Natural cross-kingdom transmission of a novel ssDNA mycovirus confers broad-spectrum resistance to plant diseases.

Cross-kingdom virus transmission between plants and fungi remains rare in nature. Here, a novel circular ssDNA virus, Diaporthe pseudophoenicicola DNA virus 1 (DpDV1), was identified from the phytopathogenic fungus Diaporthe pseudophoenicicola. DpDV1 attenuates the pathogenicity of both its original and experimental host fungi, demonstrating its potential as a biocontrol agent against fungal phytopathogens. Remarkably, DpDV1 displays bidirectional plant-fungal transmission capability without inducing observable phenotypic changes in plant hosts, overcoming the limitations imposed by vegetative

Investigating RNA Viruses Infecting Arbuscular Mycorrhizal Fungi.

Fungi are known to be frequently infected by mycoviruses, which could play an important yet underexplored role in the fungal holobiont. Mycoviruses can enhance fungal traits, such as salinity tolerance and resistance to fungicides, and may also benefit plant hosts in tripartite interactions. While research on arbuscular mycorrhizal fungi (AMF) has focused on their ecological and agricultural value, few studies have explored their virome, possibly due to the difficulty of culturing AMF in the lab. This study presents an updated bioinformatic approach for virome characterization, emphasizing RNA

A moderate level of hypovirulence conferred by a hypovirus in the avocado white root rot fungus, Rosellinia necatrix.

Two isolates of Rosellinia necatrix (Rn118-8 and Rn480) have previously obtained from diseased avocado trees in commercial orchards of the coastal area in southern Spain. Rn118-8 and Rn480 have weak virulence on avocado plants, and are infected by R. necatrix hypovirus 2 (RnHV2). In this work, the possible biological effects of the hypovirus on R. necatrix were tested. First, RnHV2 was transmitted from each of Rn118-8 and Rn480 to a highly virulent, RnHV2-free isolate of R. necatrix (Rn400) through hyphal anastomosis, using zinc compounds which attenuate the mycelial incompatibility reactions

Potential of small-molecule fungal metabolites in antiviral chemotherapy.

Various viral diseases, such as acquired immunodeficiency syndrome, influenza, and hepatitis, have emerged as leading causes of human death worldwide. Scientific endeavor since invention of DNA-dependent RNA polymerase of pox virus in 1967 resulted in better understanding of virus replication and development of various novel therapeutic strategies. Despite considerable advancement in every facet of drug discovery process, development of commercially viable, safe, and effective drugs for these viruses still remains a big challenge. Decades of intense research yielded a handful of natural and sy

Pulmonary infections in the hematopoietic cell transplant patient.

PURPOSE OF REVIEW: Despite progress in the management of pulmonary infections in the hematopoietic cell transplant (HCT) population, substantial diagnostic, and therapeutic uncertainty remains. RECENT FINDINGS: A growing HCT population reflects more transplants and improved long-term survival. We continue to learn about the epidemiologic and prognostic significance of posttransplant pulmonary infections. Mold-active triazoles have removed invasive fungal pneumonia as a barrier to transplant eligibility. Ibrutinib and respiratory viruses are newly recognized risk factors for invasive fungal di

Botrytis cinerea Protoplasts Production and Inoculation.

The absence of a cell wall turns fungal protoplasts very receptive to the uptake of exogenous genetic material and, consequently, they can be used for genetic transformation or transfection experiments. Additionally, isolated monosporic fungal strains, naturally infected or previously transfected with a mycovirus, can be used in assays of fungal protoplasts fusion. This technology will overcome the barriers of vegetative incompatibility with other Botrytis cinerea strains, or even with other fungi, and successfully allow the transference of mycoviruses from an infected to a non-infected fungal

The Viruses of Botrytis cinerea and Beyond: Molecular Characterization of RNA Viruses and Retroplasmids.

