Vascular endothelium: the battlefield of dengue viruses.

Increased vascular permeability without morphological damage to the capillary endothelium is the cardinal feature of dengue haemorrhagic fever (DHF)/dengue shock syndrome (DSS). Extensive plasma leakage in various tissue spaces and serous cavities of the body, including the pleural, pericardial and peritoneal cavities in patients with DHF, may result in profound shock. Among various mechanisms that have been considered include immune complex disease, T-cell-mediated, antibodies cross-reacting with vascular endothelium, enhancing antibodies, complement and its products, various soluble mediator

Regulatory mechanisms of the vascular endothelium: an update.

This review discusses recent experimental findings in prostacyclin, nitric oxide and endothelin research. Prostacyclin formation by endothelial cells in atherosclerosis and diabetes is reviewed and the synthesis of prostacyclin by cyclooxygenase 1 and 2 (COX-1 and COX-2) is discussed. Further work on nitric oxide describes its involvement in septic and haemorrhagic shock and its interactions with the cyclooxygenase pathway. Recent studies in endothelin research include the development of both selective and orally active receptor antagonists, characterization of endothelin converting enzymes an

Gastrointestinal inflammation: focus on the vascular endothelium.

Ischemia of the intestine predominantly affects the small bowel and colon, and may ensue from a variety of causes, ranging from radiation enteritis to the use of drugs that affect the intestinal vasculature. Gut ischemia has historically been thought of as "large-vessel" disease, but microcirculatory pathology and ischemia-reperfusion injury is increasingly being implicated in gut infections and even in chronic idiopathic inflammatory processes involving the gut, such as ulcerative colitis and Crohn's disease.

The migration of lymphocytes across vascular endothelium.

According to Morris and his colleagues the migration of lymphocytes from the blood into inflammatory sites is a morphologically similar process to the large scale migration of lymphocytes into the paracortex of lymph-nodes which occurs under normal conditions. Little is known of the nature of the selective interaction between lymphocytes and specialized endothelial cells although several investigators have tested the migratory properties of lymphocytes which have been treated in vitro with enzymes or lectins and injected i.v. Such results are difficult to interpret because decreased lymph-node

Studies on solute transfer in the vascular endothelium.

Barium chromate, Prussian blue, and cobalt-cobalticyanide can be precipitated in vivo in the endothelium of mesenteric vessels by injection of the appropriate anions into the blood stream, and topical application of the precipitating cations to the exposed mesenteries of mice and frogs. Precipitation in the endothelium occurs in the form of a diffuse fine punctate precipitate, and also as continuous lines demarcating endothelial cell outlines. A striking feature is the frequent occurrence of this type of precipitation in a tapering zone downstream from mural thrombi in the veins.

Proliferation dynamics of organotypic vascular endothelium during aging.

Healthy blood vessels are vital for organismal health. Blood vessel-lining endothelial cells (ECs) can self-replicate to maintain vascular homeostasis, yet the age-related dynamics of EC proliferation remain elusive. Employing cumulative labeling of proliferating ECs, we here present a temporal map of organotypic changes in endothelial proliferation at different stages of mouse life [juvenile (J), 1-month-old; young adult (YA), 4-month-old; old adult (OA), 14-month-old]. Comparative analyses of 12 different organs revealed a tissue-specific pattern of age-related changes in endothelial prolife

Functional status of vascular endothelium in diabetes mellitus.

Normal endothelial cells synthesize and release biologically active substances. These substances maintain homeostasis through adequate blood flow, delivery of nutrients, activation and inhibition of coagulation proteins, prevention of thrombosis and diapedesis of leukocyte. Endothelial dysfunction implies failure of vascular endothelium to perform its normal functions of vasodilatation and vasoconstriction. It results from an imbalance between endothelium derived constricting and relaxing factors. Altered endothelial cell activity predisposes to increased production of vasoconstrictors, i.e.,

A simple method for culturing mouse vascular endothelium.

Vascular endothelium is an important site for a wide array of immunological processes such as inflammation, atherosclerosis and allograft rejection. Culture methods of mouse vascular endothelium would provide an important in vitro correlate to immunological murine in vivo models. We describe a simple method to culture mouse vascular endothelium from thoracic aorta. Our cultured cells express typical phenotypic (CD105, CD31, CD106), morphological and ultrastructural (intercellular junctions, Weibel-Palade bodies) markers of vascular endothelium. They also possess functional receptors for uptake

The vascular endothelium as decision maker in lung injury.

