An indirect ELISA for the serological detection of bovine ephemeral fever virus (BEFV) infection in cattle is described in which a glycosylated protein of approximately 25 kDa (including the G1 antigenic site of the virus glycoprotein) expressed in Pichia pastoris GS115 was used as the coating antigen. The optimal concentration of coated antigen was 0.3 microg/well at a serum dilution of 1:40. The optimal positive threshold value of the assay was 1.88, as derived from receiver operating characteristic curve analysis. The test had 100% sensitivity and 96.7% specificity when compared with a micr

A novel reverse-transcription, loop-mediated isothermal amplification (RT-LAMP) assay for the detection of bovine ephemeral fever virus (BEFV) was developed and evaluated in this study. The RT-LAMP assay exhibited higher sensitivity when compared with conventional reverse-transcription polymerase chain reaction (RT-PCR) and virus isolation methods. The specificity of the assay was determined by digestion of the RT-LAMP products with restriction enzyme and detection of BEFV serogroup rabies virus (RV). Using RT-LAMP, RT-PCR and virus isolation methods, 36 blood samples were tested and the resul

Bovine ephemeral fever is a vector-borne disease of ruminants that occurs in tropical and sub-tropical regions of Africa, Asia and Australia. The disease is caused by a rhabdovirus, bovine ephemeral fever virus (BEFV), which occurs as a single serotype globally. Although several other closely related ephemeroviruses have been isolated from cattle and/or arthropods, only kotonkan virus from Nigeria and (tentatively) Mavingoni virus from Mayotte Island in the Indian Ocean have been previously associated with febrile disease. Here, we report the isolation of a novel virus (Hayes Yard virus; HYV)

Bovine ephemeral fever virus (BEFV) is a globally spread arthropod-borne RNA virus that has significant economic impacts on the cattle industry. A live attenuated commercial BEF vaccine, based on the Australian BEFV strain 919, is widely used in Israel and other countries. A previous study has suggested the high effectiveness of this vaccine (ULTRAVAC BEF VACCINE™ from Zoetis®), but anecdotal reports of high BEF morbidity among vaccinated dairy herds in Israel casted doubt on these findings. To resolve this uncertainty, a randomized controlled field vaccine effectiveness study was conducted in

Bovine Ephemeral Fever (BEF) is an arthropod-borne virus (arbovirus) that presents a significant challenge to the cattle industry due to its economic impact, primarily through the loss of milk production in dairy cows. Vaccination is the predominant strategy for managing the disease. We recently showed a vaccine effectiveness (VE) of 60 % of a vaccine based on the Australian 919 BEFV isolate, with a natural challenge occurring shortly after the administration of the second dose of the vaccine. Still, there is a lack of data regarding the duration of protective immunity after vaccination and it

Bovine ephemeral fever (BEF) is an economically important vector-borne viral disease of cattle and water buffalo in Africa, Australia and parts of Asia. The control of BEF is centred around vaccination, and therefore accurate, early identification of disease outbreaks are key to minimize its economic and welfare impact. In Africa, control programs are hampered by limited diagnostic capabilities and poor infrastructure for rapid transportation of diagnostic specimens. The primary objective of this study was to estimate the sensitivity (Se) and specificity (Sp) of four tests, namely clinical exa

Bovine ephemeral fever (BEF) is an arthropod-borne viral disease caused by the bovine ephemeral fever virus (BEFV) of the genus Ephemerovirus in the family Rhabdoviridae. BEF affects domestic and wild ruminants, particularly cattle and water buffaloes. BEF outbreaks are seasonal in Africa, Asia, the Middle East, and Australia, aligning with the summer and autumn seasons in temperate zones and during the monsoons in tropical areas. Vectors, like biting midges (Culicoides spp.) and mosquitoes (Anopheles and Culex spp.), facilitate BEFV transmission. The disease presents a spectrum of clinical si

Bovine herpesvirus type 1 (BHV-1) causes acute febrile respiratory diseases (infectious bovine rhinotracheitis, IBR), decreased milk production, weight loss and abortion. Bovine ephemeral fever virus (BEFV) causes acute febrile respiratory disease, with pulmonary emphysema and pulmonary edema as the main signs. These viruses infect domesticated herds and lead to significant economic losses. In our previous study, an inactivated BHV-1 and BEFV bivalent vaccine was formulated with water-in-oil-in-water adjuvant, and vaccine efficacy was evaluated in guinea pigs. In this study, we evaluated the e

In September and October 2015, suspected cases of bovine ephemeral fever (BEF) were reported in the mainland region of Kagoshima Prefecture and on Tanegashima Island. The genome of the BEF virus (BEFV) was detected in the diseased cows and the cows that had recovered. The serum obtained from the affected cows contained high titers of BEFV-neutralizing antibody. In total, 18 affected cows were demonstrated to be infected with BEFV during the outbreak. Our findings showed evidence that BEF occurred in mainland Japan after a 23-year absence. Phylogenetic analysis based on the surface glycoprotein

