Mechanistic Determinants of Oriented Enzyme Immobilization from Martini Simulations.

Although enzyme immobilization is widely used in biotechnology, it still poses challenges as a result of the trade-offs among stability, activity, and surface interactions. Computer simulations offer a promising aid to exploring the effects of different immobilization sites and surface chemistry on both the conformational dynamics and catalytic activity of these biomolecules. Here, we introduce a protocol based on a structure-based version of the Martini coarse-grained simulation model (Go̅Martini) to explore how surface tethering geometry influences the structure and function of immobilized B

Proton Transport as a Missing Design Variable in Immobilized Enzyme Catalysis.

Carbonic anhydrase is among the fastest known enzymes, yet its performance after immobilization remains difficult to rationalize. Conventional immobilization design emphasizes stability, enzyme loading, and mass transfer while largely neglecting proton transport. In soluble carbonic anhydrase, proton transfer can limit catalytic turnover under defined pH and buffer conditions. We propose that immobilization may introduce an additional, condition-dependent resistance at the enzyme-support interface. Surface chemistry and wettability influence interfacial water organization, potentially altering

Decoding MOF-Enzyme-Substrate Compatibility for Universal Biocatalyst Immobilization.

Metal-organic frameworks (MOFs) are widely used for enzyme immobilization due to their tunable pore structures and high stability. However, whether a relatively universal MOF carrier can support diverse enzymatic reactions remains unclear. Herein, we systematically evaluated 120 MOF-enzyme-substrate combinations using ten representative MOFs, three hydrolases (CRL, CALB, and BsEst), and four ester substrates, and then extended the identified carriers to three additional enzymes spanning catalase, oxidase, and protease reactions (GOx, Typ, and BlCAT), examining each with a single substrate. Emerging Role of Enzyme-Immobilized Nanocarriers in Osteoporosis: Advances and Challenges.

Osteoporosis (OP) is a common bone disease that involves low bone mass and high risk of fracture mainly in older men and women and perimenopausal years. Although conventional therapies provide good therapeutic effects, they have numerous limitations, including poorly targeted and systemic administration and severe side effects. Recent developments in nanotechnology enabled design of enzyme-immobilized nanocarriers as experimental platforms to enhance the delivery of therapeutic agents to bone tissue. This review pays special attention to the development of these multifunctional systems that ca

[Detection carbamate pesticides residue in cucumber by immobilized acetyicholinesterase enzyme].

A novel method was described for detection carbamate pesticides residue in cucumber by immobilizing acetylcholinesterase enzyme in a micro-screen plate with 96 wells based on GA-BSA-gelatin gels. The concentrations of BSA, GA and gelatin in enzyme immobilization solution were optimized and the best concentrations were 5%, 0.5% and 0.1%, respectively. To analyze the pesticides residue 5g cucumber sample was homogenized with 10 mL acetone then an aliquot of 5mL extract was collected in a 10 mL test tube with a cap, into which 2g sodium chloride and 2mL dichloromethane were added in sequence. Enzyme inhibitor screening by CE with an on-column immobilized enzyme microreactor created by an ionic binding technique.

Enzymes are an important class of drug targets. In the early stage of the drug discovery, the major task is to find out the inhibitors of a given enzyme target in a compound library. Herein we describe a method for screening the enzyme inhibitors in the complex mixtures (such as the natural extracts) by capillary electrophoresis with an on-column immobilized enzyme microreactor. The enzyme molecules are immobilized on the capillary wall via ionic binding with the positively charged coating, which was created by simply flushing the column with a solution of polyelectrolyte hexadimethrine bromid

Investigation of Enzyme Immobilization and Clogging Mechanisms in the Enzymatic Synthesis of Amoxicillin.

