Characterization of Geminivirus resistance in an accession of Capsicum chinense Jacq.

Pepper golden mosaic virus (PepGMV) and Pepper huasteco yellow vein virus (PHYVV), members of the Geminiviridae family, are important pathogens of pepper (Capsicum annuum L.) and other solanaceous crops. Accession BG-3821 of C. chinense Jacq. was reported earlier as resistant to mixed infection with PepGMV and PHYVV. In this work, we characterized the Geminivirus resistance trait present in BG-3821. Segregation analysis suggested that resistance depends on two genes. Our data showed that PepGMV replication in protoplast of resistant plants is approximately 70% lower when compared with the leve

Vector-enabled metagenomics reveals the first detection of the geminivirus beet curly top Iran virus in Europe.

Geminiviruses are among the most threatening emerging insect-borne viruses and are responsible for serious outbreaks worldwide. Climate change (i.e. higher temperatures) could further exacerbate their impact on crops, highlighting the need for new diagnostic approaches to manage potentially dangerous situations. vector-enabled metagenomics (VEM) exploits the natural ability of highly mobile insects to accumulate viruses acquired from plants over time and space within an ecosystem; this approach is effective for monitoring the presence of new invasive or indigenous viruses in large areas. Beet curly top virus affects vector biology: the first transcriptome analysis of the beet leafhopper.

Curly top disease, caused by beet curly top virus (BCTV), is among the most serious viral diseases affecting sugar beets in western USA. The virus is exclusively transmitted by the beet leafhopper (BLH, Circulifer tenellus) in a circulative and non-propagative manner. Despite the growing knowledge on virus-vector interactions, our understanding of the molecular interactions between BCTV and BLH is hampered by limited information regarding the virus impact on the vector and the lack of genomic and transcriptomic resources for BLH. This study unveils the significant impact of BCTV on both the pe

Bemnisia tabaci feeding induces pathogenesis-related proteins in cassava (Manihot esculenta Crantz).

Cassava (Manihot esculenta Cranzts) plants fed upon by whitefly Bemisia tabaci showed increased levels of pathogenesis-related (PR) proteins, such as beta-1, 3-glucanase, peroxidase and chitinase activities, as compared to uninfested plants. The enzymes increased in specific activities from 2 to 7 fold and protein content in leaf extracts decreased in whitefly-infested plants, compared to uninfested plants. Among the three PR proteins, B. tabaci feeding induced significantly higher beta-1, 3-glucanase activities, when compared with other two PR proteins. Study also discussed the possible appli

Metagenomic insights into viral dynamics and funcation in Baijiu.

Baijiu fermentation represents a sophisticated microbial-driven biochemical process mediated by complex microbial consortium. Despite extensive characterization of bacterial and fungal roles in fermentation systems, the virus remains a critical knowledge gap. In this study,we employed metagenomics to profile the dynamics of viral community in fermented grains across five stages (day 0, 5, 10, 20 and 30) of a Baijiu fermentation. The metagenomics revealed 101 viral families, dominated by Metaviridae, Parvoviridae, Aliceevansviridae, Herelleviridae, Geminiviridae, Iridoviridae, and Genomoviridae

How a chance finding and high-throughput sequencing helped unmask the probable causal agent of Brazilian curly top, a plant disease that disappeared over 70 years ago.

Curly top disease (CTD) affects sugar beet, tomato and other crops, resulting in stunted plants with severely curled leaves and reduced yields. The disease occurs worldwide and is caused by geographically associated monopartite geminiviruses transmitted mostly by leafhoppers. However, the aetiology of CTD in South America remains unknown because the disease disappeared in the 1950s. Here, we describe how the chance finding of tomato plants with CTD-like symptoms in Brazil in 2016 and high-throughput sequencing helped identify a novel ~2.6 kb geminivirus DNA associated with curly top disease in

Natural Defect of a Plant Rhabdovirus Glycoprotein Gene: A Case Study of Virus-Plant Coevolution.

