Alterations of gut barrier and gut microbiota in food restriction, food deprivation and protein-energy wasting.

Increasing evidence shows that gut microbiota composition is related to changes of gut barrier function including gut permeability and immune function. Gut microbiota is different in obese compared to lean subjects, suggesting that gut microbes are also involved in energy metabolism and subsequent nutritional state. While research on gut microbiota and gut barrier has presently mostly focused on intestinal inflammatory bowel diseases and more recently on obesity and type 2 diabetes, this review aims at summarizing the present knowledge regarding the impact, in vivo, of depleted nutritional sta

Vasopressin-independent regulation of collecting duct aquaporin-2 in food deprivation.

BACKGROUND: Humans and animals are frequently subjected to food deprivation or starvation. However, the adaptation of the kidney to this condition is not well understood. The purpose of these studies was to examine the effects of food deprivation on water handling by the kidney, the expression levels of collecting duct (CD) water channel aquaporin-2 (AQP2), and to determine the role of vasopressin in the adaptation of AQP2 to food deprivation. METHODS: Sprague-Dawley (SD) and Brattleboro rats were placed in metabolic cages and deprived of food but had free access to water for 72 hours. Water

Fetal alcohol and thymocyte phenotypes in offspring: response to food deprivation.

Restriction of food availability is a reliable stimulus that leads to significant hypothalamo-pituitary-adrenal (HPA) activation to which rats do not habituate. Based on our previous data that indicated that the HPA response to some, but not all, stressful stimuli is significantly greater in adult offspring of Sprague-Dawley dams exposed to 35% alcohol during the last 2 weeks of gestation than that of control rats and on the mounting neuroendocrine-immune literature that describes the role of pituitary-adrenal products in modulating cellular immunity, we hypothesized that the outcomes of food

Effect of food deprivation (fasting) on plasma gastrin levels in man.

We studied plasma gastrin levels in 12 healthy men before and after 4, 8 and 10 days of total food deprivation. The gastrin levels during the starvation period were significantly lower than the preexposure values. No such changes were observed in 6 other men, serving as controls, who were allowed to eat ad libitum during the experiment. Blood glucose levels and urinary output of catecholamines were measured concurrently. A pronounced hypoglycemia and a doubting of the urinary output of adrenaline were observed during the fasting period. Thus, depression of plasma gastrin concentrations occurs

Combined effects of food deprivation and food frequency on the amount and temporal distribution of schedule-induced drinking.

Under intermittent food schedules animals develop temporally organized behaviors throughout interfood intervals, with behaviors early in the intervals (interim) normally occurring in excess. Schedule-induced drinking (a prototype of interim, adjunctive behavior) is related to food deprivation and food frequency. This study investigated the interactions that resulted from combining different food-deprivation levels (70%, 80% or 90% free-feeding weights) with different food-occurrence frequencies (15-, 30- or 60-s interfood intervals) in a within-subjects design. Increases in food deprivation an

The medial hypothalamus is required for the feeding response to glucoprivation but not to food deprivation.

While the hypothalamus has been implicated in the regulation of energy balance, the central mechanisms and neural circuit that coordinate the feeding response to energy deficit have not been fully clarified. To better understand the role of the hypothalamus in mediating hyperphagic responses to food deprivation or glucoprivation, we examined the feeding responses in rats in which the medial hypothalamus (MH) was isolated from the rest of the brain. The isolation of the MH was performed with a Halasz's knife cut, and experiments were performed 7 days after the operation. Food consumption betwee

Effect of food deprivation on response of the mite, Dermanyssus gallinae, to heat.

Freshly blood-fed adult females of the chicken mite Dermanyssus gallinae DeGeer (Acari: Dermanysidae) were food-deprived during 1, 2-3, 8-10, 14-16 and 22-23 days. These mites were tested in groups of 10 to determine their sensitivity to a heat cue delivered for a 60 s period under controlled laboratory conditions (24 degrees C, simulated dark conditions of 2 lx). Immobile mites were videotaped and start of activation (for individual mites) and percentage of mites activated in the 60 s period were related to temperature changes. Mites were activated with temperature gradients as low as 0.003-0

Depression of antral and serum gastrin concentration by food deprivation in the rat.

In rats fasted 4 days, immunoreactive gastrin concentrations decreased to one-third of the fed levels in antral tissue and to one-eight of the fed levels in serum. The number of antral cells that reacted with fluorescent antigastrin antiserum was also correspondingly decreased. After refeeding, serum gastrin returned to normal levels in 6 days, whereas antral gastrin concentration recovered after 9 days. Normal gastrin levels were maintained in rats fed a nutritious liquid diet over a 6-day period, whereas tissue and serum hormone concentrations decreased to low levels in rats placed on a high

Sleep deprivation as a risk factor for anaphylaxis in children with food allergy.

