Expressions of angiogenesis associated matrix metalloproteinases and extracellular matrix proteins in cerebral vascular malformations.

This study aimed to compare cerebral arteriovenous malformations (cAVM) and cerebral cavernous malformations (CCM) with regard to the immunohistochemical expressions of matrix metalloproteinases (MMP) and selected extracellular matrix (ECM) proteins, which have a role in the regulation of angiogenesis. Fresh-frozen surgical specimens from patients with cAVM (n=14) and CCM (n=15) were immunohistochemically stained with antibodies for MMP-2, MMP-9, laminin, fibronectin and tenascin. To compare cAVM and CCM, expression of each protein was graded using a four-point scoring system for each histolog

Brain defence by the extracellular matrix protein Cochlin.

The vertebrate brain is protected from infection by tight barriers. However, several barrier structures, including circumventricular organs, can be breached by pathogens. Here, we show that Cochlin, an extracellular matrix-binding protein with anti-bacterial properties, is produced in barrier structures and contributes to immune defence. Transcriptome analysis and in situ hybridisation indicate that cochlin is expressed in the pineal gland, area postrema, choroid plexus and discrete regions of the meninges of zebrafish, mice and humans. The protein is present in the cerebrospinal fluid, and on

The role of the extracellular matrix protein TGFBI in cancer.

The secreted extracellular protein, transforming growth factor beta induced (TGFBI or βIGH3), has roles in regulating numerous biological functions, including cell adhesion and bone formation, both during embryonic development and during the pathogenesis of human disease. TGFBI has been most studied in the context of hereditary corneal dystrophies, where mutations in TGFBI result in accumulation of TGFBI in the cornea. In cancer, early studies focused on TGFBI as a tumor suppressor, in part by promoting chemotherapy sensitivity. However, in established tumors, TGFBI largely has a role in promo

Nε-Carboxymethyl-Lysine Modification of Extracellular Matrix Proteins Augments Fibroblast Activation.

The extracellular matrix (ECM) is a dynamic complex protein network that provides structural integrity and plays an active role in shaping fibroblast behavior both in health and disease. Despite its essential functions, the impact of age-associated post-translational modifications on ECM-driven fibroblast activities such as proliferation, survival, fibroblast-to-myofibroblast transformation (FMT), and extracellular matrix production remains largely unknown. Nε-carboxymethyl-lysine (CML) is one of the well-characterized advanced glycation end-products (AGEs) that can occur on lysine residues wi

Tissue transglutaminase colocalizes with extracellular matrix proteins in cerebral amyloid angiopathy.

Cerebral amyloid angiopathy (CAA) is a key histopathological hallmark of Alzheimer's disease (AD) and hereditary cerebral hemorrhage with amyloidosis of the Dutch type (HCHWA-D). CAA is characterized by amyloid-beta (Aβ) depositions and remodeling of the extracellular matrix (ECM) in brain vessels and plays an important role in the development and progression of both AD and HCHWA-D. Tissue transglutaminase (tTG) modulates the ECM by molecular cross-linking of ECM proteins. Here, we investigated the distribution pattern, cellular source, and activity of tTG in CAA in control, AD, and HCHWA-D ca

Involvement of extracellular matrix proteins in the course of experimental paracoccidioidomycosis.

We aimed at determining involvement of extracellular matrix proteins (ECMp) and an ECM-binding adhesin (32-kDa protein) from Paracoccidioides brasiliensis, in the course of experimental paracoccidioidomycosis. BALB/c mice were infected with P. brasiliensis conidia previously incubated with soluble laminin, fibronectin and fibrinogen or a mAb against the fungal adhesin. Inflammatory response, chitin levels and cytokine production at different postinfection periods were determined. Chitin was significantly decreased in lungs of mice infected with ECMp-treated conidia when compared with controls

Expression of extracellular matrix proteins in the fetal rat gastric mucosa.