Seven isolates of a putative cytorhabdovirus (family Rhabdoviridae, order Mononegavirales) designated as citrus-associated rhabdovirus (CiaRV) were identified in citrus, passion fruit, and paper bush from the same geographical area in China. CiaRV, bean-associated cytorhabdovirus (Brazil), and papaya virus E (Ecuador) should be taxonomically classified in the species Papaya cytorhabdovirus. Due to natural mutations, the glycoprotein (G) and P4 genes were impaired in citrus-infecting isolates of CiaRV, resulting in an atypical rhabdovirus genome organization of 3' leader-N-P-P3-M-L-5' trailer.

Characterization of Begomoviruses Sampled during Severe Epidemics in Tomato Cultivars Carrying the Ty-1 Gene.

Tomato yellow leaf curl virus (TYLCV, genus Begomovirus, family Geminiviridae) is a major species that causes a tomato disease for which resistant tomato hybrids (mainly carriers of the Ty-1/Ty-3 gene) are being used widely. We have characterized begomoviruses severely affecting resistant tomato crops in Southeast Spain. Circular DNA was prepared from samples by rolling circle amplification, and sequenced by massive sequencing (2015) or cloning and Sanger sequencing (2016). Thus, 23 complete sequences were determined, all belonging to the TYLCV Israel strain (TYLCV-IL). Massive sequencing also

Geminivirus-Induced Reprogramming of Plant Defense Mechanisms: Molecular Insights and Research Frontiers.

Geminiviruses employ multifunctional protein ammunition to evade robust plant defense pathways. Key viral proteins effectively manipulate host signaling mechanisms to create a permissive environment for viral replication. Rapid evolutionary adaptation of geminiviruses, synergized by the proliferation of insect vectors, creates a challenge for effective disease control. Current plant resistance against geminiviruses primarily relies on antiviral RNA silencing and the localized cell death mechanism as an outcome of the hypersensitive response. To win the escalating arms race between geminivirus

Ecology and Evolutionary Dynamics of the A-Strain Maize Streak Virus and the Decade-Long Strategies Targeting Streak-Resistant Maize.

The A-strain maize streak virus (MSV-A; genus Mastrevirus, family Geminiviridae) is a single-stranded DNA (ssDNA) virus that severely constrains the production of maize in sub-Saharan Africa and other places where its insect vectors thrive. In this review, we discuss the ecology- and genomics-driven evolution of this economically important virus and strategies targeting streak resistance in maize. Evolution of MSV-A is largely driven by the interplay of mutation, recombination and selective forces that may sustain fitness and result in host adaptation. In the last decade, research efforts, inc

Expression and functional characterisation of a soluble form of Tomato yellow leaf curl virus coat protein.

BACKGROUND: Tomato yellow leaf curl virus (TYLCV), a member of the genus Begomovirus within the family Geminiviridae, is an important pathogen of tomato in many tropical, subtropical and temperate regions. TYLCV is exclusively transmitted by the whitefly Bemisia tabaci in a circulative manner. The viral coat protein (CP) has been assumed to play important roles in the entry of TYLCV into the insect midgut cells. RESULTS: Testing the hypothesis that CP plays an important role in TYLCV acquisition by B. tabaci, a soluble form of the CP was expressed and purified. The purified recombinant CP mad

Molecular characterization and infectious clone construction of beet capulavirus 1 infecting tomato in Germany.

Capulavirus betae (beet capulavirus 1, BCV1) is a recently described species of the genus Capulavirus (family Geminiviridae) that was first identified in sugar beet in France. Capulaviruses are new group of geminiviruses because they are transmitted by aphids. Here, we report the identification and molecular characterization of an isolate of BCV1 infecting cultivated tomato in Germany and describe the construction of an infectious clone. In August 2025, tomato plants displaying severe virus-like symptoms, including leaf deformation, leaf curling, stunting and chlorosis were collected from two

Intra-viral protein-protein interactions could expand the functional portfolio of C4 in okra-infecting geminiviruses.

