Transglutaminase 2 function in glioblastoma tumor efferocytosis.

Glioblastoma is an aggressive and incurable type of brain cancer. Regions of tissue necrosis are a distinctive pathological feature of this cancer. These arise through thrombosis of tumor vasculature, driven by tumor-derived pro-coagulation factors. In studies of transglutaminase 2 (TGM2), we observed that TGM2 mRNA expression in glioblastoma was primarily in a subset of tumor-infiltrating myeloid cells with hypoxia gene expression signatures. Analysis of xenograft and human glioblastoma samples by immunohistochemistry showed that macrophages in the vicinity of necrotic regions expressed very

Efferocytosis fuels malignant pleural effusion through TIMP1.

Malignant pleural effusion (MPE) results from the capacity of several human cancers to metastasize to the pleural cavity. No effective treatments are currently available, reflecting our insufficient understanding of the basic mechanisms leading to MPE progression. Here, we found that efferocytosis through the receptor tyrosine kinases AXL and MERTK led to the production of interleukin-10 (IL-10) by four distinct pleural cavity macrophage (Mφ) subpopulations characterized by different metabolic states and cell chemotaxis properties. In turn, IL-10 acts on dendritic cells (DCs) inducing the prod

Consequence of alcohol intoxication-mediated efferocytosis impairment.

Alcohol ingestion is a widespread habituation that evolved along with a growing population, altering physiological conditions through immunomodulatory function. There is much research that has reported that consumption of alcohol at low and heavy levels causes different biological impacts, including cellular injury, leading to systemic dysfunction and increased inflammatory markers. In the fate of professional phagocytic cells, efferocytosis is an inevitable mechanism activated by the apoptotic cells, thus eliminating them and preventing the accumulation of cell corpses/debris in the microenvi

Immunomodulatory effect of efferocytosis at the maternal-fetal interface.

Efferocytosis is a mechanism by which phagocytes efficiently clear apoptotic cells, averting their secondary necrosis and the subsequent release of potentially immunogenic or cytotoxic substances that can trigger strong immune and inflammatory responses. During efferocytosis, the metabolic pathways of phagocytes are transformed, which, along with the catabolism of apoptotic cargo, can affect their function and inflammatory state. Extensive apoptosis occurs during placental development, and some studies reported the immunomodulatory effects of efferocytosis at the maternal-fetal interface. The

The Role of Macrophage Efferocytosis in the Pathogenesis of Apical Periodontitis.

Macrophages (Mφs) play a crucial role in the homeostasis of the periapical immune micro-environment caused by bacterial infection. Mφ efferocytosis has been demonstrated to promote the resolution of multiple infected diseases via accelerating Mφ polarization into M2 type. However, the Mφ efferocytosis-apical periodontitis (AP) relationship has not been elucidated yet. This study aimed to explore the role of Mφ efferocytosis in the pathogenesis of AP. Clinical specimens were collected to determine the involvement of Mφ efferocytosis in the periapical region via immunohistochemical and immunoflu

We are what we eat: macrophages and efferocytosis.

Liebold et al. recently revealed how the identity of dying cells drives distinct changes to the macrophages which engulf and clear them, a process known as efferocytosis. During infection with the helminth Schistosoma mansoni, liver macrophages recapitulate these phenotypes, mediated by Axl/MerTK receptors and regulating egg burdens.

Efferocytosis: A new star of atherosclerotic plaques reversal.

Efferocytosis is considered the key to eliminate apoptotic cells (ACs) under physiological and pathological conditions in vivo, mainly through different types of macrophages to achieve this process. Especially, tissue-resident macrophages (TRMs) are very significant for inflammation regression and maintenance of homeostasis in vivo. Abnormal efferocytosis will lead to the accumulation of ACs and the release of a variety of pro-inflammatory factors, which mediates the occurrence of many inflammatory diseases, including atherosclerosis (AS). AS is a chronic inflammatory vascular disease with the

After cell death: the molecular machinery of efferocytosis.