One of the major factors hindering efficient livestock production is the presence of high-impact infectious animal diseases, such as foot and mouth disease (FMD), lumpy skin disease (LSD), and bovine ephemeral fever (BEF), which are notable viral infections affecting cattle in Egypt, leading to significant economic losses. FMD is caused by the foot and mouth disease virus (FMDV) of the genus Aphthovirus in the Picornaviridae family. LSD is caused by lumpy skin disease virus (LSDV) of Capripox genus within the Poxviridae family, subfamily Chordopoxvirinae. BEF is caused by bovine ephemeral feve

Virus-host interactions are crucial for regulating viral replication. While some interactions can suppress viral spread, others may promote it, and their underlying mechanisms remain poorly understood. Here, we report that infection with bovine herpesvirus 1 (BoHV-1) or bovine ephemeral fever virus (BEFV) upregulates the transcriptional expression of Cluster of Differentiation 97 (CD97). We identified the transcription factor Krüppel-like factor 4 (KLF4), which is elevated during BoHV-1, BEFV, or vesicular stomatitis virus (VSV) infection, as a key regulator of CD97 induction. Furthermore, CD9

Australian-Indonesian collaboration in veterinary development programs has led to significant advances in the study of arboviruses. This paper reviews the resulting knowledge of arboviral infections of livestock in Indonesia. The first recognized arboviral disease of animals in Indonesia was bovine ephemeral fever. Serology indicates that the virus is widespread, as are related rhabdoviruses. Local sheep appear resistant to bluetongue disease, but imported sheep have suffered mortalities. Bluetongue viral serotypes 1, 7, 9, 12, 21 and 23 have been isolated from sentinel cattle; 1, 21 and 23 at

The effect of two anti-inflammatory drugs on the development and persistence of clinical signs in cattle experimentally infected with bovine ephemeral fever (BEF) virus was investigated by their administration, either before or after the commencement of fever. A total of 16 cattle was given phenylbutazone sodium (PBZ). The drug prevented fever and other clinical signs in six cattle when given daily during the incubation period, and at 8-h intervals for 5 days when clinical disease might be expected. When treatment with PBZ was deferred until 2-4 h after the commencement of fever, the rectal te

Lumpy skin disease virus (LSDV) is being developed as a vector for recombinant vaccines against diseases of veterinary importance. A strategy for generating viral thymidine kinase (TK) gene-disrupted recombinants which are stable and homogeneous using the South African Neethling vaccine strain of LSDV as vector has been developed. To assist with the selection process, the Escherichia coli beta-galactosidase (lacZ) visual marker gene was incorporated into the constructs. However, the use of lacZ has certain limitations. An improved strategy was then devised substituting lacZ with the enhanced g

The South African vaccine strain of lumpy skin disease virus (type SA-Neethling) is currently being developed as a vector for recombinant vaccines of economically important livestock diseases throughout Africa. In this study, the feasibility of using the viral thymidine kinase gene as the site of insertion was investigated and recombinant viruses were evaluated in animal trials. Two separate recombinants were generated and selected for homogeneity expressing either the structural glycoprotein gene of bovine ephemeral fever virus (BEFV) or the two structural glycoprotein genes of Rift Valley fe

The water buffalo (Bubalus bubalis) has great adaptability to rustic environments and more variable conditions than cattle, who generally share the habitat. Diseases carried by buffaloes are relatively unknown and ignored and could be transmissible; an imbalance occurs between pathogens, environment, and susceptible hosts, generating a severe animal health problem. Also relevant is the effect of climate change on the populations of vectors that transmit viral diseases. The discovery of new virus variants that can pass from bovine (Bos) to buffalo or vice versa or to humans has highlighted the

Bovine ephemeral fever virus (BEFV) is an economic arthropod-borne virus distributed in Africa, Asia, and Australia. Based on the sequence of the gene encoding the surface glycoprotein G, the viral antigenic determinant, BEFV has been phylogenetically classified into three clusters, including Australia, East Asia, and the Middle East. Here, we provide evidence for antigenic variations among the BEFV isolates in Iran during the period of 2012 to 2013 and also the exotic YHL strain, which are all classified into the East Asian cluster of the virus. For this propose, the entire length of the G ge

Rift Valley fever virus (RVFV) is one of the important emerging viral diseases of serious impact in public health and animal hygiene both in human and animal industries. In this study, we developed a monoclonal antibody-based competitive ELISA for the detection of antibodies to RVFV in goats and cattle. The recombinant N protein of RVFV was expressed in E. coli with a six-histidine tag, and the purified N protein was used for detecting antigen with a competitive monoclonal antibody against RVFV antibodies. The competitive ELISA (C-ELISA) could detect antibodies at 9-11 days after inoculation i

Bovine ephemeral fever (BEF) is characterized by high fever, nasal and eye discharge, excessive salivation, muscle weakness, yield losses and or with high morbidity and low mortality. The first epidemic of BEFV in Turkey occurred in 1985 since then epidemics were reported every 2-4 years in the south and southeastern regions of Turkey. Since the first detection of the virus in Turkey, the BEF virus was reported from other parts of the country except the Aegean Region. In November 2020, the possible outbreaks of BEF were reported from two different locations of the Aegean Region in Turkey. In t
