Effects of 17α-Ethinylestradiol on Development and Gonadal Differentiation in the Estuarine Mummichog, Fundulus heteroclitus.

In fish, sex determination and gonadal development are controlled by various genetic and environmental factors. In this study, experiments were conducted on the estuarine mummichog (Fundulus heteroclitus) to investigate gonadal differentiation following exposure to the synthetic estrogen, 17α-ethinylestradiol (EE2). The period of sensitivity to EE2 was explored by initiating the exposures at different times post fertilization. In Experiment 1, mummichog embryos were collected within 8 h of fertilization and then continually exposed to increasing concentrations of EE2 (up to 229 ng/L) for up to

Zinc chloride rapidly stimulates efflux transporters in renal proximal tubules of killifish (Fundulus heteroclitus).

Multidrug resistance-related protein 2 (Mrp2) is an ATP-driven efflux pump at the luminal membrane in renal proximal tubules. It acts as detoxification mechanism by transporting xenobiotics and metabolic products into urine. The trace element zinc is essential for cellular growth, differentiation and survival. It modulates immune response and is used as dietary supplement. Here, we found that 0.1-10μM ZnCl2 rapidly stimulated transport of the Mrp2 probe substrate Texas Red (TR) in isolated killifish renal proximal tubules, which provide an established model system to measure efflux transporter

Development of rhythmic melatonin secretion from the pineal gland of embryonic mummichog (Fundulus heteroclitus).

The pineal gland of vertebrates produces and secretes the hormone melatonin in response to changes in the light-dark cycle, with high production at night and low production during the day. Melatonin is thought to play an important role in synchronizing daily and/or seasonal physiological, behavioral, and developmental rhythms in vertebrates. In this study, the functional development of the pineal melatonin-generating system was examined in the mummichog, Fundulus heteroclitus, an euryhaline teleost. In this species, the pineal gland contains an endogenous oscillator, ultimately responsible for

Exogenous retinoic acid during gastrulation induces cartilaginous and other craniofacial defects in Fundulus heteroclitus.

Embryonic levels of retinoic acid (RA) and the response of cells to RA are critical to the normal development of vertebrates. To understand the effects of RA signaling in Fundulus heteroclitus, we exposed embryos to a range of RA concentrations of 2 h during gastrulation. Embryos exposed to low concentrations of RA (10(-10)-10(-7) M) develop normally, whereas those exposed to higher concentrations (5 x 10(-7)-10(-4) M) develop characteristic dose-dependent defects. We describe, in detail, four stages of development that represent morphological effects of RA on (1) cell death and defects in the

Cold acclimation allows regulation of chloride secretion in a eurythermic teleost fish Fundulus heteroclitus.

Fundulus heteroclitus (mummichog or common killifish) is an ideal model for ion transport regulation in chloride cells of the opercular epithelium (OE) and the response to thermal challenge. Mummichogs were acclimated to warm (20 °C) and cold (5 °C) seawater and opercular epithelia dissected and mounted in isolated Ussing-style epithelia chambers. The α2 adrenergic agonist clonidine inhibited the Cl(-) secretion (measured as short-circuit current, Isc), while the β-adrenergic agonist isoproterenol and 1.0mM dibutyryl cyclic adenosine monophosphate (db-cAMP) plus 0.1mM isobutyl methylxanthine (

Further consideration of phenobarbital effects on cytochrome P-450 activity in the killifish, Fundulus heteroclitus.

1. Ethoxyresorufin O-deethylase (EROD) activity, aldrin epoxidase (AE) activity, cytochrome P-450 content, and levels of cytochrome P-450E (the major BNF-inducible P-450 form and primary EROD catalyst in scup) or its homologues were measured in hepatic microsomes isolated from Fundulus heteroclitus, scup (Stenotomus chrysops) and brook trout (Salvelinus fontinalis) treated with beta-naphthoflavone (BNF) or phenobarbital (PB). 2. In all three teleost species, BNF treatment caused expected increases in P-450 content, EROD activity and P-450E level; but either no change or a slight decrease in AE

Constant temperature and fluctuating temperature have distinct effects on hypoxia tolerance in killifish (Fundulus heteroclitus).

