ABSTRACT Crown and root rot of tomato caused by Fusarium oxysporum f. sp. radicis-lycopersici is an increasing problem in Europe, Israel, Japan, and North America. The biocontrol agent Pseudomonas fluorescens strain CHA0 provides only moderate control of this disease. A one-time amendment of zinc EDTA at 33 mug of Zn(2+)/ml to hydroponic nutrient solution in soilless rockwool culture did not reduce disease when used alone, but did reduce disease by 25% in the presence of CHA0. In in vitro studies with the pathogen, zinc at concentrations as low as 10 mug/ml abolished production of the phytotox

The influence of sympathetic nervous activity on cerebral circulation and cerebrovascular CO2 reactivity was investigated through inhibition of dopamine beta-hydroxylase (DBH). A PO2 electrode, a PCO2 electrode and a plate-type thermocouple-flowmeter were placed on the pial surface of the cat brain. Cerebrocortical PO2, PCO2, cerebrocortical blood flow and arterial blood pressure were continuously recorded before, during and after intracarotid infusion of 10 mg/kg of fusaric acid, a potent DBH inhibitor. The effects of 5% CO2 inhalation and hyperventilation were measured before and after the i

A magnetic nanosensor-based method is described to screen a library of drugs for potential binding to toxins. Screening is performed by measuring changes in the magnetic relaxation signal of the nanosensors (bMR nanosensors) in aqueous suspension upon addition of the toxin. The Anthrax lethal factor (ALF) is selected as a model toxin to test the ability of our bMR nanosensor-based screening method to identify potential inhibitors of the toxin. Out of 30 molecules screened, sulindac, naproxen and fusaric acid are found to bind LF, with dissociation constants in the low micromolar range. Further

The fungi Fusarium guttiforme and Phytophthora palmivora were cultivated in four different media (Potato Dextrose Agar, Czapek, rice, and ISP2) and co-cultured to stimulate fungal interactions and enhance secondary metabolite production. Promising extracts were fractionated, yielding compounds such as the iron complex of fusaric acid (1), magnesium complex of fusaric acid (2), haematocin (4), fusarinolic acid (7), and cyclonerodiol (8). These compounds exhibited significant papain inhibitory activity, with compounds 2 and 4 showing IC50 values below 20 µM. Notable anti-Trypanosoma cruzi activi

Multitrophic interactions mediate the ability of fungal pathogens to cause plant disease and the ability of bacterial antagonists to suppress disease. Antibiotic production by antagonists, which contributes to disease suppression, is known to be modulated by abiotic and host plant environmental conditions. Here, we demonstrate that a pathogen metabolite functions as a negative signal for bacterial antibiotic biosynthesis, which can determine the relative importance of biological control mechanisms available to antagonists and which may also influence fungus-bacterium ecological interactions. Four water-soluble Fusarium metabolites (fumonisin B1, fusaric acid, butenolide and moniliformin), water-insoluble pigment (8-O-methylbostrycoidin), and an Alternaria metabolite (AAL-toxin) were tested for relative cytotoxicity to five established mammalian cell lines. Butenolide was the most cytotoxic to all five cell lines. LC50s were; 1 microgram/ml to rat hepatoma (RH) (tumors derived from parenchymal cells), 7 micrograms/ml to baby hamster kidney (BHK-21) fibroblast cells, and 15 micrograms/ml to McCoy mouse (MM) fibroblast cells: LC100s were 1 microgram/ml to Chinese hamster ovary (CHO)

Pseudomonas protegens synthesizes two major iron-chelating metabolites (siderophores): pyoverdine (Pvd) and enantio-pyochelin (E-Pch). Although iron sequestration and uptake seem to be the main biological role of these siderophores, other functions including metal homeostasis and antibiotic activity have been proposed. The aim of this study was to evaluate the contribution of Pvd and E-Pch to the survival of P. protegens in soil using wild type and isogenic mutant strains unable to produce Pvd, E-Pch or both siderophores. Survival of these strains in sterile soil microcosms, in soil microcosms

Fusarium fujikuroi causes bakanae ("foolish seedling") disease of rice which is characterized by hyper-elongation of seedlings resulting from production of gibberellic acids (GAs) by the fungus. This plant pathogen is also known for production of harmful mycotoxins, such as fusarins, fusaric acid, apicidin F and beauvericin. Recently, we generated the first de novo genome sequence of F. fujikuroi strain IMI 58289 combined with extensive transcriptional, epigenetic, proteomic and chemical product analyses. GA production was shown to provide a selective advantage during infection of the preferre

