Toward Relative Redox Potential Predictions in Flavoproteins: Treatment of a Charge-Changing Mutation in Alchemical Free Energy Simulations.

Redox enzymes play an essential role in nature and in biotechnological applications such as (photo)biocatalysis and biosensing. Understanding how a protein's sequence and structure tune its redox potential is very valuable for engineering proteins with tailored (photo)redox properties. Since protein redox potential measurements are laborious, reliable redox potential computations present an attractive alternative. However, redox calculations come with their own set of challenges, such as ensuring adequate sampling and accurate force fields. Dealing with charge-changing states introduces additi

19F NMR probes of conformational change in a bifurcating electron transfer flavoprotein.

Bifurcating electron transfer flavoproteins (bETFs) accept a pair of modestly reducing electrons and produce a more potent electron carrier based on energy derived from favorable transfer of the other electron. A domain-scale conformational change is believed to gate electron transfer within the bETF, allowing only one electron to use the favorable path. 80° rotation of the so-called head domain carries the electron transfer flavin adenine dinucleotide (ET-FAD) from near the bifurcating FAD in bETF's closed conformation to a position >35 Å away in bETF's open conformation. In this, the ET-FAD

Measurement of fluorescence changes of NAD(P)H and of fluorescent flavoproteins in saponin-skinned human skeletal muscle fibers.

Saponin-skinned human muscle fibers from M. vastus lateralis were immobilized in a quartz capillary to detect the fluorescence changes of NAD(P)H and of fluorescent flavoproteins. To get sufficient intense fluorescence signals from a small amount of muscle tissue the NAD(P)H fluorescence was excited by means of an HeCd laser at 325 nm and the flavoprotein fluorescence by an argon-ion laser at 454 nm or by the second wavelength of a HeCd laser at 442 nm. Using this experimental setup the fluorescence spectra of NAD(P)H, of alpha-lipoamide dehydrogenase and of electron-transfer flavoprotein were

Characterization of Thermal Recovery Activation in Fast Photochromic Flavoprotein Charge-Transfer Complexes.

A promising fast, negative, red-absorbing photochromic system based on an intraprotein intermolecular charge transfer (CT) complex between a flavin cofactor and a substrate-analogue inhibitor MXA (methylthioacetate or methylselenoacetate; X = S, Se) within monomeric sarcosine oxidase (MSOX) displays MXA-independent, barrierless, high-yield femtosecond photoactivation associated with CT-interaction disruption coupled to MXA isomerization. Assessing the origin of the known MXA-dependence of the nanosecond thermal recovery requires a very precise determination of activation energy barriers. Here,

Flavoprotein Fluorescence Imaging in Stargardt Disease: Linking Metabolic Stress to Structural Damage.

PURPOSE: Stargardt disease (SD) is an inherited retinal disorder that leads to progressive vision loss. Currently, no approved treatments exist. Identifying early metabolic changes in the retina could be critical for the development of new therapies. Flavoprotein fluorescence (FPF) imaging has the potential to serve as a non-invasive biomarker for detecting these early changes before structural damage is evident. Therefore, this study evaluated FPF patterns in patients with SD and correlated these findings with structural imaging modalities, specifically fundus autofluorescence (FAF) and optic

Functional Imaging of Mitochondria in Age-Related Macular Degeneration Using Flavoprotein Fluorescence.

PURPOSE: Oxidative stress-induced mitochondrial dysfunction is implicated in the pathogenesis of age-related macular degeneration (AMD). Oxidized mitochondrial flavoprotein fluorescence (FPF) may serve as a quantifiable biomarker of oxidative stress, reported as either mean score for the entire image (intensity) or variability (heterogeneity). This study examines FPF intensity and heterogeneity across a large patient cohort of various Beckman stages of AMD. METHODS: This study enrolled patients with isolated AMD and healthy control patients with no retinopathy between 2018 and 2021. Multivari

Electrochemical aptasensor detection of electron transfer flavoprotein subunit beta for leptospirosis diagnosis.

Electron transfer flavoprotein subunit beta (ETFB) of Leptospira interrogans is a biomarker for diagnosing leptospiral infection. Thus, the ETFB-specific nuclease-resistant RNA aptamer ETFB3-63 was developed and used in an electrochemical aptasensor to assay ETFB. Although the majority of reported biosensors detect various genes and antibodies of L. interrogans, this is the first attempt to construct an electrochemical biosensor to detect ETFB protein for the diagnosis of leptospiral infection. The ETFB protein can be detected without any extraction phase. In this assay, a single-stranded DNA

Carotid body type I cells engage flavoprotein and Pin1 for oxygen sensing.

Carotid body (CB) type I cells sense the blood Po2 and generate a nervous signal for stimulating ventilation and circulation when blood oxygen levels decline. Three oxygen-sensing enzyme complexes may be used for this purpose: 1) mitochondrial electron transport chain metabolism, 2) heme oxygenase 2 (HO-2)-generating CO, and/or 3) an NAD(P)H oxidase (NOX). We hypothesize that intracellular redox changes are the link between the sensor and nervous signals. To test this hypothesis type I cell autofluorescence of flavoproteins (Fp) and NAD(P)H within the mouse CB ex vivo was recorded as Fp/(Fp+NA

Studies on the inactivation of the flavoprotein D-amino acid oxidase from Trigonopsis variabilis.

Inactivation of D-amino acid oxidase occurred by different mechanisms. The enzyme showed a rapid loss of activity in the presence of micromolar amounts of Cu2+ and Hg2+. It was also sensitive to oxidative inactivation by Fe2+ and H2O2 when both reagents were added in millimolar amounts. When oxidatively inactivated D-amino acid oxidase and a corresponding non-treated control were modified with the sulfhydryl-modifying, fluorescent reagent monobromobimane and subsequently digested with endoproteinase Glu-C, Cys-298 was identified to be a target for oxidative modification according to difference

The role of tryptophan residues and hydrophobic interaction in the binding of riboflavin in egg-yolk flavoprotein.

