Effects of fructose-1,6-bisphosphate on brain polyamine biosynthesis in a model of transient cerebral ischemia.

We evaluated the effects on cerebral ischemia of a treatment with fructose-1,6-bisphosphate, a compound known to possess protective effects on acute ischemic injury in a variety of different tissues. We investigated the ability of the compound, administered either 15 minutes before or 15 minutes after the ischemic insult, in reducing the ischemia-induced changes in polyamine brain levels. The experiments were performed in adult, chloral hydrate-anesthetized Mongolian gerbils that underwent a 15 minutes ligation of the common carotid arteries followed by recirculation. Animals were sacrificed 1

Fructose-1,6-bisphosphate reverses hypotensive effect caused by L-kynurenine in Wistar male rats.

Hypotension is one of the main characteristics of the systemic inflammation, basically caused by endothelial dysfunction. Studies have shown that the amino acid L-kynurenine (KYN) causes vasodilation in mammals, leading to hypotensive shock. In hypotensive shock, when activated by the KYN, the voltage-gated potassium channel encoded by the family KCNQ (Kv7) gene can cause vasodilation. Fructose-1,6-bisphosphate (FBP) it is being considered in studies an anti-inflammatory, antioxidant, immunomodulator, and a modulator of some ion channels (Ca2+, Na+, and K+). We analyzed the effects of KYN and

In silico-screening approaches for lead generation: identification of novel allosteric modulators of human-erythrocyte pyruvate kinase.

Identification of allosteric binding site modulators have gained increased attention lately for their potential to be developed as selective agents with a novel chemotype and targeting perhaps a new and unique binding site with probable fewer side effects. Erythrocyte pyruvate kinase (R-PK) is an important glycolytic enzyme that can be pharmacologically modulated through its allosteric effectors for the treatment of hemolytic anemia, sickle-cell anemia, hypoxia-related diseases, and other disorders arising from erythrocyte PK malfunction. An in-silico screening approach was applied to identify

Age and the control of glycolysis in the rat lens.

Previous studies with lens dispersions indicated that the rate-limiting step in glycolysis shifts from hexokinase (HK) in the young lens to phosphofructokinase (PFK) in older lenses. Because the concentrations of the complex controlling factor for these enzymes could not be reproduced reliably in homogenates, the question of age-related control of glycolysis was re-examined in intact lenses. Toward this end, the levels of several metabolites of glucose were measured in fresh and incubated clear lenses. Of the substrates measured per fresh lens, only one changed significantly with age; fructose

Spectroscopic study on the reactions of bis-salophen with uranyl and then with fructose 1,6-bisphosphate and the analytical application.

The chelating reaction of bis-salophen with uranyl to form binuclear complex uranyl-bis-salophen (UBS) was studied by fluorescence spectroscopy. The coordination reaction of UBS with fructose 1,6-bisphosphate (F-1,6-BP) to form supramolecular polymer was then studied by resonance light scattering (RLS) spectroscopy. The reaction of bis-salophen with uranyl results in a remarkable enhancement of fluorescence intensity. The maximum emission wavelength of the fluorescence is at 471nm. The reaction of UBS with F-1,6-BP results in a remarkable enhancement of RLS intensity. The maximum scattering wa

Enhancement of the reductive activation of chloroplast fructose-1,6-bisphosphatase by modulators and protein perturbants.

To characterize the mechanism of chloroplast fructose-1,6-bisphosphatase activation, we have examined kinetic and structural changes elicited by protein perturbants and reductants. At variance with its well-known capacity for enzyme inactivation, 150 mM sodium trichloroacetate yielded an activatable chloroplast fructose-1,6-bisphosphatase in the presence of 1.0 mM fructose 1,6-bisphosphate and 0.1 mM Ca2+. Other sugar bisphosphates did not replace fructose 1,6-bisphosphate whereas Mg2+ and Mn2+ were functional in place of Ca2+. Variations of the emission fluorescence of intrinsic fluorophores

Alterations in 32P-labelled intermediates during flux activation of human platelet glycolysis.

