Mounting evidence highlights preeclampsia (PE) pathogenesis involves gut microbiota dysregulation and trophoblast ferroptosis. However, the mechanistic nexus bridging intestinal homeostasis and placental ferroptosis remains elusive. Here, we found that Faecalibacterium prausnitzii (F. prausnitzii) reduced blood pressure (SBP/DBP), proteinuria, and levels of anti-angiogenic (sFlt-1) and pro-inflammatory (IL-6, TNF-α) mediators in PE rats established using reduced uterine perfusion pressure (RUPP) surgery. Critically, F. prausnitzii elevated fetal survival rate, increased fetal length and weight

BACKGROUND AND AIMS: Recurrent Clostridioides difficile infection (rCDI) results from persistent microbiome dysfunction and impaired colonization resistance. Although fecal microbiota transplantation (FMT) is effective, defined and scalable alternatives are needed. We evaluated whether a single commensal strain could restore key microbiome functions and prevent recurrence.
METHODS: We assessed Faecalibacterium prausnitzii EXL01 in a murine CDI model and a multicenter, open-label single-arm phase I trial including adults with ≥3 CDI episodes. Following vancomycin preconditioning, patients rece

BACKGROUND: Faecalibacterium prausnitzii plays a crucial role in ulcerative colitis (UC) remission, but its action mechanism is unknown. Here, we aimed to explore the potential mechanisms focusing on the interaction of F. prausnitzii with host immune response and its potential modulation on gut microbiome.
METHODS: RNA-seq analysis together with 16S rRNA sequencing and metabolomics were performed in a dextran sodium sulfate (DSS)-induced colitis mouse model followed by F. prausnitzii gavage. To present evidence of sIgA involved in the anti-inflammatory effects of F. prausnitzii, we further ap

Faecalibacterium prausnitzii is prevalent in the human gut and is a potential candidate for next-generation probiotics (NGPs) or biotherapeutics. However, the biodiversity and physiological characteristics of Faecalibacterium prausnitzii remain unclear. This study isolated 26 novel F. prausnitzii strains from human feces using a combination of negative screening and prime-specific PCR amplification (NSPA). Based on a 16S rRNA gene analysis, F. prausnitzii strains can be classified into two main phylogroups (phylogroups I and II), which were further clustered into five subgroups (I-A, II-B, II-

Faecalibacterium prausnitzii, a resident anaerobic bacterium commonly found in healthy gut microbiota, has been proposed as a next generation probiotic with high potential for application in food matrices and pharmaceutical formulations. Despite its recognized health benefits, detailed information regarding its antimicrobial susceptibility profile is still lacking. However, this information is crucial to determine its safety, since the absence of acquired antimicrobial resistance is required to qualify a probiotic candidate as safe for human and animal consumption. Herein, the antimicrobial su

BACKGROUND & AIMS: Faecalibacterium prausnitzii, a highly abundant gut bacterium, has been linked to overall health, and is decreased in several pathologic conditions, such as inflammatory bowel disease (IBD). F prausnitzii has shown anti-inflammatory properties in human and mouse models, notably through the induction of interleukin 10 (IL10) signaling. Here, we investigated which cell types from human blood and intestinal tissue are responsible for producing IL10 induced by F prausnitzii and provide the first mechanistic insights.
METHODS: Immune cells isolated from human blood and intestina

BACKGROUND: Obesity is a global health issue, with sharply increasing rates due to excessive food intake and reduced physical activity, leading to an increased risk of various chronic diseases, such as type 2 diabetes and cardiovascular diseases. Fat distribution plays a significant role in health, with visceral fat being particularly associated with metabolic syndrome. Currently, computed tomography (CT) and magnetic resonance imaging (MRI) are the gold standards for measuring visceral fat, but they are costly and involve radiation risks. The gut microbiota is closely related to obesity, and

Microbiome-host interactions can influence colorectal cancer (CRC) outcomes and the effectiveness of immunotherapy treatment, but the precise mechanisms underlying this are poorly understood. Here we analyse CRC patient cohort data and observe that Facalibacterium prausnitzii abundance in faecal samples correlates with improved CRC survival outcome and immunotherapy response. In vitro assays and experiments in azoxymethane plus dextran sulfate sodium (AOM/DSS) and Apcmin/+ mouse CRC models show that F. prausnitzii extracts have anti-tumour activity. Mass spectrometry identifies F. prausnitzii

Ulcerative colitis (UC) is closely linked to intestinal barrier dysfunction and gut dysbiosis. Bacterial extracellular vesicles (bEVs) act as key mediators of bacteria-host crosstalk, with great potential in regulating host health. Recent studies have shown that EVs derived from gut commensal bacteria offer therapeutic advantages in treating UC. Herein, we explored the therapeutic effect of Faecalibacterium prausnitzii-derived EVs (PEVs) in DSS-induced colitis mice. Results showed PEVs significantly improved intestinal barrier damage, restored Th17/Treg balance and alleviated gut dysbiosis. Fu

Atopic dermatitis (AD) is characterized by a T-helper cell type 2 (Th2) inflammatory response leading to skin damage with erythema and edema. Comparative fecal sample analysis has uncovered a strong correlation between AD and Faecalibacterium prausnitzii strain A2-165, specifically associated with butyrate production. Therefore, understanding the functional mechanisms of crucial enzymes in the butyrate pathway, such as 3-hydroxybutyryl-CoA dehydrogenase of A2-165 (A2HBD), is imperative. Here, we have successfully elucidated the three-dimensional structure of A2HBD in complex with acetoacetyl-C

