Transgenerational Epigenetic Inheritance of Traumatic Experience in Mammals.

In recent years, we have seen an increasing amount of evidence pointing to the existence of a non-genetic heredity of the effects of events such as separation from parents, threat to life, or other traumatising experiences such as famine. This heredity is often mediated by epigenetic regulations of gene expression and may be transferred even across several generations. In this review, we focus on studies which involve transgenerational epigenetic inheritance (TEI), with a short detour to intergenerational studies focused on the inheritance of trauma or stressful experiences. The reviewed studi

The "evolutionary field" hypothesis. Non-Mendelian transgenerational inheritance mediates diversification and evolution.

Epigenetics is increasingly regarded as a potential contributing factor to evolution. Building on apparently unrelated results, here I propose that RNA-containing nanovesicles, predominantly small regulatory RNAs, are released from somatic tissues in the bloodstream, cross the Weismann barrier, reach the epididymis, and are eventually taken up by spermatozoa; henceforth the information is delivered to oocytes at fertilization. In the model, a LINE-1-encoded reverse transcriptase activity, present in spermatozoa and early embryos, plays a key role in amplifying and propagating these RNAs as ext

[Genetic and epigenetic aspects of celiac disease].

Genetic background of coeliac disease has been subjects to intensive research since decades. However, only results of HLA phenotyping have been taken over to routine clinical practice. Meanwhile, data on the role of epigenetical factors in the manifestation of diseases have been emerging. In coeliac disease, there are several questions both in the fields of genetics and epigenetics yet to be answered. In this review, a cross section of current knowledge on these issues is presented with special interest regarding the future clinical applications.

One-carbon metabolism and Alzheimer's disease: is it all a methylation matter?

The sporadic form of Alzheimer disease, late onset Alzheimer's disease (LOAD), is a multifactorial disease; a strong link between nutritional and genetic factors with normal aging and dementia is supported by studies on nutrition, metabolism, and neurodegeneration. Specifically, the involvement of homocysteine (HCY) and its dietary determinants (vitamins B6, B12, and folate, besides methionine) in dementia has been a topic of intense investigation. In this Commentary we would like to highlight the role of 1-carbon metabolism in epigenetics and Alzheimer's disease and evidence the co-involvemen

Beyond the variant: hereditary cancer awareness in the multi-omics era.

Hereditary cancer awareness has entered a new phase. For decades, awareness has focused on recognizing familial risk, identifying pathogenic germline variants, and helping families access counselling, surveillance and prevention. Yet in 2026, the main challenge is no longer only whether a variant can be detected, but how inherited risk can be interpreted, communicated and translated into action in the biological context of each tissue and tumor. The recent publication landscape of Hereditas illustrates this shift: cancer genetics is now inseparable from RNA regulation, epigenetics, metabolism,

Genetics Meets Metabolism: Decoding Their Role in Idiopathic Scoliosis.

Adolescent Idiopathic Scoliosis (AIS) is increasingly recognized as a multisystem disorder arising from the interplay between genetic susceptibility, metabolic dysregulation, and impaired mechanobiological signaling. Recent genomic studies highlight defects in ciliary genes, implicating disrupted ciliary structure and signaling in altered spinal growth. Concurrently, metabolic disturbances including pubertal hormones, oxidative stress, and altered IGF-1 activity may exacerbate these genetic vulnerabilities by influencing bone remodeling and extracellular matrix properties. Epigenetic regulator

Genetic and Epigenetic Mechanisms of Psoriasis.

Psoriasis is a disease involving the innate and adaptative components of the immune system, and it is triggered by environmental factors in genetically susceptible individuals. However, its physiopathology is not fully understood yet. Recent technological advances, especially in genome and epigenome-wide studies, have provided a better understanding of the genetic and epigenetic mechanisms to determine the physiopathology of psoriasis and facilitate the development of new drugs. This review intends to summarize the current evidence on genetic and epigenetic mechanisms of psoriasis.

Epigenetics of spondyloarthritis.

Spondyloarthritis (SpA) is a chronic inflammatory disorder resulting from a combination of genetic predisposition and environmental factors. Despite recent advances, a substantial fraction of its genetic basis remains poorly understood. Several mechanisms have been proposed to account for this unexplained heritability, including epigenetics which can play a role at the interface between genetic and environmental susceptibility factors. Epigenetics refers to changes in gene expression that are not encoded in the DNA sequence itself. Such mechanisms may include DNA methylation, histone modificat

Genetic contributors and soluble mediators in prediction of autoimmune comorbidity.

Comorbidities including subclinical atherosclerosis, neuropsychological aberrations and lymphoproliferation represent a major burden among patients with systemic autoimmune diseases; they occur either as a result of intrinsic disease related characteristics including therapeutic interventions or traditional risk factors similar to those observed in general population. Soluble molecules recently shown to contribute to subclinical atherosclerosis in the context of systemic lupus erythematosus (SLE) include among others B-cell activating factor (BAFF), hyperhomocysteinemia, parathormone (PTH) lev

Epigenetic-Genetic Coupling and Understanding the Molecular and Cellular Basis of Lamarckian Inheritance.

