[Morphologic changes of the cranial nerves, spinal cord ganglia and roots in botulism].

Peripheral nervous system was studied morphologically in 4 patients who had died from botulism. The results indicate considerable alterations in cranial and spinal nerves as well as in spinal cord ganglia. This may explain a neurological symptoms (radiculoneuritis, radiculoganglionitis) in botulism.

[Connections between spinal ganglia in the pathogenesis of reflex-distant pain syndromes].

Using the complex of the neurohistological research methods the author studied spinal ganglia, spinal cord (37 dogs) following the removal of groups of the spinal ganglia in the cervical and thoracic sections. Widely spread distant receptor-reflex systems of the connections between spinal ganglia accompanied by dendrites and axones were revealed. The fibres of these systems pass in the Bechterew intermediate regions of the posterior columns of the spinal cord. The straight interneuronal connections between the cervical and lumbar spinal ganglia were found. On the basis of the obtained data the

Basal ganglia-spinal cord pathway that commands locomotor gait asymmetries in mice.

The basal ganglia are essential for executing motor actions. How the basal ganglia engage spinal motor networks has remained elusive. Medullary Chx10 gigantocellular (Gi) neurons are required for turning gait programs, suggesting that turning gaits organized by the basal ganglia are executed via this descending pathway. Performing deep brainstem recordings of Chx10 Gi Ca2+ activity in adult mice, we show that striatal projection neurons initiate turning gaits via a dominant crossed pathway to Chx10 Gi neurons on the contralateral side. Using intersectional viral tracing and cell-type-specific

PlexinA4 distribution in the adult rat spinal cord and dorsal root ganglia.

PlexinsA1-A4 participate in class 3 semaphorin signaling as co-receptors to neuropilin 1 and 2, PlexinA4 being the latest member of the PlexinA subfamily to be identified. Little is known about the cellular distribution of PlexinA4 in the spinal cord and dorsal root ganglion (DRG). Here, immunohistochemical studies using antibodies to PlexinA4 revealed immunolabeling in neurons in both dorsal and, to a greater extent, ventral horns of the spinal cord. Ventral horn PlexinA4 positive neurons exhibited morphology, size, and location consistent with both motor neurons and interneurons. Labeling wa

Cystic fibrosis transmembrane conductance regulator expression in human spinal and sympathetic ganglia.

Cystic fibrosis transmembrane conductance regulator (CFTR) is a chloride channel protein, and mutations of its gene cause cystic fibrosis. CFTR is known to be expressed in epithelial cells of the respiratory, digestive and reproductive tracts. It is also present in rat neurons and heart ganglion cells. In humans, it is expressed in the hypothalamus, but has not been identified in other parts of the human nervous system. In this study, immunohistochemistry, double-staining immunofluorescence, in situ hybridization, nested reverse transcription-PCR and relative quantification of real-time PCR an

Lumbar spinal ganglia enhancement after gadolinium chelate administration: a radio-histological correlation.

The aim of the present study was to assess the frequency of enhancement of lumbar spinal ganglia after Gadolinium chelate injection in patients without radiculopathy, and to correlate the enhancement with histology. This study is based on the analysis of MR lumbar examinations conducted on 18 patients without radicular symptoms, or previous surgery of the lumbar spine, or disease of the nervous system. The patients were imaged with a 1.5 T unit. Sagittal images were first obtained with a T1-weighted turbo spin-echo. Axial and sagittal images were then obtained with a T1-weighted turbo spin-ech

Hemorrhages of dorsal root ganglia and spinal cord in congenitally hydrocephalic HTX rat.

The effects on the brain caused by hydrocephalus have been examined in detail. However, only little attention has been paid to the possibility that hydrocephalus may affect the spinal cord and the spinal ganglia via the spinal canal. Therefore, the present study focused on the pathological changes seen in the spinal cord and the dorsal root ganglia. A total of 651 congenitally hydrocephalic HTX rats were used in this study. The age ranged from postnatal day 0 to postnatal day 520. All of the HTX rats were from littermates raised in our laboratory. Macroscopic and microscopic investigations dem

Neurotrophins from dorsal root ganglia trigger allodynia after spinal nerve injury in rats.

Injury to peripheral nerves often results in chronic pain which is difficult to relieve. The mechanism underlying the pain syndrome remains largely unknown. In previous studies we showed that neurotrophins are up-regulated in satellite cells around sensory neurons following sciatic nerve lesion. In the present study, we have examined whether the neurotrophins in the dorsal root ganglia play any role in allodynia after nerve injury. Antibodies to different neurotrophins, directly delivered to injured dorsal root ganglia, significantly reduced (with different time sequences) the percentage of fo

Herpes zoster peripheral nerve complications: Their pathophysiology in spinal ganglia and nerve roots.

Varicella zoster virus (VZV) causes chickenpox at the primary infection and then becomes latent in the spinal dorsal root ganglia; VZV can reactivate with aging, immunosuppression, stress, and other factors, occurring as herpes zoster (HZ) at 1-2 skin segments. HZ peripheral nerve complications caused by VZV reactivation include Hunt syndrome, segmental HZ paresis, post-herpetic neuralgia, and Guillain-Barré syndrome (GBS). We have encountered the rare HZ complications of upper-limb paresis, myeloradiculitis, and polyradiculoneuritis: an adult woman with upper-limb paresis consistent with the

Morphometric analysis of neurons in ganglia: geniculate, submandibular, cervical spinal and superior cervical.

