Genetic background of acute anterior uveitis.

To determine the hereditary pattern of acute anterior uveitis, we examined seven families-one family with four cases, one family with three cases, and four families with two cases of acute anterior uveitis. A total of 16 patients with uveitis and 28 relatives underwent ocular examinations and serologic determination of HLA antigens, protease inhibitor alpha 1-antitrypsin typing, and X-ray examination of sacroiliac joints. Sacroiliitis was found in 11 of 16 patients (68.8%) and incomplete Reiter's disease in seven of 16 (43.8%). HLA-B27 antigen was found in all patients with uveitis and in six

Genetic background of Fuchs' heterochromic cyclitis.

We studied the genetic background of 24 patients with Fuchs' heterochromic cyclitis (FHC). Each was given a careful eye examination which included family history and serological determination of HLA antigens. Two families each had 2 cases of FHC in the same family; in addition an ancestor in the second family possibly had FHC; in both families one healthy member had simple heterochromia. One patient with FHC had congenital uveal coloboma, one pigmentary retinal dystrophy, and four had cysts of the ciliary body. The frequencies of all HLA antigens in patients with FHC compared well with the fre

HEARRING group genetic marker study: genetic background of CI patients.

BACKGROUND: While cochlear implantation (CI) and electric acoustic stimulation (EAS) have a positive outcome in most cases, their effectiveness varies depending on the etiology of the hearing loss. Among the various etiologies, genetic factors are the leading cause of hearing loss and may impact CI and EAS outcomes. AIMS/OBJECTIVES: To reveal the genetic background of the hearing loss in CI/EAS patients in each ethnic population, we undertook a multi-center study involving the genetic testing of hearing loss in CI/EAS patients from 10 centers. MATERIAL AND METHODS: Saliva samples and clinica

Genetic background of MEN1: from genetic homogeneity to functional diversity.

Multiple Endocrine Neoplasia Type 1 corresponds to a monogenic predisposition syndrome inherited as a dominant trait that affects a variety of endocrine tissues, in particular parathyroids, endocrine pancreas and anterior pituitary. It is caused by mutations in the MEN1 tumor suppressor gene that inactivate menin, the MEN1 encoded protein. Menin is involved in cell cycle control and apoptosis through its participation in functional dynamics of chromatin and regulation of transcription. In addition, genetic investigations have implicated menin in the maintenance of genomic integrity. However, t

Genetic background of high myopia in children.

OBJECTIVE: High myopia is a significant risk factor for irreversible vision loss and can occur in isolation or as a component of various syndromes. However, the genetic basis of early-onset high myopia remains poorly understood. We aimed to identify the causative genetic variants for high myopia in a cohort of Slovenian children. METHODS: The study included children referred to a tertiary paediatric ophthalmology centre at the University Eye Clinic in Ljubljana between 2010 and 2022. The participants met the following inclusion criteria: age ≤ 15 years and high myopia ≤-5.0 D before the age o

Genetic background dependent modifiers of craniosynostosis severity.

Craniosynostosis severity varies in patients with identical genetic mutations. To understand causes of this phenotypic variation, we backcrossed the FGFR2+/C342Y mouse model of Crouzon syndrome onto congenic C57BL/6 and BALB/c backgrounds. Coronal suture fusion was observed in C57BL/6 (88% incidence, p < .001 between genotypes) but not in BALB/c FGFR2+/C342Y mutant mice at 3 weeks after birth, establishing that that the two models differ in phenotype severity. To begin identifying pre-existing modifiers of craniosynostosis severity, we compared transcriptome signatures of cranial tissues from

Mouse genetic background influences the dental phenotype.

Dental enamel covers the crown of the vertebrate tooth and is considered to be the hardest tissue in the body. Enamel develops during secretion of an extracellular matrix by ameloblast cells in the tooth germ, prior to eruption of the tooth into the oral cavity. Secreted enamel proteins direct mineralization patterns during the maturation stage of amelogenesis as the tooth prepares to erupt. The amelogenins are the most abundant enamel proteins and are required for normal enamel development. Phenotypic differences were observed between incisors from individual Amelx (amelogenin) null mice that

[Genetic background of diabetes mellitus type 2].

Diabetes mellitus type 2 (DM2) is the most prevalent form of diabetes, and is caused by a combination of genetic and non-genetic factors. Genome-wide association studies have led to the identification of 20 genetic risk factors for DM2, for which the actual causal mutations are not yet known. Nevertheless, some of the genes connected with diabetes suggest that the pancreatic beta cell plays a central role in the development of the disease. In the meantime, detailed knowledge of the genetics of some specific forms of diabetes has already lead to changes in pharmacological therapy; patients coul

The genetic background of inflammatory bowel disease.

Available evidence indicates that genetic factors are essential in providing the susceptibility to the majority of the various forms of inflammatory bowel disease occurring in man. It is also clear that the genetic susceptibility to these diseases is complex, and that more than one gene may predispose (the concept of multilocus/oligogenic inheritance), and likely in different etiologic combinations (the concept of genetic heterogeneity). Paradigms are now available that should lead to the identification of a number of these predisposing genes. These paradigms include the candidate gene approac

Human genetic background in susceptibility to tuberculosis.