Over the past five years, research has progressively revealed a rich diversity of RNA viruses in Botrytis cinerea. In this study, we identified nine RNA viruses from the viromes of three B. cinerea strains, including five mitoviruses, one umbra-like virus, and three partitiviruses. Among these, Sclerotinia sclerotiorum partitivirus 1 (SsPV1) was artificially introduced in a previous study. Excluding SsPV1, we cloned the other two partitiviruses and confirmed that both belong to Gammapartitivirus and contain three genomic segments, with dsRNA3 as an RNA satellite. In addition to RNA viruses, we

[Non herpetic keratitis].

Nonherpetic keratitis includes keratitis caused by other viruses, and bacterial, fungal, and acanthamoebic keratitis. Epidemic keratoconjunctivitis, one of the most common corneal manifestations induced by nonherpetic viruses, has usually a good outcome. Conversely, non viral keratitis has more severe complications. Early clinical and microbial diagnosis are the keystones to treat successfully these keratitis. Diagnostic and therapeutic principles are detailed in this review.

The gut mycobiome is shaped by interactions with the bacterial community in twins.

The human gut microbiome comprises bacteria, viruses, and fungi, yet the fungal component (mycobiome) remains poorly characterized. Here, we investigated gut fungal composition and fungal-bacterial interactions in healthy monozygotic and dizygotic twins. Fungal communities showed substantially higher inter-individual variability than bacterial communities. Zygosity, age, and shared environment had no major influence on fungal abundance, similarity, or dominant genera. Candida was the most abundant genus (mean 5.2% in 161 individuals), followed by Geotrichum (3.7% in 132), whereas Saccharomyces

Elimination of Airborne Microorganisms Using Compressive Heating Air Sterilization Technology (CHAST): Laboratory and Nursing Home Setting.

BACKGROUND: Airborne transmission of bacteria, viruses, and fungal spores poses a major threat in enclosed settings, particularly nursing homes where residents are highly vulnerable. Compressive Heating Air Sterilization Technology (CHAST) applies compressive heating to inactivate microorganisms without reliance on filtration or chemicals. METHODS: CHAST efficacy was evaluated in laboratory and deployed for a feasibility and performance validation study of air sterilization in a nursing home environment. Laboratory studies tested prototypes (300-5000 CFM; 220-247 °C) against aerosolized surro

Longitudinal epidemiology of respiratory viruses in hospitalized patients in Oman (2017-2025).

BACKGROUND: Respiratory virus epidemiology in hospitalized patients is influenced by host susceptibility, seasonality, and health system disruptions; however, longitudinal patterns across major population-level events remain incompletely defined. METHODS: We conducted a single-center longitudinal study of 6776 hospitalized patients in Oman from 2017 to 2025. Respiratory viruses were detected using multiplex real-time PCR and analyzed descriptively by age group, season, and epidemiological period (pre-pandemic, pandemic, post-pandemic), without adjustment for testing intensity over time. Tracking Mycoviruses in Public RNAseq Datasets of Malassezia: Three Original Totiviruses Revealed.

Mycoviruses are viruses that selectively infect and multiply in fungal cells. Malassezia is the most abundant fungus on human skin and is associated with a variety of conditions, including atopic eczema, atopic dermatitis, dandruff, folliculitis, pityriasis versicolor, and seborrheic dermatitis. Here, we conducted mycovirome studies on 194 public transcriptomes of Malassezia (2,568,212,042 paired-end reads) screened against all available viral proteins. Transcriptomic data were assembled de novo resulting in 1,170,715 contigs and 2,995,306 open reading frames (ORFs) that were subsequently trac

An isolate of human blood-associated partitivirus naturally infects the phytopathogenic fungus Bipolaris maydis.

