A mixed chemical/electrical synapse can generate variable output when the strength of each synaptic component is modulated. At mixed synapses of the lobster pyloric network, the chemical component is inhibitory. Without neuromodulation, the chemical component is weak or absent and the electrical component often dominates. Dopamine reverses the sign of these mixed synaptic interactions by a reduction in the strength of electrical coupling and an enhancement of chemical inhibition, including activation of silent chemical synapses. Sign reversal at mixed synapses by neuromodulators may contribute

Repetitive transcranial magnetic stimulation (rTMS) is emerging as a groundbreaking treatment for eating disorders, such as anorexia and bulimia nervosa, though its underlying neurological mechanisms remain elusive. To shed light on the effects of rTMS on neural control of feeding behavior, we investigated the feeding responses in the marine mollusk Aplysia californica. Using in vitro preparations, we elicited feeding responses and performed dual nerve recordings to identify motor programs associated with rejection and ingestion. We applied rTMS to the cerebral ganglion, the neural center resp

Crustacean hyperglycemic hormones (CHHs) are neuropeptides involved in the regulation of hemolymph glucose. The primary source of CHHs has been identified as the neurosecretory neurons of the eyestalk X-organ and its associated neurohemal organ, the sinus gland. We have identified another source of CHH-like peptides in the nervous system. With the use of immunocytochemistry, cells in the second roots of the thoracic ganglia have been observed to stain positively for CHH-reactive material. We also identified a pair of cells in the subesophageal ganglion that contain large amounts of CHH-reactiv

Studies of hormone influences on neural circuits and behavior have primarily focused on the manipulation of individual hormones. Here, we examine the influence of behavioral (feeding) state-specific hormonal environments on the gastric mill (chewing) circuit configured by the neuropeptide Gly1-SIFamide (G-SIFamide) in the isolated Cancer borealis stomatogastric ganglion (STG). The G-SIFamide-activated gastric mill rhythm, which is similar to that driven by the G-SIFamidergic projection neuron modulatory commissural neuron 5 (MCN5), is distinguished from other gastric mill rhythms by the presen

Recent computational and experimental work has shown that similar network performance can result from variable sets of synaptic and intrinsic properties. Because temperature is a global perturbation that differentially influences every biological process within the nervous system, one might therefore expect that individual animals would respond differently to temperature. Nonetheless, the phase relationships of the pyloric rhythm of the stomatogastric ganglion (STG) of the crab, Cancer borealis, are remarkably invariant between 7 and 23°C (Tang et al., 2010). Here, we report that, when isolate

Motivated behaviors comprise appetitive actions whose occurrence results partly from an internally driven incentive to act. Such impulsive behavior can also be regulated by external rewarding stimuli that, through learning processes, can lead to accelerated and seemingly automatic, compulsive-like recurrences of the rewarded act. Here, we explored such behavioral plasticity in Aplysia by analyzing how appetitive reward stimulation in a form of operant conditioning can modify a goal-directed component of the animal's food-seeking behavior. In naive animals, protraction/retraction cycles of the

Circuit function results from both intrinsic conductances of network neurons and the synaptic conductances that connect them. In models of neural circuits, different combinations of maximal conductances can give rise to similar activity. We compared the robustness of a neural circuit to changes in their intrinsic versus synaptic conductances. To address this, we performed a sensitivity analysis on a population of conductance-based models of the pyloric network from the crustacean stomatogastric ganglion (STG). The model network consists of three neurons with nine currents: a sodium current (Na

Neuronal activity and energy supply must maintain a fine balance for neuronal fitness. Various channels of communication between the two could impact network output in different ways. Sulfonylurea receptors (SURs) are a modification of ATP-binding cassette proteins that confer ATP-dependent gating on their associated ion channels. They are widely expressed and link metabolic states directly to neuronal activity. The role they play varies in different circuits, both enabling bursting and inhibiting activity in pathological conditions. The crab, Cancer borealis, has central pattern generators (C

Ionic current levels of identified neurons vary substantially across individual animals. Yet, under similar conditions, neural circuit output can be remarkably similar, as evidenced in many motor systems. All neural circuits are influenced by multiple neuromodulators, which provide flexibility to their output. These neuromodulators often overlap in their actions by modulating the same channel type or synapse, yet have neuron-specific actions resulting from distinct receptor expression. Because of this different receptor expression pattern, in the presence of multiple convergent neuromodulators

