BETA ENDORPHINS - HOLISTIC THERAPEUTIC APPROACH TO CANCER.

AIM: Endorphins are endogenous morphine, neuropeptides, produced in the pituitary gland in response to stress and pain. There are three types of endorphins beta-endorphins, enkephalins, and dynorphins binds to mu, kappa, and delta receptors situated on nervous system and immune cells. Cancer is a major threat to mankind killing millions of people around the world annually. There has been recent advancements in the field of surgery, chemotherapy, and radiotherapy, still the prognosis of cancer patients not improved much with increasing morbidity. We can't kill cancer cells without killing norma

CSF beta-endorphins in childhood neuropsychiatric disorders.

Thirty-one children with autistic disorder, 8 with the Rett syndrome (RS), 2 with childhood disintegrative disorder and 5 with infantile spasms were compared with healthy adult controls with respect to cerebrospinal fluid (CSF) beta-endorphin levels. The autistic disorder and RS groups showed significantly lower values than the other groups. There were no age trends within the various groups. Further study of CSF beta-endorphins in these disorders and blindly examined age matched controls is warranted.

Plasma endorphins in Rett syndrome: preliminary data.

Plasma levels of beta-endorphin (beta-EP) and prolactin (PRL) were measured in 5 girls with Rett syndrome and in a control group before and after giving 10 mg metoclopramide i.v. beta-lipotropin (beta-LPH) was only measured in basal conditions. Basal values of beta-EP and beta-LPH were lower than in control individuals. The responses of plasma beta-EP to metoclopramide in Rett syndrome patients were less intense than in control individuals, while the PRL increase in girls with Rett syndrome was significantly higher than in control subjects. These preliminary data suggest a derangement of the d

The measurement of endorphins in body fluids.

The measurement of endorphins in body fluids has been an important advance in clinical research attempting to link the endogenous opioid system to psychiatric illness and symptomatology. The consideration of methodologic differences in assay technique and in clinical methods is important in evaluating results of studies. Whereas findings in early clinical studies supported the notion of increased endorphin system function in patients with schizophrenia, cumulative data from the considerable number of studies carried out throughout world centers have been unable to demonstrate a consistent abno

Enkephalins and Endorphins. Clinical, pharmacological and therapeutic implications.

During the past 8 years there has been substantial progress in our understanding of the structure, distribution and action of endogenous opioid peptides. Currently, there appear to be 2 groups of peptides; those derived from beta-lipotropin and an enkephalin-related group. Some of these peptides have been shown to be distributed widely in the central nervous system and in endocrine tissues. The activity of the peptides probably occurs at several receptors but only 1 relatively specific (mu-receptor) antagonist, naloxone, is well studies. Although there have been many clinical studies of the ac

Role of beta-endorphins in silent myocardial ischemia.

The reason for the absence of pain perception in silent myocardial ischemia is unknown. A role of increased endorphinic activity in patients with silent ischemia has been postulated. To further investigate this hypothesis, 10 men with documented coronary artery disease and previous positive electrocardiographic findings during exercise without anginal pain were studied. Six healthy volunteers served as control subjects. The protocol included 2 bicycle exercise tests, the first test serving as baseline and the second performed after administration of naloxone, a specific opiate antagonist. Plas

Endorphins in psychiatry: an overview and a hypothesis.

This article presents an overview of the biochemistry, pharmacology, and physiology of endogenous opioid peptides (endorphins). Clinical psychopharmacology of exogenous opiate agonists and antagonists is reviewed. The evidence presented in the review is compatible with a hypothesis that the level of functional endorphins may be related to psychological events, with a normal level needed for psychological homeostasis. One corollary of this hypothesis is that the level of opioids in the brains of the mentally ill may be disturbed. Therapeutic implications of this hypothesis are considered.

[Endorphins. Physiological and pharmacological aspects, and research in psychiatry].

This article gives an overview of the biochemistry, physiology and pharmacology of endogenous opioid peptides. The role of endorphins in psychiatric pathology in the last ten years, has mainly been studied through two clinical research strategies: Pharmacological, administration of opiate agonists or antagonists, or substances altering endorphins metabolism. Biological, static or dynamic dosage of opioid activity in peripheral liquids, trying to correlate those measures either with a syndrome, or with a clinical trait. These methods applied to schizophrenia, affective disorders, anxiety, addic

Bacterial lipopolysaccharide induction of leukocyte-derived corticotropin and endorphins.

Previous reports have shown that there is an endogenous opioid component associated with pathophysiological responses to endotoxin. It has been shown that these responses are alleviated by naloxone, a specific opiate antagonist. Results of another study have indicated that leukocytes may mediate some of those responses since leukocyte depletion alleviated the effects of lipopolysaccharide. In view of the above reports as well as the finding that leukocytes produce immunoreactive (ir-) endorphins and corticotropin (ACTH) when stimulated with Newcastle disease virus or ACTH-releasing factor, we

Endorphins and the central inhibition of urinary bladder motility.

