Six species of rodents from Kern County, California, were inoculated subcutaneously with western equine encephalomyelitis (WEE) virus to determine their susceptibility to infection and their potential as natural hosts. Ammospermophilus nelsoni, Citellus beecheyi, Dipodomys heermanni, Dipodomys nitratoides, Peromyscus maniculatus, and Sciurus griseus were readily infected. Infection was usually fatal in Dipodomys species, C. beecheyi, and S. griseus, but was clinically inapparent in other species. Viremic responses varied greatly in magnitude and duration in different species and with different

House finches, Carpodacus mexicanus, were experimentally infected with high and standard doses of western equine encephalomyelitis virus (WEEV) or St. Louis encephalitis virus (SLEV) to determine whether high doses would produce an elevated viremia response and a high frequency of chronic infections. Finches inoculated with approximately100,000 plaque forming units (PFU) of WEEV or SLEV produced viremia and antibody responses similar to those in finches inoculated with approximately 100 PFU of WEEV or 1000 PFU of SLEV, the approximate quantities of virus expectorated by blood-feeding Culex tar

Temporal and spatial changes in the enzootic activity of western equine encephalomyelitis (WEE) and St. Louis encephalitis (SLE) viruses were monitored at representative wetland study sites in the Coachella, San Joaquin, and Sacramento valleys of California from 1996 to 1998 using three methods: (1) virus isolation from pools of 50 host-seeking Culex tarsalis Coquillett females, (2) seroconversions in flocks of 10 sentinel chickens, and (3) seroprevalence in wild birds collected by mist nets and grain baited traps. Overall, 74 WEE and one SLE isolates were obtained from 222,455 Cx. tarsalis fe

Part of WEE virus (strain 16310-5614) genome coding for the nucleocapsid (C) protein was cloned and sequenced in two independent clones. The C gene of WEE virus is composed of 77 nucleotides, both for the BFS and 16310 strains, and is 48, 24, 33, 15, 3, and 3 nucleotides shorter than that of Venezuelan equine encephalomyelitis (VEE), Semliki Forest, Ross River, Sindbis, O'Nyong-Nyong, and Eastern equine encephalomyelitis (EEE) viruses, respectively. It contains 16 nucleotide changes in comparison with the BFS-1703 strain, four of which are significant: Ser57(BFS)-->Ala(16310), Gly63-->Cys, Lys

Adult hens, similar to those used for arbovirus surveillance, were experimentally infected with western equine encephalomyelitis (WEE) and St. Louis encephalitis (SLE) viruses to describe the viremia response, to compare serological testing methods, and to evaluate a new method of collecting whole blood onto filter paper strips from lancet pricks of the chicken comb. Young (19 weeks), but not old (38 weeks), hens developed a low-titer, transient viremia for a 1-day period. Immunoglobulin G (IgG) was detected by days 10 and 14 after infection with WEE and SLE viruses, respectively, by indirect

There are two dose-dependent barriers to the transmission of western equine encephalomyelitis virus in infected female Culex tarsalis. In the first, virus multiplies in the mesenteron, but does not invade other tissues regardless of the length of extrinsic incubation. We call this the "mesenteronal escape" barrier. In some mosquitoes virus escapes from the infected mesenteron but a second barrier prevents infection of the salivary glands and perhaps neural tissues. We designate this the "salivary gland infection" barrier. The effectiveness of the second barrier decreases with time but still is

Administration of brain suspension of intact albino mice during the 1st, 3rd and 7th days after a single, double and prolonged (5-cycle) immunization of rabbits produced no significant changes in the functional condition of the insular apparatus. A single use in the capacity of an antigen of the brain tissue containing the tick-borne encephalitis or western horse encephalomyelitis viruses also led to no marked shifts in the blood serum insulin activity. In reimmunization with the virus-containing material the value analysed decreased on the 7th day in comparison with the 1st and the 3rd days a

Mosquito population indices from California for the period 1953-1973 were analyzed to determine their association with activity of St. Louis encephalitis (SLE) and western equine encephalomyelitis (WEE) viruses. Culex tarsalis female populations, as measured by New Jersey light trap indices (LTI), correlated positively with the incidence rates of encephalitis in humans, and were a reliable means of forecasting the years of highest incidence. The critical level of C. tarsalis in urban areas below which no human cases of SLE and WEE were detected was an LTI of 0.1. Critical urban levels of C. ta

As a result of the continuing threat of Venezuelan equine encephalomyelitis (VEE), a study was made to determine if revaccination against VEE (TC-83 vaccine) was feasible and if revaccination could be incorporated into other routine vaccination practices. Of the horses given annual vaccination with bivalent western equine encephalomyelitis (WEE) and eastern equine encephalomyelitis (EEE) vaccine, 57% retained detectable serum-neutralizing (SN) antiboyd titers for VEE 18 months after the initial VEE vaccination was given. Of horses with no record of WEE-EEE vacinnation, 100% retained detectable

Historically, western equine encephalomyelitis virus (WEEV) caused large equine and human epidemics in the Americas from Canada into Argentina. Despite recent enhanced surveillance for West Nile virus, there have been few reports of equine or human cases and little documented enzootic activity of WEEV. During the past three years, WEEV has been active again in California, but without human or equine cases. In the current study, we compared host and vector competence of representative WEEV isolates made during each decade over the past 60 years using white-crowned sparrows, house sparrows, and

Western equine encephalomyelitis and St. Louis encephalitis viral RNA can be detected 20 days after death of infected Culex tarsalis in the absence of a cold chain. Viral RNA was detected with the reverse transcription-polymerase chain reaction in mosquitoes infected either parenterally or perorally in the laboratory and then killed and held for up to 20 days at 27 degrees C. Cell culture assay and in situ enzyme immunoassay did not detect infectious virus in the same mosquitoes.

The complete nucleotide sequence of a 1982 Florida strain of eastern equine encephalomyelitis (EEE) virus, and partial sequence of the nonstructural protein genes of western equine encephalomyelitis (WEE) virus, were determined. The EEE virus genome was 11,678 nucleotides in length, excluding the cap nucleotide and poly(A) tail, and the nucleotide composition was 28% A, 24% G, 25% C, and 23% U. The organization of both EEE and WEE virus genomes was like that of other alphaviruses and included a termination codon between the nsP3 and nsP4 genes. Codon usage for 10 of 20 amino acids was nonrando

The extrinsic incubation rate (inverse of the time in days from infection to median transmission) of western equine encephalomyelitis (WEE) and St. Louis encephalitis (SLE) viruses by laboratory strains of Culex tarsalis Coquillett increased as a linear function of incubation temperatures from 10 to 30 degrees C. The estimated temperatures for zero transmission thresholds (intercept of the X axis) were 10.9 and 14.9 degrees C, and the number of degree days above these thresholds required for median transmission (inverse of the slope) was 67.6 and 115.2, respectively. Although the bodies of mos
