A novel hairpin-like antimicrobial peptide from barnyard grass (Echinochloa crusgalli L.) seeds: Structure-functional and molecular-genetics characterization.

A novel plant hairpin-like defense polypeptide named EcAMP3 was isolated from latent barnyard grass (Echinochloa crusgalli L.) seeds. The native peptide and its recombinant analogue were characterized. EcAMP3 displays antifungal and antibacterial activity in vitro. The gene family encoding EcAMPs precursor protein was also characterized; the genes and pseudogenes of this family show 97-100% homology. Every member of EcAMPs precursor family contains seven identical cysteine motifs: C1XXXC2(11-13)C3XXXC4. One of those motifs corresponds to the isolated peptide. EcAMP3 is the first member of the

Effect of Germination on the Nutrient Profile, Antioxidant Components, Antinutritional Constituents, and Functional Characteristics of Barnyard Millet (Echinochloa frumentacea).

Barnyard millet (Echinochloa frumentacea) is a nutrient-dense, gluten-free grain with recognized health benefits; however, its utilization is constrained by the presence of phytate, tannins, and trypsin inhibitor activity. This study examined the influence of germination (24-72 h) on the nutritional composition, antinutritional constituents, bioactive compounds, and functional properties of barnyard millet. Germination significantly altered the nutrient composition, increasing protein content (9.21-13.36 g/100 g d.m.) and dietary fiber (11.23-13.01 g/100 g d.m.) due to enzymatic modifications

Quinclorac resistance in Echinochloa phyllopogon is associated with reduced ethylene synthesis rather than enhanced cyanide detoxification by β-cyanoalanine synthase.

BACKGROUND: Multiple herbicide resistant Echinochloa phyllopogon exhibits resistance to the auxin herbicide quinclorac. Previous research observed enhanced activity of the cyanide-detoxifying enzyme β-cyanoalanine synthase (β-CAS) and reduced ethylene production in the resistant line, suggesting β-CAS-mediated cyanide detoxification and insensitivity to quinclorac stimulation as the resistance mechanisms. To investigate the molecular mechanisms of quinclorac resistance, we characterized the β-CAS genes alongside plant transformation studies. The association of β-CAS activity and ethylene produ

Analysis of the resistance level and target site resistance mechanisms of Echinochloa crus-galli to penoxsulam from Hubei Province, China.

Echinochloa crus-galli is a grass weed that infests rice fields and causes significant crop yield losses. In this study, we surveyed 15 resistant E. crus-galli populations collected from rice fields in Hubei Province, China, and investigated the resistance levels and target site resistance mechanisms to the acetolactate synthase (ALS) inhibitor penoxsulam. The results of whole-plant bioassay experiments revealed that 15 populations presented different levels of resistance to penoxsulam. The Trp-574-Leu mutation was detected in ten resistant populations, and the Pro-197-Leu mutation was detecte

Resistance to quinclorac caused by the enhanced ability to detoxify cyanide and its molecular mechanism in Echinochloa crus-galli var. zelayensis.

Quinclorac, an auxin-type herbicide, is widely used to control barnyardgrass and some dicotyledon weeds. Echinochloa crus-galli var. zelayensis, a variety of E. crus-galli (L.) Beauv., is widespread in China and some populations have resistance to quinclorac. E. crus-galli var. zelayensis seeds with varying sensitivity to quinclorac were used in the present study. The expression of the ADP/ATP carrier protein (ANT) gene, which plays an important role in the maintenance of cellular energy balance, dramatically rose in the S biotype after exposure to quinclorac, while no change was found in two

Differential metabolic reprogramming and organ-specific antioxidant allocation in Oryza sativa and Echinochloa crus-galli under propanil-induced chemical stress.

The physiological and metabolic mechanisms by which plants manage severe chemical stress during critical reproductive stages remain poorly understood. Here, we investigated the system-level phytotoxic responses and metabolic reprogramming of a tolerant species (Oryza sativa L.) and a susceptible species (Echinochloa crus-galli) exposed to the chemical stressor propanil using GC-MS/MS and LC-MS/MS. Under severe chemical stress, O. sativa maintained relatively stable antioxidant-related metabolism in grains, with α-tocopherol and phylloquinone showing only modest decreases of 0.86- and 0.90-fold

Quinclorac-resistant Echinochloa spp. promoted growth and reproduction of Laodelphax striatellus (Hemiptera: Delphacidae) probably by providing more nutrients and stable environment.

