Endosome-lysosomes, ubiquitin and neurodegeneration.

Before the advent of ubiquitin immunochemistry and immunogold electron microscopy, there was no known intracellular molecular commonality between neurodegenerative diseases. The application of antibodies which primarily detect ubiquitin protein conjugates has shown that all of the human and animal idiopathic and transmissible chronic neurodegenerative diseases, (including Alzheimer's disease (AD), Lewy body disease (LBD), amyotrophic lateral sclerosis (ALS), Creutzfeldt-Jakob disease (CJD) and scrapie) are related by some form of intraneuronal inclusion which contains ubiquitin protein conjuga

The uncoating of EV71 in mature late endosomes requires CD-M6PR.

Enterovirus 71 (EV71) is one of the causative agents of hand-foot-and-mouth disease, which in some circumstances could lead to severe neurological diseases. Despite of its importance for human health, little is known about the early stages of EV71 infection. EV71 starts uncoating with its receptor, human scavenger receptor B2 (hSCARB2), at low pH. We show that EV71 was not targeted to lysosomes in human rhabdomyosarcoma cells overexpressing hSCARB2 and that the autophagic pathway is not essential for EV71 productive uncoating. Instead, EV71 was efficiently uncoated 30 min after infection in la

AGAP2 regulates retrograde transport between early endosomes and the TGN.

The retrograde transport route links early endosomes and the TGN. Several endogenous and exogenous cargo proteins use this pathway, one of which is the well-explored bacterial Shiga toxin. ADP-ribosylation factors (Arfs) are approximately 20 kDa GTP-binding proteins that are required for protein traffic at the level of the Golgi complex and early endosomes. In this study, we expressed mutants and protein fragments that bind to Arf-GTP to show that Arf1, but not Arf6 is required for transport of Shiga toxin from early endosomes to the TGN. We depleted six Arf1-specific ARF-GTPase-activating pro

IQGAP1 promotes CXCR4 chemokine receptor function and trafficking via EEA-1+ endosomes.

IQ motif-containing GTPase-activating protein 1 (IQGAP1) is a cytoskeleton-interacting scaffold protein. CXCR4 is a chemokine receptor that binds stromal cell-derived factor-1 (SDF-1; also known as CXCL12). Both IQGAP1 and CXCR4 are overexpressed in cancer cell types, yet it was unclear whether these molecules functionally interact. Here, we show that depleting IQGAP1 in Jurkat T leukemic cells reduced CXCR4 expression, disrupted trafficking of endocytosed CXCR4 via EEA-1(+) endosomes, and decreased efficiency of CXCR4 recycling. SDF-1-induced cell migration and activation of extracellular sig

The intact structural form of LLO in endosomes cannot protect against listeriosis.

LLO is the major immuno-dominant antigen in listeriosis and is also required for protective immunity. Two forms of LLO can be observed in endosomal membranes, a LLO intact form and a Ctsd-processed LLO(1-491) form. Endosomes obtained from resting macrophages contained only LLO intact forms, while endosomes obtained from IFN-activated macrophages contained both forms. Both types of endosomes elicited LLO(90-91)/CD8(+) and LLO(189-201)/CD4(+) specific immune responses. However, only endosomes containing the Ctsd-processed LLO(1-491) form showed significant CD4(+) and CD8(+) T cell responses simi

Endosomal Mechanisms in Heart Failure Pathophysiology.

PURPOSE OF REVIEW: To examine the role of endosomal dysfunction in heart failure pathophysiology and evaluate its potential as a therapeutic target, particularly focusing on its regulation of cardiac metabolism. RECENT FINDINGS: Endosomal dysfunction, driven by v-ATPase disassembly and loss of acidification, emerges as a key contributor to metabolic perturbations in heart failure. This dysfunction leads to uncontrolled CD36 translocation, resulting in lipotoxicity and inflammatory signaling through CD36-TLR4 complex formation. These mechanisms are especially relevant in diabetic cardiomyopath

Early Endosome Morphology in Health and Disease.