Background/Objectives: Stem base rot caused by Fusarium oxysporum is a devastating soil-borne disease, yet long-term chemical pesticide application has raised serious environmental and ecological concerns. Thymol, a plant-derived fungicide, demonstrates a favorable safety profile for humans and environmental systems while exhibiting broad-spectrum antifungal efficacy. This study aimed to evaluate the inhibitory effects of thymol against F. oxysporum and to elucidate its underlying mechanisms of action. Methods: The antifungal activity of thymol was assessed in vitro against F. oxysporum by mea

Analysis of melatonin (MEL) in pineal cell cultures by enzyme linked immunosorbent assay showed its concentration was increased by fusaric acid (FA), a mycotoxin produced by Fusarium species and associated with toxic duck and ostrich feeds. Subsequent cell culture studies demonstrated the precursors of MEL, 5-hydroxytryptophan (5HTP) and serotonin (5HT), were also affected by FA as well as other Fusarium mycotoxins. Herein we describe a technique for the analysis of 5HTP and 5HT in pineal cell cultures using HPLC with electrochemical detection (EC), and report on the effects of FA alone and in

The influence of cholinergic and dopaminergic drugs has been studied on the metabolism of GABA by measuring the levels of GABA and glutamate and the activity of glutamic acid decarbosylase and GABA-aminotransferase in mouse and rat brain and in some rat brain regions. Muscarinic agonists (oxotremorine, arecoline) induce a rapid decrease of GABA level and an increase of its destruction, indicative of an increased activity of GABA synapses in basal ganglia and brain stem. Minor changes in GABA metabolism are observed in cerebral cortex and cerebellum. On the other hand, hemicholinium-3 and the m

Early diagnosis of fungal infections in crops is essential for mitigating yield losses, yet most detection strategies rely on destructive assays that are incompatible with in situ monitoring. Among major fungal pathogens, Fusarium species pose a significant threat to global agriculture and produce the phytotoxin fusaric acid (FA), a key biomarker of early infection. Here, we develop a non-destructive, ratiometric optical nanosensor that integrates aggregation-induced emissive carbon dots within a zeolitic imidazolate framework (CD@ZIF-8) for selective FA detection in living plants. By rational

Shigella flexneri SFL124, with a deletion encompassing all, or nearly all, of the coding sequence of gene aroD was obtained after selection on a fusaric acid medium supplemented with 2,3-dihydroxybenzoic acid for tetracycline-sensitive mutants of S. flexneri SFL114 which is an aroD::Tn10 transductant. Two of 20 tetracycline-sensitive mutants tested in colony hybridization with a 32P-labelled DNA probe of approximately 1400 base pairs (comprising all except the 75 N-terminal base pairs of the coding region of gene aroD) did not hybridize. The selected mutant SFL124 is Congo-red positive, invade

Bulb rot caused by Fusarium oxysporum (F. oxysporum) is a major disease limiting the development of the Lanzhou lily industry. Fusaric acid (FA), a key virulence factor secreted by the pathogen, induces programmed cell death and accelerates bulb decay. Trace amounts of FA therefore serve as a critical biomarker for early disease warning and for screening eco-friendly biocontrol agents. To address the lack of rapid monitoring tools and reduce reliance on chemical pesticides, this study developed a molecularly imprinted electrochemical sensor for FA detection. The sensor was constructed on a chi

There is considerable similarity between paranoid schizophrenia and psychoses provoked by dopaminergic overstimulation in the central nervous system. The fact that neuroleptics are able to block dopaminergic neural activity has led to the hypothesis that there might exist a common biochemical substrate for schizophrenia and e. g. the amphetamine psychoses. Dopaminergic overstimulation may be elicited by different drugs interacting with the dopamine metabolism e. g. dopamine-beta-hydroxylase inhibition (disulfiram, fusaric acid); monoamine-oxidase-inhibition (phenelzine, tranylcypromine); dopam

Tetracyclines and bacitracin are used extensively as a growth promotant and a therapeutic, respectively in livestock. In this study, we test whether there is an interaction between bacitracin and tetracycline that can select for an increase in microbial tetracycline sensitivity, using Escherichia coli and Salmonella as a model. We found via in vitro studies that bacitracin at sublethal concentrations preferentially selects for outgrowth of tetracycline-sensitive bacteria from among a population of tetracycline-resistant and tetracycline-sensitive E. coli and Salmonella strains. Most of the bac

Strawberry root rot is a serious soil-borne root disease with a high incidence rate that is extremely difficult to control. To identify the fungal pathogens responsible for strawberry root rot, diseased plants exhibiting root rot symptoms were collected from Jining, Shandong Province, China. Strain CMGF-RB13 was isolated from the roots of infected strawberry plants, and based on morphological characteristics and molecular analyses, was identified as Fusarium verticillioides. To our knowledge, this is the first study of strawberry root rot disease caused by F. verticillioides in China. The geno
