The cellular transformation of injected colloidal iron complexes into ferritin and hemosiderin in experimental animals; a study with the aid of electron microscopy.

As revealed by electron microscopy and electron diffraction, the physical state of ferric hydroxide micelles contained in iron-dextran, saccharated iron oxide, and hydrous ferric oxide ("ferric hydroxide") differs notably from the state of the ferric hydroxide in ferritin or hemosiderin. By virtue of this difference one can trace the intracellular transformation of colloidal iron, administered parenterally, into ferritin and hemosiderin. One hour after intraperitoneal injection of iron-dextran or saccharated iron oxide into mice, characteristic deposits were present in splenic macrophages, in

Safety in iron management.

Intravenous (IV) iron therapy has become an integral part of hemodialysis management during the past several decades, and the National Kidney Foundation-Kidney Disease Outcomes Quality Initiative guidelines recognize that most patients undergoing hemodialysis will require IV iron therapy on a regular basis to reach target hemoglobin (Hgb) levels. There now are three IV iron compounds available in the United States: iron dextran, sodium ferric gluconate, and iron sucrose. Although all have been proven effective for increasing Hgb/hematocrit levels, recent data show differences in their relative

Structural and Spectroscopic Characterization of a High-Spin {FeNO}(6) Complex with an Iron(IV)-NO(-) Electronic Structure.

Although the interaction of low-spin ferric complexes with nitric oxide has been well studied, examples of stable high-spin ferric nitrosyls (such as those that could be expected to form at typical non-heme iron sites in biology) are extremely rare. Using the TMG3 tren co-ligand, we have prepared a high-spin ferric NO adduct ({FeNO}(6) complex) via electrochemical or chemical oxidation of the corresponding high-spin ferrous NO {FeNO}(7) complex. The {FeNO}(6) compound is characterized by UV/Visible and IR spectroelectrochemistry, Mössbauer and NMR spectroscopy, X-ray crystallography, and DFT c

Aza-crown compounds synthesised by the self-condensation of 2-amino-benzyl alcohol over a pincer ruthenium catalyst and applied in the transfer hydrogenation of ketones.

A well-defined PNN-Ru catalyst was revisited to self-condense 2-aminobenzyl alcohol in forming a series of novel aza-crown compounds [aza-12-crown-3 (1), aza-16-crown-4 (2) and aza-20-crown-5 (3)]. All aza-crown compounds are separated and determined by NMR, IR, and ESI-MS spectroscopy as well as X-ray crystallography, indicating the saddle structure of 1 and the twisted 1,3-alternate conformation structure of 3. These aza-crown compounds have been explored to study ferric initiation of transfer hydrogenation (TH) of ketones into their corresponding secondary alcohols in the presence of 2-prop

Recent Advances in Detection, Isolation, and Imaging Techniques for Sulfane Sulfur-Containing Biomolecules.

Hydrogen sulfide and its oxidation products are involved in many biological processes, and sulfane sulfur compounds, which contain sulfur atoms bonded to other sulfur atom(s), as found in hydropersulfides (R-S-SH), polysulfides (R-S-Sn-S-R), hydrogen polysulfides (H2Sn), etc., have attracted increasing interest. To characterize their physiological and pathophysiological roles, selective detection techniques are required. Classically, sulfane sulfur compounds can be detected by cyanolysis, involving nucleophilic attack by cyanide ion to cleave the sulfur-sulfur bonds. The generated thiocyanate

Evaluation of the pro-oxidant and antioxidant actions of L-DOPA and dopamine in vitro: implications for Parkinson's disease.

The antioxidant and pro-oxidant properties of L-DOPA and dopamine were investigated in vitro. Both compounds inhibited the peroxidation of ox-brain phospholipids, with IC50 values of 8.5 microM for dopamine and 450 microM for L-DOPA. Dopamine and L-DOPA reacted with trichloromethyl peroxyl radicals (CCl3O2.) with rate constants of 2.1 x 10(7)M-1s-1 and 1.3 x 10(7)M-1s-1 respectively. The effects of dopamine and L-DOPA on iron ion-dependent hydroxyl radical generation from H2O2 were complex. In general, low concentrations stimulated OH. formation in the presence of ferric-EDTA and, in the case

Mechanisms of myeloperoxidase catalyzed oxidation of H2S by H2O2 or O2 to produce potent protein Cys-polysulfide-inducing species.