This critical and selective review synthesizes the accumulating body of biological evidence supporting a process we term epigenetic-genetic coupling as a mechanistic basis for Lamarckian inheritance of somatically acquired adaptations. We propose that evolutionary processes in mammals and higher vertebrates can involve deaminase-driven, reverse transcriptase-mediated, RNA-templated targeted homologous recombination. We contrast well-established examples of "Soft", reversible epigenetic inheritance with historical and contemporary evidence suggestive of stable, DNA-integrated "Hard" Lamarckian

Biochemical aspects of chromatin-modifying drugs.

Epigenomic dysregulation is associated with several noncommunicable diseases (NCDs). Chromatin-modifying drugs have the ability to re-establish normal epigenetic regulation, thereby fine-tuning the epigenome. The biochemical and pharmacological mechanisms of these drugs, which target histone lysine and arginine methyltransferases, DNA methyltransferases, histone deacetylases and histone acetyltransferases, histone demethylases and bromodomain and extra-terminal motifs, serve as ideal tools for understanding the modes of action of these epigenetic regulators and the resulting biological events

Harnessing the interplay between metal toxicity and DNA methylation in plants.

Research into how plants adapt to stress through genetic and epigenetic mechanisms is a rapidly evolving field, with epigenetic flexibility serving as a critical factor for survival in heavily contaminated, high-metal environments. Cutting-edge technologies, specifically Whole Genome Bisulfite Sequencing (WGBS) and Reduced Representation Bisulfite Sequencing (RRBS), have revolutionized this area by moving beyond broad, genome-wide surveys to pinpoint specific, gene-level alterations induced by metal stress. These high-throughput methods have fundamentally transformed the landscape of environme

Microbiota-driven epigenetic programming of local immunity.

The maintenance of local immunity requires stable tissue adaptation, implying that persistent environmental cues must continuously reinforce niche-specific immune programs. Microbiota-derived metabolites are increasingly recognized as such cues, acting not merely as metabolic byproducts but as epigenetic inputs that regulate histone modifications, DNA and histone methylation, and non-coding RNA networks. Through these epigenetic mechanisms, microbial metabolites reprogram key immune populations by promoting tolerogenic or immunostimulatory states, tuning effector and memory differentiation, an

Holobiontic Intercellular Relationships Between the Oral Cavity and the Rest of the Human Organism: A Narrative Review.

The holobiont represents a fundamental concept in modern biology, defining the organism as a complex unit composed of the host and its symbionts (microbes, viruses) that live together, forming an integrated biological system in which the host and microbes collaborate and influence each other (genetically and metabolically) and evolve as a single entity, rather than the host evolving in isolation. It is recognized that the health and functioning of the host fundamentally depend on its microbiome, consolidating the entire assembly as a unit of evolutionary selection with a shared genome called t

Epigenetic Rewiring and Context-Dependent Therapeutic Vulnerabilities in Myeloproliferative Neoplasms.

Myeloproliferative neoplasms (MPN) are shaped by epigenetic rewiring that extends beyond canonical JAK2V617F-driven signaling. This review argues that context-dependent chromatin states, not merely genetic lesions, determine disease trajectory, fibrotic transformation, and leukemic progression. We synthesize recent evidence indicating that PRC2 deficiency is supported by the strongest MPN-specific evidence for BRD4 dependency, whereas PARP/BCL-2 vulnerabilities associated with TET2/IDH mutations and USP7 dependency associated with ASXL1 remain supported primarily by related myeloid malignancie

Histone modification in psoriasis: Molecular mechanisms and potential therapeutic targets.

Psoriasis is an immune-mediated, inflammatory disease. Genetic and environmental elements are involved in the nosogenesis of this illness. Epigenetic inheritance serves as the connection between genetic and environmental factors. Histone modification, an epigenetic regulatory mechanism, is implicated in the development of numerous diseases. The basic function of histone modification is to regulate cellular functions by modifying gene expression. Modulation of histone modification, such as regulation of enzymes pertinent to histone modification, can be an alternative approach for treating some

Epigenetic modifications and obsessive-compulsive disorder: what do we know?

Obsessive-Compulsive Disorder (OCD) is a chronic, severe disabling neuropsychiatric disorder whose pathophysiology is not yet well defined. Generally, the symptom onset occurs during pre-adult life and affects subjects in different life aspects, including professional and social relationships. Although robust evidence indicates the presence of genetic factors in the etiopathology of OCD, the entirely mechanisms are not totally clarified. Thus, the possible interactions between genes and environmental risk factors mediated by epigenetic mechanisms should be sought. Therefore, we provide a revie