Severity of food allergic reactions is unpredictable, and cofactors influence the development of food-induced anaphylaxis. A lot of cofactors might increase allergic reaction severity, and little is known about sleep deprivation as a possible cofactor in food allergy, especially in children. With the description of a case of a middle child, we want to emphasise the role that sleep deprivation might play as a cofactor, especially during oral immunotherapy (OIT). She was undergoing OIT for milk and experienced anaphylaxis after ingestion of milk and sleep deprivation (sleeping time 3 hours less

The effect of food deprivation on alcohol consumption in bulimic and control women.

The effect of food deprivation on alcohol self-administration was examined in five women with bulimia nervosa and five healthy controls in a semi-naturalistic experimental design. In this within-subjects study, each individual underwent two food-deprived and two non-deprived sessions. The deprivation condition consisted of no food or caloric beverages for 19 hours prior to the experimental procedure. On each of the 4 days, subjects watched a 2-hour segment of an epic movie and were permitted to drink ad libitum. No differences were observed in the number of grams of alcohol consumed under depr

Effects of food deprivation on subjective responses to d-amphetamine in humans.

The effects of 24 hours of food deprivation on the subjective response to 10 mg oral d-amphetamine were studied in 12 healthy normal volunteers. A within-subjects design was used in which subjects ingested amphetamine and placebo capsules in both a fed and a fasting state. Each of the four experimental conditions-FED/DRUG, FED/PLACEBO, FAST/DRUG, FAST/PLACEBO--was enacted twice according to a randomized block design. Three subjective effects questionnaires, the Profile of Mood States, the Addiction Research Center Inventory, and the Visual Analogue Scale, were completed prior to and 1, 3 and 6

The anorectic action of naloxone is attenuated by adaptation to a food-deprivation schedule.

Recent studies have shown that naloxone and other opiate antagonists can reduce the amounts of food and water consumed by laboratory animals, a finding consistent with a role for endogenous opioids in the control of appetite. Because there have also been some failures to observe an anorectic action of naloxone, a study was carried out in which the effects of the drug on food intake were investigated using two different experimental procedures. In naive rats deprived of food for 24 h, both naloxone (0.1, 1.0 and 10.0 mg/kg) and fenfluramine (1.0, 3.0 and 10.0 mg/kg) produced dose-related decrea

Affect regulation and food intake in bulimia nervosa: emotional responding to food cues after deprivation and subsequent eating.

Emotional responding to salient food cues and effects of food deprivation and consumption were investigated in 32 women with bulimia and 32 control women. One half of each group was food deprived before viewing unpleasant, neutral, pleasant, and food-related pictures. Then participants could eat from a buffet before viewing a parallel picture set. Women with bulimia showed a substantial potentiation of startle responses during viewing of food cues relative to control women. This startle potentiation was attenuated by food deprivation and augmented by increased food consumption. These data supp

Metabolic Profiling of the Hypothalamus of Mice during Short-Term Food Deprivation.

Nutrient availability and utilization in hypothalamic cells are directly associated with the regulation of whole-body energy homeostasis. Thus, establishing metabolic profiling in the hypothalamus in response to metabolic shift is valuable to better understand the underlying mechanism of appetite regulation. In the present study, we evaluate the alteration of lipophilic and hydrophilic metabolites in both the hypothalamus and serum of fasted mice. Fasted mice displayed an elevated ketone body and decreased lactate levels in the hypothalamus. In support of the metabolite data, we further confir

Effect of food deprivation on oxidative stress biomarkers in fish (Sparus aurata).

Oxidative stress in fish (Sparus aurata) as a consequence of food restriction and fasting, has been studied. Four groups of fish were maintained for 46 days under different conditions of food supplementation: a control group with no food restriction (ratio of food/fish of 2% w/w), two groups of animals with restricted food supplement (1 and 0.5%) and a fasting group (no meal addition). Finally, all the fish were provided with food at the same ratio as the control group for the last 7 days. Sampling and weighing of fish were carried out every week and their livers were used for the analysis of

Site of action of anorectic drugs: glucoprivic- versus food deprivation-induced feeding.

Feeding induced by 2-deoxyglucose was compared with feeding induced by food deprivation in terms of antagonism by anorectic drugs and of anatomical site of action. Glucoprivic feeding was completely blocked by microinjection of amphetamine, fenfluramine, and mazindol into the paraventricular nucleus of the hypothalamus (PVN). Deprivation-induced feeding was not blocked by amphetamine, fenfluramine, or mazindol microinjected into the PVN. Neither the feeding induced by 2-deoxyglucose nor its reversal by amphetamine were blocked by pretreatment with the beta-adrenergic antagonist, propranolol. A

Sleep deprivation engages the orexin/hypocretin system to regulate food reward seeking.

BACKGROUND: Inadequate sleep is a prevalent health issue in modern society, with unintended consequences in dysregulation of the reward system. For example, acute sleep deprivation (SD) in humans increases craving for and intake of calorie-dense foods, which lead to further health concerns. The circuit and molecular mechanisms underlying sleep regulation of reward, however, remain poorly understood. The hypothalamic orexin (also called hypocretin) system is phylogenetically conserved to dually regulate sleep/arousal and reward. Here, we tested the hypothesis that acute SD engages the orexin (O
