Bovine ephemeral fever virus (BEFV) is a member of the genus Ephemerovirus in the family Rhabdoviridae. It is an arthropod-borne virus transmitted by many species of midges and mosquitoes. It can cause severe economic consequences due to losses in milk production and the general condition of cattle and water buffalo. BEF occurs in some tropical, subtropical and warm temperate regions of Africa, Australia, the Middle East and Asia with seasonal outbreaks, but its possible spread to other areas (e.g. Europe) cannot be excluded. Therefore, using and developing rapid diagnostic methods with optima

The purpose of this study was to investigate the antibody response to the bovine ephemeral fever virus (BEFV) vaccine in Japanese Black calves. Twenty-eight Japanese Black calves, which were raised on an ordinal farm, were divided into two groups. Fifteen calves received the inactivated BEFV vaccine at 12 and 16 weeks of age (vaccination group), and 13 calves did not receive the vaccine (non-vaccination group). Blood samples were obtained at 0, 4, 8, 12, 16, 20, 24, 28, and 32 weeks of age. As the results, in the vaccination group, the antibody titers at 16, 20, 24, 28, and 32 weeks of age wer

The epitope-G1 gene, cloned from the pMD-G plasmid including G protein gene of bovine ephemeral fever virus (BEFV), was subcloned into expression vector pGEX-4T-1 to construct pGEX-G1 recombinant plasmid successfully. The pGEX-G1 was transformed into E. coli BL21(DE3) to be induced with IPTG. The optimal expression conditions for G1 gene were obtained, which included reaction temperature 16 degrees C, induction time 18h and IPTG concentration 0.1 mmol/L. The soluble target protein was purified with Glutathione Sepharose TM(4B) and the purity reached 80%. The inclusion body washed with 2% deoxy

The aim of this study was to investigate the immunogenicity of a plasmid deoxyribonucleic acid (DNA) vaccine encoding the G1 epitope of bovine ephemeral fever virus (BEFV) G glycoprotein in mice. A plasmid DNA carrying the G1 gene was constructed and designated as pcDNA3.1-G1. The expression of the target gene was confirmed in human embryonic kidney 293 (HEK 293) cells transfected with pcDNA3.1-G1 by indirect immunofluorescent staining. Immunisation experiments were intramuscularly carried out by vaccinating 6-week-old female mice in four groups, including the pcDNA3.1-G1 construct, pcDNA3.1 (

Water buffaloes in northern Australia occupy tropical wetlands where conditions favour the proliferation of arthropod vectors and the transmission of vector-borne livestock diseases. However, their role in maintaining economically important arboviruses such as Akabane virus (AKAV), bluetongue virus (BTV), and bovine ephemeral fever virus (BEFV) remains poorly understood. These three viruses cause significant production losses in cattle and pose ongoing surveillance challenges in remote areas. To assess exposure to these viruses, a convenience sample of feral water buffaloes from the Northern T

BACKGROUND: Bovine ephemeral fever (BEF) is an arthropod-borne viral disease that infects cattle and buffaloes, leading to widespread illness and notable economic losses in warm and subtropical areas, particularly in Egypt. Although the disease is self-limiting, its clinical manifestations are accompanied by profound immune, inflammatory, and metabolic disturbances. However, integrated studies linking immune response, inflammatory biomarkers, and hemato-biochemical alterations in naturally infected cattle are limited.
AIM: This study aimed to evaluate immune responses, inflammatory biomarkers

Vector-borne arboviruses produce mild to severe symptoms in domestic animals. Bovine ephemeral fever (BEF), Akabane, Aino, and Chuzan virus have been primarily attributed to reproductive disorders or febrile diseases in cattle, and Japanese encephalitis virus (JEV) is mainly associated with reproductive failures in swine. We investigated antibody titers from domestic swine against four bovine arboviruses (BEF, Akabane, Aino, and Chuzan virus) and from cattle against JEV in Korea. While the positive rates for Akabane and BEF were 37.4% and 15.7%, the positive incidence of Chuzan and Aino were r

Bovine ephemeral fever (BEF) is an arthropod-borne viral disease caused by the bovine ephemeral fever virus (BEFV) of the genus Ephemerovirus in the family Rhabdoviridae. BEF affects domestic and wild ruminants, particularly cattle and water buffaloes. BEF outbreaks are seasonal in Africa, Asia, the Middle East, and Australia, aligning with the summer and autumn seasons in temperate zones and during the monsoons in tropical areas. Vectors, like biting midges (Culicoides spp.) and mosquitoes (Anopheles and Culex spp.), facilitate BEFV transmission. The disease presents a spectrum of clinical si

The bovine ephemeral fever virus (BEFV) is an enzootic agent that affects millions of bovines and causes major economic losses. Though the virus is seasonally reported with a very high morbidity rate (80-100%) from African, Australian, and Asiatic continents, it remains a neglected pathogen in many of its endemic areas, with no proper therapeutic drugs or vaccines presently available for treatment. The RNA-dependent RNA polymerase (RdRp) catalyzes the viral RNA synthesis and is an appropriate candidate for antiviral drug developments. We utilized integrated computational tools to build the 3D

BACKGROUND: The potential tissue replication sites and specific cell types that support in vivo virus survival beyond the acute phase of bovine ephemeral fever virus (BEFV) infection have not been fully defined in cattle. To clarify the knowledge gap, tissue specimens were tested after collection from an adult steer necropsied 1 week after acute BEF.
CASE REPORT: Significant necropsy findings included fibrinoproliferative synovitis in the stifle joints and fibrin clot-laden fluid in serous body cavities. Moderate numbers of infiltrating neutrophils were demonstrated in sections of the prefemo