UNLABELLED: Preweaning diarrhea and intestinal inflammation represent leading causes of morbidity and economic loss in modern dairy calf production systems, causing impaired growth performance, increased mortality, and substantial veterinary costs that collectively constrain farm profitability. Faecalibacterium duncaniae, a key butyrate-producing commensal bacterium, is consistently depleted in diarrheic dairy calves, yet its protective effects on the ruminant intestinal epithelium remain mechanistically understood. In this study, a long-term expandable ruminant colonic organoid model was used

BACKGROUND: Faecalibacterium prausnitzii (F. prausnitzii) has attracted interest in recent research regarding its possible involvement in metabolic dysfunction-associated steatotic liver disease (MASLD). However, their causal relationship is still unclear. METHODS: Microbial biomarkers of gut microbiota associated with obesity-related phenotypes were identified from the GMrepo database. Information was retrieved on specific metabolites and host genes related to F. prausnitzii from the GutMGene database. Subsequently, a high-fat diet (HFD) was administered to C57BL/6 J mice for 16 weeks. The in

This study aimed to investigate the effects of the cell-free supernatant (CFS) of Faecalibacterium prausnitzii on biofilm formation and potential modes of action. Biofilm formed by Listeria monocytogenes poses a threat to food industry due to its resistance to disinfectants and its capacity to contaminate foods. Certain probiotics have been reported to inhibit L. monocytogenes biofilm formation. F. prausnitzii has recently emerged as a promising next generation beneficial microbe. However, its interaction with L. monocytogenes biofilm formation remains unexplored. The anti-biofilm activity of

BACKGROUND AND AIMS: Psoriasis and hidradenitis suppurativa [HS] co-occur more often with inflammatory bowel disease [IBD] than expected, due to shared pathogenic and genetic features. It is known that IBD patients harbour an altered intestinal microbiome characterised by a depletion of Faecalibacterium prausnitzii and increase of Escherichia coli. At present, it is unclear whether a similar intestinal microbiome trend can be identified in IBD-associated skin disorders. We therefore investigated the F. prausnitzii and E. coli abundance in psoriasis and HS, with and without concomitant IBD.

Faecalibacterium prausnitzii is a dominant and health-associated member of the human gastrointestinal tract that is consistently depleted in individuals with metabolic, cardiovascular, and psychiatric disorders. Despite its importance in gut health, little is known about the bacteriophages (or phages) that infect this species. Phages are increasingly recognized as key regulators of bacterial population dynamics in the gut microbiota; however, while prophages of F. prausnitzii have been described, no lytic phages targeting this bacterium have been reported. Here, we describe the isolation and c

Fatty liver hemorrhagic syndrome (FLHS) is a critical disease threatening the laying hen industry. Inulin, a widely used prebiotic, has shown promise in alleviating metabolic disorders, but its role in mitigating FLHS in laying hens is not fully understood. Here, we investigated the effects and underlying mechanisms of inulin-mediated alleviation of FLHS in a high-carbohydrate low-protein diet (HCD)-induced laying hen model. We found that inulin supplementation significantly ameliorated HCD-induced hyperlipidemia, hyperglycemia, hepatic steatosis, liver injury, and oxidative stress. These phen

Alterations in gut microbiota composition are increasingly recognized as key contributors to autoimmune disease pathogenesis. While dominant phyla such as Firmicutes and Bacteroidetes have been extensively studied at the phylum level, the immunomodulatory roles of specific members within these groups particularly the abundant but mechanistically underexplored Faecalibacterium prausnitzii (a member of Firmicutes) and Akkermansia muciniphila (of Verrucomicrobia) remain insufficiently characterized. In particular, current literature primarily focuses on associative findings, and integrated analys

To explore the pathogenic mechanism of Salmonella Enteritidis (SE) infection in chicks and the regulatory role of Faecalibacterium, we established a chick SE infection model, systematically analyzing intestinal damage, systemic inflammation, and cecal microbial community changes. Focusing on the differentially abundant Faecalibacterium, we elucidated its regulatory mechanism via metagenomics, transcriptomics, and serum metabolomics. Results showed that SE successfully colonized the chick cecum, causing significant reductions in the thickness of the intestinal mucosal and muscular layers, a dec

Obsessive-compulsive disorder (OCD) is a mental illness of unwanted recurrent thoughts and compulsive behaviors. The significance of the microbiota-gut-brain axis has been increasingly documented in the manifestation of neuropsychiatric disorders. Herein, we investigated the impact of synbiotic containing Faecalibacterium prausnitzii ATCC 27,766 (F. prausnitzii) and prebiotics [fructooligosaccharides (FOS) and galactooligosaccharides (GOS)] on quinpirole-induced OCD-like symptoms in rats. Repeated quinpirole injections led to compulsive- and anxiety-like behaviors, as rats showed reduced head-

Gut microbiome dysbiosis has been implicated in the pathogenesis of systemic lupus erythematosus (SLE). However, microbiota-targeted therapeutic strategies have been lacking. Here, we report the potential of Faecalibacterium prausnitzii (strain UT1) to ameliorate gut dysbiosis and alleviate disease progression in the B6.Sle1.Yaa male mouse model of SLE. Fecal metagenomes of patients with SLE shifted carbohydrate catabolism from dietary fibers to host glycans, coinciding with depletion of F. prausnitzii. Oral administration of UT1 partially reversed lupus-associated microbiome alterations and r
