Analysis of genes coding for small-subunit rRNA sequences in studying phylogenetics of dematiaceous fungal pathogens.

Because of their ability to display yeast-like growth forms in various environmental conditions, dematiaceous (melanized) hyphomycetes of the form-genera Exophiala, Rhinocladiella, and Wangiella have been informally termed "black yeasts." Cladistic analysis of 1,050 bp of the genes coding for small-subunit rRNA (SSU rDNA) supported a close relationship among species of these black yeasts with other dematiaceous hyphomycetes in the form-genera Fonsecaea, Phialophora, and Ramichloridium. The conventional categories of these fungi based on asexual states are not supported by phylogenetic analysis

Lipase Activation by Poly(Methyl Methacrylate) in Dispersed Solution: Mechanistic Insights by Fluorescence Spectroscopy.

We investigated the mechanisms of polymer-lipase interactions that govern the catalytic activity of lipases in the presence of polymers. Using a combination of fluorescence correlation spectroscopy (FCS), activity analysis, fluorescence spectroscopy, and computational surface analysis, three model lipases-Thermomyces lanuginosus lipase (TLL), Candida antarctica lipase B (CalB), and Bacillus subtilis lipase A (BSLA), with different degrees of hydrophobic active site exposure were studied. Low-molecular-weight poly(methyl methacrylate) (PMMA), synthesized via ARGET ATRP, was employed to study th

Co-immobilization of lipase from Thermomyces lanuginosus (TLL) and lipase B from Candida antarctica (CALB) on functionalized epoxy resin for enhanced stability, reusability, and biocatalytic efficiency.

Epoxy resin, a durable synthetic polymer with a surface rich in ligand groups, was functionalized with branched polyethyleneimine and activated with glutaraldehyde to serve as a support for the co-immobilization of lipases CALB and TLL. RE/TLL-CALB was characterized by X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Thermogravimetry (TGA), and Scanning Electron Microscopy (SEM). The Taguchi L9 method identified the ideal conditions for the co-immobilization of lipases CALB and TLL in the functionalized epoxy resin (RE): 12 h of reaction, pH 9, 100 mM buffer, and an enz

Semirational Design of Methanol-Resistant Lipase Mutants: A Pathway to Efficient Biodiesel Production.

The development of a methanol-resistant thermostable lipase is of significant importance in biodiesel synthesis. In this study, a semirational design approach was employed to combine targeted mutagenesis and the introduction of N-glycosylation modifications to enhance the activity and stability of Thermomyces lanuginosus lipase. The activity of mutant A113G was 46% higher than that of the wild type. The double mutant A113G/L74N exhibited 33% residual activity after 24 h of incubation in 50% (v/v) methanol at 50 °C. The favorable temperature stability and methanol tolerance permitted a one-step

N2O production, a widespread trait in fungi.

N2O is a powerful greenhouse gas contributing both to global warming and ozone depletion. While fungi have been identified as a putative source of N2O, little is known about their production of this greenhouse gas. Here we investigated the N2O-producing ability of a collection of 207 fungal isolates. Seventy strains producing N2O in pure culture were identified. They were mostly species from the order Hypocreales order-particularly Fusarium oxysporum and Trichoderma spp.-and to a lesser extent species from the orders Eurotiales, Sordariales, and Chaetosphaeriales. The N2O (15)N site preference

Synthetic Lethality of Fungal Catabolite Repressors Reveals Essential Coupling of Carbon Repression With Mitochondrial and Ribosomal Gene Expression.

Carbon catabolite repression (CCR) enables fungi to preferentially utilize favorable carbon sources through global transcriptional control. In filamentous fungi, CCR is classically attributed to the C2H2 zinc-finger repressor Mig1/CreA; however, residual repression following Mig1 disruption suggests additional regulatory layers in Talaromyces pinophilus (previously called Penicillium funiculosum NCIM1228). Here, we identify Mig3, a previously uncharacterized, Eurotiales-specific C2H2 transcription factor that functions redundantly with Mig1 in CCR. Mig3 contains conserved N-terminal zinc-finge

Challenge of identifying a case of chronic invasive rhinosinusitis due to Rasamsonia aegroticola.