The interaction of heme proteins with hydrogen sulfide is gaining attention as an important element in sulfide-mediated protection against oxidative stress and in regulation of redox signaling. In our previous study we reported the efficient reversible inhibition of myeloperoxidase (MPO) activity by sulfide and the kinetics of the reactions of sulfide with ferric MPO, Compound I and Compound II. Here we provide several lines of evidence that a central intermediate species in the turnover of MPO by sulfide is the Compound III state. Compound III is formed in the reactions of sulfide with ferric

Precisely Identifying the Sources of Magnetic Particles by Hierarchical Classification-Aided Isotopic Fingerprinting.

Magnetic particles (MPs), with magnetite (Fe3O4) and maghemite (γ-Fe2O3) as the most abundant species, are ubiquitously present in the natural environment. MPs are among the most applied engineered particles and can be produced incidentally by various human activities. Identification of the sources of MPs is crucial for their risk assessment and regulation, which, however, is still an unsolved problem. Here, we report a novel approach, hierarchical classification-aided stable isotopic fingerprinting, to address this problem. We found that naturally occurring, incidental, and engineered MPs hav

Beyond iron: metal-binding activity of the Pseudomonas quinolone signal-motif.

The Pseudomonas quinolone signal (PQS) is an important quorum sensing signal controlling virulence of the human pathogen Pseudomonas aeruginosa. PQS also exhibits multiple additional biological functions for P. aeruginosa, including the trapping of ferric iron. The PQS-motif has proven as privileged structure with great potential which is why we here explored the synthesis of two different types of crosslinked dimeric PQS-motifs as potential iron chelators. These compounds indeed chelated ferric iron and produced colorful and fluorescent complexes also with other metal ions. Inspired by these

Interaction of metals at the reaction center of lipoamidase.

Various metals have been shown to inhibit porcine brain lipoamidase activity at 0.1 mM, but not ferrous and ferric ions. However, in the presence of ethylenediamine tetraacetic acid (EDTA) 0.1 mM iron ions did inhibit the activity. No other metals exhibited this type of increased inhibition with the addition of EDTA. The ferric- and ferrous-EDTA compounds were equally effective. Various Fe-containing compounds also inhibited the enzyme activity, the order of inhibition being: EDTA greater than o-phenanthroline greater than azide greater than citrate. Hemin also inhibited the enzyme activity st

The metal-organic framework MIL-100(Fe) and ferric citrate as potential counteracting biomaterials against osteoarthritis-induced articular inflammation and fibrosis.

Osteoarthritis (OA) is a degenerative and deteriorating disease. The accompanying pain, stiffness, inflammation of the synovial joint, and the loss of articular function compromise the mobility, stability, and routine of patients' lives. As inflammation-associated structural destruction, calcification, and fibrosis are common features observed in OA patients, nonsteroidal anti-inflammatory drugs have been first-line analgesics for the management of OA in both humans and animals. In this study, we mimicked in vitro OA-induced articular inflammation and fibrosis and evaluated the phenotypical al

The determination of ferric iron in plants by HPLC using the microbial iron chelator desferrioxamine E.

Common methods for plant iron determination are based on atomic absorption spectroscopy, radioactive measurements or extraction with subsequent spectrophotometry. However, accuracy is often a problem due to background, contamination and interfering compounds. We here describe a novel method for the easy determination of ferric iron in plants by chelation with a highly effective microbial siderophore and separation by high performance liquid chromatography (HPLC). After addition of colourless desferrioxamine E (DFE) to plant fluids, the soluble iron is trapped as a brown-red ferrioxamine E (Fox

An invertebrate model in examining the effect of acute ferric iron exposure on proprioceptive neurons.

Iron is an essential element for plant and animal life and is found in soil, fresh waters and marine waters. The Fe3+ ion is a vital prosthetic group and cofactor to mitochondrial electron transport complexes and numerous proteins involved in normal functioning. Despite its importance to life-sustaining processes, overexposure results in toxicity. For example, ferric iron (Fe3+) accumulation in the mammalian central nervous system is associated with various neurological disorders. Although current literature addresses the long-term effects of Fe3+ overload, fewer studies exist examining the ef

Caught in the act: Monitoring OO bond cleavage in Acylperoxoferric cytochrome P450cam to form compound I in real time.

