Sickle cell disease covers a group of conditions in which pathology may be attributed to the presence of sickle hemoglobin (HbS). The identification of HbS and other variants including those in combination with HbS is commonly achieved by cellulose acetate electrophoresis at alkaline pH. Because many hemoglobin variants with similar charges have similar electrophoretic migration patterns, they are difficult to differentiate by electrophoresis. By applying sera containing various myeloma proteins to thin-layer isoelectric focusing at various pH ranges, we found that the best range of pH gradient for the detection of the band group specific for monoclonal immunoglobulin was from 5 to 8. The lowest concentration of monoclonal immunoglobulin detected by this method was approximately 0.1 mg/ml which was 8 to 32 times lower than the concentration detectable by cellulose acetate membrane electrophoresis or immunoelectrophoresis in routine laboratory use. Promastigotes from four cutaneous leishmaniasis cases from Colombia were tested by cellulose acetate electrophoresis using nine enzyme systems. The isoenzyme profiles of the Colombian isolates were indistinguishable from each other and from Panamanian Leishmania braziliensis panamensis controls, but were distinct from an isolate of Leishmania braziliensis guyanensis from Brazil and three isolates from the Leishmania mexicana complex for the enzyme phosphogluconate dehydrogenase.

A simple method for the electrophoretic separation of glycosaminoglycans from untreated urine is presented. It is shown that this rapid electrophoresis on specially shaped acetate cellulose strips gives more information than other screening tests used in laboratory for discriminating between normal and pathological samples. This technique is useful particularly with low levels of glycosaminoglycans which may give false-negative responses.

Using the modified bidimensional electrophoresis method, characteristic patterns of urinary glycosaminoglycans were obtained for nine different syndromes of mucopolysaccharidoses and normal. At the laboratory of the National Centre of Ophthalmology, Paris, tear electrophoresis on cellulose acetate support was studied in 56 Nigerian children--32 during and shortly after measles infection and 24 healthy controls. Leishmania organisms cultivated from cutaneous lesions of humans in Guatemala were characterized by cellulose acetate electrophoresis. A previously undescribed method is presented for staining citrate agarose films for electrophoresis of haemoglobin types using tetramethylbenzidine. This stain is noncarcinogenic and is considerably more sensitive than other commonly used stains, enabling abnormal haemoglobins at concentrations of 1% to be detected. The technique has been applied to screening neonates for haemoglobin S and other haemoglobinopathies after an initial test using cellulose acetate electrophoresis and Ponceau S staining has detected an abnormality.

BACKGROUND: Bisalbuminemia is a hereditary and/or acquired abnormality characterized by a double albumin (ALB) band on serum protein electrophoresis. However, there have been no epidemiological investigations of ALB variants in Chinese populations.

Sera from seventy chickens of several inbred lines were screened by cellulose acetate electrophoresis for abnormalities of immunoglobulin production. IgG levels in two sera (both from apparently healthy birds) were unusually high. The IgG proteins purified from these two sera were shown, by isoelectric focusing, to be of restricted heterogeneity.

An improved method for the estimation of haemoglobin A2, at the same time precise, simple and cheap, is proposed. Haemoglobin A is separated from haemoglobin A2 by electrophoresis on Cellogel in discontinuous buffer at alkaline pH. The strips of cellulose acetate containing the haemoglobin fractions are completely dissolved in 80% acetic acid. In the course of a study on the albumin variants in 36 unrelated subjects with genetic bisalbuminemia, we have observed, in the serum of a French blood donor, a new fast-moving variant. Cellulose acetate electrophoresis at pH 8.6 showed that this variant, so-called albumin 'Vanves', moved slowlier than albumin Gent and faster than albumin Reading.
