Compatibility between snails and schistosomes: insights from new genetic resources, comparative genomics, and genetic mapping.

The freshwater snail Biomphalaria glabrata is an important intermediate host of the parasite Schistosoma mansoni that causes human intestinal schistosomiasis. To better understand vector snail biology and help advance innovative snail control strategies, we have developed a new snail model consisting of two homozygous B. glabrata lines (iM line and iBS90) with sharply contrasting schistosome-resistance phenotypes. We produced and compared high-quality genome sequences for iM line and iBS90 which were assembled from 255 (N50 = 22.7 Mb) and 346 (N50 = 19.4 Mb) scaffolds, respectively. Using F2 o

Effect of disinfection agents and quantification of potentially viable Leptospira in fresh water samples using a highly sensitive integrity-qPCR assay.

Leptospirosis is an emerging worldwide zoonotic disease, but the general biology of the causative agents is still poorly understood. Humans are an occasional host. The main risk factors are water-associated exposure during professional or recreational activities or during outbreaks in endemic areas. Detecting the presence of pathogenic bacteria in aquatic environments and their capacity to resist various inactivation processes are research fields that need to be further developed. In addition, the methods used for detecting and enumerating Leptospira still need to be improved. We aimed to desc

The pond snail Lymnaea stagnalis.

The freshwater snail Lymnaea stagnalis has a long research history, but only relatively recently has it emerged as an attractive model organism to study molecular mechanisms in the areas of developmental biology and translational medicine such as learning/memory and neurodegenerative diseases. The species has the advantage of being a hermaphrodite and can both cross- and self-mate, which greatly facilitates genetic approaches. The establishment of body-handedness, or chiromorphogenesis, is a major topic of study, since chirality is evident in the shell coiling. Chirality is maternally inherite

A Recessive oca2 Mutation Underlies Albinism in Xiphophorus fish.

Oculocutaneous albinism (OCA) is a group of genetic disorders characterized by impaired melanin production, leading to reduced pigmentation in the skin, hair, and eyes. Xiphophorus , a genus of small freshwater fish, has been a pivotal model organism in pigmentation disorder research, providing key findings in the genetic pathways governing physiological and pathological pigment cell biology. Leveraging the well-established research framework provided by Xiphophorus , we have identified a spontaneously occurring albinism phenotype in swordtail fish Xiphophorus hellerii . Genetic mapping of alb

Why Euglenozoans.

To accompany a new collection of methods and protocols, we discuss the relevance of the microbial eukaryotes belonging to the protist phylum Euglenozoa. For those interested by Euglena, applied biology is often relevant: as a starting point for useful natural products including biofuels, nutritional supplements, and metabolites with biomedical potential, or as an environmental agent for bioremediation. Arguably the most widely studied euglenozoans are the parasitic trypanosomatids. Collectively, trypanosomatids cause several serious neglected tropical diseases and economically important diseas

C3 in Pomacea canaliculata: a conserved effector with developmental and immune roles.

C3 is the central effector of the complement system and one of the most conserved molecules of metazoan innate immunity. Despite its relevance, little is known about complement components in Lophotrochozoa. Here, we characterise the C3 ortholog of the freshwater gastropod Pomacea canaliculata (PcC3) using long-read transcriptomics, phylogenetics, expression profiling and immunohistochemistry. PcC3 displays a fully conserved domain architecture and clusters phylogenetically in agreement with metazoan relationships. Following LPS challenge, PcC3 is up-regulated in haemocytes and unexpectedly in

Siliceous non-diatom microorganisms as complementary indicators in forensic investigations of drowning deaths.

The diatom test remains one of the most widely applied laboratory methods in the post-mortem diagnosis of drowning. However, its value may be limited in environments with low diatom abundance or in atypical aquatic settings. We surveyed the forensic relevance of additional siliceous, non-diatom microorganisms (chrysophyte cysts and scales, freshwater sponge spicules, ebridians, and phytoliths) detectable using standard diatom preparation techniques based on our expertise in the fields of paleoecology and marine biology, as well as the experience gained from more than 1500 diatom analyses of su

Bithynia siamensis goniomphalos, the first intermediate host of Opisthorchis viverrini in Thailand.