Early endosomes are organelles that receive macromolecules and solutes from the extracellular environment. The major function of early endosomes is to sort these cargos into recycling and degradative compartments of the cell. Degradation of the cargo involves maturation of early endosomes into late endosomes, which, after acquisition of hydrolytic enzymes, form lysosomes. Endosome maturation involves recruitment of specific proteins and lipids to the early endosomal membrane, which drives changes in endosome morphology. Defects in early endosome maturation are generally accompanied by alterati

Effect of HSV-1 ICP0 E3 ubiquitin ligase on CIN85 endosomes.

Herpes simplex virus-1 (HSV-1) has infected more than 67% of the world population causing diseases that range in severity from benign cold sores to encephalitis. To infect and persist in the host, HSV-1 has evolved sophisticated strategies to counteract antiviral responses. The viral immediate early protein ICP0 (Infected Cells Protein 0) plays a fundamental role in this process. ICP0 is an E3 ubiquitin ligase that is required for successful onset of lytic infection and productive reactivation of viral genomes from latency. Most ICP0 studies have been focused on its nuclear functions, but late

TBK1 activity regulates the directionality of axonal transport of signalling endosomes.

The polarised and complex morphology of neurons poses massive challenges for efficient cargo delivery between the axon and soma, a process termed axonal transport. We have previously shown that the retrograde axonal transport of pro-survival, neurotrophic signalling endosomes relies on Rab7 in motor neurons, and that their trafficking is impaired in the early stages of amyotrophic lateral sclerosis (ALS) pathogenesis. Here, we report the effect of Rab7 phosphorylation on the transport of these signalling endosomes. We show that the ALS-linked kinase TBK1 phosphorylates Rab7 at S72 in neurons,

Huntingtin, signaling endosomes and neuronal survival in health and Huntington disease.

The axonal transport of organelles mediates several functions throughout the lifespan of neurons. Over the past years, the role of endosomes, a specific type of trafficking organelle, has been extensively investigated for their increasing relevance during development, maturation and maintenance of neuronal networks. Endosome formation results from membrane invagination events driven by endocytic pathways. Following their formation, endosomes undergo maturation steps, and their fate is determined by the recruitment of specific cytosolic proteins to their membrane. Early endosomes may be recycle

LAMTOR1 sorts APP to late endosomes via AP-3-mediated trafficking.

β-Amyloid precursor protein (APP), an amyloid β-peptide (Aβ) generating protein, plays a central role in Alzheimer's disease (AD). Its intracellular trafficking and proteolytic processing are precisely regulated, but the mechanisms remain elusive. Here, we identified LAMTOR1 as a novel APP intracellular domain binding partner that sorts APP from early endosomes to late endosomes. Knockout of LAMTOR1 decreased APP trafficking to late endosomes, which promoted its amyloidogenic processing on early endosomes, resulting in increased Aβ. Conditional knockout of LAMTOR1 in Camk2a-expressing neurons

Nanomedicines targeting protease-activated receptor 2 in endosomes provide sustained analgesia.

Although many internalized G protein-coupled receptors (GPCRs) continue to signal, the mechanisms and outcomes of intracellular GPCR signaling are uncertain due to the challenges of measuring organelle-specific signals and of selectively antagonizing receptors in intracellular compartments. Herein, genetically encoded biosensors targeted to the plasma membrane and early endosomes were used to analyze compartmentalized signaling of protease-activated receptor 2 (PAR2); the propensity of nanoparticles (NPs) to accumulate in endosomes was leveraged to preferentially antagonize intracellular PAR2

Altered endosomal-lysosomal biogenesis in melanoma.

Cutaneous melanoma is the deadliest form of skin neoplasm and its high mortality rates could be averted by early accurate detection. While the detection of melanoma is currently reliant upon melanin visualisation, research into melanosome biogenesis, as a key driver of pathogenesis, has not yielded technology that can reliably distinguish between atypical benign, amelanotic and melanotic lesions. The endosomal-lysosomal system has important regulatory roles in cancer cell biology, including a specific functional role in melanosome biogenesis. Herein, the involvement of the endosomal-lysosomal

The Endosomal Recycling Pathway-At the Crossroads of the Cell.