Fungal rhinosinusitis are frequently described in humans and its prevalence appears to be increasing. They mainly involve species of the genus Aspergillus. To date, conventional mycological diagnosis remains challenging, as only 30% and 50% of cultures are positive in cases of fungal balls and invasive rhinosinusitis respectively. Over the past decade, human infections due to the Rasamsonia argillacea species complex are increasingly frequent and mainly concern patients with cystic fibrosis or chronic granulomatous disease. We report here the first case of pseudotumoral R. aegroticola sinusiti

Porcine pancreas lipase: a versatile biocatalyst.

Porcine pancreatic lipase, a 1, 3-regioselective lipase, is one of the most widely used biocatalyst in academic as well as industrial processes. This lipase showed remarkable catalytic performance, often outclassed other types of lipases in producing valuable products through varieties of reactions. During hydrolysis of vegetable oil to fatty acid, mono and diacylglycerol, porcine pancreas lipase outclassed Candia rugosa and Thermomyces lanuginosus lipase. Porcine pancreas lipase efficiently catalyzed transesterification of vegetable oil to biodiesel as well. This lipase catalyzed esterificati

Active Site Studies to Explain Kinetics of Lipas`s in Organic Solvents Using Molecular Dynamics Simulations.

This study investigates the intricate dynamics underlying lipase performance in organic solvents using comprehensive molecular dynamics (MD) simulations, supported by enzyme kinetics data. The study reveals that a single criterion can neither predict nor explain lipase activity in organic solvents, indicating the need for a comprehensive approach. Three lipases were included in this study: Candida antarctica lipase B (CALB), Rhizomucor miehei lipase (RML), and Thermomyces lanuginosus lipase (TLL). The lipases were investigated in acetonitrile, methyl tert-butyl ether, and hexane with increasin

Synergistic enhancement of lipase catalysis via co-immobilization and chitosan-assisted crosslinking: a streamlined approach for EPA/DHA enrichment in fish oil.

Efficient immobilized lipase systems are essential for industrial lipid modification, yet the combined effects of enzyme loading, support properties, and post-immobilization stabilization on catalytic performance remain underexplored. In this study, Thermomyces lanuginosus lipase (TLL), Candida antarctica lipase B (CalB), and their mixture (T + C) were respectively immobilized on two structurally distinct commercial supports (A & B)—differing in hydrophobicity, surface area, and pore size—at varied loading densities. Distinguishing between long-term-stored and fresh chili pepper powder through fingerprinting of volatiles by headspace capillary-gas chromatography-ion mobility spectrometry.

Long-term storage of chili pepper powder results in physicochemical and microbiological changes that decrease its commercial value; these changes occur owing to fungal growth and production of off-flavor compounds. Herein, long-term-stored chili pepper powder (LSCPP) and fresh chili pepper powder (FCPP) were analyzed using internal transcribed spacer sequencing and volatile organic compound fingerprinting by headspace capillary-gas chromatography-ion mobility spectrometry. Fungal analysis detected only Xeromyces bisporus with high accuracy in all the analyzed LSCPP samples. However, the prolif

Effects of Nixing Pottery Storage on Microbial Community Characteristics, Quality, and Volatile Components of Liupao Tea.

Liupao tea tended to suffer from unbalanced microbial community structure during storage, which further caused quality deterioration of tea, resulting in economic losses and potential food safety risks. Nixing Pottery (NP) exhibited remarkable advantages in the storage and fresh keeping of tea. Taking glass containers (GC) as the control group, this study investigated the effects of NP on the infusion color, non-volatile internal components, volatile aromatic components, and microbial community structure of Liupao tea during a 24-month storage period. The results indicated that Liupao tea stor

Metagenomics-based insights into the microbial community succession and metabolic potential of flavor in Qingke high-temperature Daqu.

Raw grain selection profoundly influences the flavor and quality of high-temperature Daqu (HTD). However, the microbial and flavor profiles of Daqu made with Qingke (highland barley) remain unclear. This study explored how varying Qingke addition affects physicochemical indicators, volatile organic compounds (VOCs), and microbial succession, ultimately impacting the formation of characteristic flavor compounds. Results showed that Qingke high-temperature Daqu (QHTD) underwent notable physicochemical changes, with the 30%and 50% addition groups exhibiting higher moisture and total acid levels (

Culturable endophytic fungal assemblages from Styrax sumatrana and Stryax benzoin and their potential as antifungal, antioxidant, and alpha-glucosidase inhibitory resources.

