The development of genetically engineered multivalent subunit vaccines is needed to provide protection against infections caused by Clostridium septicum and multiple types of Clostridium perfringens. In this study, a pET-32a(+) plasmid encoding the C. septicum α-toxin gene and the C. perfringens β-toxin gene was constructed; a positive clone was designated pET-32a-α-β1. This plasmid, along with pET-32a-α-β2-ε-previously engineered to express a C. perfringens α-β2-ε trivalent recombinant chimeric fusion protein-was transformed into Escherichia coli DE3 for recombinant protein expression after i

The gastro-intestinal flora of clinically intact piglets on an industrialised sow unit was investigated prior to weaning (n = 12) and after weaning (n = 12). The age of the former group was 23 to 36 days and that of the latter between six and eleven days. While enormous proliferation of haemolysing E. coli was recorded from the anterior portion of the small intestine in the post-weaning group, weaning, quite, generally, was found to have only little impact upon the gastrointestinal flora of piglet. More particularly were there no indicators to the effect that post-weaning proliferation of ente

Clostridium perfringens type D is a gram-positive bacterium that causes enterotoxemia in sheep, goats, and, less frequently, other animals. This microorganism encodes 2 major toxins, alpha (CPA) and epsilon (ETX). Enterotoxemia occurs when epsilon prototoxin (pETX) is produced in the intestine and is activated by one or more proteases before being absorbed into the general circulation. Traditionally, it was believed that neonatal animals were not susceptible to type D enterotoxemia due to the trypsin-inhibitory action of colostrum in the intestinal tract and the lack of protease activation of

Clostridium perfringens type D causes enterotoxemia in sheep and goats. The disease is mediated by epsilon toxin (ETX), which affects the cerebrovascular endothelium, increasing vascular permeability and leading to cerebral edema. In the present study, we compared the distribution and severity of the cerebrovascular changes induced in lambs by C. perfringens type D strain CN1020, its isogenic etx null mutant, and the ETX-producing complemented mutant. We also applied histochemical and immunohistochemical markers to further characterize the brain lesions induced by ETX. Both ETX-producing strai

Clostridium perfringens is classified into different types, of which Type D, due to its production of epsilon toxin, can cause enterotoxemia, a fatal disease in ruminants, particularly goats. This study investigated the prevalence, isolation, molecular identification, antibiotic resistance and therapeutic response of C. perfringens Type D in goats from the Peshawar and Charsadda districts of Pakistan. A total of 5000 faecal samples (2500 from each district) were collected from clinically suspected goats and cultured on selective media. Molecular confirmation was performed by PCR targeting the

Germ-free guinea pigs died with signs and lesions of acute enterotoxemia after oral ingestion of C. perfringens types B, C, D, and E. The signs and lesions observed resembled those seen in acute enterotoxemia of sheep and cattle and the naturally occurring disease seen in "ex-germ-free" guinea pigs. C. perfringens type A was found to be innocuous. Conventional guinea pigs did not become ill after ingestion of any of the five toxigenic types.

Clostridium perfringens type D infects ruminants and causes the enterotoxemia disease by ε-toxin. A mutated ε-toxin gene lacking toxicity was designed, synthesized, and cloned into the pT1NX vector and electroporated into Lactobacillus casei competent cells to yield LC-pT1NX-ε recombinant strain. BALB/c mice, immunized orally with this strain, highly induced mucosal, humoral, and cell-mediated immune responses and developed a protection against 200 MLD/ml of the activated ε-toxin. This study showed that the LC-pT1NX-ε could be a promising vaccine candidate against the enterotoxemia disease.

Bacterial meningitis most commonly occurs in young calves secondary to septicemia. Clinical signs of hyperirritability are usually seen. Meningitis can be confirmed by cerebrospinal fluid analysis and culture or by necropsy. Intoxications by the exotoxins of Clostridium perfringens types C and D, C. botulinum, and C. tetani are difficult to confirm. The clinical signs of these intoxications vary from flaccid paralysis (botulism) to muscular rigidity (tetanus). Treatment of affected cattle has been unrewarding in botulism and enterotoxemia, whereas early aggressive treatment of tetanus cases ca

Focal symmetrical encephalomalacia (FSE) is the most prominent lesion seen in the chronic form of enterotoxemia caused by Clostridium perfringens type D in sheep. However, this lesion has not been reported in goats. The current paper reports a case of FSE in a goat from the state of Paraíba in the Brazilian semiarid region. As reported by the farmer, 30, 4-48-month-old animals from a flock of 150 goats died after showing nervous signs, including blindness and recumbence, for periods varying between 1 and 14 days. The flock was grazing native pasture supplemented with wheat and corn bran. Addit