Cells constituting a multicellular organism die in a variety of ways throughout life, and most of them die via apoptosis under normal conditions. The occurrence of apoptosis is especially prevalent during development and in tissues with a high cellular turnover rate, such as the thymus and bone marrow. Interestingly, although the number of apoptotic cells produced daily is known to be innumerable in a healthy adult human body, apoptotic cells are rarely observed. This absence is due to the existence of a cellular process called efferocytosis that efficiently clears apoptotic cells. Studies ove

Defective efferocytosis by alveolar macrophages in IPF patients.

RATIONALE: Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive fibrosing interstitial pneumonia. The pathogenicity of IPF has been widely investigated but still remains to be clarified. Efferocytosis, the specialized recognition and ingestion of apoptotic cells by phagocytes, is essential for the resolution of inflammation in the lungs and repair of injured tissues. Impaired efferocytosis contributes to the pathogenesis of chronic lung diseases such as emphysema and cystic fibrosis. We hypothesized that efferocytosis would also be reduced in alveolar macrophages isolated from subject

CD47-mediated efferocytosis in diseases: A comprehensive review.

Efferocytosis is a vital process by which phagocytes, such as macrophages, recognize and clear apoptotic cells, making it essential for maintaining tissue homeostasis and promoting the resolution of inflammation. Impaired efferocytosis can lead to the secondary necrosis of apoptotic cells and the continuous release of inflammatory factors, thereby contributing to the occurrence and development of various diseases, including tumors, atherosclerosis (AS), autoimmune diseases, and neurodegenerative diseases. CD47, a transmembrane glycoprotein widely expressed on the cell surface, binds to signal

Role of Macrophage Efferocytosis in Cardiovascular Homeostasis and Disease.

Cardiovascular diseases (CVDs) are the primary cause of mortality globally. The Global Burden of Disease Study indicates that CVDs account for one-third of all global deaths, with a notable 18.17% increase in their incidence over the last decade. Despite the widespread use and proven effectiveness of therapies like antiplatelet agents, lipid-lowering drugs, anticoagulants, and percutaneous coronary intervention (PCI) in managing risk factors, the long-term prognosis of patients remains suboptimal. The prevailing academic viewpoint asserts that chronic inflammation and dysregulated apoptosis le

[Research progress on the role of efferocytosis in liver diseases].

Efferocytosis refers to the process of phagocytes engulfing and clearing the cells after programmed cell death. In recent years, an increasing number of studies have shown that the mechanisms of efferocytosis are closely related to drug-induced liver injury, hepatic ischemia-reperfusion injury, viral hepatitis, cholestatic liver diseases, metabolic-associated fatty liver disease, alcoholic liver disease, and other liver disorders. This review summarized the research progress on the role of efferocytosis in liver diseases, with the hope of providing new targets for the prevention and treatment

Efferocytosis: the resolution of inflammation in cardiovascular and cerebrovascular disease.

Cardiovascular and cerebrovascular diseases have surpassed cancer as significant global health challenges, which mainly include atherosclerosis, myocardial infarction, hemorrhagic stroke and ischemia stroke. The inflammatory response immediately following these diseases profoundly impacts patient prognosis and recovery. Efficient resolution of inflammation is crucial not only for halting the inflammatory process but also for restoring tissue homeostasis. Efferocytosis, the phagocytic clearance of dying cells by phagocytes, especially microglia and macrophages, plays a critical role in this res

Efferocytosis: a double-edged sword in microbial immunity.

Efferocytosis is characterized as the rapid and efficient process by which dying or dead cells are removed. This type of clearance is initiated via "find-me" signals, and then, carries on by "eat-me" and "don't-eat-me" ones. Efferocytosis has a critical role to play in tissue homeostasis and innate immunity. However, some evidence suggests it as a double-edged sword in microbial immunity. In other words, some pathogens have degraded efferocytosis by employing efferocytic mechanisms to bypass innate immune detection and promote infection, despite the function of this process for the control and

Efferocytosis in heart failure: Mechanisms, dysregulation and therapeutic potential.