The endosomal recycling pathway lies at the heart of the membrane trafficking machinery in the cell. It plays a central role in determining the composition of the plasma membrane and is thus critical for normal cellular homeostasis. However, defective endosomal recycling has been linked to a wide range of diseases, including cancer and some of the most common neurological disorders. It is also frequently subverted by many diverse human pathogens in order to successfully infect cells. Despite its importance, endosomal recycling remains relatively understudied in comparison to the endocytic and

Actin-dependent endosomal receptor recycling.

Endosomes constitute major sorting compartments within the cell. There, a myriad of transmembrane proteins (cargoes) are delivered to the lysosome for degradation or retrieved from this fate and recycled through tubulo-vesicular transport carriers to different cellular destinations. Retrieval and recycling are orchestrated by multi-protein assemblies that include retromer and retriever, sorting nexins, and the Arp2/3 activating WASH complex. Fine-tuned control of actin polymerization on endosomes is fundamental for the retrieval and recycling of cargoes. Recent advances in the field have highl

The late endosome is essential for mTORC1 signaling.

The multisubunit mTORC1 complex integrates signals from growth factors and nutrients to regulate protein synthesis, cell growth, and autophagy. To examine how endocytic trafficking might be involved in nutrient regulation of mTORC1, we perturbed specific endocytic trafficking pathways and measured mTORC1 activity using S6K1 as a readout. When early/late endosomal conversion was blocked by either overexpression of constitutively active Rab5 (Rab5CA) or knockdown of the Rab7 GEF hVps39, insulin- and amino acid-stimulated mTORC1/S6K1 activation were inhibited, and mTOR localized to hybrid early/l

Zn2+ depletion blocks endosome fusion.

Fusion among endosomes is an important step for transport and sorting of internalized macromolecules. Working in a cell-free system, we previously reported that endosome fusion requires cytosol and ATP, and is sensitive to N-ethylmaleimide. Fusion is regulated by monomeric and heterotrimeric GTP-binding proteins. We now report that fusion can proceed at very low Ca2+ concentrations, i.e. < 30 nM. Moreover, fusion is not affected when intravesicular Ca2+ is depleted by preincubation of vesicles with calcium ionophores (5 microM ionomycin or A23187) in the presence of calcium chelators (5 mM EGT

Niemann-Pick C1 maintains cholesterol homeostasis in late endosomes to drive hepatitis B virus endosomal escape.

Hepatitis B virus (HBV) infection poses a major global health burden, as current nucleos(t)ide analogs and interferon-alpha (IFN-α) cannot eradicate the viral reservoir. Host factors play an indispensable regulatory role in enabling HBV to establish persistent infection. While host factors governing early HBV infection processes-including attachment, endocytic internalization, endosomal trafficking, and covalently closed circular DNA (cccDNA) formation-are largely elucidated, the regulators mediating HBV release from late endosomes remain elusive. This study investigates the role of Niemann-Pi

Enhanced generation of intraluminal vesicles in neuronal late endosomes in the brain of a Down syndrome mouse model with endosomal dysfunction.

Down syndrome (DS) is a human genetic disease caused by trisomy of chromosome 21 and characterized by early developmental brain abnormalities. Dysfunctional endosomal pathway in neurons is an early event of DS and Alzheimer's disease. Recently, we have demonstrated that exosome secretion is upregulated in human DS postmortem brains, in the brain of the trisomic mouse model Ts[Rb(12.1716 )]2Cje (Ts2) and by DS fibroblasts as compared with disomic controls. High levels of the tetraspanin CD63, a regulator of exosome biogenesis, were observed in DS brains. Partially blocking exosome secretion by

Late Endosomal/Lysosomal Cholesterol Accumulation Is a Host Cell-Protective Mechanism Inhibiting Endosomal Escape of Influenza A Virus.

To transfer the viral genome into the host cell cytoplasm, internalized influenza A virus (IAV) particles depend on the fusion of the IAV envelope with host endosomal membranes. The antiviral host interferon (IFN) response includes the upregulation of interferon-induced transmembrane protein 3 (IFITM3), which inhibits the release of the viral content into the cytosol. Although IFITM3 induction occurs concomitantly with late endosomal/lysosomal (LE/L) cholesterol accumulation, the functional significance of this process is not well understood. Here we report that LE/L cholesterol accumulation i