The geniculate ganglia (GG), the submandibular ganglion (SG), the cervical spinal ganglion (CSG) and the superior cervical ganglion (SCG) are clinically important. But details of these ganglia have not been included in textbook descriptions. We conducted research to bring precise figures to light. We prepared serial sections of these ganglia in five Japanese adults after precise dissections to count the numbers, measure the perimeters and the areas, and calculate the circularity ratios (CRs) of the ganglion cells. The numbers of neurons were in the following order: SCG > CSG > SG > GG, and the

Evaluation of the bilateral cardiac afferent distribution at the spinal and vagal ganglia by retrograde labeling.

The identity of sensory neurons innervating the heart tissue and the extent of information reported to the brain via these neurons are poorly understood. In order to evaluate the multidimensional distribution and abundance of the cardiac spinal and vagal afferents, we assessed the retrograde labeling efficiency of various tracers, and mapped the cardiac afferents qualitatively and quantitatively at the bilateral nodose ganglia (NGs) and dorsal root ganglia (DRGs). From the five different retrograde tracers evaluated, Di-8-ANEPPQ yielded reproducibly the highest labeling efficiency of cardiac a

TDP43 pathology in the brain, spinal cord, and dorsal root ganglia of a patient with FOSMN.

OBJECTIVE: To describe the histopathologic features of a case of facial-onset sensory and motor neuronopathy (FOSMN). METHODS: We describe a postmortem examination performed on a 54-year-old man with FOSMN associated with personality change. RESULTS: Postmortem examination revealed TAR DNA-binding protein (TDP) 43 proteinopathy with widespread distribution. TDP43 pathology was seen in the neurons and glial cells and was most pronounced in the subthalamic nucleus followed by the spinal cord, including dorsal root ganglia, brainstem, and other deep cerebral nuclei. In the medial temporal lobe,

Neuroimmune and Neuropathic Responses of Spinal Cord and Dorsal Root Ganglia in Middle Age.

Prior studies of aging and neuropathic injury have focused on senescent animals compared to young adults, while changes in middle age, particularly in the dorsal root ganglia (DRG), have remained largely unexplored. 14 neuroimmune mRNA markers, previously associated with peripheral nerve injury, were measured in multiplex assays of lumbar spinal cord (LSC), and DRG from young and middle-aged (3, 17 month) naïve rats, or from rats subjected to chronic constriction injury (CCI) of the sciatic nerve (after 7 days), or from aged-matched sham controls. Results showed that CD2, CD3e, CD68, CD45, TNF

Infection of organotypic cultures of spinal cord and dorsal root ganglia with Trypanosoma cruzi.

Although the involvement of the nervous system in Chagas' disease is well described, the mechanism of the neuronal destruction is unclear. Immunologic, toxic mechanisms and direct invasion have been advocated. Organotypic cultures of spinal cord and dorsal root ganglion derived from Swiss outbred mice were infected with the Brazil strain of Trypanosoma cruzi. Light microscopic and ultrastructural studies were performed at regular intervals. It was found that trypomastigotes were rapidly taken up by glial and other supporting cells. Neurons were rarely parasitized and demyelination was not evid

Characteristics of neurite outgrowth from rat spinal ganglia: effects of serum and segmental level.

The outgrowth of neurites from spinal ganglia in tissue culture may be affected by many variables including medium constituents and the age of ganglia. If E15 rats were used, the rate of outgrowth from ganglion explants in response to nerve growth factor was independent of the cranial-caudal position from which the ganglia were derived. This contrasts with the cranial-to-caudal gradient present during embryonic development. The effect of serum on this outgrowth was also studied and ten of 17 lots of human placental serum, two of three lots of pooled human placental serum, and one of two lots o

Distinct functional types of macrophage in dorsal root ganglia and spinal nerves proximal to sciatic and spinal nerve transections in the rat.

Inflammation proximal to a peripheral nerve injury may be responsible for ectopic discharge and/or death of sensory neurones, factors thought to contribute to the development and/or maintenance of neuropathic pain. Here, ED1+, ED2+ and major histocompatibility complex class II (MHC II)+ macrophages in dorsal root ganglia (DRGs) and spinal nerve roots have been compared quantitatively in adult rats following transection of one sciatic or one spinal nerve, using double labelling immunohistochemistry. In control DRGs, all ED2+ cells expressed ED1 and some also MHC II. One week after either lesion

Translational profiling of dorsal root ganglia and spinal cord in a mouse model of neuropathic pain.

Acute pain serves as a protective mechanism, guiding the organism away from actual or potential tissue injury. In contrast, chronic pain is a debilitating condition without any obvious physiological function. The transition to, and the maintenance of chronic pain require new gene expression to support biochemical and structural changes within the pain pathway. The regulation of gene expression at the level of mRNA translation has emerged as an important step in the control of protein expression in the cell. Recent studies show that signaling pathways upstream of mRNA translation, such as mTORC

Critical evaluation of the expression of gastrin-releasing peptide in dorsal root ganglia and spinal cord.

There are substantial disagreements about the expression of gastrin-releasing peptide (GRP) in sensory neurons and whether GRP antibody cross-reacts with substance P (SP). These concerns necessitate a critical revaluation of GRP expression using additional approaches. Here, we show that a widely used GRP antibody specifically recognizes GRP but not SP. In the spinal cord of mice lacking SP (Tac1KO), the expression of not only GRP but also other peptides, notably neuropeptide Y (NPY), is significantly diminished. We detectedGrpmRNA in dorsal root ganglias using reverse transcription polymerase
