Effects of cadmium and high temperature on some parameters of calcium metabolism in the killifish (Aphanius fasciatus).

This study aims to investigate the influence of high temperature on cadmium (Cd) toxicity in Aphanius fasciatus (Pisces: Cyprinodontidae). For this reason, Cd, mineral, and organic content in the vertebral column as well as the histological structure of gills and bone were compared in fishes exposed for 30 days to Cd (2 mg/L CdCl2) and/or high temperature (26 °C). Cd exposure caused a negative correlation between Cd and Ca concentrations (r = 0.98, p < 0.05), as well as a significant decrease in inorganic components (p < 0.05) and ash weight/dry weight ratio (p < 0.05) in the vertebral column.

In Vitro Fertilization of the African Turquoise Killifish Nothobranchius furzeri.

The ability to perform in vitro fertilization, together with sperm cryopreservation, greatly facilitates the long-term laboratory maintenance of wild-type and transgenic model organisms and helps prevent genetic drift. It is also useful in cases where reproduction may be compromised. In this protocol, we present a method for in vitro fertilization of the African Turquoise killifish Nothobranchius furzeri that is compatible with the use of fresh or cryopreserved sperm.

A scalable and tunable platform for functional interrogation of peptide hormones in fish.

Pituitary hormones play a central role in shaping vertebrate life history events, including growth, reproduction, metabolism, and aging. The regulation of these traits often requires precise control of hormone levels across diverse timescales. However, fine tuning circulating hormones in-vivo has traditionally been experimentally challenging. Here, using the naturally short-lived turquoise killifish (N. furzeri), we describe a high-throughput platform that combines loss- and gain-of-function of peptide hormones. Mutation of three primary pituitary hormones, growth hormone (gh1), follicle stimu

Quantification of noradrenergic-, dopaminergic-, and tectal-neurons during aging in the short-lived killifish Nothobranchius furzeri.

Parkinson's disease (PD) is characterized by phosphorylation and aggregation of the protein α-Synuclein and ensuing neuronal death progressing from the noradrenergic locus coeruleus to midbrain dopaminergic neurons. In 2019, Matsui and colleagues reported a spontaneous age-dependent degeneration of dopaminergic neurons and an even greater neurodegeneration of the noradrenergic neurons in the short-lived killifish Nothobranchius furzeri. Given the great possible relevance of a spontaneous model for PD, we assessed neurodegeneration of noradrenergic and dopaminergic neurons in two further labora

Upper thermal tolerance of the euryhaline fish Fundulus heteroclitus is lower in freshwater and shaped by environmental salinity-acclimation time interaction.

This study investigates whether salinity affects the upper thermal tolerance (CTmax) of the euryhaline mummichog (Fundulus heteroclitus). Mummichogs were acclimated to seawater (38 ppt, SW) or freshwater (<1 ppt, FW) at 18.5°C (Tacc), then transferred to the opposite salinity. CTmax and physiological profiles were assessed on days 0, 1, 3, 6, and 13 post-transfer. CTmax was consistently lower in FW than in SW, regardless of prior acclimation. Compared to the Tacc controls, CTmax exposure reduced plasma ion levels in FW-transferred fish and increased them in SW-transferred fish. In both salinit

Hypoxia-inducible factor 1 alpha protein increases without changes in mRNA during acute hypoxic exposure of the Gulf killifish, Fundulus grandis.

The hypoxia inducible factor 1 (HIF1) is a central regulator of the molecular responses of animals to low oxygen. While the hypoxia-responsiveness of HIF1 is generally attributed to the stabilization of the alpha protein subunit (HIF1α) at low oxygen, several studies on fish report increased tissue levels of HIF1A mRNA during hypoxia, suggesting transcriptional regulation. In the current study, HIF1α protein and HIF1A mRNA were determined in parallel in tissues of Gulf killifish, Fundulus grandis, exposed to short-term hypoxia (24 h at 1 mg O2 l-1). HIF1α protein was higher in brain, ovary, an

Long-term exposure to a pharmaceutical pollutant affects geotaxic behaviour in the adult but not juvenile life stage of killifish.

