A comprehensive review on pharmaceutical excipients.

The effectiveness of pharmaceutical drugs depends not only on their active components and manufacturing processes, but also on the role played by pharmaceutical excipients. The traditional definition of excipients as inactive and cost-effective substances has evolved significantly. They are now recognized as essential elements of drug formulations, constituting 80-90% of the final product. The rapid advancements in delivery systems, along with scientific, regulatory, financial and technological developments in biopharmaceutics, have generated renewed interest in the use and functionality of ex

Ethylenediamine, profile of a sensitizing excipient.

Ethylenediamine is an excipient with many industrial and pharmaceutical uses. It is included in creams as a stabilizer and in aminophylline as the counter ion of theophylline. Ethylenediamine is one of the most frequent contact sensitizers, producing local and generalized reactions. Besides, many cases of systemically induced dermatitis have also been described both after oral, rectal and intravenous use. Inhalation of ethylenediamine or aminophylline dust may provoke rhinitis and asthmatic reactions. In contrast to these delayed reactions only one immediate reaction of the urticarial type aft

Nano-Enabled Toxicity of Pharmaceutical Excipients.

Nanotechnology has brought revolutionary advances in the pharmaceutical field. Among drug delivery systems, nanocarriers, which enhance the solubility, stability and membrane permeability of numerous drugs, have become indispensable. However, nanocarriers not only amplify the therapeutic potential of drugs but can also increase the toxic potential of excipients used in their formulation. For example, commonly used excipients such as polyethylene glycol, cationic polymers (e.g., polyethylenimine), cationic lipids, metal-based materials, and cyclodextrins may exhibit altered, and often enhanced,

[Hypersensitivity reactions to excipients in patches].

The most common side effects of transdermal patches are mechanical reactions caused by applying or removing the transdermal patch, or by excessive perspiration under the patch. Allergic contact dermatitis (type IV allergic reaction) is the most commonly occurring hypersensitivity reaction and can be caused by the active substance or by excipients. Type I allergic reactions such as urticaria, bronchospasm and angioedema are rare and usually caused by the active substance in the patch. Allergy testing to determine the allergen is indicated following a type I allergic reaction, or after a type IV

Thermal properties of compacted pharmaceutical excipients.

Thermal properties of powders are critical material attributes that control temperature rise during tableting and roll compaction. In this study, various analytical methods were used to measure the thermal properties of widely used pharmaceutical excipients including microcrystalline cellulose (MCC) of three different grades (Avicel PH 101; Avicel PH 102 and Avicel DG), lactose and mannitol. The effect of relative density on the measured thermal properties was investigated by compressing the powders into specimen of different relative densities. Differential thermal analysis (DTA) was employed

Excipient Taxonomy for the 21st Century.

The performance of pharmaceutical dosage forms relies heavily on the characteristics of the excipients that are incorporated into the drug product during the manufacturing process. Therefore, it is imperative that formulators are able to accurately and completely specify the key chemical and physical properties of those excipients. Current approaches to describing excipients are outdated and inadequate for the needs of the 21st century and in this article we highlight the benefits of a more systematic and comprehensive approach to specifying and controlling excipient properties. We hope that t

Advances in polysaccharide nanocrystals as pharmaceutical excipients.

Polysaccharide nanocrystals (PNs) are attractive pharmaceutical excipients due to their abundant surface hydroxyl groups, high surface charges, prominent mechanical properties, excellent fluidity, and good swelling properties. In this review, we summarize three kinds of PNs, including cellulose nanocrystals (CNCs), starch nanocrystals (SNCs), and chitin nanocrystals (ChNCs). We introduce the applications of PNs as stabilizers, adsorbents, film-forming materials, gel materials, disintegrants, and ointment matrices. We focus on the advantages of PNs to improve mechanical properties, thermal stab

Injectable Excipients as Novel Influenza Vaccine Adjuvants.

Influenza outbreaks can be either seasonal or pandemic. Vaccination is an effective strategy to control influenza; however, the efficacy of the currently available inactivated influenza virus vaccines is suboptimal, especially in the elderly. Vaccine efficacy can be improved by the addition of adjuvants, but few adjuvants have been approved for human vaccines. To explore novel, safe, and effective adjuvants for influenza vaccines, here we used a mouse model to screen 46 injectable drug additives approved in Japan. Of these 46 candidates, we identified 20 compounds that enhanced the efficacy of

Viscoelastic interactions between polydeoxyribonucleotide and ophthalmic excipients.

