Magnetic "fishing" assay to screen small-molecule mixtures for modulators of protein-protein interactions.

Protein-protein interactions are an intricate part of biological pathways and have become important targets for drug discovery. Here we present a two-stage magnetic bead assay to functionally screen small-molecule mixtures for modulators of protein-based interactions, with simultaneous affinity-based isolation of active compounds and identification by mass spectrometry. Proteins of interest interact in solution prior to the addition of Ni(II)-functionalized magnetic beads to recover an intact protein-protein complex through affinity capture of a polyhistidine-tagged primary target ("protein-co

Doublecortin in the Fish Visual System, a Specific Protein of Maturing Neurons.

Doublecortin (DCX) is a microtubule associated protein, essential for correct central nervous system development and lamination in the mammalian cortex. It has been demonstrated to be expressed in developing-but not in mature-neurons. The teleost visual system is an ideal model to study mechanisms of adult neurogenesis due to its continuous life-long growth. Here, we report immunohistochemical, in silico, and western blot analysis to detect the DCX protein in the visual system of teleost fish. We clearly determined the expression of DCX in newly generated cells in the retina of the cichlid fis

Parathyroid hormone-related protein is a factor in normal fish pituitary.

Using antibodies to the amino-terminal region of human parathyroid hormone-related protein (PTHrP) we have demonstrated PTHrP immunoreactivity in pituitaries and plasma of the sea bream (Sparus aurata). Pituitary cells at two distinct locations contained immunodetectable PTHrP; an anterior group in the rostral pars distalis which also contained immunoreactive thyroid stimulating hormone (TSH), and a posterior group lying at the border of the pars intermedia and proximal pars distalis between cells which stained with antibody to human corticotrophin-like intermediate lobe peptide. Fish protein hydrolysates as a health-promoting ingredient-recent update.

Dietary habits and lifestyle-related diseases indicate that food has a direct impact on individual health. Hence, a diet containing essential nutrients is important for healthy living. Fish and fish products are important in diets worldwide because of their nutritional value, especially their easily digestible proteins with essential amino acids. Similarly, fish protein hydrolysate (FPH) obtained from fish muscle and by-products has been reported to exhibit various biological activities and to have functional properties, which make FPH a suitable nutraceutical candidate. This review focuses on

Abundant fish protein inhibits α-synuclein amyloid formation.

The most common allergen in fish, the highly-abundant protein β-parvalbumin, forms amyloid structures as a way to avoid gastrointestinal degradation and transit to the blood. In humans, the same amyloid structures are mostly associated with neurodegenerative disorders such as Alzheimer's and Parkinson's. We here assessed a putative connection between these amyloids using recombinant Atlantic cod β-parvalbumin and the key amyloidogenic protein in Parkinson's disease, α-synuclein. Using a set of in vitro biophysical methods, we discovered that β-parvalbumin readily inhibits amyloid formation of

Freezing of a fish antifreeze protein results in amyloid fibril formation.

Amyloid is associated with a number of diseases including Alzheimer's, Huntington's, Parkinson's, and the spongiform encephalopathies. Amyloid fibrils have been formed in vitro from both disease and nondisease related proteins, but the latter requires extremes of pH, heat, or the presence of a chaotropic agent. We show, using fluorescence spectroscopy, electron microscopy, and solid-state NMR spectroscopy, that the alpha-helical type I antifreeze protein from the winter flounder forms amyloid fibrils at pH 4 and 7 upon freezing and thawing. Our results demonstrate that the freezing of some pro

Development of DNA vaccines encoding ribosomal proteins (RplL and RpsA) against Nocardia seriolae infection in fish.

Nocardia seriolae, a Gram-positive pathogen, has been identified as the causative agent of fish nocardiosis. DNA vaccination has been proven to be effective in conferring protection against bacterial infection in fish. The 30S ribosomal protein S1 (RpsA) and 50S ribosomal protein L7/L12 (RplL) were identified to be the common immunodominant antigens of three fish pathogenic Nocardia (N. seriolae, N. salmonicida and N. asteroids) by immunoproteomics profiling in our previous study. In current study, the immunogenicity and protective efficacy of two DNA vaccines encoding RplL and RpsA were evalu

Expression of the Major Vault Protein (MVP) and Cellular Vault Particles in Fish.

Cellular vaults are ubiquitous 13 mega Da multi-subunit ribonuceloprotein particles that may have a role in nucleocytoplasmic transport. Seventy percent of the vault's mass consists of a ≈100 kDa protein, the major vault protein (MVP). In humans, a drug resistance-associated protein, originally identified as lung resistance protein in metastatic lung cancer, was ultimately shown to be the previously described MVP. In this study, a partial MVP sequence was cloned from channel catfish. Recombinant MVP (rMVP) was used to generate a monoclonal antibody that recognizes full length protein in distan

Revalorization of fish viscera as a sustainable source of proteins, lipids and polysaccharides in the food industry.

