UNLABELLED: We have studied the effects of handgrip on plasma levels of catecholamines, neuropeptide Y (NPY), and leu-enkephalin before and after hemodialysis of uremic patients. A cuprophan dialyzer was used. We found, that dopamine level was higher in uremia group before hemodialysis both during rest (0.38 +/- 0.39 pmol/ml) and handgrip (1.13 +/- 1.00 pmol/ml) compared to control (0.17 +/- 0.19, and 0.66 +/- 0.83 pmol/ml respectively). The influence of methionine-enkephalin and leucine-enkephalin on human detrusor and on pig detrusor, trigone, bladder neck and urethral smooth muscle was investigated in vitro. The enkephalins inhibited the smooth muscle contractility presynaptically. Methionine-enkephalin was about 40% more potent than leucine-enkephalin. Phentolamine-resistant contractions were less inhibited by the enkephalins than atropine-resistant contractions and contractions that were not blocked. The inhibitory nerve responses of the trigone, bladder neck and urethra were unaffected.

Leucine and methionine enkephalins were measured by biological assay in normal colon, adenocarcinoma of the colon, carcinoma of the caecum, and in normal and inflamed appendix. Methionine enkephalin levels in both the adenocarcinomas and inflamed appendices were significantly higher than in normal controls. No significant change occurred in leucine enkephalin levels. We report on a 23-year-old patient with periodic hypersomnia. Electroencephalographic (EEG) background activity for this individual was slightly slowed in the EEG during an episode of hypersomnia, and intermittent slow activity was found in addition. Usual laboratory parameters were normal; however, leucine-enkephalin was markedly elevated in the plasma at that time, whereas free cysteine could not be demonstrated. Clinical findings were normal in the following years, and the EEG background activity returned to normal; leucine-enkephalin and cysteine also returned to normal values.
