Extracellular vesicles in neurodegenerative diseases.

Extracellular vesicles (EVs) are released by all neural cells, including neurons, oligodendrocytes, astrocytes, and microglia. The lack of adequate technology has not halted neuroscientists from investigating EVs as a mean to decipher neurodegenerative disorders, still in search of comprehensible pathogenic mechanisms and efficient treatment. EVs are thought to be one of ways neurodegenerative pathologies spread in the brain, but also one of the ways the brain tries to displace toxic proteins, making their meaning in pathogenesis uncertain. EVs, however do reach biological fluids where they ca

Protocol for isolating human extracellular vesicles from nephrotic urine and enriching autoimmunoglobulin-triggered extracellular vesicles.

This protocol describes the isolation of total urinary extracellular vesicles (EVs) from nephrotic patient urines and enrichment of AutoImmunoglobulin-Triggered Extracellular Vesicles (AIT-EVs). Procedures for urine collection, sample processing, buffer preparation, antibody coupling to DynaBeads, urine ultrafiltration, and total EV isolation are provided. The protocol further details EV concentration measurement and selective AIT-EV capture and elution using anti-human IgG4 antibodies. This approach enables reproducible enrichment of AIT-EVs and can be adapted to isolate other EV subtypes by

Vesicle-MaNiA: extracellular vesicles in liquid biopsy and cancer.

Normal and tumor cells shed vesicles to the environment. Within the large family of extracellular vesicles, exosomes and microvesicles have attracted much attention in the recent years. Their interest ranges from mediators of cancer progression, inflammation, immune regulation and metastatic niche regulation, to non-invasive biomarkers of disease. In this respect, the procedures to purify and analyze extracellular vesicles have quickly evolved and represent a source of variability for data integration in the field. In this review, we provide an updated view of the potential of exosomes and mic

Extracellular Vesicles and Their Role in Osteogenesis.

Extracellular vesicles (EVs) have emerged as promising cell-free therapeutic agents for bone regeneration due to their ability to modulate osteogenesis, angiogenesis, and immune responses. This review summarizes the current knowledge on EV biology, including their classification, biogenesis, and molecular cargo, with a particular focus on their role in bone healing. EVs derived from mesenchymal stromal cells and other cell types promote osteoblast differentiation, inhibit osteoclast activity, enhance vascularization, and modulate inflammation through the transfer of bioactive proteins and non-

Extracellular vesicles for the treatment of preeclampsia.

Preeclampsia is a common pregnancy complication, characterized by hypertension, proteinuria, or other organ or system lesions that appear after 20 weeks of gestation. Preeclampsia, a systemic multisystem disease of placental origin, is also one of the leading causes of maternal mortality. Serious complications such as HELLP syndrome and placental abruption that occur in patients with preeclampsia without active treatment will be life threatening. In recent years, extracellular vesicles, especially those derived from mesenchymal stem cells, have been widely used in the treatment of various dise

Extracellular Vesicles and the Oviduct Function.

In mammals, the oviduct (or the Fallopian tube in humans) can be divided into the infundibulum (responsible for oocyte pick-up), ampulla (site of fertilization), isthmus (where preimplantation embryos develop), and uterotubal junction (where embryos transit to the uterus). The oviductal fluid, as well as extracellular vesicles produced from the oviduct epithelial cells, referred to as oEVs, have been shown to improve the fertilization process, prevent polyspermy, and aid in embryo development. oEVs contain molecular cargos (such as miRNAs, mRNAs, proteins, and lipids) that can be delivered and

Extracellular Vesicles in Mycobacteria and Tuberculosis.

Tuberculosis (TB) remains a public health issue causing millions of infections every year. Of these, about 15% ultimately result in death. Efforts to control TB include development of new and more effective vaccines, novel and more effective drug treatments, and new diagnostics that test for both latent TB Infection and TB disease. All of these areas of research benefit from a good understanding of the physiology of Mycobacterium tuberculosis (Mtb), the primary causative agent of TB. Mtb secreted protein antigens have been the focus of vaccine and diagnosis research for the past century. Recen

Osteoarthritis (OA) is a type of chronic joint disease that is characterized by the degeneration and loss of articular cartilage and hyperplasia of the synovium and subchondral bone. There is reasonable knowledge about articular cartilage physiology, biochemistry, and chondrocyte metabolism. However, the etiology and pathogenesis of OA remain unclear and need urgent clarification to guide the early diagnosis and treatment of OA. Extracellular vesicles (EVs) are small membrane-linking particles that are released from cells. In recent decades, several special biological properties have been foun

CRISPR/Cas9 Delivery Using Extracellular Vesicles.