The involvement of endogenous opioid mechanisms in the central neurogenic control of urinary bladder function has been examined in anesthetized rats. Intracerebroventricular (ICV) microinjections of beta-endorphin (0.5-2.0 micrograms) produced powerful inhibition of rhythmic bladder contractions initiated by central reflex activity. The peptide fragments gamma-endorphin and alpha-endorphin (4-16 micrograms), formed by the processing of beta-endorphin by membrane homogenates of brain, were less active than the parent compound. The inhibitory effects of beta-endorphin was reversed by ICV naloxon

Endorphins and the regulations of the human menstrual cycle.

In order to assess a possible influence of endogenous opioids upon gonadotrophin secretion in women, we examined the effects of i.v. administration of 10 mg naloxone, a specific opiate antagonist, in ten normal menstruating women, in thirteen women with amenorrhoea and/or hyperprolactinaemia and in two women with putative deficiency of gonadotrophin-releasing hormone (GnRH). In thirteen subjects, a saline vehicle control study (randomized order of administration) was also performed. In the normal women, naloxone failed to elicit changes in serum gonadotrophin levels when administered during th

Endorphins in experimental spinal injury: therapeutic effect of naloxone.

An experimental model of cat spinal injury was used to investigate the hypotheses that endorphins are involved in the pathophysiology of spinal cord injury and that the opiate antagonist naloxone, by blocking the effects of endorphins, may improve physiological and neurological recovery. Experimental trauma to the cervical spine caused a reduction in blood pressure and spinal cord blood flow (SCBF) associated with increased plasma levels of beta-endorphin-like immunoreactivity; both blood pressure and SCBF were significantly improved by naloxone treatment. Naloxone-treated animals also showed

[The possibility of endorphins playing a role in psychic disturbances].

1. The release of endorphins appears to play a role as a reserve-mechanism in stress situations. 2. The application of high doses of the specific opiate antagonist 12-allyl-7,7a,8,9-tetrahydro-3,7a-dihydroxy-(4aH)-(8,9-c)-iminoethanophenanthro(4,5-b,c,d)furan-5(6H)-one (naloxone) to schizophrenic patients induces a partial reversal of hallucinations and actual delusional experience within 2--7 h after injection. 3. The blockade of opiate receptors by naloxone is possibly also effective against catatonic states of schizophrenia. 4. In affective psychoses no curative action of naloxone could be

Beta-endorphin in pregnancy.

Concentrations of maternal plasma beta-endorphin (beta-EP) as measured by radioimmunoassay decline during pregnancy, reaching a nadir during the second trimester, rise during labor, remain elevated during the early postpartum period and are increased prior to elective cesarean section in the absence of labor. They decline in response to epidural anesthesia during labor and increase during induction of general but not regional anesthesia for cesarean section. Umbilical venous plasma beta-EP levels are not affected by the route or mode of delivery nor the presence or absence of labor, but rise i

[The role of endorphins in the etiopathogenesis of circulatory diseases].

At the beginning of the 70-ties various studies showed the existence of endogenous opiates in central nervous system. The chemical structure of first endogenous pentapeptides called enkephalins (met-enkephalin and leu-enkephalin) and their receptor was already known in 1975. Further investigations confirmed the existence of opiates (endorphins) in brain, pituitary gland, spinal cord and other tissues. Endorphins may exert various effects: analgesic (especially dynorphin and b-endorphin), antidiuretic, depressive on respiratory center, constipative, they also cause physical and mental dependenc

The role of endorphins and vasopressin in canine endotoxin shock.

Chemical antagonists were used to assess the role of beta-endorphin and arginine-vasopressin (AVP) in canine endotoxin shock. Fifteen awake dogs were given Escherichia coli endotoxin IV. Within 5 min, CO decreased to 28%, LV dP/dt to 46%, and MAP to 52% baseline. Fifteen minutes after endotoxin, five dogs each received naloxone, AVP antagonist, or no treatment. Control (untreated) animals exhibited persistent cardiovascular depression, with CO 49%, LV dP/dt 69%, and MAP 91% of baseline after 45 min. Naloxone improved CO to 69%, LV dP/dt to 94%, and MAP to 91% by 30 min after treatment. AVP blo

Opiate receptors and endorphins in the pathophysiology of hemorrhagic shock.

We investigated the hypothesis that endorphins released by stress act on opiate receptors to depress cardiovascular function during hemorrhagic shock. Anesthetized adult mongrel dogs were bled into a heparinized reservoir to achieve a mean arterial pressure (MAP) of 45 mm Hg. The reservoir was adjusted to maintain MAP for 1 hour and then clamped for 1 hour, at the end of which time the shed blood was reinfused. While the reservoir was clamped we treated the animals with an intravenous bolus followed by 3-hour infusion of either 0.9% NaCl (as control) or the specific opiate receptor antagonist