This study investigated the interaction between polydeoxyribonucleotide (PDRN) and several ionic and nonionic isotonic agents, thickeners and a preservative that were employed as excipients in ophthalmic preparations. Interaction of each individual excipient and PDRN aqueous solution was evaluated by analyzing their rheological properties. Rheological properties of PDRN solutions were evaluated by dynamic oscillatory shear tests and values of elastic modulus (G'), viscous modulus (G″) and loss tangent (tan δ) were used to assess the relative changes in viscoelastic properties. Polymeric plant-derived excipients in drug delivery.

Drug dosage forms contain many components in addition to the active pharmaceutical ingredient(s) to assist in the manufacturing process as well as to optimise drug delivery. Due to advances in drug delivery technology, excipients are currently included in novel dosage forms to fulfil specific functions and in some cases they directly or indirectly influence the extent and/or rate of drug release and absorption. Since plant polysaccharides comply with many requirements expected of pharmaceutical excipients such as non-toxicity, stability, availability and renewability they are extensively inves

Managing the microbiological quality of pharmaceutical excipients.

The microbiological quality of the pharmaceutical excipients used to manufacture pharmaceutical and over-the-counter drug products may significantly affect the outcome of individual processing steps and the microbiological attributes of the final drug products. Unlike active pharmaceutical ingredients, excipients are purchased from multiple suppliers and in many cases are produced for the food, cosmetics, consumer products, photographic, and paint industries and not specifically for the pharmaceutical industry, so the management of their microbiological quality is less straight- forward. This

Are excipients really inert ingredients? A review of adverse reactions to excipients in oral dermatologic medications in Canada.

BACKGROUND: Whereas several literature reviews have discussed the role of excipients in drug-related reactions, no article has focused specifically on those found in oral dermatologic medications. METHODS: The Compendium of Pharmaceuticals and Specialties (CPS) was used to reference the inert ingredients found in oral dermatologic medications. An extensive literature review was subsequently conducted using PubMed and MEDLINE to document adverse reactions to these excipients. RESULTS: Sixty-three oral dermatologic medications were reviewed. Lactose was commonly used as a filler. Several medic

Nitrite testing in excipients - Industry best practices.

NNitrosamine risk assessment and control are essential components of pharmaceutical drug development and product evaluation. Nitrites present in excipients pose a risk to vulnerable amines that are present in APIs, API impurities or other process related amines, as they can be nitrosated to form N-nitrosamines. Detection and quantification of nitrite in excipients is an essential undertaking within the pharmaceutical industry to inform nitrosamine risk assessment and related risk mitigation strategies. An industry consortium and Lhasa Nitrites database was established to collaborate on this ch

Pharmaceutical excipients - quality, regulatory and biopharmaceutical considerations.

Practically all medications contain excipients, which are added for the purpose of production enhancement, patient acceptability, improving stability, controlling release etc. Typically excipients are the major components of a drug product, with the active molecule only present in relatively small amounts. Historically, excipients were termed inactive components. However, as highlighted in the present paper; excipients can have an impact on the absorption, distribution, metabolism and elimination (ADME) processes of the co-administered drug, which is important information when selecting excipi

What are excipients doing in medicinal products?

Medicinal products contain not only active drugs but also other ingredients included for a variety of purposes and collectively known as excipients (from the Latin word excipere, meaning 'to take out or receive'). Prescribers may wrongly overlook or dismiss excipients on the assumption that these are inactive. In reality, such substances are not always inert and may have risks. Here we consider why excipients are present in medicines, their potential unwanted effects, how they are regulated and where to find out more about them.

Evaluation of hydroperoxides in common pharmaceutical excipients.

While the physical properties of pharmaceutical excipients have been well characterized, impurities that may influence the chemical stability of formulated drug product have not been well studied. In this work, the hydroperoxide (HPO) impurity levels of common pharmaceutical excipients are measured and presented for both soluble and insoluble excipients. Povidone, polysorbate 80 (PS80), polyethylene glycol (PEG) 400, and hydroxypropyl cellulose (HPC) were found to contain substantial concentrations of HPOs with significant lot-to-lot and manufacturer-to-manufacturer variation. Much lower HPO l

Three-dimensional aspects of formulation excipients in drug discovery: a critical assessment on orphan excipients, matrix effects and drug interactions.

Formulation/pharmaceutical excipients play a major role in formulating drug candidates, with the objectives of ease of administration, targeted delivery and complete availability. Many excipients used in pharmaceutical formulations are orphanized in preclinical drug discovery. These orphan excipients could enhance formulatability of highly lipophilic compounds. Additionally, they are safe in preclinical species when used below the LD50 values. However, when the excipients are used in formulating compounds with diverse physico-chemical properties, they pose challenges by modulating study result