At gestational day 16 the epithelium of the rat stomach consists of a stratified layer of undifferentiated cells, and two days later glandular structures appear. The present study was carried out to identify extracellular matrix proteins that could be involved in the epithelial cell proliferation and differentiation processes that occur in the fetal rat stomach during this period. For comparative purposes the expression of the same components in the adult gastric mucosa was examined. Pregnant Sprague-Dawley rats received an intraperitoneal injection of 5-bromo-2'-deoxyuridine to label prolifer

Structural and functional aspects of calcium binding in extracellular matrix proteins.

Ca2+ ions play crucial roles in many matrix-matrix, cell-matrix and cell-cell contacts. Recent X-ray and NMR structure determinations have revealed an intriguing diversity of Ca(2+)-binding sites in extracellular proteins, ranging from the stabilization of isolated domains to intimate involvement in the superstructure of macromolecular assemblies. The central role of Ca2+ in extracellular proteins is illustrated by the molecular characterization of hereditary connective tissue disorders in humans. Point mutations of Ca(2+)-binding residues in fibrillin and cartilage oligomeric matrix protein a

Expression of extracellular matrix and adhesion proteins in pelvic organ prolapse.

Pelvic organ prolapse (POP) is a common disorder in women. It is characterized by the descent of the vaginal wall with consequent drop of pelvic organs. Pregnancy, labour and childbirth seem to be important events leading to the development of POP, since they are associated with prolonged stretch and mechanical stress of muscles, ligaments and connective tissue supporting pelvic organs. In pubocervical fascia, we explored the expression level of extracellular matrix and adhesion molecules. Tissue samples were obtained from twenty patients with POP who underwent cystocele repair, and from twent

RAGE-mediated extracellular matrix proteins accumulation exacerbates HySu-induced pulmonary hypertension.

AIMS: Extracellular matrix (ECM) proteins accumulation contributes to the progression of pulmonary arterial hypertension (PAH), a rare and fatal cardiovascular condition defined by high pulmonary arterial pressure, whether primary, idiopathic, or secondary to other causes. The receptor for advanced glycation end products (RAGE) is constitutively expressed in the lungs and plays an important role in ECM deposition. Nonetheless, the mechanisms by which RAGE mediates ECM deposition/formation in pulmonary arteries and its roles in PAH progression remain unclear. METHODS AND RESULTS: Expression of

Matrilin-2 interacts with itself and with other extracellular matrix proteins.

Matrilin-2 is a component of extracellular filamentous networks. To study the interactions by which it can be integrated into such assemblies, full-length and truncated forms of matrilin-2 were recombinantly expressed in HEK-293 cells and purified from conditioned medium. The recombinant proteins, when used in interaction assays, showed affinity to matrilin-2 itself, but also to other collagenous and non-collagenous extracellular matrix proteins. The interaction between matrilin-2 and collagen I was studied in greater detail and could be shown to occur at distinct sites on the collagen I molec

Adhesion of MNC to extracellular matrix proteins following in vitro photochemotherapy.

BACKGROUND: Photochemotherapy is a safe and effective treatment for patients with drug-resistant severe GvHD. The technique involves the exposure of MNC to psoralen and UVA light (PUVA). We have investigated the effect of in vitro PUVA on MNC adhesion to extracellular matrix (ECM) proteins. METHODS: MNC were isolated from peripheral blood (PB) and umbilical cord blood (UCB), and treated with PUVA. After labeling by a chemiluminescent probe, MNC were plated on ECM proteins (collagen, fibronectin, vitronectin and laminin) and the number of adherent cells was measured. RESULTS: Untreated MNC fr

Changes in extracellular matrix proteins and actin during corneal endothelial growth.