The Motor Ganglion (MG) is a small collection of neurons that control the swimming movements of the tunicate tadpole larva. Situated at the base of the tail, molecular and functional comparisons suggest that may be a homolog of the spinal cord and/or hindbrain ("rhombospinal" region) of vertebrates. Here we review the most current knowledge of the development, connectivity, functions, and unique identities of the neurons that comprise the MG, drawn mostly from studies in Ciona spp. The simple cell lineages, minimal cellular composition, and comprehensively mapped "connectome" of the Ciona MG a

Motor networks such as the pyloric network of the stomatogastric ganglion often require descending neuromodulatory inputs to initiate, regulate, and modulate their activity and their synaptic connectivity to manifest physiologically appropriate output. Prolonged removal of these descending inputs often results in a compensatory response that alters the inputs themselves, their targets, or both. Using the pyloric network of the crab, Cancer borealis, we investigated whether isolation of motor networks would result in alterations that change the responses of these networks to restored modulatory

How are stereotyped behaviors organized in a simple nervous system? A new study in the Drosophila larva reports that the foraging routine can be performed in the absence of any input from the brain.

Neuronal release of modulatory substances provides motor pattern generating circuits with a high degree of flexibility. In vitro studies have characterized the actions of modulatory projection neurons in great detail in the stomatogastric nervous system, a model system for neuromodulatory influences on central pattern generators. Less is known about the activities and actions of modulatory neurons in fully functional and richly modulated network settings, i.e., in intact animals. It is also unknown whether their activities contribute to the motor patterns in different behavioral conditions. He

The neurons innervating muscles that deliver the enormous power enabling froghopper insects to excel at jumping were revealed by backfilling the nerves from those muscles. The huge trochanteral depressor muscle (M133) of a hind leg consists of four parts. The two largest parts (M133b,c) occupy most of the metathorax and are innervated by the same two motor neurons that have small, laterally placed somata in the metathoracic ganglion and axons in nerve N3C(2). They are also supplied by three dorsal unpaired median (DUM) neurons with the largest diameter somata in the central nervous system. A s

Marine invertebrates, such as lobsters and crabs, deal with a widely and wildly fluctuating temperature environment. Here, we describe the effects of changing temperature on the motor patterns generated by the stomatogastric nervous system of the crab, Cancer borealis. Over a broad range of "permissive" temperatures, the pyloric rhythm increases in frequency but maintains its characteristic phase relationships. Nonetheless, at more extreme high temperatures, the normal triphasic pyloric rhythm breaks down, or "crashes". We present both experimental and computational approaches to understanding

Cardiac sac network activity (cycle period tens of seconds to minutes) has long been known to alter pyloric network activity (cycle period approximately 1 s), but these effects have not been quantified. Some pyloric muscles extract cardiac sac timed variations in pyloric motor neuron firing, and consequently produce cardiac sac timed movements even though no cardiac sac neurons innervate them. Determining pyloric behavior therefore requires detailed description of cardiac sac effects on pyloric neural output. Pyloric muscle activity correlates well with motor neuron overall spike frequency (OS

The motor pattern that drives coordinated movements of swimmerets in different segments during forward swimming characteristically begins with a power-stroke by the most posterior limbs, followed progressively by power-strokes of each of the more anterior limbs. To explain this caudal-to-rostral progression, the hypothesis was proposed that the neurons that drive the most posterior swimmerets are more excitable than their more anterior counterparts, and so reach threshold first. To test this excitability-gradient hypothesis, I used carbachol to excite expression of the swimmeret motor pattern

It is now commonly accepted that the arthropod nervous system has evolved only once, and so homologies between crustacean and insect nervous systems can be meaningfully sought. To do this, we have examined the distribution of serotonin (5-hydroxytryptamine)-like immunoreactive neurons in the central nervous system (CNS) of four common British isopods. Two species of terrestrial woodlouse, Oniscus asellus and Armadillidium vulgare, the littoral sea slater, Ligia oceanica, and the aquatic water hoglouse, Asellus meridianus, all possess approximately 40 pairs of serotonin-like immunoreactive neur

Our understanding of the nervous tissues that affect the wing flapping of insects mainly focuses on the brain, but wing flapping is a rhythmic movement related to the central pattern generator in the ventral nerve cord. To verify whether the neural activity of the abdominal ganglion of the honeybee (Apis mellifera) affects the flapping-wing flight, we profiled the response characteristics of indirect flight muscles to abdominal ganglion excitation. Strikingly, a change in the neural activity of ganglion 3 or ganglion 4 has a stronger effect on the electrophysiological activity of indirect flig
