Understanding the toxicity mechanism of gelsemine in zebrafish.

Gelsemium elegans (GE), also known as Duanchangcao, is a plant associated with toxic symptoms related to the abdomen; however, the toxicity caused by GE remains unknown. Gelsemine (GEL) is an alkaloid extracted from GE and is one of the most toxic alkaloids. This study used zebrafish as an animal model and employed high-throughput gene sequencing to identify genes and signaling pathways related to GEL toxicity. Exposure to GEL negatively impacted heart rate, swim bladder development, and activity in zebrafish larvae. Transcriptomics data revealed the enrichment of inflammatory and phagocyte si

Modulation of GABAA receptors and of GABAergic synapses by the natural alkaloid gelsemine.

The Gelsemium elegans plant preparations have shown beneficial activity against common diseases, including chronic pain and anxiety. Nevertheless, their clinical uses are limited by their toxicity. Gelsemine, one of the most abundant alkaloids in the Gelsemium plants, have replicated these therapeutic and toxic actions in experimental behavioral models. However, the molecular targets underlying these biological effects remain unclear. The behavioral activity profile of gelsemine suggests the involvement of GABAA receptors (GABAARs), which are the main biological targets of benzodiazepines (BDZ

Integrative metabolomics and machine learning reveal diagnostic biomarkers for gelsenicine intoxication.

ETHNOPHARMACOLOGICAL RELEVANCE: Gelsemium elegans Benth. (G. elegans) is a highly toxic medicinal plant traditionally used to treat pain and inflammatory disorders. Accidental ingestion or misuse often causes severe poisoning and death, primarily due to respiratory depression. Its most toxic alkaloid, gelsenicine, undergoes rapid metabolism in vivo. The lack of specific biomarkers hinders timely and accurate assessment of poisoning severity. AIM OF THE STUDY: This study combined untargeted metabolomics with machine learning (ML) to distinguish gelsenicine-induced fatal intoxication (GFI) from

The immunomodulatory effect of koumine on B cells under dependent and independent responses by T cells.

Dysregulated activation of polyclonal B cells and production of pathogenic antibodies are involved in the development of rheumatoid arthritis (RA). Therefore, targeted B cell therapy is effective against RA. Gelsemium elegans (Gardn. & Champ.) Benth., a toxic plant widely distributed in Southeast Asia, has been used for treating rheumatoid pain, neuropathic pain, spasticity, skin ulcers, and cancers for many years in traditional Chinese medicine. Koumine, an alkaloid monomer from Gelsemium elegans Benth., exerts therapeutic effects against RA. However, whether koumine affects B cells remains u

The analgesic effect and possible mechanisms by which koumine alters type II collagen-induced arthritis in rats.

Gelsemium elegans Benth. is a toxic plant that has been used as an ancient Chinese herbal remedy for rheumatoid arthritis (RA) and nervous pain, spasticity, skin ulcers, and cancers. Koumine, one of its representative alkaloids, shows numerous promising pharmacological activities, including anti-inflammatory and analgesic activities. Here, we investigated the analgesic effect of koumine on the collagen-induced arthritis (CIA) rat model of RA and explored the potential pharmacological mechanisms underlying the analgesia. In the CIA rats, repeated koumine treatments significantly reduced pain co

Koumine inhibits osteoclastogenesis and prevents ovariectomy-induced bone loss via suppression of the MAPK signaling pathways.

Excessive osteoclast formation drives osteolytic bone diseases such as osteoporosis. Koumine (KM), an alkaloid derived from Gelsemium elegans, exhibits various bioactivities; however, its role in bone homeostasis remains unknown. This study investigated the effects of KM on RANKL-induced osteoclastogenesis in bone marrow-derived macrophages (BMMs). Cell viability, differentiation (TRAcP staining), and function (F-actin ring formation, bone resorption pit assay) were assessed. The underlying mechanisms were explored using Western blot and qPCR. The in vivo efficacy of KM was evaluated in an ova

Astrocytes in the ventral posterolateral nucleus modulate diabetic neuropathic pain in male rats and are inhibited by the analgesic koumine.

Diabetic neuropathic pain (DNP) is a common and intractable complication of diabetes mellitus that is relatively poorly managed by current pharmacotherapies. The ventral posterolateral nucleus (VPL) is a key thalamic subnucleus involved in nociceptive transmission, but the role of VPL astrocytes in DNP remains unclear. In this study, we observed a significant increase in VPL astrocyte activity 14 days after injections of streptozotocin in male rats. Short-term inhibition of VPL astrocytes with designer receptors exclusively activated by designer drugs (DREADDs) alleviated mechanical allodynia

Regulation of microglia related neuroinflammation contributes to the protective effect of Gelsevirine on ischemic stroke.

Stroke, especially ischemic stroke, is an important cause of neurological morbidity and mortality worldwide. Growing evidence suggests that the immune system plays an intricate function in the pathophysiology of stroke. Gelsevirine (Gs), an alkaloid from Gelsemium elegans, has been proven to decrease inflammation and neuralgia in osteoarthritis previously, but its role in stroke is unknown. In this study, the middle cerebral artery occlusion (MCAO) mice model was used to evaluate the protective effect of Gs on stroke, and the administration of Gs significantly improved infarct volume, Bederson

Koumine enhances spinal cord 3α-hydroxysteroid oxidoreductase expression and activity in a rat model of neuropathic pain.

BACKGROUND: Koumine is an alkaloid monomer found abundantly in Gelsemium plants. It has been shown to reverse thermal hyperalgesia and mechanical allodynia induced by sciatic nerve chronic constriction injury (CCI) in rats in a dose-dependent manner. Interestingly, this effect is mediated by elevated allopregnanolone levels in the spinal cord (SC). Since 3α-hydroxysteroid oxidoreductase (3α-HSOR), the key synthetase of allopregnanolone, is responsible for allopregnanolone upregulation in the SC, the objective of the present study was to investigate the role of its expression in the SC in koumi

Anti-allodynic and neuroprotective effects of koumine, a Benth alkaloid, in a rat model of diabetic neuropathy.

Diabetic neuropathy is characterized by progressive degeneration of nerve fibers associated with diabetes mellitus. Antidepressants and anticonvulsants are the mainstay of pharmacological treatment, but are often limited in effectiveness against the core clinical feature of pain. In the current study, we examined the potential effects of koumine, a Gelsemium elegans Benth alkaloid, using a rat model of diabetic neuropathy. Rats were administered intraperitoneally a single dose of streptozocin (60 mg/kg) to induce type 1 diabetes. Koumine was given at a dose range of 0.056-7 mg/kg subcutaneousl

Toxic compounds in honey.

There is a wealth of information about the nutritional and medicinal properties of honey. However, honey may contain compounds that may lead to toxicity. A compound not naturally present in honey, named 5-hydroxymethylfurfural (HMF), may be formed during the heating or preservation processes of honey. HMF has gained much interest, as it is commonly detected in honey samples, especially samples that have been stored for a long time. HMF is a compound that may be mutagenic, carcinogenic and cytotoxic. It has also been reported that honey can be contaminated with heavy metals such as lead, arseni