This study investigated the blocking mechanism of immobilized penicillin G acylase (PGA) during the enzymatic synthesis of amoxicillin. Laboratory observations revealed that the primary cause of clogging was the crystallization of the substrate and product on the enzyme surface. Adjusting key parameters can significantly reduce clogging and improve catalytic efficiency. Methanol can decrease enzyme activity, but isopropyl alcohol cleaners can effectively remove clogs and protect enzyme activity. These findings provide an experimental foundation for optimizing the PGA immobilization process, wh

Rational design of biocatalysts based on covalent immobilization of acylase enzymes.

Acylases catalyze the hydrolysis of amide bonds. Penicillin G acylase (PGA) is used for the semi-synthesis of penicillins and cephalosporins. Although protein immobilization increases enzyme stability, the design of immobilized systems is difficult and usually it is empirically performed. We describe a novel application of our strategy for the Rational Design of Immobilized Derivatives (RDID) to produce optimized acylase-based immobilized biocatalysts for enzymatic bioconversion. We studied the covalent immobilization of the porcine kidney aminoacylase-1 onto aldehyde-based supports. Predictio

Recent Advances of Macromolecular Hydrogels for Enzyme Immobilization in the Food Products.

Enzymes are one of the main biocatalysts with various applications in the food industry. Stabilization of enzymes on insoluble carriers is important due to the low reuse, low operational stability, and high cost in applications. The immobility and the type of carrier affect the activity of the immobile enzyme. Hydrogels are three-dimensionally cross-linked macromolecular network structures designed from various polymers. Hydrogels can provide a matrix for an immobile enzyme due to their extraordinary properties such as high water absorbing capacity, carrier of bioactive substances and enzymes,

Enzyme immobilization on glass fiber membrane for detection of halogenated compounds.

Enzyme immobilization using inorganic membranes has enticed increased attention as they not only improve enzyme stability, but also furnish user-friendly biodevices that can be tailored to different applications. Herein, we explored the suitability of the glass fiber membrane for enzyme immobilization and its application for halocarbon detection. For this, halohydrin dehalogenase (HheC) and bovine serum albumin were crosslinked and immobilized on a glass fiber membrane without membrane functionalization. Immobilized HheC exhibited higher storage stability than its free counterpart over 60 days

[Enzyme immobilization via silochromes, modified by transition metal salts].

The immobilization of pectoawamorine G 10X, pronase E and P, protease from Bac. mesentericus and rat liver microsomal fraction was performed by sylochromes modified by Ti, Zr and Hf salts. The pectinesterase, caseinolytic, esterase and hydroxylating activity of the immobilized preparations, their stability during storage, thermostability and pH optimum are determined. Of greatest interest for practical application are preparations of pectoawamorine. G 10X and pronase E and P immobilized by HfOCl2.

Positive effects of composite material immobilized enzymes in 2,4,6-trichlorophenol degradation on soil properties and plant growth.

2,4,6-Trichlorophenol (2,4,6-TCP) is recognized as a bio-toxic compound which is widely present in water and soil, and immobilized enzymes technology is widely used to degrade 2,4,6-TCP efficiently. However, previous studies have primarily focused on the degradation capability of immobilized enzymes towards 2,4,6-TCP, while the impacts on soil after degradation remain largely unexplored. In this study, sodium alginate/hydroxyapatite/chitosan microspheres immobilized with enzymes were used for 2,4,6-TCP degradation, and the impacts of degradation on soil properties and plant growth were explore

Enzyme inhibitor screening by capillary electrophoresis with an on-column immobilized enzyme microreactor created by an ionic binding technique.

A novel strategy for screening the enzyme inhibitors from the complex mixtures by capillary electrophoresis with an on-column immobilized enzyme microreactor created by an ionic binding technique is reported. The enzyme microreactor was prepared in two steps: First, the capillary wall was dynamically coated with a polycationic electrolyte hexadimethrine bromide (HDB) by simply flushing the column using the HDB solution. Subsequently, a plug of the enzyme solution was injected and incubated for 5 min to permit the enzyme molecules to immobilize on the positively charged coating via ionic bindin

Coated bacterial enzymes: a one-step approach for enzymatic purification and immobilization.

