The development and appearance of antibody was studied in the intestine and serum from histocompatible GB1 chickens orally infected with oocysts of Eimeria acervulina (restricted to the duodenum) or Eimeria tenella (restricted to the caeca). The local immune response was measured as the specific antibody levels in the supernatants of intestinal fragments (duodenum and caecum) maintained in culture for 16 h at 41 degrees C, 5% CO2, 95% air. Specific IgM was detected 1 week after E. acervulina infection, and the specific IgA and IgG contents of the duodenum and caecum were significantly elevated

ABSTRACTEimeria are globally enzootic parasites that can cause coccidiosis in chickens. Until recently, remarkably little had changed over the last 40 years in the fundamental biology that underpins detection and control of Eimeria. Tools such as microscopy and lesion scoring remain central to diagnosis, and control still relies on routine supplementation of diets with anticoccidial drugs or application of live vaccines. However, refocusing on aspects of economics, molecular biology, and bacteriology that relate to coccidiosis has prompted considerable change in dogma. The cost of coccidiosis

An attenuated line of Eimeria caviae was produced by selection for early maturation of oocysts during serial passage in guinea pigs. The prepatent period of the parasite was reduced from 11 to 6 days. Parasites of the precocious line and the parent strain were found in the crypts of the epithelial cells of caecum and colon and were morphologically indistinguishable. The period of development of type I, type II and type III merozoites of the E. caviae precocious line in guinea pigs was similar to that of the parent strain until the 3rd day. The time of the transition from gametocytes to oocysts

Coccidiosis, a serious intestinal parasitic disease caused by Eimeria spp., can result in huge annual economic losses to the poultry industry worldwide. At present, coccidiosis is mainly controlled by anticoccidial drugs. However, drug resistance has developed in Eimeria because of the long-term and unreasonable use of the drugs currently available. In our previous study, RNA-seq showed that the expression of methionine aminopeptidase1 (EtMetAP1) was up-regulated in diclazuril-resistant (DZR) and maduramicin-resistant (MRR) strains compared to drug-sensitive (DS) strain of Eimeria tenella. Chicken coccidiosis caused by Eimeria spp. has serious adverse effects on the global poultry industry. Previous studies have found Eimeria infection disrupts the absorption of nutrients and the efficiency of utilization of protein and energy in the diet. However, the effects of Eimeria infection on the absorption and utilization of dietary lipids are poorly understood. This study aimed to investigate the effects of Eimeria infection on dietary lipid absorption and to reveal possible mechanism influencing lipid metabolism in chicken. Forty-five 28-day-old yellow-feather chickens were allocated

Ribosomal RNAs (rRNAs), essential components of ribosomes involved in protein synthesis, are encoded by ribosomal DNAs (rDNAs), which are typically organized as tandem repeats within rDNA clusters. The internal transcribed spacer (ITS) region of rDNA, characterized by low intraspecific but high interspecific divergence, has been a focus of phylogenetic and diagnostic studies. However, comprehensive analysis of rDNA clusters remains challenging due to their repetitive nature and polymorphisms. In apicomplexan genomes, rDNA structure and function remain poorly explored. Here, we developed a pipe

BACKGROUND: Species of the genus Eimeria cause coccidiosis in chickens, resulting in a huge burden to the poultry industry worldwide. Eimeria tenella is one of the most prevalent chicken coccidia in China, and E. tenella infection causes hemorrhagic cecitis.
METHODS: Using an established model of coccidiosis in chickens combined with necropsy, imaging of pathological tissue sections, and other techniques, we evaluated the gross and microscopic lesions of cecal tissue within 15 days after inoculation with sporulated oocysts and described the endogenetic developmental process and relationship b

Eimeria necatrix, an apicomplexan protozoa of the genus Eimeria, causes intestinal coccidiosis that can reduce growth performance of poultry and result in high mortality in older chickens. In this report, the whole sporozoite proteins of E.necatrix were studied by two-dimensional electrophoresis (2-DE) and Western blotting using hyper-immune chicken serum containing E.necatrix-specific antibodies. Approximately 680 protein spots for E.necatrix sporozoite were detected by 2-DE with silver staining, where 98 spots were cross-reacted with the E. necatrix-specific immune sera. Out of the 56 spots