Climate change is leading to rapid change in aquatic environments, increasing the mean and variability of temperatures, and increasing the incidence of hypoxia. We investigated how acclimation to constant temperatures or to diel temperature fluctuations affects hypoxia tolerance in mummichog killifish (Fundulus heteroclitus). Killifish were acclimated to constant cool (15°C), constant warm (25°C) or a diel temperature cycle (15°C at night, 25°C during day) for 6 weeks. We then measured hypoxia tolerance (time to loss of equilibrium in severe hypoxia, tLOE; critical O2 tension, Pcrit), whole-an

Toxicological effects of micronized tire crumb rubber on mummichog (Fundulus heteroclitus) and fathead minnow (Pimephales promelas).

Recent studies on the distribution of microplastics in the Charleston Harbor, SC, USA revealed that a large part of the microplastic particles that are found in the intertidal sediments are tire wear particles. These particles originate from the wear of tire treads on roadways, and wash into the estuary during rain events. The abundance of these particles has raised questions about potential toxicity to aquatic organisms that ingest these particles. The synthetic rubber in car tires consists of a large variety of chemicals, which can vary between manufacturers, but usually contains styrene-but

Temporal variations in embryotoxicity of Lake Ontario American eel (Anguilla rostrata) extracts to developing Fundulus heteroclitus.

The recruitment of American eel (Anguilla rostrata) juveniles to Lake Ontario (LO), Canada has declined significantly since the 1980s. To investigate the possible contribution of maternally-transferred persistent organic pollutants (POPs) to this decline, this study measured temporal variations in the toxicity of complex organic mixtures extracted from LO American eels captured in 1988, 1998 and 2008 to developing Fundulus heteroclitus exposed by intravitelline (IVi) injection. The 1988 and 1998 eel extracts were most toxic, causing a pattern of sublethal embryotoxic responses similar to those

Rapid and reversible modulation of blood haemoglobin content during diel cycles of hypoxia in killifish (Fundulus heteroclitus).

We investigated whether fish can make dynamic haematological adjustments to support aerobic metabolism during repeated cycles of hypoxia-reoxygenation. Killifish were acclimated to normoxia, constant hypoxia (2 kPa O2), or intermittent cycles of nocturnal hypoxia (12 h of normoxia: 12 h of 2 kPa O2 hypoxia) for 28 days. Normoxia-acclimated fish were sampled in the daytime in normoxia and after exposure to a single bout of nocturnal hypoxia. Each hypoxia acclimation group were sampled at the PO2 experienced during acclimation during both the day and night. All acclimation groups had increased b

Mechanisms of Ca2+ uptake in freshwater and seawater-acclimated killifish, Fundulus heteroclitus, and their response to acute salinity transfer.

Killifish (Fundulus heteroclitus) has been extensively used as a model for ion regulation by euryhaline fishes. Na+ and Cl- dynamics have been well studied in killifish, but few studies have addressed that of Ca2+. Therefore, this study aimed to characterize Ca2+ fluxes in freshwater (FW) and seawater (SW)-acclimated killifish, their response to salinity transfer, and to elucidate the mechanisms of Ca2+ influx in FW and SW. SW killifish displayed a significantly higher Ca2+ influx rate than that of FW fish, while Ca2+ efflux rates were comparable in both salinities. Ca2+ influx was saturable i

The effects of short-term hypoxia on hypoxia-inducible factor 1 alpha mRNA and microRNA in Fundulus heteroclitus.

Hypoxia-inducible factor 1 (Hif1) is responsible for coordinating and regulating much of the response to acute hypoxia in vertebrates. This complex response is multifactorial, involving a large network of regulators. Within this network, epigenetic modulators, such as microRNA (miRNA), have garnered increasing interest due to their ability to regulate and fine-tune a myriad of cellular processes. Thus, we aimed to determine the effects of acute mild hypoxia on four known hypoxia-responsive miRNAs, along with three hypoxia-related transcripts. To do this, we exposed Atlantic killifish, Fundulus

Effect of Cu(ATSM) on the expression and activity of ABC export proteins in killifish (Fundulus heteroclitus) kidney tubules.