The purification of industrial enzymes typically relies on costly, multi‑step chromatographic protocols. To address this, we developed a novel platform termed Coated Bacterial Enzymes (CBEs), which enables one‑step purification and immobilization of recombinant proteins fused to the SlpA cell wall binding domain. As a proof of concept, we used a β‑galactosidase from Bifidobacterium bifidum of dairy relevance. The chimeric enzyme BbgII‑SlpA was expressed in Escherichia coli and captured from crude lysate onto glutaraldehyde‑inactivated Bacillus subtilis cells via SlpA domain. Binding was charac

Expanding the high-pH range of the sucrose synthase reaction by enzyme immobilization.

The glycosylation of an alcohol group from a sugar nucleotide substrate involves proton release, so the reaction is favored thermodynamically at high pH. Here, we explored expansion of the alkaline pH range of sucrose synthase (SuSy; EC 2.4.1.13) to facilitate enzymatic glycosylation from uridine 5'-diphosphate (UDP)-glucose. The apparent equilibrium constant of the SuSy reaction (UDP-glucose + fructose ↔ sucrose + UDP) at 30 °C increases by ∼4 orders of magnitude as the pH is raised from 5.5 to 9.0. However, the SuSy in solution loses ≥80 % of its maximum productivity at pH ∼7 when alkaline r

Changes in ficin specificity by different substrate proteins promoted by enzyme immobilization.

Ficin extract has been immobilized using different supports: glyoxyl and Aspartic/1,6 hexamethylenediamine (Asp/HA) agarose beads. The latter was later submitted to glutaraldehyde modification to get covalent immobilization. The activities of these 3 kinds of biocatalysts were compared utilizing 4 different substrates, casein, hemoglobin and bovine serum albumin and benzoyl-arginine-p-nitroanilide at pH 7 and 5. Using glyoxyl-agarose, the effect of enzyme-support reaction time on the activity versus the four substrates at both pH values was studied. Reaction time has been shown to distort the

SnO2 hollow nanotubes: a novel and efficient support matrix for enzyme immobilization.

A major challenge in the industrial use of enzymes is maintaining their stability at elevated temperatures and in harsh organic solvents. In order to address this issue, we investigated the use of nanotubes as a support material for the immobilization and stabilization of enzymes in this work. SnO2 hollow nanotubes with a high surface area were synthesized by electrospinning the SnCl2 precursor and polyvinylpyrrolidone (dissolved in dimethyl formamide and ethanol). The electrospun product was used for the covalent immobilization of enzymes such as lipase, horseradish peroxidase, and glucose ox

Angiotensin converting enzyme immobilized on magnetic beads as a tool for ligand fishing.

Angiotensin converting enzyme (ACE) presents an important role in blood pressure regulation, since that converts angiotensin I to the vasoconstrictor angiotensin II. Some commercially available ACE inhibitors are captopril, lisinopril and enalapril; due to their side effects, naturally occurring inhibitors have been prospected. In order to endorse this research field we have developed a new tool for ACE ligand screening. To this end, ACE was extracted from bovine lung, purified and chemically immobilized in modified ferrite magnetic beads (ACE-MBs). The ACE-MBs have shown a Michaelian kinetic

A biocatalytic reticular framework via enzyme immobilization for environmental pollutant monitoring.

The growing prevalence of anthropogenic pollutants demands the advancement of environmental monitoring technologies. Biocatalytic sensing, leveraging enzymatic specificity and efficiency, offers a promising alternative, yet the structural fragility and instability of free enzymes limit its practical application. A transformative strategy involves immobilizing enzymes within engineered porous frameworks, such as metal-organic frameworks (MOFs) and covalent organic frameworks (COFs). These crystalline materials offer ultrahigh surface areas, tunable pore structures, and versatile surface chemist
