ETHNOPHARMACOLOGICAL RELEVANCE: Polygonum multiflorum Radix (PMR) is the dried root tuber of Polygonum multiflorum Thunb., which has been used in the clinic for a variety of pharmacological activities. However, Polygonum multiflorum Radix-induced liver injury (PMR-ILI) has been reported in recent years, which has limited its clinical use to some extent. The occurrence of PMR-ILI is not universal, so finding the different metabolic characteristics between PMR-ILI and Polygonum multiflorum Radix-tolerance group (PMR-T) is very important for the PMR rational clinical application and PMR-ILI early

Polygonum multiflorum (PM) has been reported to cause immune-mediated idiosyncratic liver injury. 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside (THSG) has been identified as a hepatotoxic constituent responsible for PM-induced hepatotoxicity. Covalent protein modification by reactive metabolites plays a crucial role in herb and drug-induced liver injury. Whether it is essential for the hepatotoxicity of THSG needs further clarification. THSG can be hydrolyzed into its aglycone 2,3,5,4'-tetrahydroxystilbene (THS). This study aims to investigate the impact of THS on liver injury and elucidate

Hawthorn (Crataegus pinnatifida Bge.) and Polygonum multiflorum are medicinal and edible plants, with their major constituents being ursolic acid (UA) and 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside (THSG), respectively. Prolonged or high-dose intake of THSG may induce hepatotoxicity. While these plants are commonly combined in dietary formulations, the mechanism behind their compatibility is unclear. This study investigated how UA influences the absorption and disposition of THSG to clarify this compatibility. In vitro intestinal perfusion experiments demonstrated that the hawthorn-proces

This study aimed to optimize the extraction and purification of TSG-enriched Polygonum multiflorum extract and evaluate its therapeutic potential against androgenetic alopecia. P. multiflorum Thunb. has long been used in traditional Chinese medicine to promote hair growth and preserve hair pigmentation. Microwave-assisted extraction (MAE) was optimized to obtain a 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside (TSG)-enriched P. multiflorum extract (PME). The crude extract was further purified by medium-pressure liquid chromatography and semipreparative high-performance liquid chromatography.

2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside (TSG) is the most abundant constituent of Polygonum multiflorum and is exclusively found in this herb. This renowned herbal medicine has been documented to lead to liver damage in humans. The present study demonstrated that TSG underwent 2-step metabolic activation to generate a reactive metabolite, involving both intestinal and hepatic metabolisms. TSG was hydrolyzed to its aglycone 2,3,5,4'-tetrahydroxystilbene (TS) in the intestine, and then, the 2,3 catechol of 2,3,5,4'-tetrahydroxystilbene was metabolized to an ortho-benzoquinone intermediat

ETHNOPHARMACOLOGICAL RELEVANCE: Polygonum multiflorum Thunb. (PM) is a herb, extracts of which have been used as Chinese medicine for years. Although it is believed to be beneficial to the liver, heart, and kidneys, it causes idiosyncratic drug-induced liver injury (DILI).
AIM OF THE STUDY: We propose that the intrinsic DILI caused by natural products in PM (NPPM) is an important complementary mechanism to PM-related herb-induced liver injury, and aim to identify the ingredients with high DILI potential by machine learning methods.
MATERIALS AND METHODS: One hundred and ninety-seven NPPM wer

Safety concerns surrounding skin-lightening agents have intensified following chemical leukoderma linked to rhododendrol. Here, we performed an in vitro safety and hazard profiling comparison of raspberry ketone (RK) and a total anthraquinone fraction from Fallopia multiflora var. cillinerve (Polygonum cillinerve) using an immortalized keratinocyte-melanocyte co-culture model (human HaCaT keratinocytes and murine B10.BR melanocytes, 3:1). Rhododendrol and arbutin were included as contextual references. Following viability-guided range finding, cells were exposed for 48 h and evaluated for mela

Polygonum multiflorum Thunb. (PM) is a traditional Chinese medicine, commonly used to treat a variety of diseases. However, the hepatotoxicity associated with PM hampers its clinical application and development. In this study, we refined the zebrafish hepatotoxicity model with regard to the following endpoints: liver size, liver gray value, and the area of yolk sac. The levels of alanine aminotransferase, aspartate transaminase, albumin, and microRNAs-122 were evaluated to verify the model. Subsequently, this model was used to screen different extracts, components, and constituents of PM, incl

BACKGROUND: Polygoni Multiflori Caulis (PMC), the dried stem of Pleuropterus multiflorus (Thunb.) Turcz. ex Nakai (PM), is a fundamental traditional Chinese medicine utilized for treating sleep disorders. Despite its medicinal importance, species confusion exists due to the taxonomic synonymy of Polygonum multiflorum var. hypoleucum (PMH) and Pleuropterus angulatus (PA) with PM. Furthermore, conventional quality standards often restrict PMC collection to specific stem diameters. This study evaluatedthe chemical and genetic diversity of PMC and its related taxa and assessed the authenticity of

ETHNOPHARMACOLOGICAL RELEVANCE: The liver toxicity of Reynoutria multiflora (Thunb.) Moldenke. (Polygonaceae) (Polygonum multiflorum Thunb, PM) has always attracted much attention, but the related toxicity materials and mechanisms have not been elucidated due to multi-component and multi-target characteristics. In previous hepatotoxicity screening, different components of PM were first evaluated and the hepatotoxicity of component D [95% ethanol (EtOH) elution] in a 70% EtOH extract of PM (PM-D) showed the highest hepatotoxicity. Furthermore, the main components of PM-D were identified and the

In recent years, liver injury induced by Traditional Chinese Medicines (TCMs) has gained increasing attention worldwide. Assessing the hepatotoxicity of compounds in TCMs is essential and inevitable for both doctors and regulatory agencies. However, there has been no effective method to screen the hepatotoxic ingredients in TCMs available until now. In the present study, we initially built a large scale dataset of drug-induced liver injuries (DILIs). Then, 13 types of molecular fingerprints/descriptors and eight machine learning algorithms were utilized to develop single classifiers for DILI,

The processing of traditional Chinese medicine (TCM) plays an important role in the clinical application, which usually has the function of "increasing efficiency and reducing toxicity". Polygonum multiflorum (PM) has been reported to induce hepatotoxicity, while it is believed that the toxicity is reduced after processing. Studies have shown that the hepatotoxicity of PM is closely related to the changes in chemical components before and after processing. However, there is no comprehensive investigation on the chemical changes of PM during the processing progress. In this research, we establi

Macrophage polarization has been linked to hepatotoxicity induced by raw Polygonum multiflorum (RPM) and Polygonum multiflorum praeparata (PMP), but the regulatory mechanisms behind this remain unclear. This study aims to investigate the regulatory effects of RPM and PMP on M2 macrophages and the potential mechanisms. Sprague-Dawley rats were exposed to RPM and PMP under lipopolysaccharide (LPS) stimulation. RAW264.7 cells induced with IL-4 were treated with RPM and PMP. Under LPS stimulation, both RPM and PMP increased serum enzyme levels and pro-inflammatory factor levels and induced histopa
