Light microscopic, enzyme biochemical and steroid analytical investigations of follicular atresia in the ovary of domestic goose.

Follicular atresia in the ovary of the domestic goose was investigated by light microscopic, steroid RIA and lysosomal enzyme activity measuring methods during the spring reproduction cycle. Degenerative processes are associated with the transformation and proliferation of granulosa, internal and external thecal cells in the follicle. Seven types of atretic follicles were identified on the basis of the presence, absence or dominance of cells containing lipids and synthesizing steroids. Conclusive evidence for the relation between cell type and hormone content was found only in one type: in typ

Long-term treatment with dehydroepiandrosterone may lead to follicular atresia through interaction with anti-Mullerian hormone.

BACKGROUND: Hyperandrogenism is the primary manifestation of polycystic ovary syndrome (PCOS), which appears to be caused by excess exposure to androgen. As such, androgenized animal models have been developed and investigated to study the etiology of PCOS. Anti-Mullerian hormone (AMH) is known to be associated with follicle growth, and its levels are two to three times higher in women with PCOS than in those with normal ovaries. We studied how duration of androgen administration affects folliculogenesis and AMH expression. METHODS: We divided 30 immature (3-week-old) Sprague Dawley rats into

Prominent Follicular Keratosis in Multiple Intestinal Atresia with Combined Immune Deficiency Caused by a TTC7A Homozygous Mutation.

Multiple intestinal atresia with combined immune deficiency (MIA-CID) is an autosomal recessive syndrome due to mutations in the TTC7A gene implicated in the polarization of intestinal and thymic epithelial cells. MIA-CID is lethal in the first year of life in the majority of patients. Dermatological manifestations have been reported in a few cases. We describe a child affected with MIA-CID due to a previously unreported TTC7A homozygous missense mutation. Surgery for bowel occlusion was performed in the first days of life. The patient was totally dependent on parenteral nutrition since birth

Gremlin 1 regulates granulosa cell apoptosis through the TGF-β/SMAD signaling pathway during bovine follicular atresia.

Gremlin 1 is a highly conserved secretory glycoprotein of the DAN family that plays significant roles in human and mouse follicular development and ovulation through both BMP-dependent and BMP-independent pathways. However, its function and mechanisms in bovine follicular development and atresia remain unclear. This study investigated its specific molecular mechanism during follicular development in bovines. Gremlin 1 protein is expressed in ovarian follicles at various developmental stages, with both its mRNA and protein levels increasing in correlation with the progression of follicular atre

Selenized Yeast Protects Against Cadmium-Induced Follicular Atresia in Laying Hens by Reducing Autophagy in Granulosa Cells.

Cadmium (Cd) exposure can induce follicular atresia and laying performance reduction in hens, which is linked to autophagy within the granulosa cells. Selenium (Se) can influence autophagy and counteract Cd toxicity. This study aimed to investigate the protective effect of Se on Cd-induced follicular atresia in laying hens. Sixty-four laying hens were randomly allocated into 4 treatments: control group: basal diet; Se group: basal diet + 0.4 mg/kg Se from selenized yeast; Cd group: basal diet + 25 mg/kg Cd from CdCl2; and Cd+Se group: basal diet + 25 mg/kg Cd + 0.4 mg/kg Se. Biochemical identification of apoptosis (programmed cell death) in granulosa cells: evidence for a potential mechanism underlying follicular atresia.

In the present study, we examined the possibility that granulosa cell death during ovarian follicular atresia occurs by apoptosis (programmed cell death). To investigate this possibility, atresia was induced in immature female rats by injecting 15 IU PMSG. Controls received either vehicle or no treatment. PMSG-treated animals were killed on days 1-5 post-injection while controls were killed on days 1 or 5. The onset of atresia was assessed histologically by light microscopic inspection of 5 microns tissue sections and functionally by quantification of serum progesterone and estrogen levels. Ap

Tissue-type plasminogen activator in rat oocytes: expression during the periovulatory period, after fertilization, and during follicular atresia.

The regulation of tissue-type plasminogen activator (tPA) in rat oocytes during the periovulatory period, in early embryos, and in oocytes during induced follicular atresia was studied using a quantitative chromogenic substrate assay. Oocytes and early embryos were collected from three ovulation models: 1) intact immature female rats treated with PMSG, followed by hCG 48 h later; 2) hypophysectomized immature rats treated with PMSG, followed by a GnRH agonist (GnRHa) 56 h later; and 3) adult cyclic rats on the mornings of proestrus and estrus and up to 5 days after fertilization. Investigation of NAD+ metabolism alterations during follicular atresia in porcine ovaries and their regulatory mechanisms in granulosa cell apoptosis.

Follicular atresia plays a significant role in the depletion of the ovarian reserve and the decline of overall ovarian function. Understanding its molecular mechanisms is essential to preventing ovarian aging and maintaining female reproductive health. Although NAD + depletion is known to induce apoptosis in various cell types, the dynamic changes in NAD + metabolism during follicular atresia remain unclear. Moreover, its specific impact on granulosa cells has not been fully elucidated. In this study, we examined the metabolic alterations of NAD+ during follicular atresia. We also explored the

[Expression of oncogenes, growth factors and their receptors in follicular growth, regression and atresia: their roles in granulosa cell proliferation and differentiation].