Heart failure (HF) remains one of the leading health problems globally. It is often characterized by unresolved inflammation, adverse cardiac remodeling, and tissue damage. Efferocytosis, the process by which phagocytes clear apoptotic cells, is a key process that helps to resolve inflammation and maintain tissue homeostasis. In the healthy heart, efferocytosis ensures the proper removal of apoptotic cardiac cells (e.g., cardiac fibroblasts and cardiomyocytes) and promotes inflammation resolution and secretion of anti-inflammation cytokines. However, emerging evidence suggests that this proces

Macrophage efferocytosis promotes inflammation resolution and accelerates wound healing.

Skin trauma, particularly chronic non-healing wounds, imposes significant economic and physical burdens on patients and their families. A defining feature of these wounds is persistent dysregulated inflammation, with macrophage efferocytosis playing a critical role. Efferocytosis-the programmed removal of apoptotic cells (ACs) by macrophages-is vital for resolving inflammation and facilitating tissue repair. In chronic wounds associated with systemic diseases such as diabetes, impaired macrophage efferocytosis hinders AC clearance, leading to prolonged inflammation and delayed healing. This re

MERTK coordinates efferocytosis by regulating integrin localization and activation.

Efferocytosis is mediated by MERTK in many tissues, but the signaling pathway and molecular mechanisms used by MERTK to engulf apoptotic cells is largely unknown. Here, using mass spectrometry and super-resolution microscopy, we identified 180 nm receptor complexes comprised of MERTK, β2 integrins and several associated signaling molecules. Efferocytosis was found to be dependent on both MERTK and β2 integrins, with MERTK inducing the conformational change of β2 integrins from low to high-affinity via a PI3K-dependent pathway, with the active integrins then mediating the expansion of an effero

Role of efferocytosis in chronic pain -- From molecular perspective.

The complex nature of pain pathophysiology complicates the establishment of objective diagnostic criteria and targeted treatments. The heterogeneous manifestations of pain stemming from various primary diseases contribute to the complexity and diversity of underlying mechanisms, leading to challenges in treatment efficacy and undesirable side effects. Recent evidence suggests the presence of apoptotic cells at injury sites, the distal dorsal root ganglia (DRG), spinal cord, and certain brain regions, indicating a potential link between the ineffective clearance of dead cells and debris and pai

RNA N-glycosylation enables immune evasion and homeostatic efferocytosis.

Glycosylation is central to the localization and function of biomolecules1. We recently discovered that small RNAs undergo N-glycosylation2 at the modified RNA base 3-(3-amino-3-carboxypropyl) uridine (acp3U)3. However, the functional significance of N-glycosylation of RNAs is unknown. Here we show that the N-glycans on glycoRNAs prevent innate immune sensing of endogenous small RNAs. We found that de-N-glycosylation of cell-culture-derived and circulating human and mouse glycoRNA elicited potent inflammatory responses including the production of type I interferons in a Toll-like receptor 3- a

Efferocytosis induces macrophage proliferation to help resolve tissue injury.

Apoptotic cell clearance by macrophages (efferocytosis) promotes resolution signaling pathways, which can be triggered by molecules derived from the phagolysosomal degradation of apoptotic cells. We show here that nucleotides derived from the hydrolysis of apoptotic cell DNA by phagolysosomal DNase2a activate a DNA-PKcs-mTORC2/Rictor pathway that increases Myc to promote non-inflammatory macrophage proliferation. Efferocytosis-induced proliferation expands the pool of resolving macrophages in vitro and in mice, including zymosan-induced peritonitis, dexamethasone-induced thymocyte apoptosis, a
