Deubiquitinating Enzyme USP2 Alleviates Muscle Atrophy by Stabilizing PPAR-γ.

Insulin resistance, a hallmark of type 2 diabetes, accelerates muscle breakdown and impairs energy metabolism. However, the role of ubiquitin specific peptidase 2 (USP2), a key regulator of insulin resistance, in sarcopenia remains unclear. Peroxisome proliferator-activated receptor γ (PPAR-γ) plays a critical role in regulating muscle atrophy. The role of deubiquitinase USP2 in mitigating muscle atrophy was investigated. Our findings revealed reduced USP2 expression in skeletal muscles of patients with type 2 diabetes. In mouse models of diabetes- and dexamethasone (DEX)-induced muscle atroph

Effect of Storage Temperature and Time on Stability of Liver Enzymes in Blood Serum.

BACKGROUND: It is increasingly common to collect and store specimens for future unspecified research. However, the effects of prolonged storage on the stability and quality of analytes in serum have not been well investigated. We aimed to determine whether the stability of liver enzymes extracted from frozen bio-samples stored at the baseline is affected by storage conditions. METHODS: A total of four liver enzymes in the sera of 400 patients were examined following storage. After deter-mining the baseline measurements, the serum of each patient was aliquoted and stored at -70°C for three and

pH Controlled Activation and Stabilization of Enzymes Using Responsive Polymer-Bioconjugates.

Stimuli-responsive bioconjugates are developed to control enzymatic activity and stability using a pH-responsive polymer based on N,N-dimethyl aminopropylacrylamide. Prior work has shown that conjugating N-isopropylacrylamide (NIPAm) polymer to theCandida antarctica lipase B (CalB) enzyme has an inhibitory effect. Due to the similar hydrophobicity to the substrate of CalB, NIPAm polymer may block the enzyme's catalytic site, thereby inhibiting the enzyme's activity. This study shows that conjugating pDMAPA DP40-b-NIPAm DP10 to CalB has a pH-responsive effect on the catalytic property of CalB,

The deubiquitinating enzyme ATXN3 promotes hepatocellular carcinoma progression by stabilizing TAZ.

Hepatocellular carcinoma (HCC) was a primary cause of cancer-related morbidity and mortality in China. ATXN3 was a deubiquitinase (DUB) associated with spinocerebellar ataxia, widely expressed in the cytoplasm and nucleus of cells in the central nervous system and other tissues. The crucial role of the Hippo pathway in tumorigenesis has been established, among which TAZ served as one of the key molecules. However, the mechanisms underlying the deubiquitinase and TAZ in HCC remained largely unknown. In the present study, we explored that ATXN3 was overexpressed in HCC. ATXN3 promoted the prolif

The combinatorial effects of osmolytes and alcohols on the stability of pyrazinamidase: Methanol affects the enzyme stability through hydrophobic interactions and hydrogen bonds.

Inside the cells, proteins are surrounded by mixtures of different osmolytes. However, our current understanding of the combinatorial effects of such mixtures on the stability of proteins remains elusive. In the present study, the stability and structure of recombinant pyrazinamidase (PZase) from Mycobacterium tuberculosis were analyzed in the presence of stabilizing osmolytes (sorbitol, sucrose and glycerol) and alcohols (methanol, ethanol, isopropanol and n-propanol). The far-UV and near-UV circular dichroism (CD), intrinsic fluorescence and thermostability results indicated that methanol, u

Disease-linked mutations in the phosphatidylcholine regulatory enzyme CCTα impair enzymatic activity and fold stability.

CTP:phosphocholine cytidylyltransferase (CCT) is the key regulatory enzyme in phosphatidylcholine (PC) synthesis and is activated by binding to PC-deficient membranes. Mutations in the gene encoding CCTα (PCYT1A) cause three distinct pathologies in humans: lipodystrophy, spondylometaphyseal dysplasia with cone-rod dystrophy (SMD-CRD), and isolated retinal dystrophy. Previous analyses showed that for some disease-linked PCYT1A variants steady state levels of CCTα and PC synthesis were reduced in patient fibroblasts, but other variants impaired PC synthesis with little effect on CCT levels. The deubiquitinating enzyme UCHL3 promotes bladder cancer progression and glycolysis through enhancing PKM2 protein stability.

Pyruvate kinase M2 (PKM2) is a critical rate-limiting enzyme in glycolysis and is frequently upregulated in bladder cancer (BCa), contributing to the Warburg effect and tumor progression. However, the mechanism supporting abnormal PKM2 stabilization via deubiquitination in BCa remains to be characterized. Here, we identified ubiquitin C-terminal hydrolase L3 (UCHL3) as a bona fide deubiquitylase of PKM2 in BCa. UCHL3 stabilized PKM2 in a deubiquitylation activity-dependent manner. UCHL3 depletion significantly decreased PKM2 protein levels, suppressed glycolytic activity, and inhibited BCa cel

Investigating thermal stability based on the structural changes of lactase enzyme by several orthogonal methods.

Thermal stability of lactase (β-galactosidase) enzyme has been studied by a variety of physico-chemical methods. β-galactosidase is the main active ingredient of medications for lactose intolerance. It is typically produced industrially by the Aspergillus oryzae filamentous fungus. Lactase was used as a model to help understand thermal stability of enzyme-type biopharmaceuticals. Enzyme activity (hydrolyzation of lactose) of β-galactosidase was determined after storing the solid enzyme substance at various temperatures. For a better understanding of the relationship between structure and activ

Stabilization of Enzymes via Covalent Tethering to Appropriately Tuned Random Copolymer Brushes.