Ovarian folliculogenesis is a dynamic and complex process during which follicles undergo tremendous growth and maturation. Beginning as a single layer of pregranulosa cells surrounding the immature oocyte, granulosa cells actively proliferate and differentiate until the time of ovulation. The proliferation of granulosa cells is necessary not only for follicular growth but also for creation of the unique micro-environment for oocyte maturation. Although the primary role of follicle-stimulating hormone (FSH) in granulosa cell growth and function is well established, detailed mechanisms regulatin

Lemongrass (Cymbopogon citratus) Extracts Ameliorate Polycystic Ovary Syndrome in Rats by Inhibiting Follicular Atresia and Modulating Ghrelin and Leptin.

Polycystic ovary syndrome (PCOS) accelerates ovarian follicular depletion through atresia. This study investigated the therapeutic potential of aqueous (AE) and hydroethanolic (HEE) extracts of Cymbopogon citratus in mitigating PCOS-induced ovarian damage (specifically ferroptosis, apoptosis, and oxidative stress) in a rat model of PCOS. PCOS was induced by oral administration of letrozole (1 mg/kg/day for 21 days), followed by treatment with AE or HEE (100, 200, or 400 mg/kg/day for 20 days) with metformin (200 mg/kg) as a positive control. The results showed that HEE contains 5 times more al

Effect of High dose Gonadotropin Stimulation on Follicular Atresia through Light Chain 3B and Voltage Dependent Anion Channel 2.

BACKGROUND: Follicle development takes place under the control of hormonal and environmental stimuli. It suggested that to improve in vitro fertilisation outcomes in poor responders increasing gonadotropin doses be used. Excessive gonadotropin leads to atresia and impairs follicular development, but the molecular mechanisms of follicular atresia remain largely unknown. Recently, it was suggested that autophagy may be an alternative mechanism involved in follicle depletion. AIMS: In this study, we aimed to clarify the role of autophagic markers such as light chain (LC) 3B and voltage dependent

Oxidative Stress and SIRT1-Nrf2 Anti-Ferroptotic Pathways in Granulosa Cells: A Molecular Key to Follicular Atresia and Ovarian Aging.

The functional deterioration of granulosa cells (GCs), essential for follicular growth, steroidogenesis, and oocyte competence, indicates ovarian aging and reduced fertility. An expanding corpus of research indicates that oxidative stress is a primary molecular contributor to granulosa cell dysfunction, culminating in mitochondrial impairment, reduced metabolic support for oocytes, and the activation of regulated apoptotic pathways that end in follicular atresia. Ferroptosis, an emergent type of iron-dependent lipid peroxidation, has been identified as a crucial mechanism contributing to chemo

Expression of Mitochondria-Associated Genes (PPARGC1A, NRF-1, BCL-2 and BAX) in Follicular Development and Atresia of Goat Ovaries.

Most follicles undergo atresia during the developmental process. Follicular atresia is predominantly regulated by apoptosis of granulosa cells, but the mechanism underlying apoptosis via the mitochondria-dependent apoptotic pathway is unclear. We aimed to investigate whether the mitochondria-associated genes peroxisome proliferator-activated receptor-gamma, coactivator1-alpha (PPARGC1A), nuclear respiratory factor-1 (NRF-1), B-cell CLL/lymphoma 2 (BCL-2) and BCL2-associated X protein (BAX) played a role in follicular atresia through this pathway. The four mitochondria-associated proteins (PGC-

Involvement of angiopoietin-tie system in bovine follicular development and atresia: messenger RNA expression in theca interna and effect on steroid secretion.

Angiogenesis is involved in the local mechanisms that regulate follicular development and ovulation. Recently, the angiopoietin (ANPT)-Tie system has been shown to be required to regulate angiogenesis and blood vessel regression. Expression of the ANPT-Tie system in the cyclic ovary suggests that the relative changes in the expression of ANPT-1 and ANPT-2 influence the stability of ovarian blood vessels. In this study, we investigated 1) the mRNA expression for ANPT-1, ANPT-2, and endothelial cell-specific receptors Tie1 and Tie2 in the theca interna (TI) of the bovine developing, mature, and

Upregulation of miR-33 Exacerbates Heat-Stress-Induced Apoptosis in Granulosa Cell and Follicular Atresia of Nile Tilapia (Oreochromis niloticus) by Targeting TGFβ1I1.

High temperature affects egg quality and increases follicular atresia in teleosts. The present study aimed to explore the regulated mechanism of ovary syndrome of Nile tilapia (Oreochromis niloticus) exposed to heat stress. To this end, we conducted histological and biochemical analyses and integrated miRNA-target gene analyses. The histochemical analyses confirmed that heat stress promoted the apoptosis of granulosa cell and therefore resulted in increased follicular atresia in the ovary. Heat stress led to the differential expression of multiple miRNAs (miR-27e, -27b-3p, -33, -34a -133a-5p,