Chicken coccidiosis inflicts severe economic losses on the poultry industry. The mechanisms underlying Eimeria tenella (E. tenella) invasion, colonization, and damage to host cells remain incompletely elucidated. This study was designed to elucidate EtMIC2-interacting proteins and the mechanism that EtMIC2 regulates E. tenella invasion and host cell apoptosis. His pull-down-MS and KEGG enrichment results showed apoptosis-related proteins including ITGAV (Integrin alpha-V), PlexinB2, MAP2K6, GNA11, TRK, CASP7, PAK1, and RAP1b. Only the interaction model between EtMIC2 and ITGAV met the AlphaFol

The nutria (Myocastor coypus) is a semiaquatic rodent that originally inhabited South America. However, the animals have spread to different continents as alien species, and their numbers are quickly increasing, especially in North America, Europe, and Eastern Asia including Japan. Although nutrias have been suggested to serve as reservoirs for pathogens, including parasites, there have been few reports on this subject. In the present study, we surveyed the gastrointestinal parasites in nutrias living in Japan to better understand their prevalence in nutrias. We collected 72 samples of intesti

The purpose of this study was to determine if Eimeria oocyst concentrations and species composition in commercial broiler house litter changed during different cycles of anticoccidial drug (ACD) or live Eimeria oocyst vaccine (VAC) control programs and if there was a correlation between Eimeria oocyst levels and broiler performance. Litter samples were collected from a total of 15 different broiler farms encompassing a total of 45 individual houses during at least one complete grow-out cycle over a 21-mo period. Of these 15 broiler farms, three were followed for the entire 21-mo period spannin

We measured the in vitro production of interleukin-1 (IL-1) and tumor necrosis factor-alpha (TNF- ) by macrophages from chickens during and immediately following an infection with one of two different species of Eimeria, E. maxima and E. tenella. Quantitatively, the amounts of IL-1 produced during each infection were nearly identical regardless of the oocyst dose of each parasite. TNF production followed a biphasic pattern of increased production with the first peak associated with the pathogenesis of disease and the second peak associated with the development of protective immunity. These exp

Protein phosphorylation is an important post-translational modification (PTM) involved in diverse cellular functions. It is the most prevalent PTM in both Toxoplasma gondii and Plasmodium falciparum, but its status in Eimeria tenella has not been reported. Herein, we performed a comprehensive, quantitative phosphoproteomic profile analysis of four stages of the E. tenella life cycle: unsporulated oocysts (USO), partially sporulated (7 h) oocysts (SO7h), sporulated oocysts (SO), and sporozoites (S). A total of 15,247 phosphorylation sites on 9514 phosphopeptides corresponding to 2897 phosphopro

Coccidiosis is a disease caused by Eimeria, which represents the first parasitic disease in poultry farming. Among them, E. tenella is a virulent species which specifically colonizes the caecum. The inflammatory response to infection is associated to numerous host proteases including cysteine cathepsins that can be deleterious for tissue and innate immunity integrity. Here, germ-free and conventional chickens were used as models to find out whether the microbiota could modify the intestinal expression of host cysteine cathepsins during coccidiosis. The basal caecal peptidase activity primarily

Innate immune reactions conducted by macrophages may affect the outcome of primary infections and are crucial for the transition to adaptive immune responses. In bovine coccidiosis little is known on early monocyte/macrophage-mediated responses. We therefore investigated in vivo, in vitro and ex vivo reactions of monocytes and macrophages against Eimeria bovis, one of the most pathogenic Eimeria species in cattle. Macrophages significantly infiltrate the gut mucosa of E. bovis-infected calves, particularly after challenge infection. Furthermore, peripheral monocytes of infected animals, as pre
