Regulation of cell shape in Euglena gracilis. III. Involvement of stable microtubules.

The role of cytoplasmic microtubules in a recently reported biological clock-controlled rhythm in cell shape of the alga Euglena gracilis (strain Z) was examined using indirect immunofluorescence microscopy. The resulting fluorescent patterns indicated that, unlike many other cell systems, Euglena cells apparently change from round to long to round cell shape without associated cytoplasmic microtubule assembly and disassembly. Instead, the different cell shapes were correlated with microtubule patterns, which suggested that movement of stable microtubules to accomplish cell shape changes. In l

Spontaneous mutation rate estimation in the large-genome unicellular eukaryote Euglena gracilis.

The spontaneous mutation rate per nucleotide (µ) is a fundamental parameter shaping genome evolution, yet it remains unknown for many eukaryotic groups. Here, we estimated µ in the freshwater unicellular eukaryote Euglena gracilis, which possesses a 2.4 Gb genome, among the largest reported for unicellular species. We conducted a mutation accumulation experiment using 10 independent MA lines propagated through repeated single-cell bottlenecks and then identified 153 de novo nucleotide mutations. After correction for false negatives, the spontaneous mutation rate was estimated at µ=6.60 × 10⁻¹⁰

Characterization of sulfur-compound metabolism underlying wax-ester fermentation in Euglena gracilis.

Euglena gracilis is a microalga, which has been used as a model organism for decades. Recent technological advances have enabled mass cultivation of this species for industrial applications such as feedstock in nutritional foods and cosmetics. E. gracilis degrades its storage polysaccharide (paramylon) under hypoxic conditions for energy acquisition by an oxygen-independent process and accumulates high amount of wax-ester as a by-product. Using this sequence of reactions referred to as wax-ester fermentation, E. gracilis is studied for its application in biofuel production. Although the wax-es

Chromium induced stress conditions in heterotrophic and auxotrophic strains of Euglena gracilis.

Oxidative stress parameter and antioxidant defense compound as well as enzyme activity were studied in relation to different Cr(VI) concentrations (0, 10, 20, 40 μM) in two strains of Euglena gracilis, one isolated from a polluted river (MAT) and the other acquired from a culture collection (UTEX). Chromium toxicity was measured in the auxotrophic and obligated heterotrophic variants of the two strains. Chromium uptake was higher in auxotrophic cultures, reflected by their higher cell proliferation inhibition and lower IC50 levels compared to heterotrophic ones. In the Cr(VI) treatments a redu

Simultaneous Cd2+, Zn2+, and Pb2+ uptake and accumulation by photosynthetic Euglena gracilis.

The ability of Euglena gracilis to simultaneously remove and accumulate Zn2+, Cd2+, and Pb2+ from culture up- to media was evaluated. E. gracilis was able to remove up to 80% of the Cd2+ present in the medium when cultured with 20 or 50 microM CdCl2. Higher external Cd2+ concentrations increased Cd2+ accumulation per cell but decreased cell growth, thus decreasing the capacity of the cell culture to remove Cd2+. E. gracilis removed 70% to 80% of the Zn2+ present in the medium when cultured with 5 to 50 microM ZnSO4. Zn2+ did not affect Cd2+ removal capacity. E. gracilis was much less efficient

Time-course development of the Cd2+ hyper-accumulating phenotype in Euglena gracilis.

To determine the onset of the Cd2+-hyperaccumulating phenotype in Euglena gracilis, induced by Hg2+ pretreatment (Avilés et al. in Arch Microbiol 180:1-10, 2003), the changes in cellular growth, Cd2+ uptake, and intracellular contents of sulfide, cysteine, gamma-glutamylcysteine, glutathione and phytochelatins during the progress of the culture were analyzed. In cells exposed to 0.2 mM CdCl2, the Cd2+-hyperaccumulating phenotype was apparent only after 48 h of culture, as indicated by the significant increase in cell growth and higher internal contents of sulfide and thiol-compounds, along wit

Genotoxicity and subchronic toxicity evaluation of dried Euglena gracilis ATCC PTA-123017.

Euglena gracilis is a microalga capable of synthesizing various nutrients of interest in human and animal nutrition. When cultivated aerobically in the dark, Euglena synthesize paramylon, a storage polysaccharide comprised of high molecular weight beta-1,3-D-glucose polymers organized in cytoplasmic granules. Beta-glucans have been shown to have immune modulation effects, including anti-microbial, anti-tumor, and anti-oxidant properties, and metabolic effects, such as regulation of cholesterol and blood sugar levels. Preparations of E. gracilis and paramylon may therefore have potential utilit

Molecular changes in phenolic compounds in Euglena gracilis cells grown under metal stress.

