitor of 20-hydroxyecdysone (20E) signaling by promoting ubiquitin-proteasome-dependent degradation of the ecdysone receptor (EcR). In both Drosophila melanogaster and Ostrinia furnacalis, ailanthone treatment inhibited insect growth and metamorphosis. Mechanistically, in D. melanogaster, these developmental defects were attributable to disruption of 20E signaling, as evidenced by

Tissue-specific requirement of Polr1D in the prothoracic gland for ecdysone-mediated developmental transitions in Drosophila melanogaster.

BACKGROUND: POLR1D is a shared subunit of RNA Polymerases I and III, which transcribe the rRNA incorporated into ribosomes. Mutations in POLR1D cause Treacher Collins syndrome, a craniofacial disorder that arises from impaired ribosome biogenesis in neural crest cells. Previously, we found that RNAi knockdown of Polr1D in several non-neural Drosophila tissues caused developmental defects that phenocopy mutations affecting ecdysone signaling. Ecdysone is a steroid hormone produced in the prothoracic gland (PG) of insects that triggers developmental transitions. Here, we show that Polr1D is requ

Hormone-dependent activation and repression of microRNAs by the ecdysone receptor in the dengue vector mosquito Aedes aegypti.

Female mosquitoes transmit numerous devastating human diseases because they require vertebrate blood meal for egg development. MicroRNAs (miRNAs) play critical roles across multiple reproductive processes in female Aedes aegypti mosquitoes. However, how miRNAs are controlled to coordinate their activity with the demands of mosquito reproduction remains largely unknown. We report that the ecdysone receptor (EcR)-mediated 20-hydroxyecdysone (20E) signaling regulates miRNA expression in female mosquitoes. EcR RNA-interference silencing linked to small RNA-sequencing analysis reveals that EcR not

An ecdysone and tetracycline dual regulatory expression system for studies on Rac1 small GTPase-mediated signaling.

Regulated expression systems are invaluable for studying gene function, offer advantages of dosage-dependent and temporally defined gene expression, and limit possible clonal variation when toxic or pleiotropic genes are overexpressed. Previously, establishment of inducible expression systems, such as tetracycline- and ecdysone-inducible systems, required assessment of the inducible characteristics of individual clones by tedious luciferase assays. Taking advantage of a green fluorescent protein (GFP) reporter controlled by tetracycline- or ecdysone-responsive element and fluorescence-activate

A Phloroglucinol from Dryopteris crassirhizoma Acts as a 20E Antagonist with Insecticidal Activity by Targeting the Ecdysone Receptor.

The 20-hydroxyecdysone (20E) signaling pathway is a key target for developing insect growth regulators, yet few effective 20E antagonists have been identified. Through bioassay-guided fractionation using a 20E-responsive reporter assay in Drosophila Kc cells, a symmetric dimeric phloroglucinol, methylene-bis-phlorobutyrophenone (compound A3), was isolated from the rhizomes of Dryopteris crassirhizomaNakai. Compound A3 potently inhibited 20E-induced transcriptional activation without intrinsic agonistic activity. Two-hybrid and microscale thermophoresis (MST) assays suggested that A3 might comp