Basement membranes influence growth, shape and differentiation of cells and tissues. However, the role and influence of Descemet's membrane during corneal development is not understood. To address this question, the relationships between cell growth and fibronectin, laminin and actin distribution in the developing rat corneal endothelium in vivo has been examined. During fetal development, rat corneal endothelial cells undergo DNA synthesis and mitosis. However, at day 14 of gestation both processes begin to decline and neither can be detected in endothelium of 1-month-old animals. By this tim

Matrix gla protein: An extracellular matrix protein regulates myostatin expression in the muscle developmental program.

AIM: Skeletal muscle development involves interactions between intracellular and extracellular factors that act in concert to regulate the myogenic process. Matrix gla protein (MGP), a well-known inhibitor of calcification in soft tissues, has been reported to be highly up-regulated during myogenesis. Our interest in the regulation of muscle satellite cells (MSCs) by extracellular matrix (ECM) led us to investigate the effects of MGP during the progression of myogenesis. METHODOLOGY: Participation of MGP in the myogenic process was investigated in vitro using C2C12 cells, and knockdown of its

Characterization of Pseudogymnoascus destructans conidial adherence to extracellular matrix: Association with fungal secreted proteases and identification of candidate extracellular matrix binding proteins.

Pseudogymnoascus destructans is the etiological agent of white-nose syndrome (WNS), a fungal skin infection of hibernating bats. Pathophysiology of the disease involves disruption of bat metabolism and hibernation patterns, which subsequently causes premature emergence and mortality. However, information on the mechanism(s) and virulence factors of P. destructans infection is minimally known. Typically, fungal adherence to host cells and extracellular matrix (ECM) is the critical first step of the infection. It allows pathogenic fungi to establish colonization and provides an entry for invasio

Melanocyte differentiation and mechanosensation are differentially modulated by distinct extracellular matrix proteins.

Melanocyte dysfunctions can lead to pigmentation disorders or melanoma. Melanocytes interact context-dependently with various types of ECM, including collagens and fibronectin. Alterations in ECM composition and stiffness can impact cell behavior, but their specific roles for melanocyte functions remain unclear. We here exposed melanocytes to different ECM proteins and varying substrate stiffnesses, and identified MITF, a key regulator of melanocyte differentiation and function, as an ECM- and mechanosensitive transcription factor. Moreover, distinct ECM proteins and substrate stiffness engage

Molecular-level studies of extracellular matrix proteins conducted using atomic force microscopy.

Extracellular matrix (ECM) proteins provide anchorage and structural strength to cells and tissues in the body and, thus, are fundamental molecular components for processes of cell proliferation, growth, and function. Atomic force microscopy (AFM) has increasingly become a valuable approach for studying biological molecules such as ECM proteins at the level of individual molecules. Operational modes of AFM can be used to acquire the measurements of the physical, electronic, and mechanical properties of samples, as well as for viewing the intricate details of the surface chemistry of samples. Expression of extracellular matrix-degrading proteins in classic, atypical, and anaplastic meningiomas.

Although the majority of meningiomas, commonly benign tumors (WHO I), are amenable to surgical resection, a percentage of up to 3% will recur as higher-grade meningiomas with potential brain invasion. Our study aims at the in situ identification of proteolytic, extracellular matrix-degrading enzymes in a broad spectrum of meningiomas. We examined 80 meningiomas (50 classic meningiomas WHO I, 19 meningiomas WHO II, including atypical, chordoid, and clear cell types, as well as 11 anaplastic meningiomas WHO III) for the immunohistochemical expression patterns of cathepsin D and metalloproteinase

[Immunohistochemical detection of extracellular matrix proteins in the irradiated rat submandibular gland].

BACKGROUND: Extracellular matrix proteins (EMP) form the main components of basement membranes (BM) and associated structures. Basement membranes influence tissue structure, exchange of substances, and cell growth and differentiation. While BM changes have been reported in malignant tumors and in inflammatory diseases, systematic studies of BM changes following irradiation are rare. Because functional and histomorphological damage to the salivary glands is a well known sequela of radiotherapy in humans, our goal was to describe possible BM changes using an experimental model and immunohistoche