Extracellular vesicles (EVs), particles released from cells, have the potential to become an important in vivo delivery vehicle for CRISPR machinery. Unlike viral vectors such as Adeno-associated virus (AAV), EVs reduce risks associated with immunogenicity, long-term expression, and potential off-target effects. EVs can efficiently deliver CRISPR/Cas9 ribonucleoprotein (RNP) complexes, which provide transient, ready-to-function editing machinery with reduced off-target risk compared with plasmid DNA or mRNA delivery. RNP loading into EVs can occur without specific targeting signals, although s

Extracellular vesicles in viral disease management.

INTRODUCTION/BACKGROUND: Extracellular vesicles (EVs) are non-replicating lipid-bilayered bodies that are naturally released by a cell that aid in various biological functions including cell-to-cell communication. They resemble the cells that they originate from, mimicking their composition and contents. The shared Endosomal Sorting Complex Required for Transport (ESCRT) mechanism between virions and EVs allows EVs to aid in the dispersion and infection of viruses. SCOPE/OBJECTIVES: The aim of this review is to encapsulate important studies that highlight the potential use of EVs in diagnosis

Extracellular Vesicles and Pathological Cardiac Hypertrophy.

Pathological cardiac hypertrophy is a well-recognized risk factor for cardiovascular diseases (CVDs). Although lots of efforts have been made to illustrate the underlying molecular mechanisms, many issues remain undiscovered. Recently, intercellular communication by delivering small molecules between different cell types in the progression of cardiac hypertrophy has been reported, including bioactive nucleic acids or proteins. These extracellular vesicles (EVs) may act in an autocrine or paracrine manner between cardiomyocytes and noncardiomyocytes to provoke or inhibit cardiac remodeling and

Extracellular Vesicles and Ischemic Cardiovascular Diseases.

Characterized by coronary artery obstruction or stenosis, ischemic cardiovascular diseases as advanced stages of coronary heart diseases commonly lead to left ventricular aneurysm, ventricular septal defect, and mitral insufficiency. Extracellular vesicles (EVs) secreted by diverse cells in the body exert roles in cell-cell interactions and intrinsic cellular regulations. With a lipid double-layer membrane and biological components such as DNA, protein, mRNA, microRNAs (miRNA), and siRNA inside, the EVs function as paracrine signaling for the pathophysiology of ischemic cardiovascular diseases

Primary Aldosteronism, Aldosterone, and Extracellular Vesicles.

Primary aldosteronism (PA) is an endocrine related condition leading to arterial hypertension due to inappropriately high and unregulated aldosterone concentration. Recently, a broad spectrum of PA has been recognized, which brings new challenges associated with early identification of this condition that affect renal epithelial and extrarenal tissues. Reports have shown the potential role of extracellular vesicles (EVs) and EV cargo as novel and complementary biomarkers in diagnosis and prognosis of PA. In vivo and in vitro studies have identified specific EV surface antigens, EV-proteins, an

Extracellular Vesicles in Amyotrophic Lateral Sclerosis.

Amyotrophic Lateral Sclerosis is a progressive neurodegenerative disease and is the most common adult motor neuron disease. The disease pathogenesis is complex with the perturbation of multiple pathways proposed, including mitochondrial dysfunction, RNA processing, glutamate excitotoxicity, endoplasmic reticulum stress, protein homeostasis and endosomal transport/extracellular vesicle (EV) secretion. EVs are nanoscopic membrane-bound particles that are released from cells, involved in the intercellular communication of proteins, lipids and genetic material, and there is increasing evidence of

Extracellular Vesicles, Inflammation, and Cardiovascular Disease.

Cardiovascular disease is a leading cause of death worldwide. The underlying mechanisms of most cardiovascular disorders involve innate and adaptive immune responses, and extracellular vesicles are implicated in both. In this review, we describe the mechanistic role of extracellular vesicles at the intersection of inflammatory processes and cardiovascular disease. Our discussion focuses on atherosclerosis, myocardial ischemia and ischemic heart disease, heart failure, aortic aneurysms, and valvular pathology.

Extracellular vesicles and domestic animal reproduction.

Embryo implantation is a complex process in which significant changes occur continually in both the corpora lutea and in the endometrium of females and which varies depending on the embryonic, pre-implantation, or fetal stages. However, at all stages, correct maternal-embryonic communication is essential. In the last few years, a new intercellular communication tool, mediated by extracellular vesicles (EVs), has emerged. Many authors agree on the relevant role of EVs in correct communication between the mother and the embryo, as a fundamental system for the pregnancy to reach term and embryoni