A novel reverse-transcription, loop-mediated isothermal amplification (RT-LAMP) assay for the detection of bovine ephemeral fever virus (BEFV) was developed and evaluated in this study. The RT-LAMP assay exhibited higher sensitivity when compared with conventional reverse-transcription polymerase chain reaction (RT-PCR) and virus isolation methods. The specificity of the assay was determined by digestion of the RT-LAMP products with restriction enzyme and detection of BEFV serogroup rabies virus (RV). Using RT-LAMP, RT-PCR and virus isolation methods, 36 blood samples were tested and the resul

An indirect ELISA for the serological detection of bovine ephemeral fever virus (BEFV) infection in cattle is described in which a glycosylated protein of approximately 25 kDa (including the G1 antigenic site of the virus glycoprotein) expressed in Pichia pastoris GS115 was used as the coating antigen. The optimal concentration of coated antigen was 0.3 microg/well at a serum dilution of 1:40. The optimal positive threshold value of the assay was 1.88, as derived from receiver operating characteristic curve analysis. The test had 100% sensitivity and 96.7% specificity when compared with a micr

Bovine ephemeral fever (BEF), caused by the bovine ephemeral fever virus (BEFV), is associated with an acute febrile infection in cattle and widespread in tropical and subtropical areas, leading to great economic losses to cattle and milk industry. However, no efficacious BEF vaccine is currently available in China. Herein, we generated a recombinant rabies virus (RABV) expressing BEFV glycoprotein (LBNSE-BG), utilizing a reverse genetics system based on the recombinant rabies virus strain LBNSE. It was found that mice immunized with LBNSE-BG produced robust neutralizing antibodies against bot

Bovine ephemeral fever virus (BEFV) is a globally spread arthropod-borne RNA virus that has significant economic impacts on the cattle industry. A live attenuated commercial BEF vaccine, based on the Australian BEFV strain 919, is widely used in Israel and other countries. A previous study has suggested the high effectiveness of this vaccine (ULTRAVAC BEF VACCINE™ from Zoetis®), but anecdotal reports of high BEF morbidity among vaccinated dairy herds in Israel casted doubt on these findings. To resolve this uncertainty, a randomized controlled field vaccine effectiveness study was conducted in

The prevalence of antibody to the viruses of bovine virus diarrhoea (BVD), bovine herpes virus typel (BHV1), rift valley fever (RVF), bovine ephemeral fever (BEF) and bluetongue (BT) and to Leptospira sp. was determined in wildlife populations in Zimbabwe. Evidence of infection with BVD virus was found in 14 of the 16 species examined but was greatest in eland Taurotragus oryx, nyala Tragelaphus angasi and bushbuck Tragelaphus scriptus. Persistent infection with BVD virus was found in 1 of 303 antibody-free eland but not in the smaller sample of 102 antibody-free buffalo Syncerus caffer. Antib

Bovine ephemeral fever is a vector-borne disease of ruminants that occurs in tropical and sub-tropical regions of Africa, Asia and Australia. The disease is caused by a rhabdovirus, bovine ephemeral fever virus (BEFV), which occurs as a single serotype globally. Although several other closely related ephemeroviruses have been isolated from cattle and/or arthropods, only kotonkan virus from Nigeria and (tentatively) Mavingoni virus from Mayotte Island in the Indian Ocean have been previously associated with febrile disease. Here, we report the isolation of a novel virus (Hayes Yard virus; HYV)

Bovine Ephemeral Fever (BEF) is an arthropod-borne virus (arbovirus) that presents a significant challenge to the cattle industry due to its economic impact, primarily through the loss of milk production in dairy cows. Vaccination is the predominant strategy for managing the disease. We recently showed a vaccine effectiveness (VE) of 60 % of a vaccine based on the Australian 919 BEFV isolate, with a natural challenge occurring shortly after the administration of the second dose of the vaccine. Still, there is a lack of data regarding the duration of protective immunity after vaccination and it

Bovine ephemeral fever (BEF) is an economically important vector-borne viral disease of cattle and water buffalo in Africa, Australia and parts of Asia. The control of BEF is centred around vaccination, and therefore accurate, early identification of disease outbreaks are key to minimize its economic and welfare impact. In Africa, control programs are hampered by limited diagnostic capabilities and poor infrastructure for rapid transportation of diagnostic specimens. The primary objective of this study was to estimate the sensitivity (Se) and specificity (Sp) of four tests, namely clinical exa

Bovine ephemeral fever (BEF) is characterized by high fever, nasal and eye discharge, excessive salivation, muscle weakness, yield losses and or with high morbidity and low mortality. The first epidemic of BEFV in Turkey occurred in 1985 since then epidemics were reported every 2-4 years in the south and southeastern regions of Turkey. Since the first detection of the virus in Turkey, the BEF virus was reported from other parts of the country except the Aegean Region. In November 2020, the possible outbreaks of BEF were reported from two different locations of the Aegean Region in Turkey. In t

A novel ephemeral fever rhabdovirus and a CHeRI orbivirus of a previously unidentified genetic lineage were isolated in mosquito cell line C6/36 cells as co-infecting agents from the spleen tissue of a dead farmed white-tailed deer (WTD; Odocoileus virginianus) in Florida. We designated the ephemeral fever rhabdovirus as Hardee County ephemerovirus 1, strain CHeRI ephemerovirus 1. The genetic sequences of the CHeRI orbivirus isolated in this work differ significantly from those of three previously described CHeRI orbivirus lineages. We designated this new virus as CHeRI orbivirus 4, strain CHe