ABC transporters are important for excretion of xenobiotics and metabolites into urine. They are sensitive to metallic pollutants like cadmium, mercury, zinc, or arsenic. Here, we show that copper (Cu(ATSM)) stimulates ABC transporter-mediated export in isolated renal proximal tubules from Atlantic killifish (Fundulus heteroclitus) with main focus on Mrp2. Transporter stimulation was reduced by cycloheximide (CHX), an inhibitor of translation, suggesting that it is partially caused by induced expression. Functional activation was reversed by modulators of the endothelin receptor (ETB)/nitric o

The influence of salinity on acute nickel toxicity to the two euryhaline fish species, Fundulus heteroclitus and Kryptolebias marmoratus.

Nickel (Ni) is a common pollutant found in aquatic environments and may be harmful at elevated concentrations. Increasing salinity has been shown to decrease the bioavailability and toxicity of other metals to aquatic organisms. In the present study, acute Ni toxicity experiments (96-h) were conducted at various salinities (0-36 ppt) to determine the effects of salinity on Ni toxicity to 2 euryhaline fish species, Kryptolebias marmoratus and Fundulus heteroclitus. Nickel concentrations causing lethality to 50% of the fish ranged from 2 mg/L in moderately hard freshwater to 66.6 mg/L in 36 ppt

Bioaccumulation of As, Cd, Cr, Hg(II), and MeHg in killifish (Fundulus heteroclitus) from amphipod and worm prey.

Elevated metal levels in fish are a concern for the fish themselves, their predators, and possibly humans who consume contaminated seafood. Metal bioaccumulation models often rely on assimilation efficiencies (AEs) of ingested metals and loss rate constants after dietary exposure (k(ef)s). These models can be used to better understand processes regulating metal accumulation and can be used to make site-specific predictions of metal concentrations in animal tissues. Fish often consume a varied diet, and prey choice can influence these two parameters. We investigated the trophic transfer of As,

Aryl hydrocarbon receptor ligands increase ABC transporter activity and protein expression in killifish (Fundulus heteroclitus) renal proximal tubules.

Many widespread and persistent organic pollutants, for example, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and some polychlorinated biphenyls, activate the aryl hydrocarbon receptor (AhR) causing it to translocate to the cell nucleus where it transactivates target genes, increasing expression of a number of xenobiotic metabolizing enzymes as well as some transporters. AhR's ability to target transporters within the kidney is essentially unexplored. We show here that exposing isolated killifish (Fundulus heteroclitus) renal proximal tubules to micromolar β-naphthoflavone (BNF) or nanomolar TCDD

Can diversifying selection be distinguished from history in geographic clines? A population genomic study of killifish (Fundulus heteroclitus).

A common geographical pattern of genetic variation is the one-dimensional cline. Clines may be maintained by diversifying selection across a geographical gradient but can also reflect historical processes such as allopatry followed by secondary contact. To identify loci that may be undergoing diversifying selection, we examined the distribution of geographical variation patterns across the range of the killifish (Fundulus heteroclitus) in 310 loci, including microsatellites, allozymes, and single nucleotide polymorphisms. We employed two approaches to detect loci under strong diversifying sele

Altered lipid homeostasis in a PCB-resistant Atlantic killifish (Fundulus heteroclitus) population from New Bedford Harbor, MA, U.S.A.

Sentinel species such as the Atlantic killifish (Fundulus heteroclitus) living in urban waterways can be used as toxicological models to understand impacts of environmental metabolism disrupting compound (MDC) exposure on both wildlife and humans. Exposure to MDCs is associated with increased risk of metabolic syndrome, including impaired lipid and glucose homeostasis, adipogenesis, appetite control, and basal metabolism. MDCs are ubiquitous in the environment, including in aquatic environments. New Bedford Harbor (NBH), Massachusetts is polluted with polychlorinated biphenyls (PCBs), and, as

Effects of temperature, ionic strength and pH on the function of skeletal muscle myosin from a eurythermal fish, Fundulus heteroclitus.

The mummichog, Fundulus heteroclitus, is an intertidal fish that exhibits little change in swimming ability despite large and rapid variations in environmental parameters. We therefore tested the hypothesis that this nearly constant function is due to Fundulus myosin being intrinsically insensitive to changes of temperature, ionic strength and pH. In vitro motility assays were used to quantify the speed of unregulated actin filaments on myosin purified from F. heteroclitus glycolytic skeletal muscle. Filament speed was 2.07+/-0.17 microm s(-1) at 26 degrees C, ionic strength (Gamma/2) of 0.08