Using a newly developed isotopic tracer technique for the measurement of 32P-labelled intermediates in glycolysis and nucleotide metabolism in platelets, we studied the variations in 32P-labelled intermediates during activation of the glycolytic flux by cyanide and platelet-activating agents. The major variations occurred in [32P]Fru-1,6-P2, dihydroxy acetone phosphate, ATP and Pi. There was a quantitative covariance between the increase in lactate production and the rise in [32P]Fru-1,6-P2 induced by different platelet-activating agents. In contrast, cyanide induced weaker activation of the f

Presence of a truncated M-type subunit and altered kinetic properties of 6-phosphofructo-1-kinase isozymes in the brain of a dog affected by glycogen storage disease type VII.

6-Phosphofructo-1-kinase (PFK) activity in the brain of a dog affected by glycogen storage disease type VII was only 31% of the PFK activity in the normal dog brain. PFK in the normal dog brain was composed of L-type, M-type and C-type subunits with apparent molecular weights of 78,000, 86,000, and 88,000, respectively, and subunit proportions (L:M:C) of 27:49:24. PFK in the affected dog brain was composed of nearly equal levels of the normal L-type and C-type subunits, but a normal M-type subunit was not detected. Using antidog muscle PFK IgG, immunoblots of gels containing partially purified

In vitro effects of fructose-1,6-diphosphate on oxygen affinity and electrolyte equilibrium in human blood.

The in vitro effects of a solution of fructose-1,6-diphosphate on hemoglobin-oxygen affinity and electrolyte distribution between plasma and red cells were evaluated. Fructose-1,6-diphosphate increased intracellular pH and decreased extracellular pH. There were no shifts in Na+, K+, Ca++ and Cl- between plasma and red cells. Red cell content of 2,3-diphosphoglycerate remained unchanged, while P50 was reduced (as a consequence of the intracellular alkalosis). The therapeutical implications of these results are discussed.

[The activity of certain enzymes of gluconegenesis in rabbit brain and liver intoxicated by CS2].

The carbon disulphide (CS) administrated chronic to rabbit produced an important inhibition of glucose-6-phosphate phosphohyrolase and fructose-1,6-diphosphate phosphoydrolase activity in brain and liver. The decrease of theses two enzymes of gluconeogenetic cycle seem to justify certain physiological and clinical changes of the neuropsychic and hepatic activity observed in the intoxication by CS2.

Fructose 1,6-bisphosphate is anticonvulsant and improves oxidative glucose metabolism within the hippocampus and liver in the chronic pilocarpine mouse epilepsy model.

Glucose metabolism is altered in epilepsy, and this may contribute to seizure generation. Recent research has shown that metabolic therapies including the ketogenic diet and medium chain triglycerides can improve energy metabolism in the brain. Fructose 1,6-bisphosphate (F16BP) is an intermediate of glycolysis and when administered exogenously is anticonvulsant in several rodent seizure models and may alter glucose metabolism. Here, we showed that F16BP elevated the seizure threshold in the acute 6-Hz mouse seizure model and investigated if F16BP could restore impairments in glucose metabolism

Effect of chronic uremia on fructose 2,6-bisphosphate glycolytic and gluconeogenic enzymes in rat liver.

The level of fructose 2,6-bisphosphate and the maximal activities of key gluconeogenic and glycolytic enzymes were determined in the liver of a rat model of chronic uremia and in ad libitum-fed control and pair-fed control animals. Fructose 2,6-bisphosphate was decreased in uremia and its level negatively correlated with the concentration of blood urea nitrogen. The changes in gluconeogenic enzymes in uremic rats were not different from those in the pair-fed controls. However, pyruvate kinase was decreased in uremia when compared to both controls. These studies offer a possible mechanism for t

Fructose 2,6-bisphosphate and its phosphorothioate analogue. Comparison of their hydrolysis and action on glycolytic and gluconeogenic enzymes.

