Repeated-doses and reproductive toxicity studies of the monoterpene 1,8-cineole (eucalyptol) in Wistar rats.

1,8-cineole (eucalyptol) is widely used as an excipient in the pharmaceutical industry and as a food flavoring agent, thus providing significant potential for human exposure to the compound. We investigated the preclinical toxicity and reproductive toxicity of 1,8-cineole (CIN). In the repeated-doses toxicity study for 50 days, CIN (100, 500 or 1000 mg/kg) did not produce any signs of toxicity or deaths, but affected body weight gain during the first week of treatment. The hematological and biochemical profiles did not show significant differences except for increase in the MCV, platelet and u

1,8-Cineole (Eucalyptol), a mosquito feeding and ovipositional repellent from volatile oil ofHemizonia fitchii (Asteraceae).

The mosquito feeding and ovipositional repellency of the major monoterpenoid present in the volatile oil ofHemizonia fitchii (Asteraceae), i.e., 1,8-cineole, was investigated. Although 1,8-cineole did not exhibit any significant mosquito larvicidal activity, it was moderately effective as a feeding repellent and highly effective as an ovipositional repellent against adultAedes aegypti (yellow fever mosquito). The ovipositional repellency of 1,8-cineole, coupled with the presence of severalHemizonia chromenes previously shown to possess mosquito larvicidal activity, may therefore account in lar

Eucalyptol Ameliorates Retinal Microvascular Defects through Modulating ER Stress and Angiopoietin-Tie Signaling in Diabetic Eyes.

Loss of the inner blood-retinal barrier (BRB) integrity is a main feature of ocular diseases such as diabetic macular edema. However, there is a lack of clarity on how inner BRB function is modulated within the diabetic retina. The current study examined whether eucalyptol inhibited inner BRB destruction and aberrant retinal angiogenesis in 33 mM glucose-exposed human retinal microvascular endothelial (RVE) cells and db/db mice. This study further examined the molecular mechanisms underlying endothelial dysfunction including retinal endoplasmic reticulum (ER) stress and angiopoietin (Ang)/Tie

Suppression of Propionibacterium acnes-Induced Skin Inflammation by Laurus nobilis Extract and Its Major Constituent Eucalyptol.

Acne is an inflammatory skin disorder in puberty with symptoms including papules, folliculitis, and nodules. Propionibacterium acnes (P. acnes) is the main anaerobic bacteria that cause acne. It is known to proliferate within sebum-blocked skin hair follicles. P. acnes activates monocytic cell immune responses to induce the expression of proinflammatory cytokines. Although the anti-inflammatory function of the Laurus nobilis (L. nobilis) extract (LNE) on several immunological disorders have been reported, the effect of LNE in P. acnes-mediated skin inflammation has not yet been explored. Anti-biofilm properties of eucalyptol in combination with antifungals against Candida albicans isolates in patients with hematological malignancy.

Oral candidiasis is a fungal infection caused mainly by Candida albicans and it is a major problem among hematologic malignancy patients. Biofilm formation is an attributable factor to both virulence and drug resistance of Candida species. The aim of the study was to evaluate the biofilm-producing ability of oral C. albicans isolates and to evaluate the inhibitory activity of eucalyptol on Candida biofilm, alone and in combination with antifungal agents. Samples were collected from the oral cavity of 106 patients with hematologic malignancy. The isolated yeasts were identified by PCR-sequencin

Effects of Eucalyptol in respiratory system mechanics on acute lung injury after exposure to short-term cigarette smoke.

Eucalyptol is a compound that has demonstrated antioxidant, anti-inflammatory and bronchodilator effects, but there are no investigations about the effects of this constituent on the respiratory system mechanics in relation to acute lung injury caused by short-term cigarette smoke (CS) exposure. In view of the above, this work investigated the effects of Eucalyptol on the mechanics of the respiratory system of mice in short-term CS exposure. For this, we used data from respiratory mechanics in vivo, and histopathology and lung parenchymal morphometry analysis in vitro. The experiments were per

Eucalyptol-based dispersive liquid-liquid microextraction in conjunction with GC-MS for phthalate esters determination in bottled water samples.

BACKGROUND: Endocrine-disrupting compounds (EDCs) rank among the most hazardous chemicals to human health. Accordingly, the accurate determination and continuous monitoring of EDCs in everyday products are critical to ensuring public health and safety. These efforts require advanced analytical procedures to detect specific EDCs in samples with diverse matrix compositions at extremely low concentration levels. Most of the analytical methods developed to date rely on the use of toxic organic solvents for sample preparation. However, current trends in analytical chemistry, in accordance with the

Magnoliae flos (Shin-i) Essential Oil and Its Principal Component Eucalyptol Inhibit JAK1 to Counteract the LPS-Induced RAW264.7 Inflammation.

Magnoliae flos (Shin-i) has been traditionally utilized in Chinese medicine for rhinitis, sinusitis, and headaches. However, the underlying anti-inflammatory mechanisms of Shin-i essential oil (SIEO), especially its effects on the Janus kinase/signal transducer and activator of transcription (JAK-STAT) signaling pathway, remain inadequately elucidated. The chemical composition of SIEO was characterized using gas chromatography-mass spectrometry (GC-MS). Network pharmacology and gene ontology (GO) enrichment analyses predicted that SIEO may target the JAK-STAT signaling pathway. Cellular experi

Repellency of Eucalyptol and DEET Against Triatoma infestans, a Chagas Disease Vector: A Proof-of-Concept Study Under a Human Odor Background.

Triatomines are vectors of Chagas disease. Control of Triatoma infestans, one of its main vectors, relies largely on the use of pyrethroid insecticides. However, due to resistance to them as well as environmental concerns, alternative control strategies are required. Here, we evaluated the volatile repellent activity of eucalyptol, carvacrol, carvone, and DEET (N, N-diethyl-3-methylbenzamide) under conditions that prevented the contact of the insects with the compounds, and under a human odor background, providing a realistic assessment of repellency for human protection. Importantly, eucalypt

Eucalyptol Ameliorates Neuroendocrine Stress-Aggravated Rheumatoid Arthritis Through Anti-inflammatory, Antioxidant, and Chondroprotective Mechanisms Associated with NF-κB and MAPK Signaling Pathways.

BACKGROUND: Rheumatoid arthritis (RA) is a lingering autoimmune disorder typified by synovial inflammation, oxidative imbalance, and advanced articular damage. Psychological and physical stress are known to exacerbate disease progression by enhancing inflammatory signaling and disrupting redox homeostasis. Although methotrexate remains a first-line therapy, its long-term use is accompanied by adverse effects, necessitating safer adjunctive strategies. Eucalyptol (1,8-cineole), a naturally occurring monoterpene, has demonstrated anti-inflammatory and antioxidant potential; however, its role in