Rice is an important agricultural crop that faces serious challenges from pathogens, pests, and weeds during growth stages. Meanwhile, these organisms would interact with each other to increase the level of destruction. The previous studies showed that barnyard grass (Echinochloa spp) could be used as a temporary host to increase infestation of small brown planthopper (SBPH, Laodelphax striatellus), which is one of the main polyphagous pests. Herbicides are widely used to control weeds that induce resistance development. However, little is known about the effects of increased weed resistance o

A novel Phe-206-Leu mutation in acetolactate synthase confers resistance to penoxsulam in barnyardgrass (Echinochloa crus-galli (L.) P. Beauv).

BACKGROUND: Barnyardgrass (Echinochloa crus-galli (L.) P. Beauv) has evolved resistance to the acetolactate synthase (ALS) inhibitor penoxsulam which is used to control weeds in rice fields in China. The present study is conducted to identify the target-site resistance (TSR) mechanisms conferring resistance in a penoxsulam-resistant population. RESULTS: The ALS sensitivity in vitro of the resistant population was sixfold lower to penoxsulam than that of the sensitive population. ALS sequencing revealed that no known mutation conferring ALS herbicide resistance was detected. However, a novel m

Up-regulation of bZIP88 transcription factor is involved in resistance to three different herbicides in both Echinochloa crus-galli and E. glabrescens.

The resistance of weeds to herbicides poses a major threat to agricultural production, and non-target-site resistance (NTSR) is often a serious problem as its mechanisms can in some cases confer resistance to herbicides with different modes of action. In this study, we hypothesized that bZIP transcription factors (TFs), which regulate abiotic stress responses in many plants, play a regulatory role in NTSR. Whole-plant assays indicated that the wild grasses Echinochloa crus-galli and E. glabrescens are resistant to the herbicides penoxsulam, cyhalofop-butyl, and quintrione. Transcriptome sequen

Trp-574-Leu and the novel Pro-197-His/Leu mutations contribute to penoxsulam resistance in Echinochloa crus-galli (L.) P. Beauv.

Recently, due to the widespread use of the acetolactate synthase (ALS)-inhibiting herbicide penoxsulam in paddy fields in China, Echinochloa crus-galli (L.) P. Beauv. has become a problematic grass weed that is frequently not controlled, posing a threat to weed management and rice yield. There are many reports on target-site mutations of ALS inhibiting herbicides; however, the detailed penoxsulam resistance mechanism in E. crus-galli remains to be determined. Greenhouse and laboratory studies were conducted to characterize target-site resistance mechanisms in JL-R, AH-R, and HLJ-R suspected re

Enhanced Herbicide Metabolism and Target Site Mutation Enabled the Multiple Resistance to Cyhalofop-butyl, Florpyrauxifen-benzyl, and Penoxsulam in Echinochloa crus-galli.

This study investigated the multiple herbicide resistance (MHR) mechanism of one Echinochloa crus-galli population that was resistant to florpyrauxifen-benzyl (FPB), cyhalofop-butyl (CHB), and penoxsulam (PEX). This population carried an Ala-122-Asn mutation in the acetolactate synthase (ALS) gene but no mutation in acetyl-CoA carboxylase (ACCase) and transport inhibitor response1 (TIR1) genes. The metabolism rate of PEX was 2-fold higher, and the production of florpyrauxifen-acid and cyhalofop-acid was lower in the resistant population. [Release of mercury from soil and plant in water-level-fluctuating zone of the Three Gorges Reservoir area and its accumulation in zebrafish].

To investigate the production, distribution and bioavailability of methylmercury (MMHg) in soil and plants of the water-level-fluctuating zone (WLFZ) of the Three Gorges Reservoir area, simulation experiments were conducted in laboratory. Results indicated that the level of total mercury (THg) in soil decreased with the lengthening of submerging time while that in water increased obviously. The level of MMHg in inundated soil and water increased, especially in the water treated by Echinochloa crusgalli and soils. And the MMHg level in that treatment was 2.52 times higher than that treated only

Distribution, frequency and molecular basis of penoxsulam, metamifop and florpyrauxifen-benzyl resistance in Echinocloa spp. from rice fields across Jiangsu Province, China.