Viruses must rewire the cells they infect to promote their own multiplication, a task they accomplish through the deployment of multifunctional virus-encoded proteins. However, the limited coding capacity imposed by small viral genomes constrains their functional repertoire. Recently, it has been proposed that proteins encoded by plant-infecting geminiviruses can physically interact with one another, forming intra-viral protein complexes that may acquire novel activities and thereby expand viral functionality. Here, we test this concept by focusing on the C4 proteins of two okra-infecting gemi

Tomato yellow leaf curl virus (TYLCV) in Kuwait and global analysis of the population structure and evolutionary pattern of TYLCV.

Tomato yellow leaf curl virus (Family Geminiviridae, Genus Begomovirus) is a serious menace in the cultivation of tomato causing Tomato leaf curl disease (ToLCD). Recently, we presented the TYLCV isolates having additional genomic features (nucleotides insertions) characterized from the tomato fields of Kuwait adding up to the genetic diversity repertoire of these viruses. The widespread prevalence of disease in tropics across the continents, emergence of genetic variants and ever increasing complete genome sequences of virus isolates in public database warrant a global analysis to infer the g

Geminiviral C2 proteins promote virus transmission by subverting plant defenses upon infestation of non-vector herbivores.

BACKGROUND: Insect vectors are responsible for transmitting the majority of plant viruses. While the interplays between insect vectors and viruses have been extensively investigated, how other organisms associated with the shared host plants of the insect and virus impact virus transmission dynamics, and the corresponding viral countermeasures, remain largely unknown. RESULTS: We explored the modulation of whitefly-mediated transmission of geminiviruses by two non-vector herbivores and the role of viral C2 proteins. The infestation of cotton bollworm induced the accumulation of jasmonic acid

Whitefly Bemisia tabaci (Homoptera: Aleyrodidae) infestation on cassava genotypes grown at different ecozones in Nigeria.

Large-scale screening of cassava, Manihot esculenta Crantz, genotypes for resistance to infestation by whitefly Bemisia tabaci Gennadius, the vector of cassava mosaic geminiviruses, is limited. A range of new cassava elite clones were therefore assessed for the whitefly infestation in the 1999/2000 and 2000/2001 cropping seasons in experimental fields of International Institute of Tropical Agriculture, Ibadan, Nigeria. On each scoring day, between 0600 and 0800 hours when the whiteflies were relatively immobile, adult whitefly populations on the five topmost expanded leaves of cassava cultivar

Mutating the cysteine residues within the WCCH motif of tobacco curly shoot virus (TbCSV) V2 protein attenuates viral infection and confers cross-protection against infection by severe strain.

BACKGROUND: Tobacco curly shoot virus (TbCSV, Begomovirus, Geminiviridae) is a viral agent that poses a serious threat to economically important crops like tomato and tobacco in Southwest China. The geminiviral V2 protein has been demonstrated to play multifunctional roles essential for viral infection and pathogenicity. This study comprehensively investigated the biological importance of five cysteines within the conserved 'WCCH' motif of the TbCSV V2 protein, and evaluated the effects of cysteine mutations on viral pathogenicity and the efficacy for cross-protection against wild-type severe

Begomovirus Transmission to Tomato Plants Is Not Hampered by Plant Defenses Induced by Dicyphus hesperus Knight.

Plants can respond to insect infestation and virus infection by inducing plant defenses, generally mediated by phytohormones. Moreover, plant defenses alter host quality for insect vectors with consequences for the spread of viruses. In agricultural settings, other organisms commonly interact with plants, thereby inducing plant defenses that could affect plant-virus-vector interactions. For example, plant defenses induced by omnivorous insects can modulate insect behavior. This study focused on tomato yellow leaf curl virus (TYLCV), a plant virus of the family Geminiviridae and genus Begomovir