Benzoin resin, produced by the native Indonesian trees Styrax sumatrana and Styrax benzoin, has been incorporated into medical practices to treat wounds, erythema, and many other conditions for centuries. Endophytic fungi that reside within medicinal plants have antimicrobial, antioxidant, and α-glucosidase inhibitory capacities, contributing to plant health and derivative products. In this study, we determined the antifungal, antioxidant, and α-glucosidase inhibitory capacities of endophytic fungal isolates from three different tissues (leaves, bark, and stems) of S. sumatrana and S. benzoin

Rasamsonia argillacea species complex: taxonomy, pathogenesis and clinical relevance.

Since 2010, colonizations/infections by Rasamsonia argillacea species complex, previously known as Geosmithia argillacea, have been regularly reported in literature. We reviewed all available cases focusing on pathogenesis and clinical relevance. The number of cases may be underestimated, as these fungi are frequently misidentified as Penicillium or Paecilomyces species. Major underlying conditions that predispose for infections by the R. argillacea species complex include cystic fibrosis (CF) and chronic granulomatous disease (CGD). While the pathogenic role of the colonization of CF lungs is

[Rasamsonia argillacea in Patient with Immunodeficiency: First Suspected Case in Türkiye].

Rasamsonia species are thermophilic fungi that primarily cause infection in immunocompromised patients. R.argillacea species complex is frequently misidentified as a pathogen due to its morphological similarity to the genera Penicillium and Paecilomyces and its incidence remains undetermined. This situation further complicates the process of timely diagnosis and treatment. Rasamsonia species can be identified using internal transcribed spacer (ITS) sequence analysis, which is considered the primary fungal barcode. Accurate species identification is imperative for the administration of appropri

Rasamsonia argillacea pulmonary and aortic graft infection in an immune-competent patient.

Rasamsonia argillacea (formerly known as Geosmithia argillacea) is a fungus recently recognized as a pathogen of immunocompromised patients. Here we report the first case of Rasamsonia infection in an immunocompetent host, presenting as a pulmonary and aortic graft infection. Its morphological similarity to nonpathogenic Penicillium species delayed the diagnosis and initiation of appropriate treatment.

Isolation of Neosartorya pseudofischeri from blood: first hint of pulmonary Aspergillosis.

We report a case of invasive pulmonary aspergillosis caused by Neosartorya pseudofischeri S. W. Peterson [anamorph Aspergillus thermomutatus (Paden) S. W. Peterson]. The diagnosis was initially based on a positive blood culture for a strain isolated from a neutropenic patient by means of a BACTEC 9050 blood culture system. The final diagnosis was established based on X-ray and computer tomography scan results as well as the detection of Aspergillus antigen in the patient's serum.

Culturable Fungal Community of Pterocladiella capillacea in Keelung, Taiwan: Effects of Surface Sterilization Method and Isolation Medium.

Fungi associated with macroalgae are less known when compared with those on wood in the marine environment. In this study, we assessed the diversity of fungi associated with the red alga Pterocladiella capillacea at Chao-Jin Park, Keelung, Taiwan. Algal segments of healthy and dead thalli were washed/sterilized with different solutions (sterile artificial seawater, 70% ethanol, and 4% sodium hypochlorite), plated on three different media (glucose-yeast extract-peptone seawater agar (GYPS), potato dextrose seawater agar (PDAS), and artificial seawater agar (SA)), and isolated as pure cultures.

Discovery of a fungal Tc toxin complex and functional mycoserpin through a novel two-by-two comparative genomics approach.

INTRODUCTION: Fungi have been a rich source of pharmaceuticals such as antibiotics, immunosuppressants, and cholesterol-lowering drugs; however, their therapeutic potential remains largely untapped due to difficulties in culturing and elucidating the genetic basis of beneficial traits. Fungi contain 'cryptic' genes that are expressed under certain, and often obscure, growth conditions and can produce complex compounds that are difficult to synthesize economically. Developments in genome sequencing and DNA-synthesis technologies offer new opportunities to produce such compounds using biotechnol