Barnyard grasses (Echinochloa spp.) are the most troublesome weeds in rice ecosystems of Jiangsu Province, China, and long-term use of herbicides inevitably cause the development of resistance. In this study, 122 field-sampled populations were tested for resistance to penoxsulam (PEN), metamifop (MET), florpyrauxifen-benzyl, (FPB), propanil, and tripyrasulfone. Field-dose assays revealed that 51, 7, and 6 populations have either developed or were developing resistance to penoxsulam, metamifop and florpyrauxifen-benzyl, and all 122 populations were still sensitive to propanil and tripyrasulfone

Glutathione S-transferase activity facilitates rice tolerance to the barnyard grass root exudate DIMBOA.

BACKGROUND: In paddy fields, the noxious weed barnyard grass secretes 2,4-dihydroxy-7-methoxy-2H-1,4-benzoxazin-3(4H)-one (DIMBOA) to interfere with rice growth. Rice is unable to synthesize DIMBOA. Rice cultivars with high or low levels of allelopathy may respond differently to DIMBOA. RESULTS: In this study, we found that low concentrations of DIMBOA (≤ 0.06 mM) promoted seedling growth in allelopathic rice PI312777, while DIMBOA (≤ 0.08 mM) had no significant influence on the nonallelopathic rice Lemont. DIMBOA treatment caused changes in the expression of a large number of glutathione S-t

Defensive Molecules Momilactones A and B: Function, Biosynthesis, Induction and Occurrence.

Labdane-related diterpenoids, momilactones A and B were isolated and identified in rice husks in 1973 and later found in rice leaves, straws, roots, root exudate, other several Poaceae species and the moss species Calohypnum plumiforme. The functions of momilactones in rice are well documented. Momilactones in rice plants suppressed the growth of fungal pathogens, indicating the defense function against pathogen attacks. Rice plants also inhibited the growth of adjacent competitive plants through the root secretion of momilactones into their rhizosphere due to the potent growth-inhibitory acti

Novel Galactosyl Moiety-Conjugated Furylchalcones Synthesized Facilely Display Significant Regulatory Effect on Plant Growth.

The expansion of weed infestation has increased the demand on new herbicides. A series of novel galactosyl moiety-conjugated furylchalcones was facilely synthesized in which the furyl group (A ring) was combined with the substituted benzene group (B ring), and a galactosyl moiety was introduced. All these galactosyl furylchalcones were predicted to be phloem-mobile. Most of the galactosyl furylchalcones significantly promoted early seedling growth of sorghum and barnyardgrass under dark conditions, but all of them revealed considerable anti-growth ability on illuminated pot plants; especially,

Tracing the Origin and Evolutionary History of Pyricularia oryzae Infecting Maize and Barnyard Grass.

Blast disease is a notorious fungal disease leading to dramatic yield losses on major food crops such as rice and wheat. The causal agent, Pyricularia oryzae, encompasses different lineages, each having a different host range. Host shifts are suspected to have occurred in this species from Setaria spp. to rice and from Lolium spp. to wheat. The emergence of blast disease on maize in Iran was observed for the first time in the north of the country in 2012. We later identified blast disease in two additional regions of Iran: Gilan in 2013 and Golestan in 2016. Epidemics on the weed barnyard gras

Synthesis and herbicidal activities of acylalanine-containing 3-(pyridin-2-yl)benzothiazol-2-ones.

BACKGROUND: 3-(Pyridin-2-yl)benzothiazol-2-one derivatives are a novel class of protoporphyrinogen oxidase (PPO) inhibitors. Although they demonstrate extraordinary herbicidal activity against broadleaf weeds, their efficacy against grass weeds remains limited. To develop broad-spectrum herbicide candidates, a series of 3-(pyridin-2-yl)benzothiazol-2-one derivatives bearing acylalanine moiety were designed in this study.