While monitoring the reaction of ferric cytochrome P450cam (Cyp101) with substituted peroxybenzoic acids using rapid-scanning, stopped-flow (RSSF) spectroscopy, an intermediate appears en route to formation of the high-valent moiety known as Compound I [Fe(IV)=O/porphyrin radical cation] that is thought to be the key catalytic species for O-atom transfer to substrate. We have previously suggested (Spolitak, T., Dawson, J.H., Ballou, D.P., J. Biol. Chem.2005, 280, 20,300-20,309) that this species is an acylperoxo-ferric heme adduct that subsequently undergoes OO bond cleavage to generate Compou

Ferric Pyrophosphate in Iron Deficiency Anemia Management: An Updated Review of Current Practices, Bioavailability Enhancement Techniques, and Future Directions.

According to the World Health Organization, Anemia is a health concern that impacts a substantial number of individuals globally, with 50% of cases due to iron deficiency and the remaining 50% being caused by other conditions and vitamin deficiencies. Iron deficiency anemia can cause several health issues, such as weakness, exhaustion, poor cognitive function, and a higher chance of pregnancy difficulties. Iron supplementation, particularly through dietary sources and supplement formulations, is fundamental in addressing this condition and is favored for managing mild to moderate cases. Ferrou

An open-label, crossover study of a new phosphate-binding agent in haemodialysis patients: ferric citrate.

BACKGROUND: Hyperphosphataemia contributes to secondary hyperparathyroidism and renal osteodystrophy in patients with end-stage renal disease (ESRD). Calcium salts are widely employed to bind dietary phosphate (P) but they may promote positive net calcium balance and metastatic calcification. We recently reported that ferric compounds bind intestinal phosphate in studies of normal and azotemic rats. METHODS: To extend this observation, we performed an open-label, random order, crossover comparison study of ferric citrate and calcium carbonate in haemodialysis patients from two teaching hospit

Colloidal stability and chemical reactivity of complex colloids containing Fe³⁺.

The reactivity of iron contained within insoluble colloidal metal-pyrophosphate salts was determined and compared to the reactivity of a soluble iron salt (FeCl3). As a model system for the reactivity of iron in food products, the formation of an iron-polyphenol complex was followed with spectrophotometry. Three types of systems were prepared and their colloidal stability and reactivity studied: Fe(3+) pyrophosphate, protein-coated Fe(3+) pyrophosphate and mixed-metal pyrophosphates containing Fe(3+) and a second cation M. The additional cation used was either monovalent (sodium) or divalent (

Investigating Soil Organic Matter Complexation in Natural Analog Systems Using Geoelectrical Methods.

Soil organic matter (SOM) interactions with minerals, such as ferrihydrite, are fundamental to processes that control carbon sequestration, nutrient cycling, and maintaining soil health. This study investigates the sensitivity of geoelectrical methods, such as induced polarization (IP), to the formation of organo-mineral complexes, focusing on ferrihydrite (a common mineral in soils), and pentaglycine (a component of the SOM). Multiscale experiments (millifluidic system and column) using ferrihydrite-coated sand, ceramic beads, and a polydimethylsiloxane (PDMS) pore network show a large increa

Ligand binding to the haem-copper binuclear catalytic site of cytochrome bo, a respiratory quinol oxidase from Escherichia coli.

The Escherichia coli quinol oxidase, cytochrome bo, is closely related to the cytochrome-c oxidase, cytochrome aa3 and reacts with ligands to the high-spin ferric haem or the high-spin ferric-cupric binuclear catalytic site in similar ways. Cyanide reacts with the isolated enzyme to give a low-spin complex, manifested by a red shift in the Soret band, the loss of an absorption band at 630 nm and the appearance of a low-spin ferric haem EPR resonance at g = 3.3. Sulphide also elicits a low-spin complex, whereas azide gives a mixture of low-spin and high-spin species. Formate and fluoride (and a