Ecosystems around the world are increasingly polluted with pharmaceutical compounds that may perturb wildlife behaviour. Because many pharmaceuticals are continuously present in the aquatic environment, animals are often exposed to them across several life stages or even their entire life. Despite a large body of literature showing various impacts of exposure to pharmaceuticals on fish, hardly any long-term studies across different life stages have been conducted which makes it hard to accurately estimate the ecological outcomes of pharmaceutical pollution. Here, we performed a laboratory expe

Untargeted Metabolomic Analysis in the Short-Lived Killifish Using HILIC Ultra Performance Liquid Chromatography Method Coupled to a Q Exactive Orbitrap Mass Spectrometer.

Over the past decade, advancements in liquid chromatography-mass spectrometry (LC-MS) have transformed untargeted metabolomic analysis. These improvements have led to biomarker discoveries, enabling scientists to deepen their understanding of biological systems in disease contexts. Achieving reproducibility, robustness, and reliable quantification is essential for confident metabolite identification. In this chapter, we present an untargeted metabolomic approach to analyze metabolites in the optic nerve tissue of adult Nothobranchius furzeri, an established biogerontology model that is increas

Alternative social and reproductive niches linked to intra-sexual color variation in a facultatively protogynous North American annual killifish, Millerichthys robustus.

Intra-sexual variation in coloration is often linked to comprehensive differences in reproductive behavior and life histories. Both males and females can express intrapopulation variation in coloration, and facultative hermaphrodism adds further complexity to the social and reproductive system. In replicated experimental tanks, we studied social interactions and reproductive behavior in males, females and sex-changing (transitional) intermediate individuals of the Mexican annual killifish, Millerichthys robustus, a facultative protogynous hermaphrodite with distinct intraspecific variation in

Development of rhythmic melatonin secretion from the pineal gland of embryonic mummichog (Fundulus heteroclitus).

The pineal gland of vertebrates produces and secretes the hormone melatonin in response to changes in the light-dark cycle, with high production at night and low production during the day. Melatonin is thought to play an important role in synchronizing daily and/or seasonal physiological, behavioral, and developmental rhythms in vertebrates. In this study, the functional development of the pineal melatonin-generating system was examined in the mummichog, Fundulus heteroclitus, an euryhaline teleost. In this species, the pineal gland contains an endogenous oscillator, ultimately responsible for

Rapid and reversible modulation of blood haemoglobin content during diel cycles of hypoxia in killifish (Fundulus heteroclitus).

We investigated whether fish can make dynamic haematological adjustments to support aerobic metabolism during repeated cycles of hypoxia-reoxygenation. Killifish were acclimated to normoxia, constant hypoxia (2 kPa O2), or intermittent cycles of nocturnal hypoxia (12 h of normoxia: 12 h of 2 kPa O2 hypoxia) for 28 days. Normoxia-acclimated fish were sampled in the daytime in normoxia and after exposure to a single bout of nocturnal hypoxia. Each hypoxia acclimation group were sampled at the PO2 experienced during acclimation during both the day and night. All acclimation groups had increased b

Aging Reduces Intestinal Stem Cell Activity in Killifish and Intermittent Fasting Reverses Intestinal Gene Expression Patterns.

The process of aging is associated with a decline in cell, tissue, and organ function, leading to a range of health problems. Increasing evidence indicates that dietary restriction can counteract age-dependent effects and improve health and longevity in whole organisms, but less is known about the influence of aging and the impact of nutrition on individual organs of an organism. In this study, we examined the intestine of the very short-lived aging model system, the African turquoise killifish (Nothobranchius furzeri), throughout its lifetime. We investigated the effects of age and nutrition

Protogynous functional hermaphroditism in the North American annual killifish, Millerichthys robustus.