Metal presence in the aquatic ecosystem has increased and diversified over the last decades due to anthropogenic sources. These contaminants cause abiotic stress on living organisms that lead to the production of oxidizing molecules. Phenolic compounds are part of the defense mechanisms countering metal toxicity. In this study, the production of phenolic compounds by Euglena gracilis under three different metal stressors (i.e. cadmium, copper, or cobalt) at sub-lethal concentration was assessed using an untargeted metabolomic approach by mass spectrometry combined with neuronal network analysi

Comprehensive genomic and transcriptomic analysis of inositol phosphate-metabolizing enzymes in Euglena gracilis.

Euglena gracilis possesses a dynamic inositol metabolic network, with enzyme expression varying under different growth conditions and stressors, enabling future synthetic engineering. Inositol and its phosphate derivatives play pivotal roles in energy metabolism and cellular signaling. Although inositol was detected in Euglena gracilis (E. gracilis) decades ago, the enzymatic machinery governing its metabolic conversion remains poorly characterized. In this study, we conducted a comprehensive bioinformatic analysis of inositol phosphate-metabolizing enzymes and identified 25 enzyme entries (en

Sequential UV mutagenesis and CO2 acclimation reprogram metabolic carbon allocation in Euglena gracilis.

Euglena gracilis is promising for CO2 bioutilization, but its metabolic response to elevated CO2 is poorly characterized. Here, the LE-ZW mutant, obtained via UV mutagenesis and CO2 acclimation, was evaluated under 5% and 10% CO2. LE-ZW exhibited 7% higher cell density and a 61% increase in photosynthetic carbon fixation at 10% CO2 compared with 5% CO2. Carbon allocation shifted with CO2 concentration: lipid accumulation peaked at 20.57% under 5% CO2, while paramylon content reached 37.37% under 10% CO2. Protein levels increased by 4% under high CO2 (vs 5% CO2 in LE-ZW). Transcriptomic and met

Different growth response of Euglena gracilis to Hg, Cd, Cr and Ni compounds.

The toxicity of inorganic mercury, nickel, chromium and cadmium on the unicellular photosynthetic flagellate Euglena gracilis, strain Z (E.g.) has been tested. Under the conditions used each metal impaired the growth rate of E.g., and had a very strong effect on cell motility. The degree of cytotoxicity and motility decreased from mercury iodide to cadmium chloride to cadmium nitrate to potassium dichromate to nickel sulfate. No mutagenic effects of the metals investigated have been observed. Adverse effects of metal compounds can be tested on the eukaryotic species of Euglena gracilis used as

Control of chloroplast formation in Euglena gracilis. Antagonism between carbon and nitrogen sources.

1. Cells of Euglena gracilis grown in the dark on high ratios of carbon source to nitrogen source (;high-carbon cells') are unable to form chlorophyll during a subsequent incubation in the light; cells grown in the dark on low ratios of carbon to nitrogen (;low-carbon cells') synthesize chlorophyll at a rapid rate during the subsequent incubation in the light. High-carbon cells will form chlorophyll rapidly if supplied with a nitrogen source during the incubation in the light: of the nitrogen sources tested, ammonium sulphate was the most effective at overcoming the block in chlorophyll synthe

Pyruvate:NADP+ oxidoreductase is stabilized by its cofactor, thiamin pyrophosphate, in mitochondria of Euglena gracilis.

Pyruvate:NADP(+) oxidoreductase (PNO) is a thiamin pyrophosphate (TPP)-dependent enzyme that plays a central role in the respiratory metabolism of Euglena gracilis, which requires thiamin for growth. When thiamin was depleted in Euglena cells, PNO protein level was greatly reduced, but its mRNA level was barely changed. In addition, a large part of PNO occurred as an apoenzyme lacking TPP in the deficient cells. The PNO protein level increased rapidly, without changes in the mRNA level, after supplementation of thiamin into its deficient cells. In the deficient cells, in contrast to the suffic

Regulation of cell shape in Euglena gracilis. IV. Localization of actin, myosin and calmodulin.