Opisthorchiasis caused by Opisthorchis viverrini (O. viverrini) remains as medically important problem in Thailand especially in the north-eastern part. Infection with this parasite can lead to cholangiocarcinoma improvement. The highest prevalence of O. viverrini infection has been found in the Northeast Thailand and is associated with the high incidence rate of cholangiocarcinoma. To complete the life cycle of O. viverrini, the freshwater snails namely Bithynia funiculata, Bithynia siamensis siamensis and Bithynia siamensis goniomphalos (B. s. goniomphalos) are required to serve as the first

Contrasting population genetic structures of freshwater eels (Anguilla marmorata, A. bicolor, A. mossambica) from Madagascar.

Freshwater eels (genus Anguilla) are of major biological and commercial interest due to their complex catadromous life cycle and as a high-value fishery resource, but the biology of tropical species is still poorly understood. This study investigates the genetic integrity, diversity and population structure of three species of tropical eels (Anguilla marmorata, A. bicolor and A. mossambica) in the western Indian Ocean, based on partial nucleotide sequences of the mtDNA control region (n = 107 samples) and the nuclear GTH2b gene (n = 91 samples) for individuals collected from two freshwater sys

Comments on "was hydrogen peroxide present before the arrival of oxygenic photosynthesis? The important role of iron(II) in the archean ocean".

Recent research has hypothesized that hydrogen peroxide (H2O2) may have emerged from abiotic geochemical processes during the Archean eon (4.0-2.5 Ga), stimulating the evolution of an enzymatic antioxidant system in early life. This eventually led to the evolution of cyanobacteria, and in turn, the accumulation of oxygen on Earth. In the latest issue of Redox Biology, Koppenol and Sies (vol. 29, no. 103012, 2024) argued against this hypothesis and suggested instead that early organisms would not have been exposed to H2O2 due to its short half-life in the ferruginous oceans of the Archean. We f

Status Quo and Future Perspectives of Molecular and Genomic Studies on the Genus Biomphalaria-The Intermediate Snail Host of Schistosoma mansoni.

Schistosomiasis, or also generally known as bilharzia or snail fever, is a parasitic disease that is caused by trematode flatworms of the genus Schistosoma. It is considered by the World Health Organisation as the second most prevalent parasitic disease after malaria and affects more than 230 million people in over 70 countries. People are infected via a variety of activities ranging from agricultural, domestic, occupational to recreational activities, where the freshwater snails Biomphalaria release Schistosoma cercariae larvae that penetrate the skin of humans when exposed in water. Human gnathostomiasis: a neglected food-borne zoonosis.

BACKGROUND: Human gnathostomiasis is a food-borne zoonosis. Its etiological agents are the third-stage larvae of Gnathostoma spp. Human gnathostomiasis is often reported in developing countries, but it is also an emerging disease in developed countries in non-endemic areas. The recent surge in cases of human gnathostomiasis is mainly due to the increasing consumption of raw freshwater fish, amphibians, and reptiles. METHODS: This article reviews the literature on Gnathostoma spp. and the disease that these parasites cause in humans. We review the literature on the life cycle and pathogenesis

Biology and systematics of heterokont and haptophyte algae.

In this paper, I review what is currently known of phylogenetic relationships of heterokont and haptophyte algae. Heterokont algae are a monophyletic group that is classified into 17 classes and represents a diverse group of marine, freshwater, and terrestrial algae. Classes are distinguished by morphology, chloroplast pigments, ultrastructural features, and gene sequence data. Electron microscopy and molecular biology have contributed significantly to our understanding of their evolutionary relationships, but even today class relationships are poorly understood. Haptophyte algae are a second

Study Protocol for the Daasanach Human Biology Project: A Longitudinal Study of the Impact of Drinking Water Salinity and Heat Stress on Water Insecurity, Hydration, Energetics, Hypertension, and Kidney Function in Northern Kenya.

