The human oral microbiome is the second largest microbial community in humans, harboring over 700 bacterial species, which aid in digestion and protect from growth of disease-causing pathogens. One such oral pathogen, Tannerella forsythia, along with other species, contributes to the pathogenesis of periodontitis. T. forsythia is unable to produce its own N-acetylmuramic acid (NAM) sugar, essential for peptidoglycan biosynthesis and therefore must scavenge NAM from other species with which it cohabitates. Here, we explore the recycling potential of T. forsythia for NAM uptake with a bioorthogo

Forsythia Fruit (FF), the fruit of Forsythia suspensa, has been used since ancient times as an herbal medication in East Asia to treat inflammation, gonorrhea, and pharyngitis. However, the efficacy of FF against liver damage due to inflammation has not been studied. Here, we explored the protective effects of FF in a mouse hepatitis model induced by lipopolysaccharide (LPS)/D-galactosamine (GalN) treatment. We measured inflammatory cytokine and aminotransferase levels in mouse blood and analyzed the effects of FF on inflammatory gene and protein expression levels in liver tissue. Our results

This study developed a stable Forsythia essential oil (FEO) microemulsion (FEO-ME) using a self-microemulsifying drug delivery system (SMEDDS). The FEO-ME effectively reduced the volatility of essential oil and inhibited bioactive component degradation. By constructing pseudoternary phase diagrams, the optimal formulation was identified, utilizing the surfactant EL-40 and cosurfactant ethanol. The resulting microemulsion exhibited an average droplet size of 21.58 nm and an encapsulation efficiency of 85.6%. FEO-ME demonstrated excellent storage stability over 15 days, with minimal changes in d

Tannerella forsythia infection was common in oral diseases but less reported in lung diseases. This report presents a patient with bronchiectasis who was infected by Tannerella forsythia and subsequently hospitalized with symptoms including fever, progressive cough, sputum production, and shortness of breath. A chest computed tomography (CT) scan revealed multiple bilateral pulmonary bronchiectasis with signs of infection. Metagenomic next-generation sequencing (mNGS) of the bronchoalveolar lavage fluid primarily detected Tannerella forsythia. Treatment with Piperacillin-tazobactam and ornidaz

Phenylethanoid glycosides, the main active ingredients in Fructus Forsythiae extract possesses strong antibacterial, antioxidant and antiviral effects, and their contents were higher largely than that of other ingredients such as lignans and flavones, but their absolute bioavailability orally was significantly low, which influenced clinical efficacies of its oral preparations seriously. In the present study, the absorption mechanism of phenylethanoid glycosides was studied using in vitro Caco-2 cell model. And the effect of chito-oligosaccharide (COS) on the intestinal absorption of phenyletha

Tannerella forsythia is an anaerobic, Gram-negative bacterium involved in the so-called "red complex," which is associated with severe and chronic periodontitis. The surface layer (S-layer) of T. forsythia is composed of cell surface glycoproteins, such as TfsA and TfsB, and is known to play a role in adhesion/invasion and suppression of proinflammatory cytokine expression. Here we investigated the association of this S-layer with serum resistance and coaggregation with other oral bacteria. The growth of the S-layer-deficient mutant in a bacterial medium containing more than 20% non-heat-inact

Tannerella forsythia is a gram-negative bacterium strongly associated with the development and/or progression of periodontal disease. Here, we have shown that a newly characterized matrix metalloprotease-like enzyme, referred to as karilysin, efficiently cleaved the antimicrobial peptide LL-37, significantly reducing its bactericidal activity. This may contribute to the resistance of T. forsythia to the antibacterial activity of LL-37, since their vitality was found not to be affected by LL-37 at concentrations up to 2.2 muM. Furthermore, proteolysis of LL-37 by karilysin not only abolished it

Cadmium (Cd(II)) contamination threatens agricultural systems and the safety of traditional Chinese medicinal herbs. While beneficial microbes can alleviate Cd(II) stress in plants, the synergistic mechanisms underlying microbial co-inoculation in medicinal plant species remain unclear. This study investigated the individual and combined effects of Rhodopseudomonas palustris and Bacillus subtilis on Forsythia suspensa under Cd(II) stressed. The results showed that co-inoculation achieved markedly better effects than single-strain inoculation. Specifically, the HRB treatment enhanced plant heig

Periodontitis, a chronic inflammatory disease of periodontal tissue, is often associated with a group of pathogenic bacteria known as the "red complex", including Tannerella forsythia. Previous papers showed that T. forsythia induces many kinds of inflammatory cytokines including interleukin (IL)-1β regulated by inflammasome activation. However, the physiological function of periodontitis and the mechanism to induce inflammasome activation by T. forsythia infection are poorly understood. In this study, we demonstrate that the Nod-like receptor pyrin domain containing 3 (NLRP3) and caspase-4 ar

AIM: To determine the sensitivity of Porphyromonas gingivalis, Prevotella intermedia, and Tannerella forsythia to triclosan, and determine if these bacteria develop resistance to triclosan upon prolonged exposure.
MATERIALS AND METHODS: Susceptibility to triclosan was tested against three periodontal pathogens P. gingivalis, P. intermedia, and T. forsythia. Escherichia coli strains sensitive and resistant to triclosan were used as biological controls to confirm the efficacy of triclosan in the assays. Agar plates were prepared locally with vitamin K and hemin-supplemented medium.