Purified chicken liver 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase was phosphorylated either from fructose 2,6-bis[2-32P]phosphate or fructose 2-phosphoro[35S]thioate 6-phosphate. The turnover of the thiophosphorylated enzyme intermediate as well as the overall phosphatase reaction was four times faster than with authentic fructose 2,6-bisphosphate. Fructose 2-phosphorothioate 6-phosphate was 10-100-fold less potent than authentic fructose 2,6-bisphosphate in stimulating 6-phosphofructo-1-kinase and pyrophosphate:fructose 6-phosphate phosphotransferase, but about 10 times more potent

Regulation of hepatic gluconeogenesis and glycogenolysis by phosphorylated glycerol and glycolytic intermediates in diabetic and control Chinese hamsters.

Metabolic regulation of gluconeogenesis and glycogenolysis by two phosphorylated derivatives of glycerol, G3P, and DHAP, and by F2,6BP, was assessed in vitro in liver homogenates obtained from Chinese hamsters (C. griseus) of two types: diabetic animals from sublines with consistent glycosuria and hyperglycemia, and normoglycemic controls. Only FBPase was sensitive to inhibition by the phosphorylated metabolites. G3P was weakly inhibitory of FBPase. Addition of 7 X 10(-3) M DHAP halved FBPase activity in the diabetic hamsters and 4 X 10(-3) M DHAP produced the same effect in the controls. The

The effect of organic solvents on the activation and the activity of spinach chloroplast fructose-1,6-bisphosphatase.

A two-stage assay was used to study the effect of organic solvents on the activation of and the catalysis by chloroplast fructose-1,6-bisphosphatase. Irrespective of chemical structure, all the organic solvents tested had a dual effect on the enzyme. In the activation they stimulated and inhibited at low and high concentrations, respectively, in a process that required dithiothreitol, fructose 1,6-bisphosphate, and Ca2+. Conversely, organic solvents inhibited catalysis. The enhancement in fructose-1,6-bisphosphatase activity did not arise from a change in the molecular weight of the enzyme and

Red cell pyruvate kinase deficiency: an optimised assay.

As a result of using a variety of assay techniques, rather different values for the rate of the pyruvate kinase (PK) reaction were obtained. Kinetic measurements performed with red cell PK using the stopped-flow as well as the "classical" spectrophotometric method demonstrated that only the initiation of the reaction with ADP yields reproducible results in the standard assay. Moreover, the influence of substrate concentrations, activator concentrations, ionic strength, buffer system, temperature and duration of preincubation were thoroughly investigated and were shown to play an important role

6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase from frog skeletal muscle: purification, kinetics and immunological properties.

Fructose 2,6-bisphosphate is the most potent activator of 6-phosphofructo-1-kinase, a key regulatory enzyme of glycolysis in animal tissues. This study was prompted by the finding that the content of fructose 2,6-bisphosphate in frog skeletal muscle was dramatically increased at the initiation of exercise and was closely correlated with the glycolytic flux during exercise. 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase, the enzyme system catalyzing the synthesis and degradation of fructose 2,6-bisphosphate, was purified from frog (Rana esculenta) skeletal muscle and its properties were c

Role of fructose 2,6-bisphosphate in the control of heart glycolysis.

The aim of this work was to study whether changes in fructose 2,6-bisphosphate concentration are correlated with variations of the glycolytic flux in the isolated working rat heart. Glycolysis was stimulated to different extents by increasing the concentration of glucose, increasing the workload, or by the addition of insulin. The glycolytic flux was measured by the rate of detritiation of [2-3H]- and [3-3H]glucose. Under all the conditions tested, an increase in fructose 2,6-bisphosphate content was observed. The glucose- or insulin-induced increase in fructose 2,6-bisphosphate content was re

Activation of rat heart phosphofructokinase-2 by insulin in vivo.

Fifteen minutes after the intravenous injection of overnight starved, anaesthetized rats with insulin, the concentration of fructose 2,6-biphosphate was increased more than 2-fold in the hearts of these animals. Insulin injection also caused a 2-3 fold increase in the Vmax of phosphofructokinase-2 with no detectable change in Km values. The effect persisted after precipitation of the enzyme with polyethylene glycol or after gel filtration through Sephadex G-25.
