Postharvest fungal diseases significantly reduce the quality and shelf life of produce, leading to major economic losses. This study evaluated the antifungal potential of an extract of fresh Gracilaria coronopifolia (GCE), a red seaweed rich in sulfated polysaccharides, against postharvest fungal pathogens. Chemical analysis revealed that the extract contained 66.07 ± 1.98% total sugar, an anhydrogalactose-to-galactose ratio of 1:1.84, 5.90 ± 0.05% sulfate content, a degree of sulfation of 0.37, and a molecular weight of 25.094 kDa. GCE exhibited concentration- and species-dependent antifungal

Pathogens are often present alongside other microbial species in chronic infections. The mold Aspergillus fumigatus and bacteria Pseudomonas aeruginosa are both prevalent opportunistic pathogens in cystic fibrosis (CF) patients, and their interactions play a role in the pathogenesis of chronic airway disease. In this study, their interactions were investigated during the formation of A. fumigatus-P. aeruginosa mixed biofilms. P. aeruginosa is capable of differentially modulating mixed biofilm formation, shifting from suppressing fungal growth to promoting robust mixed biofilm formation as the

PURPOSE: The applicability of radioimmunotherapy with organism-specific monoclonal antibodies to treatment of infectious disease in experimental models has been recently shown for fungal, bacterial, and viral infections. To identify the best delivery vehicle for radioimmunotherapy of human pathogenic fungus Cryptococcus neoformans (CN), we have done comparative evaluation of capsular polysaccharide-specific antibodies with IgG1 and IgM isotypes and F(ab')2 and Fab fragments.
EXPERIMENTAL DESIGN: 18B7 IgG1 and 13F1 IgM and their isotype-matching controls were radiolabeled with 188Re, and their

Climate and environmental changes have modified the habitats of fungal pathogens, inflicting devastating effects on livestock and crop production. Additionally, drug-resistant fungi are increasing worldwide, driving the urgent need to identify new molecular scaffolds for the development of antifungal agents for humans, animals, and plants. Poacic acid (PA), a plant-derived stilbenoid, was recently discovered to be a novel molecular scaffold that inhibits the growth of several fungi. Its antifungal activity has been associated with perturbation of the production/assembly of the fungal cell wall

Polysaccharides from fungi are good free radical scavengers. However, there are no enzymes digesting these polysaccharides in the human body, which limits the use of fungal polysaccharides. Therefore, it is of great significance to study the preparation methods of fungal polysaccharides to improve the utilization rate of fungal polysaccharides. In this paper, the acidic polysaccharide of Tremella fuciformis was extracted by boiling and precipitated by ethanol. The total sugar content obtained by freeze-drying after ion exchange chromatography purification was 93.6%. It is mainly composed of ma

Methodologies to identify epitopes or ligands of the fungal cell wall polysaccharides influencing the immune response of human pathogens have to date been imperfect. Using the galactomannan (GM) of Aspergillus fumigatus as a model, we have shown that synthetic oligosaccharides of distinct structures representing key fragments of cell wall polysaccharides are the most precise tools to study the serological and immunomodulatory properties of a fungal polysaccharide.

Fungal cell wall polysaccharides - such as β-glucans, chitin, and mannans - are key immunogenic structures recognized by Toll-like receptors and C-type lectin receptors on innate immune cells, initiating phagocytosis, cytokine production, and antigen presentation, and shaping downstream adaptive immunity. CD4+ T cells differentiate into Th1, Th2, Th17, and Treg subsets to maintain immune balance, whereas B cells contribute to fungal clearance and memory through species-specific antibodies. The boundary between protective and pathogenic immunity varies with fungal species, infection stage, and

Both fungi and bacteria contribute to the efficiency of activated sludge processes. However, fungal degradability has historically been overlooked in the anaerobic fermentation of waste-activated sludge (WAS). This study investigated the role of an enriched microbial consortium of degrading fungi (MCDF) in WAS fermentation. MCDF was initially enriched using a mixture of WAS and two mesophilic anaerobic sludges with chitin as the substrate. Fungal diversity and key cell wall polysaccharides, including chitin, mannan, and glucans, were identified in WAS. The addition of enriched MCDF to WAS sign

Fungal polysaccharides have garnered attention for their antidiabetic potential. Here, we evaluated five edible fungal polysaccharides-Agaricus bisporus (ABP), Pleurotus eryngii (PEP), Boletus spp. (BP), Lentinus edodes (LEP), and Pleurotus ostreatus (POP) in type 2 diabetic mice. All alleviated diabetic symptoms to varying degrees, with ABP showing the most pronounced effects. ABP improved body weight, reduced hyperphagia and polydipsia, lowered fasting glucose, and enhanced glucose tolerance and insulin sensitivity. Histological analysis showed that ABP better preserved pancreatic islets and

More than 90% of the cell wall of the filamentous fungus Aspergillus fumigatus comprises polysaccharides. Biosynthesis of the cell wall polysaccharides is under the control of three types of enzymes: transmembrane synthases, which are anchored to the plasma membrane and use nucleotide sugars as substrates, and cell wall-associated transglycosidases and glycosyl hydrolases, which are responsible for remodeling the de novo synthesized polysaccharides and establishing the three-dimensional structure of the cell wall. For years, the cell wall was considered an inert exoskeleton of the fungal cell.