Sex change (sequential hermaphroditism) has evolved repeatedly in teleost fishes when demographic conditions mediate fundamentally different sex-specific returns for individuals of particular age and size. We investigated the conditions for potential sex change in an annual killifish (Millerichthys robustus) from temporary pools in Mexico. In natural populations, we detected adults with intersex colouration and gonads. Therefore, we experimentally tested whether this apparent sex change can be generated by manipulation of ecological and social conditions, rather than being caused by environmen

Regional Distribution of a Brain-Encysting Parasite Provides Insight on Parasite-Induced Host Behavioral Manipulation.

Some parasite species alter the behavior of intermediate hosts to promote transmission to the next host in the parasite's life cycle. This is the case for Euhaplorchis californiensis, a brain-encysting trematode parasite that causes behavioral changes in the California killifish (Fundulus parvipinnis). These manipulations increase predation by the parasite's final host, piscivorous marsh birds. The mechanisms by which E. californiensis achieves this manipulation remain poorly understood. As E. californiensis cysts reside on the surface of the killifish's brain, discerning regional differences

Diversity and distribution of gynogenetic hybrids between Fundulus diaphanus and Fundulus heteroclitus in Porter's Lake (Nova Scotia) in relation to salinity.

Unisexual hybrids between Fundulus diaphanus and Fundulus heteroclitus were found in low proportions at intermediate salinity in Porter's Lake, Nova Scotia. One clone accounted for 72% of the hybrids, with most other hybrids being different at a single microsatellite allele. This clone thrives over a wide range of salinities, suggesting a general-purpose genotype.

Parasites alter host community structure in a natural experiment.

Parasites can profoundly alter host communities. However, the impact of parasites can vary from one community to another. Understanding why the impact of parasites varies across communities is challenging because it requires (i) separating the direct effects of the parasite on the host species from the indirect effects it exerts through the ecological interactions among the hosts and (ii) identifying how the presence of one host alters rates of infection in another. Freshwater fish communities in Trinidad have recently been invaded by a generalist parasitic nematode. This event, combined with

A NEW MYXIDIUM SPECIES FROM THE LEAST KILLIFISH HETERANDRIA FORMOSA IN LOUISIANA.

In a parasitological survey of the least killifish, Heterandria formosa (Girard), from Ward Creek in Louisiana, we encountered a myxozoan parasite in the gallbladders of this host bearing myxospores consistent with a Myxidium species. Prevalence was 72.2% in 18 hosts examined, with infection characterized by the presence of sheetlike to discoid plasmodia, approximately 1-2 mm across, floating freely in the gallbladder. Myxospores were observed in the plasmodia and found floating freely in the bile and were elliptoid and fusiform in valvular and sutural views respectively. The myxospore poles w

Olfactory and gustatory chemical sensor systems in the African turquoise killifish: Insights from morphology.

Smell and taste are extensively studied in fish species as essential for finding food and selecting mates while avoiding toxic substances and predators. Depending on the evolutionary position and adaptation, a discrete variation in the morphology of these sense organs has been reported in numerous teleost species. Here, for the first time, we approach the phenotypic characterization of the olfactory epithelium and taste buds in the African turquoise killifish (Nothobranchius furzeri), a model organism known for its short lifespan and use in ageing research. Our observations indicate that the o

EXPERIMENTAL INFECTIONS WITH EUHAPLORCHIS CALIFORNIENSIS AND A SMALL CYATHOCOTYLID INCREASE CONSPICUOUS BEHAVIORS IN CALIFORNIA KILLIFISH (FUNDULUS PARVIPINNIS).

Some parasites manipulate their host's phenotype to enhance predation rates by the next host in the parasite's life cycle. Our understanding of this parasite-increased trophic transmission is often stymied by study-design challenges. A recurring difficulty has been obtaining uninfected hosts with a coevolutionary history with the parasites, and conducting experimental infections that mimic natural processes. In 1996, Lafferty and Morris provided what has become a classic example of parasite-increased trophic transmission; they reported a positive association between the intensity of a brain-in