Human blood-associated partitivirus (HuBPV) was first identified through metagenomic analysis of serum samples from two Peruvians, but its natural host remains unknown. Here, we report the detection of an HuBPV strain (HuBPV-Bm) in the phytopathogenic fungus Bipolaris maydis strain HN11 in Hubei Province, China. The dsRNA1 and dsRNA2 of HuBPV-Bm show more than 97.6% and 98.8% nucleotide sequence identity, respectively, to those from the metagenomically discovered HuBPV strain (HuBPV-M). Notably, HuBPV-Bm contains a third dsRNA segment that was not reported for HuBPV-M. All mycelia derived from

Breaking Through to the Other Side: How Plant Viruses Modify Plasmodesmata and the Plant Cell Wall.

Viruses are obligate intracellular parasites that rely on their hosts for the cellular machinery necessary for their replication and transmission to new hosts. Viruses are therefore invaluable for illuminating numerous facets of their hosts' biology. Plant viruses take advantage of the cell-to-cell connectivity provided by plasmodesmata for their movement from sites of replication to the vasculature for eventual systemic dissemination in their plant hosts, a strategy distinct from viruses that use animal and fungal hosts. Given their reliance on plasmodesmata for successful infection and sprea

The impact of the host microbiota on Candida albicans infection.

The human microbiota is a complex microbial ecosystem populated by bacteria, fungi, viruses, protists, and archaea. The coexistence of fungi alongside with many billions of bacteria, especially in the gut, involves complex interactions, ranging from antagonistic to beneficial, between the members of these two kingdoms. Bacteria can impact fungi through various means, such as physical interactions, secretion of metabolites, or alteration of the host immune response, thereby affecting fungal growth and virulence. This review summarizes recent progress in this field, delving into the latest under

Known unknowns: how might the persistent herpesvirome shape immunity and aging?

The microbial community that colonizes all living organisms is gaining appreciation for its contributions to both physiologic and pathogenic processes. The virome, a subset of the overall microbiome, large and diverse, including viruses that persistently inhabit host cells, endogenous viral elements genomically or epigenomically integrated into cells, and viruses that infect the other (bacterial, protozoan, fungal, and archaeal) microbiome phylla. These viruses live in the organism for its life, and therefore are to be considered part of the aging process experienced by the organism. This revi

Biodiversity of microorganisms in the Baltic Sea: the power of novel methods in the identification of marine microbes.

Until recently, the data on the diversity of the entire microbial community from the Baltic Sea were relatively rare and very scarce. However, modern molecular methods have provided new insights into this field with interesting results. They can be summarized as follows. (i) Although low salinity causes a reduction in the biodiversity of multicellular species relative to the populations of the North-East Atlantic, no such reduction occurs in bacterial diversity. (ii) Among cyanobacteria, the picocyanobacterial group dominates when considering gene abundance, while filamentous cyanobacteria dom

The therapeutic potential of phage-based antifungal treatment: strategies, mechanisms, and prospects.

Human fungal infections are increasingly being recognized as a significant global health threat. The burden of fungal diseases is escalating, primarily due to the rising number of at-risk individuals, compounded by the limited availability of antifungal therapies that are both effective and minimally toxic. Phages, viruses that specifically infect and kill bacteria, have long been investigated for their therapeutic potential. However, despite their success in bacteriology, the applications of phages in antifungal therapy are under active research. Particularly, phages could be used to treat fu

Brilacidin, a host defense peptide mimetic, potentiates ibrexafungerp antifungal activity against the human pathogenic fungus Aspergillus fumigatus.

UNLABELLED: Aspergillus fumigatus is the primary etiological agent of aspergillosis. Here, we show that the host defense peptide mimetic brilacidin (BRI) can potentiate ibrexafungerp (IBX) against clinical isolates of A. fumigatus. BRI + IBX can inhibit the growth of A. fumigatus voriconazole- and caspofungin-resistant clinical isolates. BRI is a small molecule host defense peptide mimetic that has previously exhibited broad-spectrum immunomodulatory/anti-inflammatory activity against viruses, bacteria, and fungi. In vitro, combination of BRI + IBX plays a fungicidal role, increases the fungal