Polymers, mainly polysaccharides, are a class of macromolecules that have a variety of structural and functional roles in biological systems. Recently, research on polysaccharides, especially exopolysaccharides (EPS) from microbial sources, has gained significant interest. These substances are valued for their biocompatibility, biodegradability, and wide range of applications across the pharmaceutical, biomedical, and industrial sectors. Endophytic microorganisms, which live inside healthy plant tissues, are a newly discovered sustainable source of exopolysaccharides. This article reviews rese

Over the past several decades, research on the synthesis and organization of the cell wall polysaccharides of Aspergillus fumigatus has expanded our knowledge of this important fungal structure. Besides protecting the fungus from environmental stresses and maintaining structural integrity of the organism, the cell wall is also the primary site for interaction with host tissues during infection. Cell wall polysaccharides are important ligands for the recognition of fungi by the innate immune system and they can mediate potent immunomodulatory effects. The synthesis of cell wall polysaccharides

Bacteria and fungi have developed resistance to the existing therapies such as antibiotics and antifungal drugs, and multiple mechanisms are mediating this resistance. Among these, the formation of an extracellular matrix embedding different bacterial cells, called biofilm, is an effective strategy through which bacterial and fungal cells are establishing a relationship in a unique environment. The biofilm provides them the possibility to transfer genes conferring resistance, to prevent them from desiccation and to impede the penetration of antibiotics or antifungal drugs. Biofilms are formed

Solid state cultivation of Ganoderma lucidum biomass, strain BFWS Gal 4, originally isolated from the Slovenian forest, was studied in a horizontal stirred tank reactor. Periodic mixing of N = 80 rpm, 2 min/day was used. Production of fungal polysaccharides and fungal biomass on solid substrate based on beech sawdust, olive oil, and mineral salts was studied. Optimal moisture of the solid matrix was in the range of 80% to 74%. When the moisture content dropped below 57%, the growth of the mycelium and polysaccharide production stopped, but it revived when wet air was applied in further process

Myeloid-derived suppressor cells (MDSCs) are a heterogeneous group of immune cells that play a central role in regulating host immune responses during fungal infections. Their recruitment is mediated by pathogen recognition receptors, particularly Dectin-1 and CARD9 signaling, which promote the production of reactive oxygen species (ROS) and IL-1β. Once activated, MDSCs suppress T-cell and natural killer cell functions through immunosuppressive cytokines like IL-10 and TGF-β, as well as enzymes such as arginase-1 and indoleamine 2,3-dioxygenase 1 (IDO-1). This review explores the role of MDSCs

Multiple sclerosis (MS) is a chronic, inflammatory disease of the central nervous system, whose causes are still unknown. We have proposed that MS, as well as some ophthalmologic diseases, are associated with fungal infection. In the present study, we closely monitored a patient with MS over a three-year period. Antibodies against different Candida spp. were detected in peripheral blood serum, although the titer of these antibodies fluctuated. The presence of fungal macromolecules, such as proteins, polysaccharides, and DNA, was also tested. In several sera samples, antigens related to C. fama

Silver nanoparticles (AgNPs) synthesized via fungal-mediated biosynthesis have gained attention as eco-friendly alternatives to chemically produced nanomaterials, with broad biomedical potential. Fungi represent particularly attractive systems because their secretomes contain diverse biomolecules, including enzymes, proteins, polysaccharides, and secondary metabolites, capable of reducing silver ions and stabilizing the resulting nanoparticles. Despite extensive investigation, the molecular mechanisms underlying fungal-mediated AgNP formation remain poorly defined. This review critically exami

Radiation exposure can lead to reproductive damage (RD), for which there is currently no effective treatment. Natural compounds, particularly fungal polysaccharides, have shown promising therapeutic potential for RD. Due to limited availability of effective polysaccharides, research has turned to alternative sources from edible mushrooms. This study evaluated polysaccharides from Pleurotus eryngii, Agaricus bisporus, Coprinus comatus, and Pleurotus citrinopileatus for their effects on male mice subjected to X-ray radiation. Histological analysis demonstrated significant damage to testicular ti

BACKGROUND: Fungal cell wall polysaccharides maintain the integrity of fungi and interact with host immune cells. The immunomodulation of fungal polysaccharides has been demonstrated in previous studies. However, the effect of chitin-rich heteroglycan extracted from Sporothrix schenckii sensu stricto on the immune response has not been investigated.
RESULTS: In this study, chitin-rich heteroglycan was extracted from S. schenckii sensu stricto, and immunomodulation was investigated via histopathological analysis of skin lesions in a mouse model of sporotrichosis and evaluation of the phagocyti