Egg-yolk flavoprotein has 7.2 tryptophan residues exposed, while the apoprotein shows an apparent exposure of 80 percent of these (5.7 residues) with dimethylsulphoxide as the perturbant. In the apoprotein at pH 6.9 only 4 groups are perturbed to ethylene glycol, 3.2 to glycerol and 1.4 to sucrose. Diminishing estimates of exposure obtained with increasing molecular diameter of the perturbant suggests that part of indole chromophores of apoprotein are located in "crevices" of the protein molecule. The apoprotein was treated with 2-hydroxy-5-nitrobenzyl bromide, H2O2 and N-bromosuccinimide unde

Putative Mechanisms for Initial Reaction of Anaerobic Benzene Degradation Presumably Involving a Flavoprotein.

Biodegradation of benzene is most challenging among the plethora of hydrocarbons due to its high chemical stability. While aerobic bacteria employ O2-dependent oxygenases to overcome the high energy barrier, anaerobic benzene degradation remains enigmatic. Here we achieve an anaerobic enrichment, which is clonal with respect to the benzene-degrading, sulphate-reducing, marine bacterium BzS1, from a previously reported enrichment culture. We investigate this clonal enrichment culture by metabolite and proteogenomic analyses. BzS1 has a tailor-made catabolic network for utilising benzene, toluen

Flavoprotein fluorescence elevation is a marker of mitochondrial oxidative stress in patients with retinal disease.

PURPOSE: Recent studies of glaucoma, age-related macular degeneration, and diabetic retinopathy have demonstrated that flavoprotein fluorescence (FPF) can be utilized non-invasively as an indicator of mitochondrial oxidative stress in the retina. However, a comprehensive assessment of the validity and reliability of FPF in differentiating between healthy and diseased eyes across multiple disease states is lacking. Here, we evaluate the sensitivity and specificity of FPF in discriminating between healthy and diseased eyes in four leading causes of visual impairment worldwide, one of which has n

Functional imaging of mitochondria in genetically confirmed retinal dystrophies using flavoprotein fluorescence.

BACKGROUND: Whether by indirect oxidative stress or direct genetic defect, various genetic retinal dystrophies involve mitochondrial stress. Mitochondrial flavoprotein fluorescence (FPF), reported as either average signal intensity or variability (heterogeneity), may serve as a direct, quantifiable marker of oxidative stress. MATERIALS AND METHODS: This observational study enrolled patients with genetically confirmed retinal dystrophies between January and December 2021. Patients with concomitant maculopathy and ocular hypertension were excluded. Patients were FPF imaged with OcuMet Beacon® t

Discovery of chalcone derivatives as antileishmanial agents: phenotypic screening, exploratory docking and enzymatic assays on trypanothione reductase and related flavoproteins.

Leishmaniasis is a neglected tropical disease caused by intracellular protozoa of the genus Leishmania. As current treatments of leishmaniasis are limited by toxicity, drug resistance, and difficult administration, novel therapies targeting parasite-specific pathways are needed. In this context, 25 flavonoid-related compounds, including 19 chalcone analogues, were evaluated for their antiparasitic activity against Leishmania. Among the tested compounds, chalcones 1, 2, 5, and 12 showed potent inhibitory effects on the growth of promastigotes and amastigotes of Leishmania infantum as well as ot

Electron Transfer Flavoprotein Subunit Beta Is a Candidate Endothelial Cell Autoantigen in Behçet's Disease.

Behçet's disease (BD) is a chronic inflammatory disease with multisystem involvement, and it is listed as a rare disease in the United States but is common in the Middle East, China, and Japan. The aim of this study was to identify novel autoantigens in Chinese patients with BD. First, the candidate autoantigens were screened by Western blotting, and the sequences of putative antigens were identified by LC-MALDI-TOF/TOF mass spectrometry. Next, the screened protein was cloned, expressed and purified. Then, an optimized ELISA was developed, and the serological criteria were evaluated using a la

Correlation of Mitochondrial Flavoprotein Fluorescence and Sectoral OCT in Patients with Glaucoma, Suspects, and Ocular Hypertension.

PURPOSE: Flavoprotein fluorescence (FPF) can be measured using a noninvasive investigational device, the OcuMet Beacon (OcuSciences). The Beacon uses blue light to stimulate oxidized mitochondrial flavoproteins, which subsequently emit green autofluorescence. As mitochondrial metabolic stress may reflect axonal damage in glaucomatous eyes, this cross-sectional, exploratory study investigated relationships between optical coherence tomography (OCT) structural parameters and FPF values in patients with glaucoma, ocular hypertension, and glaucoma suspects. Alternative pyrimidine biosynthesis protein ApbE is a flavin transferase catalyzing covalent attachment of FMN to a threonine residue in bacterial flavoproteins.

Na(+)-translocating NADH:quinone oxidoreductase (Na(+)-NQR) contains two flavin residues as redox-active prosthetic groups attached by a phosphoester bond to threonine residues in subunits NqrB and NqrC. We demonstrate here that flavinylation of truncated Vibrio harveyi NqrC at Thr-229 in Escherichia coli cells requires the presence of a co-expressed Vibrio apbE gene. The apbE genes cluster with genes for Na(+)-NQR and other FMN-binding flavoproteins in bacterial genomes and encode proteins with previously unknown function. Experiments with isolated NqrC and ApbE proteins confirmed that ApbE i