Clostridium perfringens vegetative cells cause both histotoxic infections (e.g., gas gangrene) and diseases originating in the intestines (e.g., hemorrhagic necrotizing enteritis or lethal enterotoxemia). Despite their medical and veterinary importance, the molecular pathogenicity of C. perfringens vegetative cells causing diseases of intestinal origin remains poorly understood. However, C. perfringens beta toxin (CPB) was recently shown to be important when vegetative cells of C. perfringens type C strain CN3685 induce hemorrhagic necrotizing enteritis and lethal enterotoxemia. Additionally,

Clostridium perfringens uses its large arsenal of protein toxins to produce histotoxic, neurologic and intestinal infections in humans and animals. The major toxins involved in diseases are alpha (CPA), beta (CPB), epsilon (ETX), iota (ITX), enterotoxin (CPE), and necrotic B-like (NetB) toxins. CPA is the main virulence factor involved in gas gangrene in humans, whereas its role in animal diseases is limited and controversial. CPB is responsible for necrotizing enteritis and enterotoxemia, mostly in neonatal individuals of many animal species, including humans. ETX is the main toxin involved i

Clostridium perfringens is a gram-positive anaerobic rod that is classified into 5 toxinotypes (A, B, C, D, and E) according to the production of 4 major toxins, namely alpha (CPA), beta (CPB), epsilon (ETX) and iota (ITX). However, this microorganism can produce up to 16 toxins in various combinations, including lethal toxins such as perfringolysin O (PFO), enterotoxin (CPE), and beta2 toxin (CPB2). Most diseases caused by this microorganism are mediated by one or more of these toxins. The role of CPA in intestinal disease of mammals is controversial and poorly documented, but there is no dou

Disorders of the intestinal vasculature in animals include arteriovenous malformations, telangiectasia (ectasia, angiodysplasia), hamartomas, and neoplasia (hemangiomas, hemangiosarcomas). Two, 2-d-old, female Nigerian dwarf goat kids born to different dams on the same farm had severe and progressive abdominal distension and were euthanized. The autopsies revealed segmental circumferential dark-red areas that caused focal constriction of the mid-jejunum. The constriction led to obstruction of the intestinal lumen and proximal dilation of the jejunum and duodenum in both cases. Clostridium perf

Data related to rates of disease and their associated costs were collected for 12 months from 2 sheep feedlots in northern Colorado. There was an apparent seasonal occurrence of many of the diseases. Pneumonia, enterotoxemia, acidosis, and transport tetany accounted for most of the diseases seen in these feedlots and were responsible for most of the economic losses. There was a large difference in the incidence of diseases between the 2 feedlots and in the expenditures for disease prevention between the 2 feedlots.

In the goblet cells in the caecum of control rabbits only sialo- and sulphomucins were produced and the same glycoproteins were in the brush border of the surface epithelium. By enterotoxaemia there was a mixture of both acid and neutral glycids or mixed ones in some goblet cells. In lumina of the crypts and on surface of the mucosa, the neutral mucins were predominantly present. By enterotoxaemia binding of soybean agglutinin (SBA) and Ricinus communis agglutinin I (RCA I) in the brush border and in the Golgi apparatus was diminished or abolished. Enterotoxaemia of sheep and goats occurs worldwide, but the condition in goats is poorly understood. The disease in goats is mostly caused by Clostridium perfringens type D, although the role of the toxins of this microorganism in the pathogenesis of the disease is not fully understood. The disease occurs in three forms, peracute, acute and chronic, the cardinal clinical sign of the acute and chronic forms being diarrhoea. The main biochemical alterations are hyperglycaemia and glycosuria, while at necropsy the disease is often characterized by haemorrhagic colitis. The typical histological ch

Pyoderma is a purulent skin infection usually caused by bacteria and can be divided into primary and secondary categories based on histology. In the present study, an 18-month-old female mixed breed sheep was examined for pyoderma at the injection site of the enterotoxemia vaccine. After routine bacteriology and histopathology procedures, secondary pyoderma caused by Proteus mirabilis was diagnosed. The bacterium analysed using genome sequencing and new strain called AJJ 2021 was diagnosed. This is the first report of pyoderma caused by Proteus mirabilis in sheep.