The vascular endothelium is the largest organ in the human body, capable of performing a wide range of cellular signaling and synthetic functions. It is also subjected to considerable mechanical stress due to the shear forces generated by blood flow, which amounts to approximately 10,000 L per day. The endothelial layer plays a crucial role in regulating vascular tone locally and controlling the extravasation of certain blood components. Additionally, it is integral to the coagulation process. The endothelium serves as the entry point for immune cells, which migrate from the bloodstream to sur

[Effect of sleep apnea syndrome on the vascular endothelium].

Obstructive sleep apnoea (OSA) is a common disorder associated with an increased risk of cardiovascular disease and stroke. As it is strongly associated with known cardiovascular risk factors, including obesity, insulin resistance, and dyslipidemia, OSA is an independent risk factor for hypertension. Although the association between OSA and the metabolic syndrome tends to confound studies of the independent effects of OSA on vascular disease, recent evidences from basic science to epidemiological and clinical studies suggest that OSA may add worsening pathophysiological conditions to obesity.

[Antibodies to phospholipids and the vascular endothelium in nodular polyarteritis].

Clinical significance of antibodies to phospholipids (aPL) and vascular endothelium (aVE) was evaluated in 20 patients (9 women and 11 men aged 36 +/- 10.8 years) with nodular polyarteritis (NP) corresponding to classification criteria of the USA Rheumatology College. Antibodies to cardiolipin (aCL) (IgG and IgM) and to beta 2-glycoprotein (beta 2-GP1) (IgG) were titered by solid-phase enzyme immunoassay. Total serum level of aVE (IgG + IgM + IgA) was measured by solid-phase enzyme immunoassay using Eahy. 926 endothelial hybrydoma cell culture. Anticardiolipin antibodies were detected in 11 (5

HIV Infection, Antiretroviral Drugs, and the Vascular Endothelium.

Endothelial cell activation, injury, and dysfunction underlies the pathophysiology of vascular diseases and infections associated with vascular dysfunction, including human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome. Despite viral suppression with combination antiretroviral therapy (ART), people living with HIV (PLWH) are prone to many comorbidities, including neurological and neuropsychiatric complications, cardiovascular and metabolic diseases, premature aging, and malignancies. HIV and viral proteins can directly contribute to the development of these comorbidities.

Emerging Roles of Vascular Endothelium in Metabolic Homeostasis.

Our understanding of the role of the vascular endothelium has evolved over the past 2 decades, with the recognition that it is a dynamically regulated organ and that it plays a nodal role in a variety of physiological and pathological processes. Endothelial cells (ECs) are not only a barrier between the circulation and peripheral tissues, but also actively regulate vascular tone, blood flow, and platelet function. Dysregulation of ECs contributes to pathological conditions such as vascular inflammation, atherosclerosis, hypertension, cardiomyopathy, retinopathy, neuropathy, and cancer. The clo

Small fibronectin fragments induce endothelium-dependent vascular relaxations.

Strips of rabbit thoracic aorta precontracted with phenylephrine relaxed when exposed to selected synthetic peptides derived from the cell attachment domain of fibronectin. The relaxations elicited by both acetylcholine and the hexapeptide Gly-Arg-Gly-Asp-Ser-Pro (GRGDSP) were dependent on the presence of an intact endothelium, were resistant to indomethacin, and were inhibited by hemoglobin. The structure-activity relationship of four oligopeptides derived from fibronectin was in fair agreement with their ability to prevent fibronectin-mediated cell adhesion in other experimental systems. A Window into the Vascular Endothelium in Covid-19: Nails.

INTRODUCTION: Endothelial damage is associated with acute and long-term coronavirus disease 2019 (COVID-19) complications. Proximal nailfold capillaries and nail beds give important clues to microvascular changes associated with endothelial dysfunction. OBJECTIVE: We aimed to use dermoscopy to examine the proximal nailfold capillaries and nail bed of COVID-19 patients and identify microvascular changes. METHODS: A prospective study was designed to evaluate the dermoscopic features of proximal nail fold capillaries and nail bed in mild-to-moderate COVID-19 patients and healthy controls betwee

Aldosterone: Essential for Life but Damaging to the Vascular Endothelium.

The renin angiotensin aldosterone system is a key regulator of blood pressure. Aldosterone is the final effector of this pathway, acting predominantly via mineralocorticoid receptors. Aldosterone facilitates the conservation of sodium and, with it, water and acts as a powerful stimulus for potassium excretion. However, evidence for the pathological impact of excess mineralocorticoid receptor stimulation is increasing. Here, we discussed how in the heart, hyperaldosteronism is associated with fibrosis, cardiac dysfunction, and maladaptive hypertrophy. In the kidney, aldosterone was shown to cau