Tuberculosis (TB), especially in developing countries, is a major threat to human health. The pathogenesis of TB remains poorly understood, and <5%-10% of individuals infected with Mycobacterium tuberculosis (MTB) will develop clinical disease. The human genetic factors contributing to susceptibility or resistance to TB pathogenesis have been investigated by high-throughput and low-throughput association studies. Genetic polymorphisms of several genes including TLR, IGRM, VDR, ASAP1, AGMO, FOXP1, and UBLCP1 effect on the disease phenotype and also the outcome of TB treatment. Recently, microRN

[Genetic background of human lactoferrin transgenic goats].

The genetic background such as copy number, integration site and chromosome karyotype of exogenous genes of transgenic animals obtained by random integration is still unclear. There may be some problems such as silent integration, invalid integration, toxic integration and unpredictable expression level of exogenous genes. In this study, six primary (F0) and their corresponding offspring (F1) of human lactoferrin (hLF) transgenic goats were selected as the research objects, and blood samples were collected from jugular vein and DNA were extracted. The genetic background and expression level of

Genetic background of autoimmune hepatitis in Japan.

Autoimmune hepatitis (AIH) is an organ-specific autoimmune disease characterized by chronic inflammation of the liver. Several studies from ethnically different countries have clarified that the genetic predisposition to type 1 AIH is linked mainly to human leukocyte antigen (HLA)-class II genes. Recently, molecular analysis using polymerase chain reaction (PCR)-based DNA typing has revealed that susceptibility to type 1 AIH is primarily associated with the HLA class II DRB1 locus, which encodes a polymorphic β chain of the HLA-DR antigen. However, additional susceptibility genes (either HLA o

The genetic background of uremic secondary hyperparathyroidism.

Many studies have shown how gene mutations and genetic polymorphisms could influence secondary uremic hyperparathyroidism (HPTH), modulating parathyroid (PT) function and hyperplasia. Parathyroid hormone (PTH) gene expression and hormone secretion is regulated mainly by serum calcium, with a post-transcriptional effect, and by vitamin D with a transcriptional effect. PT cells retain the ability to proliferate and to apoptose. Hyperphosphatemia, hypocalcemia and vitamin D deficiency all stimulate PT cell proliferation. In the early stage of chronic uremia, PT proliferation is polyclonal, as in

Genetics of myositis - distinct backgrounds of subtypes.

Myositis is a heterogeneous group of inflammatory muscular disorders. Although the main etiology is autoimmune chronic inflammation, the underlying pathomechanism remains unclear. Advances in genetic technology have provided important insights into its complex pathophysiology. Large genetic studies on myositis have advocated a relationship with several HLA loci and possible disease susceptibility genes in non-HLA genes. Idiopathic inflammatory myopathy, or autoimmune myositis, was originally divided into polymyositis and dermatomyositis. However, this classification has recently been revised b

[Genetic background of dyslexia and dysgraphy in children].

The review is devoted to one of the current problems of pediatric neurology - reading and writing disorders in children as part of a partial developmental disorder. With the development of neuroscience, the paradigm of «brain damage» in the understanding of a number of pathological conditions was replaced by the concept of «evolutionary neurology». The dominance of the ontogenetic approach caused the appearance of a new section in ICD-11 - «Neurodevelopmental disorders». Twenty-one genes associated with the acquisition of reading and writing skills have been identified. Modern studies demonstr

PFAPA syndrome in siblings. Is there a genetic background?

UNLABELLED: "PFAPA syndrome" is an autoinflammatory entity composed of periodic fever, aphthous stomatitis, pharyngitis, and cervical adenitis. There have been many reports of children with the disease, but only occasionally have been described in siblings, and no specific genetic mutation has been determined yet. Corticosteroids are the mainstay in the treatment of the acute attacks. The role of surgery in long-term follow-up (tonsillectomy with or without adenoidectomy) is controversial. We report two brothers affected with the syndrome, in whom corticosteroids as the only treatment led to a

Genetic background of obesity: a case-control study.

PURPOSE: The rising prevalence of obesity and type 2 diabetes mellitus (T2DM) constitutes a major global public health concern. Increasing evidence indicates that genetic polymorphisms may modulate the risk of T2DM in individuals with obesity, providing valuable insight into the biological pathways that connect these interrelated disorders. A better understanding of the genetic determinants underlying obesity-associated T2DM could enhance disease characterization and inform risk stratification strategies. The genetic architecture linking obesity and T2DM has, however, not yet been fully elucid

Decomposing the genetic background of chronic back pain.

Chronic back pain (CBP) is a disabling condition with a lifetime prevalence of 40% and a substantial socioeconomic burden. Because of the high heterogeneity of CBP, subphenotyping may help to improve prediction and support personalized treatment of CBP. To investigate CBP subphenotypes, we decomposed its genetic background into a shared one common to other chronic pain conditions (back, neck, hip, knee, stomach, and head pain) and unshared genetic background specific to CBP. We identified and replicated 18 genes with shared impact across different chronic pain conditions and two genes that wer