The immunofluorescence patterns for actin, myosin, calmodulin and tubulin were observed in Euglena gracilis Klebs strain Z during the biological clock-controlled shape changes observed with division-synchronized cells, and during two shock responses that induce cell rounding. The fluorescence patterns for actin, myosin, calmodulin and tubulin show a high degree of coincidence and are visualized as lines running parallel to, and having the same spacing as, the pellicle strips beneath the plasma membrane. The fluorescence patterns remain intact during the daily shape changes, implying that the s

Light-triggered organization of the stigma in dark-grown nondividing cells of Euglena gracilis.

Dark-grown cells of Euglena gracilis Klebs var. bacillaris Cori contain amorphous stigma material. When these cells are placed on resting medium for 3 days in darkness, the cells cease division; the organization of a normal stigma from the amorphous material requires continuous illumination for 72-96 hr. We have now found that if dark-grown cells are placed on resting medium for 8 days, a 40-min light pulse is sufficient to cause normal organization of the stigma in a subsequent 72-hr dark period. Thus stigma development is light-dependent at 3 days of resting but becomes light-triggered at 8

Strategic Carbon Source Selection Enhances Biomass and Paramylon Yields in Mixotrophic Euglena gracilis Cultivation.

Euglena gracilis's mixotrophic metabolism offers biotechnological potential. This study investigated how glucose, sodium acetate, ethanol, and propanetriol regulate its growth, photosynthesis, and paramylon production. All carbon sources boosted paramylon yield versus photoautotrophic controls. Ethanol and glucose were both highly effective, supporting the highest biomass accumulation (5.71 and 4.42-fold increases, respectively) and paramylon content without a significant difference between them. Ethanol supplementation enhanced chlorophyll b via coupled TCA cycle/glyoxylate shunt activity, wh

Non-Proliferative Immune Restoration by Euglena gracilis EK Powder in Cyclophosphamide-Induced Immunosuppressed Mice.

Cytotoxic chemotherapy frequently induces immunosuppression, impairing the innate cellular and humoral immune responses. Therefore, immunomodulatory agents that restore immune function without indiscriminate leukocyte expansion are required. This study examined whether Euglena gracilis EK powder (EKP), a paramylon-rich β-1,3-glucan preparation, supports immune recovery under cyclophosphamide (CPA)-induced immunosuppression. Male BALB/c mice received EKP (80, 200, or 500 mg/kg/day for 21 days) under a dual-temporal CPA design: acute phase (days 7 and 9) and established phase (days 14 and 17). T

Carboxymethylation of paramylon derived from Euglena gracilis and its hypoglycemic mechanism in diabetic mice.

Paramylon is a polysaccharide primarily composed of β-1,3-glucan, characterized by its high crystallinity and insolubility in water. Enhancing its water solubility through structural modifications presents an effective strategy to unlock its biological activity. In this study, carboxymethylation was employed to produce carboxymethylated paramylon (CEP) with varying carboxyl concentrations. The successful introduction of carboxyl groups led to a notable improvement in water solubility. In vivo experiments demonstrated that CEP reduced fasting blood glucose levels by 24.42 %, improved oral gluco

An atypical heme-binding structure of cytochrome c1 of Euglena gracilis mitochondrial complex III.

Complex III was purified from submitochondrial particles prepared from Euglena gracilis. The purified complex consisted of 10 subunits and lost antimycin sensitivity. The Euglena complex III showed an atypical difference absorption spectrum for cytochrome c1 with its alpha-band maximum at 561 nm. The pyridine ferrohemochrome prepared from covalently bound heme in the Euglena complex III had an alpha-peak at 553 nm. This wavelength is the same as that of pyridine ferrohemochrome prepared from Euglena mitochondrial cytochrome c (c-558), the heme of which is linked to only a single cysteine resid

Metabolomic Insights into the Adaptations and Biotechnological Potential of Euglena gracilis Under Different Trophic Conditions.

Euglena gracilis is a metabolically versatile microalga capable of thriving under photoautotrophic (light, no ethanol), mixotrophic (light with 1% v/v ethanol), and heterotrophic (dark with 1% v/v ethanol) conditions. Here, we applied untargeted LC-MS metabolomics (Agilent 1290 LC, 6545XT QTOF-MS; Agilent Technologies, Santa Clara, California, USA) to investigate its trophic-mode-dependent metabolic adaptations and assess its biotechnological potential. Metabolites were separated on a C18 column and analyzed in both positive and negative ion modes. Multivariate analyses (PCA and sPLS-DA) revea