Fish viscera, a fish processing by-product, is currently underutilized, despite its complex composition of high value-added components, such as proteins, lipids, and polysaccharides. This work explores the potential for revalorizing fish viscera as a sustainable source of proteins, lipids and polysaccharides in the food industry. Furthermore, their potential food applications and future perspectives are discussed. Fish viscera, constituting 12-18% of the total fish weight, is abundant in proteins, lipids, and polysaccharides. Protein hydrolysates derived from fish viscera exhibit diverse bioac

Development of liquid chromatography-tandem mass spectrometry methods for the quantitation of Anisakis simplex proteins in fish.

The parasite Anisakis simplex is present in many marine fish species that are directly used as food or in processed products. The anisakid larvae infect mostly the gut and inner organs of fish but have also been shown to penetrate into the fillet. Thus, human health can be at risk, either by contracting anisakiasis through the consumption of raw or under-cooked fish, or by sensitisation to anisakid proteins in processed food. A number of different methods for the detection of A. simplex in fish and products thereof have been developed, including visual techniques and PCR for larvae tracing, an

Natural occurrence of metallothioneinlike proteins in liver tissues of four fish species from the northeast Mediterranean Sea.

Fish with different ecological needs were captured from the northeast Mediterranean Sea to determine metallothioneinlike proteins in their livers. Heat-treated liver samples were run on a column packed with Sephadex G-75 (Sigma, Germany) for subsequent determination of metals, sulfhydryl, and absorbance at 254 and 280 nm. Liver cytosols of Sparus auratus and Mullus barbatus had three heat-stable protein peaks, whereas Mugil cephalus and Atherina hepsetus had two. The third protein peak, containing metallothionein-like proteins (5000 to 6000 Da), was present in all fish. Only high- and low-mole

Cryoprotective effects of fish skin collagen peptide on frozen gluten proteins: From conformational changes to functional properties.

Fish skin collagen peptides (FSCP) are mainly extracted from fish processing residues and exhibit outstanding biological activity and excellent functional properties. This study conducted a systematic evaluation of the effects of FSCP incorporation (0-2.0%, w/w) on the multiscale structural characteristics and functional performance of gluten proteins during freezing. Results demonstrated that FSCP exhibited pronounced ice recrystallization inhibition activity, effectively suppressing water migration, reducing the content of freezable water, and preserving the structural integrity of gluten pr

SGIV and RGNNV regulate fish innate immune responses by mediating ubiquitinated modifications of proteasome and ribosomal proteins.

Singapore grouper iridovirus (SGIV) and red-spotted grouper nervous necrosis virus (RGNNV) have been identified as the predominant DNA and RNA viral pathogens, respectively, affecting grouper populations. Protein ubiquitination, a crucial post-translational modification, plays pivotal regulatory roles in both host innate immune responses and viral replication processes. In the present study, we employed quantitative proteomics approaches to systematically identify and characterize alterations in ubiquitination-modified proteins in grouper spleen (Epinephelus akaara, GS) cells following SGIV an

2D DIGE/MS to investigate the impact of slaughtering techniques on postmortem integrity of fish filet proteins.

Two-dimensional difference gel electrophoresis (2D DIGE) was applied to investigate the impact of slaughtering on the postmortem integrity of muscle tissue proteins in European sea bass (Dicentrarchus labrax). Three different slaughtering techniques were evaluated: asphyxia in air (AA), asphyxia in ice (AI), and spinal cord severance (SCS). Principal components analysis (PCA) revealed a significant divergence of SCS samples, whereas AA and AI samples, although grouped separately, were less divergent and could be included in a single asphyxia cluster. In terms of single proteins, the most signi

Nutritional evaluation of fish protein hydrolysate and its application in piglet production.

This study was conducted to determine the digestible energy (DE), metabolizable energy (ME), and ileal digestibility of amino acids (AA) of fish protein hydrolysate (FPH), and to evaluate the effects of FPH on the performance of piglets. In Exp. 1, the available energy content of FPH was determined by difference methods: 12 barrows with an initial body weight (BW) of 40.0 ± 2.1 kg were randomly assigned into two dietary treatments with six replicates per treatment, the treatments contained a corn basal diet and an experimental diet in which 20% corn was replaced by FPH. Autoimmune hepatitis associated with the odour of fish food proteins: a causal relationship or just a mere association?

We present the case of a 15-year-old boy [HLA phenotype: A 1, 25 (10); B 18, 8; C 7; DR 17 (3), 6] with classic (type 1) autoimmune hepatitis presumably caused by a long-term exposure to the strong odour of food fed to a large number of tropical fish which that the boy kept in tanks in his bedroom. The boy presented with a history of recent symptoms of common cold, and a high cytomegalovirus-IgG titer, both known to activate proinflammatory cytokines. Separation and identification of ACE inhibitory peptides from lizard fish proteins hydrolysates by metal affinity-immobilized magnetic liposome.

Purification of peptides responsible for angiotensin I-converting enzyme (ACE) inhibitory activity from highly complex protein hydrolysates is difficult. Affinity chromatography is a powerful method for purification of peptides. In this study, a metal affinity-immobilized magnetic liposome (MA-IML) was prepared using lipid, N-hexadecyl iminodiacetic acid (HIDA) and magnetic nanoparticles made of FeCl3·6H2O and FeCl2·4H2O as main materials. MA-IML was used to adsorb ACE inhibitory peptides from lizard fish proteins hydrolysates. The optimal pH of adsorption solution was 8.5. The peptide sample