INTRODUCTION: Increasing salinity of groundwater is a global environmental health threat that has been overlooked in human biology and may increase the risk of water insecurity, dehydration, chronic kidney disease (CKD), and hypertension in humans. However, no health standards exist for salinity levels in drinking water. This protocol paper describes the Daasanach Human Biology Project, which is testing how water salinity exposure and heat stress impact hydration, kidney disease, and blood pressure among Daasanach pastoralists in a hot-arid environment in northern Kenya. METHODS/DESIGN: This

Structure and function of the digestive system in molluscs.

The phylum Mollusca is one of the largest and more diversified among metazoan phyla, comprising many thousand species living in ocean, freshwater and terrestrial ecosystems. Mollusc-feeding biology is highly diverse, including omnivorous grazers, herbivores, carnivorous scavengers and predators, and even some parasitic species. Consequently, their digestive system presents many adaptive variations. The digestive tract starting in the mouth consists of the buccal cavity, oesophagus, stomach and intestine ending in the anus. Several types of glands are associated, namely, oral and salivary gland

Remodeling of Uterine Tissues During Gestation of Potamotrygon wallacei (Elasmobranchii), a Neotropical Freshwater Stingray Endemic to the Negro River, Central Amazonia.

Neotropical freshwater stingrays of the subfamily Potamotrygoninae exhibit aplacental viviparity with uterine trophonemata. In this reproductive mode, females nourish and provide oxygenation to the embryo via the mucosa of the uterine wall. The aim of this study was to describe and histologically quantify the tissue components of the gravid uterus in an Amazonian freshwater stingray. Adult females of Potamotrygon wallacei were studied in different reproductive periods: resting stage, pregnant, and postpartum. During reproductive rest, the left ovary has numerous follicles compared to the right

Beyond Genetic Indicators: How Reproductive Mode and Hybridisation Challenge Freshwater Mussel Conservation.

Genetic diversity is a fundamental aspect of biodiversity, yet it is rarely assessed and monitored in conservation practice. Unionid freshwater mussels exemplify the dramatic loss of biodiversity in freshwater ecosystems, yet genomic data for these ecologically important species remain scarce. Here, we conducted a high-resolution population genomics study of all Anodonta species in Switzerland, with a focus on two species with contrasting reproductive strategies. After generating draft genomes of the hermaphroditic Anodonta cygnea and the gonochoric Anodonta anatina, we performed whole-genome

New tools to uncover old tricks: an update on the knowledge on the most successful invasive freshwater helminth, Schyzocotyle acheilognathi.

The Asian fish tapeworm (AFT), Schyzocotyle acheilognathi, is a highly invasive and pathogenic parasite of freshwater fishes, present on all continents except Antarctica. Globally, 310 + definitive fish host species have been recorded, with Mexico showing the highest number of reports. Here, we summarize the current knowledge about the biology, distribution, and ecological determinants of this parasite, highlighting key knowledge gaps that could guide future research. Considering Mexico has quite comprehensive records of this parasite (both in host and geographical terms), we propose that this

Detection and quantification of the crayfish plague agent in natural waters: direct monitoring approach for aquatic environments.

Aphanomyces astaci, a specialised parasite of North American freshwater crayfish, is the disease agent of crayfish plague that is lethal to European freshwater crayfish. The life cycle of A. astaci has been inferred from experimental laboratory studies, but less is known about its natural sustainability and ecology. To address such questions, tools for monitoring of A. astaci directly in aquatic environments are needed. Here, we present an approach for detecting and quantifying A. astaci directly from water samples using species-specific TaqMan minor groove binder real-time PCR. Samples of a 1