T. forsythia is a subgingival periodontal bacterium constituting the subgingival pathogenic polymicrobial milieu during periodontitis (PD). miRNAs play a pivotal role in maintaining periodontal tissue homeostasis at the transcriptional, post-transcriptional, and epigenetic levels. The aim of this study was to characterize the global microRNAs (miRNA, miR) expression kinetics in 8- and 16-week-old T. forsythia-infected C57BL/6J mouse mandibles and to identify the miRNA bacterial biomarkers of disease process at specific time points. We examined the differential expression (DE) of miRNAs in mous

ETHNOPHARMACOLOGICAL RELEVANCE: Forsythiae Fructus (called Lianqiao in Chinese), the fruit of Forsythia suspensa (Thunb.) Vahl, is utilized as a common traditional medicine in China, Japan and Korea. It is traditionally used to treat pyrexia, inflammation, gonorrhea, carbuncle and erysipelas. Depending on the different harvest time, Forsythiae Fructus can be classified into two forms, namely Qingqiao and Laoqiao. The greenish fruits that start to ripen are collected as Qingqiao, while the yellow fruits that are fully ripe are collected as Laoqiao. Both are applied to medical use. This review a

Tannerella forsythia is a bacteria associated with severe periodontal disease. This study reports identification and characterization of a membrane-associated serine protease from T. forsythia. The protease was isolated from T. forsythia membrane fractions and shown to cleave both gelatin and type I collagen. The protease was able to cleave both substrates over a wide range of pH values, however optimal cleavage occurred at pH 7.5 for gelatin and 8.0 for type I collagen. The protease was also shown to cleave both gelatin and type I collagen at the average reported temperature for the gingival

Forsythia suspensa (Thunb.) Vahl. has been widely used in traditional medicines in Asia to treat gonorrhoea, erysipelas, inflammation, pyrexia, ulcer and other diseases. Recently the investigation has been focused on the antioxidant and antibacterial activity of this plant. However, limited scientifically proven information is available. We isolated two compounds (forsythiaside and forsythin) from this plant. The aims of this investigation, therefore, were to assay antioxidant activity and antibacterial properties of the two main and distinctive compounds isolated and to exploit antioxidants a

Sesame (Sesamum indicum), an important oilseed crop, is well known for its therapeutic values. It is one of the richest sources of polyunsaturated fatty acids and health beneficiary lignans. However, nutraceutical potential is restricted due to the presence of secoisolariciresinol diglucoside (SDG), a potent anticarcinogen, in imperceptible amount (~ 0.03 mg/25 g of seeds). Consequently, a logical strategy was implemented to augment the SDG content in sesame by heterologous expression of the pinoresinol-lariciresinol reductase (Fiplr) gene from Forsythia intermedia. Four individual transgenic

Periodontitis is a chronic inflammatory disease driven by reactive oxygen species (ROS) overproduction and immune dysregulation. Carbon dots derived from traditional Chinese medicine (TCM) integrate the functional chemical features of herbal precursors with the favorable physicochemical properties of nanomaterials. As an exemplary case, we hydrothermally synthesized Forsythia suspensa-derived carbon dots (Fo-CDs), which exhibited a high superoxide dismutase (SOD)-like activity of 340 U/mg, along with catalase-mimicking and broad-spectrum free radical scavenging capacities. Mechanistically, Fo-

Tannerella forsythia is a fastidious S-layer Gram-negative, strict anaerobic, and highly pleomorphic bacterium associated with periodontal inflammatory processes, bone loss, cardiovascular mortality, systemic sequelae, sepsis, and preterm birth. One of the purposes of this study was to characterize, for the first time, Tannerella forsythia (ATCC 43037) using Fourier transform infrared micro-spectroscopy. The analysis aimed to establish a suitable methodology for obtaining reproducible spectra for the characterization of this microorganism, as well as to assign the vibrational bands using techn

The bacterial cell wall consists of a three-dimensional peptidoglycan layer, composed of peptides linked to the sugars N-acetylmuramic acid (MurNAc) and GlcNAc. Unlike other bacteria, the pathogenic Tannerella forsythia, a member of the red complex group of bacteria associated with the late stages of periodontitis, lacks biosynthetic pathways for MurNAc production and therefore obtains MurNAc from the environment. Sugar kinases play a crucial role in the MurNAc recycling process, activating the sugar molecules by phosphorylation. In this study, we present the first crystal structures of a MurN