Immobilizing enzymes can often cause their inactivation due to enzyme-support interactions and changes in the enzyme's microenvironment. As such, the design of novel supports remains largely empirical, and a rational approach may avoid time-consuming and tedious experiments. This chapter describes a method for stabilizing enzymes via immobilization on heterogeneous polymer brush supports. By varying the composition of the brush, the enzyme-brush interface may be finely tuned to prevent enzyme unfolding and rescue denatured enzymes via a chaperone-like mechanism. The design of polymer brush sup

Deubiquitinating enzyme PSMD7 promotes bladder cancer development: Involvement of RAB1A stabilization.

BACKGROUND: Proteasome 26S subunit, non-ATPase 7 (PSMD7) is a deubiquitinating enzyme that is involved in the stability of ubiquitinated proteins and participates in the development of multiple types of cancer. The roles of PSMD7 and its potential mechanisms in bladder cancer (BC) remain elusive. METHODS: In this study, we identified that PSMD7 was overexpressed in BC tissues based on gene expression omnibus (GEO) database and TNMplot web. To investigate the functional role of PSMD7, two BC cell lines, T24 and 5637, were selected. The cells were transfected with vectors containing short hairp

Capillary-Mediated Vitrification: A Novel Approach for Improving Thermal Stability of Enzymes and Proteins.

Capillary-mediated vitrification (CMV) is a novel method for stabilizing biological molecules and complexes. CMV leverages capillary evaporation to enable rapid desiccation of aqueous solutions while avoiding both freezing and boiling. In the CMV process, an aqueous solution containing the biological material of interest and common excipients is applied to a solid, porous support, referred to as the scaffold, and desiccated under vacuum. The pores within the scaffold accelerate drying by increasing surface area while preventing boiling through the interaction of the vapor pressure, capillary f

Enzyme replacement therapy stabilized white matter lesion progression in Fabry disease.

BACKGROUND: The central nervous system manifestations in Fabry disease (FD) include progressive white matter lesions (WMLs) and stroke. Due to progressive microvascular involvement, men and women with FD over 35 years of age develop WMLs. Moreover, the prevalence of stroke has been estimated to be 12 times higher in FD compared with the general population. Enzyme replacement therapy (ERT) is available and has shown beneficial effects on renal, cardiac, and peripheral nerve function in FD, but the ERT effect on the progression of WMLs, or the reduction in cerebrovascular events, remains unknown

Metal sulfide nanoparticles synthesized via enzyme treatment of biopolymer stabilized nanosuspensions.

Nanoparticles of CuS, Cu(x)S, Ag(2)S and CdS were successfully prepared using a novel general and green synthetic process to give dextran biopolymer stabilised metal sulfifde nanosuspensions. Following preparation, dextranase enzyme was used to remove the bulk of the bound dextran to give pure stable metal sulfide nanocrystals for application in for example aspects of medicine, photonics and solar cells. Particles of good homogeneity were obtained and the CuS nanoparticle size was controlled to 9-27 nm by adjusting the reaction conditions. Cu(2)S nanoparticles were 14 nm, Ag(2)S nanoparticles

Synthesis of (3S,4S,5S)-trihydroxylpiperidine derivatives as enzyme stabilizers to improve therapeutic enzyme activity in Fabry patient cell lines.

A series of 3S,4S,5S-trihydroxylated piperidines bearing structural diversity at C-2 or C-6 positions has been synthesized and tested to determine their ability to stabilize the activity of recombinant human α-Galactosidase A (rh-α-Gal A). Hit molecules were identified by rapid inhibitory activity screening, and then further investigated for their ability to protect this enzyme from thermo-induced denaturation and enhance its activity in Fabry patient cell lines. Our study resulted in the identification of a new class of small molecules as enzyme stabilizers for the potential treatment of Fabr

Hierarchical enzyme immobilization: encapsulating mesoporous polymer nanospheres in covalent organic frameworks for enhanced reusability and stability.

Enzymes, as efficient and specific natural catalysts, are limited in industrial applications by their instability and non-recyclability under extreme conditions. Combining enzymes with solid supports can address separation issues and enhance stability. This study constructed an enzyme nanoreactor by co-loading glucose oxidase (GOx) and horseradish peroxidase (HRP) into mesoporous melamine-formaldehyde (MF) resin microspheres (denoted as Ps) and covalent organic frameworks (COFs). Specifically, enzymes were immobilized on mesoporous MF resin microspheres via electrostatic adsorption, followed b

Defining the therapeutic corridor of stability in enzyme replacement therapy for Pompe disease: a position statement.

BACKGROUND: Pompe disease, also known as glycogen storage disease type II, is a rare, progressive lysosomal storage disorder caused by pathogenic variants in the GAA gene. Enzyme replacement therapy has transformed the natural history of the disease; however, with three agents now approved - alglucosidase alfa, avalglucosidase alfa, and cipaglucosidase alfa plus miglustat - clinicians lack a consensus quantitative framework for determining when a patient has left the zone of therapeutic stability and requires treatment reassessment, escalation, or switching. METHODS: A structured synthesis wa

Stability of alglucosidase alfa in 0.9% sodium chloride for enzyme replacement therapy in patients with Pompe disease: insights from enzyme activity and cellular uptake measurements.

OBJECTIVES: Enzyme replacement therapy (ERT) with alglucosidase alfa is the cornerstone of treatment for Pompe disease, a rare disorder caused by acid α-glucosidase (GAA) deficiency. Since 2008, home infusions have been provided in the Netherlands. However, the short shelf-life of the ready-to-administer infusions poses challenges for manufacturing and dispensing pharmacies. This study assessed the stability of ready-to-administer alglucosidase alfa infusions by determining enzyme activity and cellular uptake of two concentrations during 11 days of storage. METHODS: Alglucosidase alfa infusio
